Functional Analysis of Wheat TaPaO1 Gene Conferring Pollen Sterility Under Low Temperature

        Changping Zhao,Guoliang Yuan,Yukun Wang,Shaohua Yuan,Peng Wang,Wenjing Duan,Jianfang Bai,Hui Sun,Na Wang,Fengting Zhang,Liping Zhang 한국식물학회 2018 Journal of Plant Biology Vol.61 No.1

Thermosensitive male sterility plays an important role in wheat fertility and production. As a key enzyme for chlorophyll degradation, pheophorbide a oxygenase (PaO) can suppress cell death in plants. We cloned the wheat gene TaPaO1 from the thermosensitive genetic male sterile (TGMS) line BS366; it encodes a typical PaO protein, containing a conserved Rieske [2Fe-2S] iron–sulphur motif, a mononuclear non-heme iron-binding motif, and a C-terminal CxxC motif. TaPaO1 was expressed in all tissues and was upregulated during the meiosis stage of BS366 anthers under low temperature. Subcellular localization of TaPaO1 specifically labelled the surrounding of chloroplasts. TaPaO1 regulated by RD29A promoter which responded to low temperature led to pollen sterility in transgenic tobacco. Expression analysis showed that TaPaO1 exhibited a higher level of expression in the anther than in other tissues in transgenic tobacco plants during low temperature treatment. We propose that the higher senescence-related activity of TaPaO1 may lead to the cell death of anthers, which happens at an early developmental stage under low temperature. These results provide new insights into the function of PaO during the early developmental stage of anthers. PaO is closely related to cell death regardless of whether it exhibits increased activity or inactive.

Identification and Mapping of a Thermo-Sensitive Genic Self-Incompatibility Gene in Maize

        Xin Ge Lin,Hui Ling Xie,Zhang Ying Xi,Yan Min Hu,Guang Yuan Zhao,Liu Jing Duan,Zong You Hao,Zong Hua Liu,Ji Hua Tang 한국유전학회 2009 Genes & Genomics Vol.31 No.3

In this study, we describe a novel ecological self-incompatibility (SI) line HE97 in maize. The main environmental factors influencing the inbred line characteristics were identified through field sowing trials during a two-year study period (2001 and 2002). The results showed that daily minimum temperature had the greatest effect on floral morphology and breeding system of the SI line. In staminate floret differentiation, when the daily minimum temperature exceeded 24℃, the line exhibited complete self-compatibility; however SI was observed when the daily minimum temperature was below 20℃. Therefore, we characterized the line as exhibiting thermo-sensitive genic self-incompatibility (TGSI). A set of F2 and F2:3 populations, derived from the inbred lines HE97 and Z58, were evaluated for two years to elucidate the TGSI line patterns of inheritance. Classical genetic analyses and QTL mapping results revealed that HE97 self-incompatibility was governed by a single allele, named here as tgsi1. The tgsi1 gene was mapped to chromosome 2 between SSR markers nc131 and bnlg1633, with a distance of 2.40 cM from nc131 and 2.44 cM from bnlg1633.

Exploring the Key Genes and Pathways of Osteoarthritis in Knee Cartilage in a Rat Model Using Gene Expression Profiling

        Yi-Ming Ren,Xin Zhao,Tao Yang,Yuan-Hui Duan,Yun-Bo Sun,Wen-Jun Zhao,Meng-Qiang Tian 연세대학교의과대학 2018 Yonsei medical journal Vol.59 No.6

Purpose: To compare differentially expressed genes (DEGs) mediating osteoarthritis (OA) in knee cartilage and in normal kneecartilage in a rat model of OA and to identify their impact on molecular pathways associated with OA. Materials and Methods: A gene expression profile was downloaded from the Gene Expression Omnibus database. Analysis ofDEGs was carried out using GEO2R. Enrichment analyses were performed on the Gene Ontology (GO) and Kyoto Encyclopedia ofGenes and Genomes pathway using the Search Tool for the Retrieval of Interacting Genes database (http://www.string-db.org/). Subsequently, the regulatory interaction network of OA-associated genes was visualized using Cytoscape software (version 3.4.0;www.cytoscape.org). Results: In the gene expression profile GSE103416, a total of 99 DEGs were identified. Among them, 76 DEGs (76.77%) were overexpressed, and the remaining 23 DEGs (23.23%) were underexpressed. GO and pathway enrichment analyses of target geneswere performed. Using gene-gene interaction network analysis, relevant core genes, including MET, UBB, GNAI3, and GNA13,were shown to hold a potential relationship with the development of OA in cartilage. Using quantitative real-time PCR, the Gna13/cGMP-PKG signaling pathway was identified as a potential research target for therapy and for further understanding the developmentof OA. Conclusion: The results of the present study provide a comprehensive understanding of the roles of DEGs in knee cartilage in relationto the development of OA.

