        Establishment of a special pathogen free Chinese Wuzhishan Minipigs Colony

        To meet the increasing demands of specific pathogen free (SPF) minipigs in biomedical researches, 8 pregnant Chinese Wuzhishan minipigs (WZSP) sows with clear background were chosen to obtain SPF WZSP by hysterectomy. At 111 ± 2 days of the pregnancy, piglets were aseptically taken out from the sows and artificially suckled for 40 to 45 days in the positive isolators. Then, the piglets defined as F0 were transferred to barrier environment and fed with standard feeds. The original SPF colony was formed for breeding by selected piglets from F0 group of 6-8 months old. Biological characteristics of SPF WZSP were collected and further compared to those of conventional (CV) WZSP, including growth performance, reproductive performance, hematology and blood biochemistry, and major pathogens detection. As a result, 61 F0 piglets were obtained from 8 candidate sows, and 55 out of them survived. After strictly selection, 35 F0 piglets were used to form the original SPF colony, which produced 14 litters of SPF piglets defined as F1. Piglet survival rates, growth performance, and reproductive performance of SPF WZSP were similar to CV WZSP. Some hematology and blood biochemistry parameters showed significant differences between SPF and CV WZSP. 18 kinds of pathogens were identified to be free in F0 and F1 SPF colony by repeated pathogen detections. In conclusion, we established a satisfied SPF WZSP colony maintaining original characteristics, free of controlled diseases, and being proved to be a suitable laboratory animal.

        Traditional Chinese medicine for COVID-19 pandemic and emerging challenges: An online cross-sectional survey in China

        Background We aimed to investigate use of infection control behaviours, preventative and therapeutic interventions, and outcomes among respondents to an online survey during the COVID-19 pandemic in China. Methods The survey was designed by an international team, translated and adapted to simplified Chinese, including 132 kinds of traditional Chinese medicine (TCM) preparation recommended by guidelines. It was distributed and collected from February to May 2021, with data analysed by WPS spreadsheet and wjx.cn. Descriptive statistics were used to describe demographics and clinical characteristics, diagnosis, treatments, preventative behaviours and interventions, and their associated outcomes. Results The survey was accessed 503 times with 341 (67.8%) completions covering 23 provinces and four municipalities in China. Most (282/341, 82.7%) respondents reported no symptoms during the pandemic and the majority (290/341, 85.0%) reported having a SARS-CoV-2 PCR test at some point. Forty-five (13.2%) reported having a respiratory infection, among which 19 (42.2%) took one or more categories of modern medicine, e.g. painkillers, antibiotics; 16 (35.6%) used TCM interventions(s); while seven respondents combined TCM with modern medicine. All respondents reported using at least one behavioural or medical approach to prevention, with 22.3% taking TCM and 5.3% taking modern medicines. No respondents reported having a critical condition related to COVID-19. Conclusion We found evidence of widespread use of infection control behaviours, modern medicines and TCM for treatment and prevention of COVID-19 and other respiratory symptoms. Larger scale studies are warranted, including a more representative sample exploring TCM preparations recommended in clinical guidelines.         Effect of wind time on the dispersal capacity of Trichogramma dendrolimi Matsumura (Hymenoptera Trichogrammatidae)

        The efficacy of Trichogramma against pests basically depends on the dispersal capacity of the wasps. A releaserecapture method was used in this study to evaluate the effect of relative wind speed (RWS) during the nighttime and the daytime on the dispersal capacity of postrelease population of Trichogramma dendrolimi Matsumura. Two repeated measurements were carried out, using sentinel cards with the host eggs and yellow sticky card traps on the third and the sixth day after wasp release. The results showed that: (1) both the number of recaptured wasps and parasitized eggs increased with the RWS in the daytime during the first three days. The number of parasitized eggs also increased with the RWS in the daytime over the latter three days. The distances that encompassed 98% recaptured T. dendrolimi adults (X 98 ) at the first three days was quadratically increased by the RWS of the daytime. (2) The number of wasps recaptured decreased with the distance from the releasing point during the first three days, but it was not influenced by the distance from the releasing point during the latter three days. (3) The average dispersal radius of the wasps during the first three days was shorter than during the latter three days. The present results will be helpful to improve release techniques based on the wind effects at different times.

