      • Analysis of Indoleamine 2-3 Dioxygenase (IDO) and EGFR Co-expression in Breast Cancer Tissue by Immunohistochemistry

        Background: To determine the amount of co-expression of IDO and EGFR in breast cancer patients. Materials and Methods:In order to obtain the distribution of co-expression of IDO and EGFR in breast cancer, we tested 110 breast cancer paraffin tissue blocks with immunohistochemical methods. Then we investigated the relationship between the diagnostic and pathologic characteristics (tumor size, lymph node status, histologic grade, the gene expression of ER, PR, HER2, p53, Ki67 and PCNA) with the situation of co-expression of IDO and EGFR by reviewing the medical records of 32 breast cancer patients. Results: Among 110 breast cancers, 32 cases demonstrated IDO and EGFR co-expression (29.1%), IDO and EGFR synchronous co-expression being found in 19.1% and asynchronous in 10.0%. Conclusions: IDO and EGFR were co-expressed in breast cancer, including synchronous and asynchronous co-expression. The results suggest that considering IDO and EGFR as two indicators for breast cancer treatment or prognosis analysis provides a potential option of individual treatment for the portion of breast cancer patients with co-expression of IDO and EGFR.

        Enhanced interfacial adhesion between polypropylene and carbon fiber by graphene oxide/polyethyleneimine coating

        In order to improve the interfacial properties in polypropylene/carbonfiber (PP/CF) composites,graphene oxide and brached polyethyleneimine were coated onto the surface of carbonfiber by layer-by-layer assembly in this work. Compared with the origin PP/CF composite, the composites reinforced by PP/CF–GO showed significant enhancement not only in tensile strength but also in elongation at break. Theimprovedfiber–matrix adhesion was proved by fracture morphology observation of scanning electronmicroscopy and almost unaffected mechanical properties of thefiber itself during the coating process. The optimal assembly time was found to be 10 for enhancing the overall composite mechanicalperformance.

      • Towards Social Recommendation based on Probabilistic Matrix Factorization

        As an important tool to help users filter Internet information, recommender system has played a very important role wherever in academia or in industrial area. During the past years, different recommendation approaches based on the social network have been proposed with the rapid development of online social networks. Different from the traditional ones which assume all the users are independent and identically distributed, these approaches follow the intuition that a person’s implicit or explicit social network will affect his behaviors on the Web. In this paper, on the basis of the existing work, we fuse a baseline predictor model with an improved social recommendation model and propose a social recommendation algorithm based on probability matrix factorization. The experimental result shows that our method outperforms the existing approaches in accuracy.

      • Vibration-based delamination detection of composites using modal data and experience-based learning algorithm

        In this paper, a vibration-based method using the change ratios of modal data and the experience-based learning algorithm is presented for quantifying the position, size, and interface layer of delamination in laminated composites. Three types of objective functions are examined and compared, including the ones using frequency changes only, mode shape changes only, and their combination. A fine three-dimensional FE model with constraint equations is utilized to extract modal data. A series of numerical experiments is carried out on an eight-layer quasi-isotropic symmetric (0/-45/45/90)s composited beam for investigating the influence of the objective function, the number of modal data, the noise level, and the optimization algorithms. Numerical results confirm that the frequency-and-mode-shape-changes-based technique yields excellent results in all the three delamination variables of the composites and the addition of mode shape information greatly improves the accuracy of interface layer prediction. Moreover, the EBL outperforms the other three state-of-the-art optimization algorithms for vibration-based delamination detection of composites. A laboratory test on six CFRP beams validates the frequency-and-mode-shape-changesbased technique and confirms again its superiority for delamination detection of composites.

        A general closed-form solution to a Timoshenko beam on elastic foundation under moving harmonic line load

        In this paper, a general closed-form solution for evaluating the dynamic behavior of a Timoshenko beam on elastic foundation under a moving harmonic line load is formulated in the frequency-wavenumber domain and in a moving coordinate system. It is found that the characteristic equation is quartic with real coefficients only, and its poles can be presented explicitly. This enables the substitution of these poles into Cauchy's residue theorem, leading to the general closed-form solution. The solution can be reduced to seven existing closed-form solutions to different sub-problems and a new closed-form solution to the subproblem of a Timoshenko beam on an elastic foundation subjected to a moving quasi-static line load. Two examples are included to verify the solution.