Fishbone Oscillations in the Experimental Advanced Superconductivity Tokamak

        Liqing Xu,Liqun Hu,Yi Yuan,Yingying Li,Guo-Qiang Zhong,Hai-Qing Liu,Kaiyun Chen,Tong-Hui Shi,Yan-Min Duan 한국물리학회 2018 THE JOURNAL OF THE KOREAN PHYSICAL SOCIETY Vol.72 No.6

A fishbone oscillation was observed in the neutral beam injection plasma at Experimental Ad- vanced Superconductivity Tokamak (EAST). This m = 1/n = 1 (m, n: poloidal, toroidal mode numbers, respectively) typical internal kink mode travels in the ion-diamagnetism direction in the poloidal section with a rotation speed close to the ion diamagnetic drift frequency. A high thermal plasma beta and high amounts of energetic ions are necessary for the mode to develop. Fishbone oscillations can expel heavy impurities in the core, which favors sustaining a high-performance plasma. The born frequency of the fishbone oscillation is the ion diamagnetic drift frequency and the chirping down of the frequency during the initial growth phase is the result of a drop in ion- diamagnetic drift frequency. The excitation energy is thought to be due to the thermal plasma pressure gradient; however, the development of a fishbone oscillation is related to energetic ions. Effect of wind time on the dispersal capacity of Trichogramma dendrolimi Matsumura (Hymenoptera Trichogrammatidae)

        Jin-cheng Zhou,Qian-jin Dong,Tong-shu Zhang,Li-jia Duan,Su-fang Ning,Quanquan Liu,Yuan-yuan Li,Chun-xue Li,Hui Dong 한국응용곤충학회 2019 Journal of Asia-Pacific Entomology Vol.22 No.3

The efficacy of Trichogramma against pests basically depends on the dispersal capacity of the wasps. A releaserecapture method was used in this study to evaluate the effect of relative wind speed (RWS) during the nighttime and the daytime on the dispersal capacity of postrelease population of Trichogramma dendrolimi Matsumura. Two repeated measurements were carried out, using sentinel cards with the host eggs and yellow sticky card traps on the third and the sixth day after wasp release. The results showed that: (1) both the number of recaptured wasps and parasitized eggs increased with the RWS in the daytime during the first three days. The number of parasitized eggs also increased with the RWS in the daytime over the latter three days. The distances that encompassed 98% recaptured T. dendrolimi adults (X 98 ) at the first three days was quadratically increased by the RWS of the daytime. (2) The number of wasps recaptured decreased with the distance from the releasing point during the first three days, but it was not influenced by the distance from the releasing point during the latter three days. (3) The average dispersal radius of the wasps during the first three days was shorter than during the latter three days. The present results will be helpful to improve release techniques based on the wind effects at different times.

      • Effect of Xeroderma Pigmentosum Complementation Group F Polymorphisms on Gastric Cancer Risk and Associations with H.pylori Infection

Zhang, Ji-Shun,Zhang, Chuan,Yan, Xue-Yan,Yuan, Zhi-Fang,Duan, Zhuo-Yang,Gao, Hui Asian Pacific Journal of Cancer Prevention 2013 Asian Pacific journal of cancer prevention Vol.14 No.3

We conducted a hospital case-control study by genotyping four potential functional single nucleotide polymorphisms (SNPs) to assess the association of Xeroderma pigmentosum complementation group F (XPF) with gastric cancer susceptibility, and role of XPF polymorphisms in combination with H.pylori infection in risk definition. A total of 331 patients with gastric cancer and 355 controls were collected. Four SNPs of XPF, rs180067, rs1799801, rs2276466 and rs744154, were genotyped by Taqman real-time PCR method with a 7900 HT sequence detector system. The gastric cancer patients were more likely to have smoking habit, a family history of cancer and H.pylori infection. We did not find any significant difference in the genotype distributions of XPF rs180067, rs1799801, rs2276466 and rs744154 between cases and controls. However, multivariate logistic analysis showed a non-significant decreased risk in patients carrying rs180067 G allele, rs1799801 T allele or rs2276466 T allele genotypes. A non-significant increased risk of gastric cancer was found in individuals carrying the rs744154 GG genotype. Stratification by H.pylori infection and smoking was not significantly different in polymorphisms of XPF rs180067, rs1799801, rs2276466 and rs744154. The four XPF SNPs did not show significant interaction with H.pylori infection and smoking status (P for interaction was 0.35 and 0.18, respectively). Our study indicated that polymorphisms in rs180067, rs1799801, rs2276466 and rs744154 may affect the risk of gastric cancer but further large sample size studies are needed to validate any association.
