The Phytotoxic Effects of Selenium–Mercury Interactions on Root Growth in Brassica rapa (LvLing)

Rapid industrial and agricultural development has dramatically increased the emission of selenium (Se) and mercury (Hg) into the environment. Combined soil pollution by Se and Hg poses a potential threat to crop production. However, no toxic effects of Hg–Se interactions on plants have been reported previously. In this study, we investigated the effects of Hg–Se interactions on biochemical and physiological indices in the roots of Brassica rapa (LvLing). Seedlings were treated hydroponically with solutions of mercury chloride (1 μM), sodium selenite (4 μM), or a combination of the two. Combined Hg+Se treatment significantly inhibited root growth, reduced root biomass, and enhanced reactive oxygen species (ROS) and malondialdehyde accumulation and led to a loss of plasma membrane integrity. The combined treatment increased glutathione peroxidase, glutathione S-transferase, and peroxidase activity, reduced superoxide dismutase activity, and had no effect on catalase activity. In addition, we detected increased glutathione concentrations in root tips and reduced ascorbic acid concentrations in the presence of Hg+Se relative to individual treatments with these elements. Thus, Hg–Se interactions enhanced oxidative injury, cell death, and phytotoxicity in B. rapa roots.
Ghost Cell Odontogenic Carcinoma Arising from Calcifying Cystic Odontogenic Tumor: A Case Report

Ghost cell odontogenic carcinoma (GCOC) is an exceptionally rare and malignant odontogenic tumor with aggressive growth characteristics. We describe a case of GCOC which was considerably derived from a previously resected calcifying cystic odontogenic tumor (CCOT). Cellular atypia, mitotic activity, Ki-67 labeling index and matrix metalloprotease-9 positive expression rate were all increased in the currently resected specimen compared to the initial one. This is a rare case of malignant transformation of CCOT to GCOC with respect to its histopathological and immunohistochemical findings.
Broadband Spectrum Sensing of Distributed Modulated Wideband Converter Based on Markov Random Field

The Distributed Modulated Wideband Converter (DMWC) is a networking system developed from the Modulated Wideband Converter, which converts all sampling channels into sensing nodes with number variables to implement signal undersampling. When the number of sparse subbands changes, the number of nodes can be adjusted flexibly to improve the reconstruction rate. Owing to the different attenuations of distributed nodes in different locations, it is worthwhile to find out how to select the optimal sensing node as the sampling channel. This paper proposes the spectrum sensing of DMWC based on a Markov random field (MRF) to select the ideal node, which is compared to the image edge segmentation. The attenuation of the candidate nodes is estimated based on the attenuation of the neighboring nodes that have participated in the DMWC system. Theoretical analysis and numerical simulations show that neighboring attenuation plays an important role in determining the node selection, and selecting the node using MRF can avoid serious transmission attenuation. Furthermore, DMWC can greatly improve recovery performance by using a Markov random field compared with random selection.

Two isocoumarin derivatives, stoloniferol A (1) and B (2), a known 5α, 8α-epidioxy-23-methyl- (22E, 24R)-ergosta-6, 22-dien-3β-ol (3), and a known dihydrocitrinone (4) were isolated from the ethyl acetate extract of the sea squirt-derived fungus, Penicillium stoloniferum QY2-10, and a halophilic fungus, Penicillium notatum B-52, respectively. Their structures were elucidated by spectroscopic methods and optical rotation. The stereochemistry of 2 was determined on the basis of different NOE experiments and chemical transformation. Compound 3 showed cytotoxicity against P388 cells, with an IC50 value of 4.07 µM.

The production of L-tryptophan was investigated in a recombinant strain Escherichia coli W3110-ZDrr. It was observed that phosphate and feeding strategies are key factors to ensure the good cell growth and high production of L-tryptophan. The simple exponential feeding strategy could only produce 10.6 g/L L-tryptophan due to the improper feeding rate; while the manual glucose-feedback feeding approach could effectively control the substrate and inhibit the formation of acetate, and thus improvemed the Ltryptophan production to 25.5 g/L. The modified exponential feeding approach avoided overfeeding or underfeeding and achieved high production of L-tryptophan. Moreover, the addition of Tween 60 or PL61 could enhance the cell growth and the production of L-tryptophan in the fed-batch mode. Subsequent metabolic flux analysis showed that more carbon flux was distributed into the biosynthesis of Ltryptophan when Tween 60 or PL61 was supplied. The present work presents one base for further large-scale production of this important amino acid.
Cat NPC2, a Newly Identified Allergen, With High Cross-Reactivity to Can f 7

Purpose: Pet-derived allergens are the common indoor inhalant allergens. Among them, cat and dog allergens constitute more than 80% of animal allergic patients, which greatly affect the quality-of-life of patients and increase the burden of social health care. The aim of this study was to identify Cat-Niemann pick type C2 (NPC2) protein, a homologue of Can f 7, as a new allergen. Methods: Cat-NPC2 complementary DNA (cDNA) was cloned and optimized for amplification and expression in Escherichia coli. Then, recombinant Cat-NPC2 (rCat-NPC2) was purified by Ni2+ affinity chromatography. The allergenicity was assessed by enzyme-linked immunosorbent assay (ELISA), western blot and basophil activation test (BAT). Based on the sequence similarity, the cross-reactivity between Cat-NPC2 and Can f 7 was investigated by inhibition ELISA. Circular dichroism spectroscopy and homology modeling were used to characterize the structure of Cat-NPC2. Results: The cDNA sequence of Cat-NPC2 was cloned with a 450-bp open reading frame coding for 149 amino acids (GenBank MN_737596). The condon-optimized NPC2 gene was subcloned and expressed in E. coli with a molecular weight of 18.9 kDa. The native Cat-NPC2 was detected in cat dander extracts. The allergenicity determined by ELISA, western blot and BAT suggested at least 14.5% cat-allergic patients displayed high specific immunoglobulin E (IgE) recognition of Cat-NPC2. The predicted structure of Cat-NPC2 was found to consist of 7 β-strands arranged in 2 β-sheets. An ELISA based assay showed that rCat-NPC2 bound to cholesterol in a dose dependent manner. Based on the structure and sequence similarities, IgE cross-reactivity was demonstrated between Cat-NPC2 and Can f 7/Der f 2. Conclusions: In the study, a novel cat allergen, belonging to the NPC2 protein family, was identified and characterized at both molecular and immunological levels. The study will offer a deeper understanding of cat allergens and improve a component-resolved diagnosis in pet allergy.
Abietane Diterpenoids from Perovskia atriplicifolia and Their Anti-HBV Activities

Bioassay-guided phytochemical investigation on the 90% EtOH extract of Perovskia atriplicifolia resulted in the isolation of eight abietane diterpenoids, including three new ones (1–3). Based on spectroscopic methods involving 1D and 2D NMR spectroscopy techniques, mass spectrometry, and optical rotation, the structures of the new compounds (1–3) were unambiguously characterized. Compounds 1–2 and 4–8 were evaluated for their anti-HBV (hepatitis B virus) activity in HepG 2.2.15 cell line. Results suggested rosmadial (8) had the most anti-HBV potency, suppressing the secretion of HBsAg and HBeAg, with IC50 values of 0.09 and 0.34 mM, respectively.