        Experimental investigation of lateral displacement of PVD-improved deposit

        Laboratory model tests were conducted to investigate the effect of surcharge loading rate on the magnitude of lateral displacement of prefabricated vertical drains (PVDs) improved deposit. The test results indicate that under the condition that the system had sufficient factor of safety (FS) ($FS{\geq}1.2$), for the similar model ground under the same total applied surcharge load, the lateral displacement increases with the increase of loading rate. The test results have been used to check the validity of a previously proposed method for predicting the maximum lateral displacement, and it shows that the data points are around the middle line of the predicted range, which supports the usefulness of the proposed method. The basic idea of the prediction method is an empirical relationship between the normalized lateral displacement (NLD) and a ration of load to the undrained shear strength of the deposit (RLS). The model test results offer some modifications of the NLD-RLS relationship: (1) instead of a bilinear relationship, NLD-RLS relationship may be entirely nonlinear; (2) the upper bound value of RLS for the proposed method can be used may be limited to 2.1 instead of the originally proposed value of 3.0.

      • Clinicopathological and Prognostic Characteristics of Triple-Negative Breast Cancer (TNBC) in Chinese Patients: A Retrospective Study

        Aims: To determine the clinical, pathological and prognostic features associated with triple-negative breast cancer (TNBC). Methods: Clinical and histologic data of 21,749 breast cancer patients who were treated at Tianjin Medical University Cancer Institute and Hospital between July 2002 and December 2011 were collected. Patients were divided into two groups: those with TNBC and those with other types of breast cancer. Patients and tumor characteristics were compared between the two groups using the Chi-square test. The prognostic results of 9,823 patients in the study population were also analyzed to determine long-term survival rates in the two groups of breast cancer patients. Results: Among the breast cancer patients treated in our hospital between 2003 and 2011, 10.4%-13.5% of them had triple-negative breast cancers. Data analyses revealed significant differences in disease onset age, family history of breast cancer, tumor size, tumor histologic grade, lymph note positivity and metastatic status between TNBC and non-TNBC patients. There were also significant differences in 5-year, 7-year and 9-year disease-free and 7-year and 9-year overall survival probability between the groups. Conclusions:TNBC are associated with younger disease onset age, larger tumor size, higher rate of axillary lymph node positivity, and higher tumor histologic grade. TNBC is also related to family history of breast cancer, increased metastatic risk and poor prognosis.

        A Proteomic Analysis of Leaf Responses to Enhanced Ultraviolet-B Radiation in Two Rice (Oryza sativa L.) Cultivars Differing in UV Sensitivity

        To determine the proteomic response to UV irradiation, two cultivars, i.e., Lemont (UV tolerant) and Dular (UV sensitive), were exposed to natural and enhanced ultraviolet-B (UV-B) irradiation for 1, 7, and 14 days, and two-dimensional gel electrophoresis in combination with mass spectrometry (MS) and bioinformatics were used to compare the different proteomic responses in the leaves of the two cultivars. Thirty-nine proteins were up- or downregulated following the UV-B treatments. Among them, 30 increased or decreased more than 1.5-fold in abundance. They were further tested by using matrix-assisted laser desorption/ionization time of flight MS and performed a database search. Twentyfour proteins were thus identified. These identified proteins were mostly upregulated in Lemont, whereas only 14 of them upregulated in Dular. Nine proteins involved in glycometabolism and fatty acid metabolisms, signal transduction, and protein synthesis and folding in Dular were not changed. These results suggest that there was a complex regulative mechanism on the proteomes in rice leaves upon UV-B exposure.