        What are the Individual`s Real Cares to Switch Personal Cloud Services?

        As the fast development of information technology (IT), abundant attractive alternatives have showed up and challenged to the traditional information technology (IT). Cloud services, the hottest representative among these alternatives, has attracted the attention from all walks of life. Considering the diversity of cloud services, this study attempts to find out the important factors affected on individual`s switching intention from incumbent IT to cloud services mainly based on two-factor theory (switching enablers and switching inhibitors) and explore the direct influences of these factors on individual`s switching intention. According to our findings, individual`s switching intention to cloud services is not only positively influenced by switching enablers but also negatively influenced by switching inhibitors. All the switching enablers in this study (perceived omnipresence, perceived collaboration support and perceived compatibility of cloud services) have positively significant influences on individual`s switching intention as well as the switching inhibitors (usage habit of incumbent IT and perceived risk of cloud services) have negatively significant influences on individual`s switching intention. It`s noteworthy that satisfaction of incumbent IT has insignificant influence on individual`s switching intention in this study. Moreover, as we expected, both social influence and personal innovativeness have significant influences on the generation of individual`s switching intention.

        Metagenomics analysis of the gut microbiome in healthy and bacterial pneumonia forest musk deer

        Background The forest musk deer (FMD, Moschus berezovskii) is an threatened species in China. Bacterial pneumonia was found to seriously restrict the development of FMD captive breeding. Historical evidence has demonstrated the relationship between immune system and intestinal Lactobacillus in FMD. Objective We sought to elucidate the diferences in the gut microbiota of healthy and bacterial pneumonia FMD. Methods The bacterial pneumonia FMD was demonstrated by bacterial and pathological diagnosis, and the gut microbiome of healthy and bacterial pneumonia FMD was sequenced and analysed. Results There are three pathogens (Pseudomonas aeruginosa, Streptococcus equinus and Trueperella pyogenes) isolated from the bacterial pneumonia FMD individuals. Compared with the healthy group, the abundance of Firmicutes and Proteobacteria in the pneumonia group was changed, and a high level of Proteobacteria was found in the pneumonia group. In addition, a higher abundance of Acinetobacter (p=0.01) was observed in the population of the pneumonia group compared with the healthy group. Several potentially harmful bacteria and disease-related KEGG subsystems were only found in the gut of the bacterial pneumonia group. Analysis of KEGG revealed that many genes related to type IV secretion system, type IV pilus, lipopolysaccharide export system, HTH-type transcriptional regulator/antitoxin MqsA, and ArsR family transcriptional regulator were signifcantly enriched in the metagenome of the bacterial pneumonia FMD. Conclusion Our results demonstrated that the gut microbiome was signifcantly altered in the bacterial pneumonia group. Overall, our research improves the understanding of the potential role of the gut microbiota in the FMD bacterial pneumonia.

      • Autophagy Inhibition Promotes Gambogic Acid-induced Suppression of Growth and Apoptosis in Glioblastoma Cells

        Objective: To investigate the effects of gambogic acid (GA) on the growth of human malignant glioma cells. Methods: U251MG and U87MG human glioma cell lines were treated with GA and growth and proliferation were investigated by MTT and colony formation assays. Cell apoptosis was analyzed by annexin V FITC/PI flow cytometry, mitochondrial membrane potential assays and DAPI nuclear staining. Monodansylcadaverine (MDC) staining and GFP-LC3 localisation were used to detect autophagy. Western blotting was used to investigate the molecular changes that occurred in the course of GA treatment. Results: GA treatment significantly suppressed cell proliferation and colony formation, induced apoptosis in U251 and U87MG glioblastoma cells in a time- and dose-dependent manner. GA treatment also lead to the accumulation of monodansylcadaverine (MDC) in autophagic vacuoles, upregulated expressions of Atg5, Beclin 1 and LC3-II, and the increase of punctate fluorescent signals in glioblastoma cells pre-transfected with GFP-tagged LC3 plasmid. After the combination treatment of autophagy inhitors and GA, GA mediated growth inhibition and apoptotic cell death was further potentiated. Conclusion: Our results suggested that autophagic responses play roles as a self-protective mechanism in GA-treated glioblastoma cells, and autophagy inhibition could be a novel adjunctive strategy for enhancing chemotherapeutic effect of GA as an anti-malignant glioma agent.