        Effects of IFN-γ on IL-18 Expression in Pregnant Rats and Pregnancy Outcomes

        The present study focused on establishing the effects of interferon-gamma (IFN-${\gamma}$) on interleukin-18 (IL-18) expression patterns and pregnancy outcomes in pregnant rats. Pregnant rats at the post-implantation stage were randomized into control, low IFN-${\gamma}$ (L-IFN-${\gamma}$) and high IFN-${\gamma}$ groups (H-IFN-${\gamma}$) that received normal saline, 100 IU/g of IFN-${\gamma}$ and 500 IU/g of IFN-${\gamma}$ vaginal muscular injection, respectively. The effects of IFN-${\gamma}$ on IL-18 expression and pregnancy outcomes were assessed systematically using several methods, including immunohistochemistry streptavidin-perosidase (SP), image pattern analysis, enzyme-linked immune-sorbent assay (ELISA), whole blood count (WBC) count, microscopy and visual observation. IL-18 was detected in the uteri of all pregnant rats, and mainly distributed in the endometrium, decidual cells, vascular endothelium and myometrium. Immunohistochemistry and image pattern analyses revealed significantly lower IL-18 expression in the H-IFN-${\gamma}$ group compared to the L-IFN-${\gamma}$ and control groups (p<0.01), indicating that high doses of IFN-${\gamma}$ induce downregulation of IL-18 in the uterus of pregnant rats. ELISA results disclosed that IL-18 expression in peripheral blood of the H-IFN-${\gamma}$ group was lower than that of the L-IFN-${\gamma}$ group (p<0.05), and significantly reduced compared to the control group (p<0.01). Moreover, the number of peripheral leukocytes in the H-IFN-${\gamma}$ group was significantly higher than those in the control and L-IFN-${\gamma}$ groups (p<0.01). Morphology analysis showed no evident differences between the L-IFN-${\gamma}$ and control groups. However, for the H-IFN-${\gamma}$ group, uterine mucosa bleeding, necrosis and excoriation were observed using microscopy. Visual observation revealed marroon, swelling, crassitude and no embryo in the uterus, which are obvious indicators of abortion. These results indicate that IFN-${\gamma}$ plays a regulatory role in IL-18 expression in the uterus and peripheral blood of pregnant rats at the post-implantation stage. Moreover, high levels (500 IU/g) of IFN-${\gamma}$ influence normal pregnancy at the early stages in rats by downregulating IL-18 expression in the uterus and peripheral blood and increasing the number of peripheral leukocytes, consequently triggering termination of pregnancy.

        In general, most of the function and using of building is for single purpose. However, current buildings combine several functions that causes a lot of problems not on firefighting only but also on environment engineering. Because of hard integration on different fields that causes a lot of conflict. That wastes cost and time. That also threaten the safety of firefighting. This search focuses on the drawing verification and field inspection on firefighting. These two items both remain paper work. To complete the current work, it needs to bring a great amount of drawing papers in the field. By BIM, integrated data can be extracted. It makes the drawing verification and field inspection easier and increases the efficiency. That is the main point of this research.

      • Expression of Arabidopsis thaliana SIK (Stress Inducible Kinase) Gene in a Potato Cultivar (Solanum tuberosum L. Taedong Valley )

        Osmotic stress is one of major limiting factors in crop production. In particular, seasonal drought often causes the secondary disease in the field, resulting in severe reduction in both quality and productivity. Recent efforts have revealed that many genes encoding protein kinases play important roles in osmotic stress signal transduction pathways. Previously, the AtSIK (Arabidopsis thaliana Stress Inducible Kinase) mutants have shown to enhance tolerance to abiotic stresses, accompanying with higher expression of abiotic stress-related genes than did the wild-type plants. In this study, we have transformed potato (cv. Taedong Valley) with the AtSIK expression cassette. Both PCR and RT-PCR using AtSIK-specific primers showed stable integration and expression of the AtSIK gene in individual transgenic lines, respectively. Foliar application of herbicide (Basta(R)) at commercial application rate (0.3% (v/v)) revealed another evidence of stable gene introduction of T-DNA which includes the bar gene for herbicide resistance. Overexpression of the AtSIK gene under dual CaMV35S promoter increased sensitivity to salt stress (300 mM NaCl), which was demonstrated by the reduction rate of chlorophyll contents in leaves of transgenic potato lines. These results suggest that possible increase of osmotic tolerance in potato plants may be achieved by antisense expression of AtSIK gene.

        Use of In Vivo-Induced Antigen Technology to Identify In Vivo-Expressed Genes of Campylobacter jejuni During Human Infection

        Campylobacter jejuni is a prevalent foodborne pathogen worldwide. Human infection by C. jejuni primarily arises from contaminated poultry meats. Genes expressed in vivo may play an important role in the pathogenicity of C. jejuni. We applied an immunoscreening method, in vivo-induced antigen technology (IVIAT), to identify in vivo-induced genes during human infection by C. jejuni. An inducible expression library of genomic proteins was constructed from sequenced C. jejuni NCTC 11168 and was then screened using adsorbed, pooled human sera obtained from clinical patients. We successfully identified 24 unique genes expressed in vivo. These genes were implicated in metabolism, molecular biosynthesis, genetic information processing, transport, and other processes. We selected six genes with different functions to compare their expression levels in vivo and in vitro using real-time RT-PCR. The results showed that the selected six genes were significantly upregulated in vivo but not in vitro. In short, these identified in vivo-induced genes may contribute to human infection of C. jejuni, some of which may be meaningful vaccine candidate antigens or diagnosis serologic markers for campylobacteriosis. IVIAT may present a significant and efficient method for understanding the pathogenicity mechanism of Campylobacter and for finding targets for its prevention and control.
