        Treatment of bladder cancer by geoinspired synthetic chrysotile nanocarrier-delivered circPRMT5 siRNA

        Background: Circular RNAs (circRNAs) have important functions in many fields of cancer biology. In particular, we previously reported that the oncogenic circRNA, circPRMT5, has a major role in bladder cancer progression. Therapy based on circRNAs have good prospects as anticancer strategies. While anti-circRNAs are emerging as therapeutics, the specific in vivo delivery of anti-circRNAs into cancer cells has not been reported and remains challenging. Methods: Synthesized chrysotile nanotubes (SCNTs) with a relatively uniform length (~ 200 nm) have been designed to deliver an siRNA against the oncogenic circPRMT5 (si-circPRMT5) inhibit circPRMT5. In addition, the antitumor effects and safety evaluation of SCNTs/si-circPRMT5 was assessed with a series of in vitro and in vivo assays. Results: The results showed that SCNTs/si-circPRMT5 nanomaterials prolong si-circPRMT5’s half-life in circulation, enhance its specific uptake by tumor cells, and maximize the silencing efficiency of circPRMT5. In vitro, SCNTs encapsulating si-circPRMT5 could inhibit bladder cancer cell growth and progression. In vivo, SCNTs/si-circPRMT5 inhibited growth and metastasis in three bladder tumor models (a subcutaneous model, a tail vein injection lung metastatic model, and an in situ model) without obvious toxicities. Mechanistic study showed that SCNTs/sicircPRMT5 regulated the miR-30c/SNAIL1/E-adherin axis, inhibiting bladder cancer growth and progression. Conclusion: The results highlight the potential therapeutic utility of SCNTs/si-circPRMT5 to deliver si-circPRMT5 to treat bladder cancer.

      • Topic Sentiment Analysis in Chinese News

        Sentiment analysis in news is different from normal text sentiment analysis. News usually have a specific topic, a focus semantic emotion, therefore, this paper, based on the principal of using Emotion Dependency Tuple (EDT) as the basic unit of news emotion analysis, resolves topic sentiment analysis in news into three progressive sub-problem, namely, topic sentence recognition, EDT extraction and topic sentiment analysis. We use an improved TF-IDF and cross entropy to extract feature set of topics. Then, based on space vector model, calculate the topic association of a sentence and extract topic sentence. Finally, we construct topic sentence based on EDT and complete clustering of news topic sentiment. This method is evaluated using COAE2014 dataset, and differential means shows that our results close to the best results. This shows that the topic based EDT could effectively improve performance of sentiment analysis in news.

        Universal Indicators for Oil and Gas Prospecting Based on Bacterial Communities Shaped by Light-Hydrocarbon Microseepage in China

        Light hydrocarbons accumulated in subsurface soil by long-term microseepage could favor the anomalous growth of indigenous hydrocarbon-oxidizing microorganisms, which could be crucial indicators of underlying petroleum reservoirs. Here, Illumina MiSeq sequencing of the 16S rRNA gene was conducted to determine the bacterial community structures in soil samples collected from three typical oil and gas fields at different locations in China. Incubation with n-butane at the laboratory scale was performed to confirm the presence of "universal microbes" in light-hydrocarbon microseepage ecosystems. The results indicated significantly higher bacterial diversity in next-to-well samples compared with background samples at two of the three sites, which were notably different to oil-contaminated environments. Variation partitioning analysis showed that the bacterial community structures above the oil and gas fields at the scale of the present study were shaped mainly by environmental parameters, and geographic location was able to explain only 7.05% of the variation independently. The linear discriminant analysis effect size method revealed that the oil and gas fields significantly favored the growth of Mycobacterium, Flavobacterium, and Pseudomonas, as well as other related bacteria. The relative abundance of Mycobacterium and Pseudomonas increased notably after n-butane cultivation, which highlighted their potential as biomarkers of underlying oil deposits. This work contributes to a broader perspective on the bacterial community structures shaped by long-term light-hydrocarbon microseepage and proposes relatively universal indicators, providing an additional resource for the improvement of microbial prospecting of oil and gas.

        Solvothermal Synthesis of TiO2/CNFs Heterostructures with Photocatalytic Activity

        Titanium dioxide nanoparticles (TiO2 NPs) embedded carbon nanofibers (CNFs) have been successfully prepared by electrospinning technique, solvothermal synthesis and high-temperature calcination processes. The as-obtained products were characterized by field-emission scanning electron microscopy (FE-SEM), transmission electron microscopy (TEM), X-ray diffraction (XRD), UV-Vis diffuse reflectance spectra (DRS) and Fourier transform infrared (FTIR) spectra. The results of characterization revealed that solvothermal treatment and high-temperature calcination were beneficial to the synthesis of anatase TiO2. Compared to the as-fabricated TiO2 powder, TiO2 NPs contained in the TiO2/CNFs were successfully grown on the surface of CNFs without aggregation. The photocatalytic activities of the TiO2/CNFs heterostructures were evaluated by the degradation of methyl orange (MO) under UV light irradiation for 14 h. The TiO2/CNFs exhibited higher photocatalytic activities than pure TiO2 powder, which were attributed to the small size of TiO2 NPs, electron transfer, high adsorption properties of CNFs and the formation of heterostructures between TiO2 and CNFs. Moreover, in aqueous solution, the as-prepared TiO2/CNFs could be easily recycled for the application of CNFs.

        Nonenzymatic Electrochemical Immunosensor Using Ferroferric Oxide–Manganese Dioxide–Reduced Graphene Oxide Nanocomposite as Label for α-Fetoprotein Detection

        A novel nonenzymatic electrochemical immunosensor was fabricated for quantitative detection of α-fetoprotein (AFP). The immunosensor was constructed by modifying gold electrode with electrochemical reduction of graphene oxide-carboxyl multi-walled carbon nanotube composites (ERGO–CMWCNTs) and electrodeposition of gold nanoparticles (AuNPs) for effective immobilization of primary antibody (Ab1). Ferroferric oxide–manganese dioxide–reduced graphene oxide nanocomposites (Fe3O4@MnO2–rGO) were designed as labels for signal amplification. On one hand, the excellent electroconductivity and outstanding electron transfer capability of ERGO–CMWCNTs/AuNPs improved the sensitivity of the immunosensor. On the other hand, introduction of rGO could not only increase the specific surface area for immobilization of secondary antibody (Ab2) but also build a synergetic effect to reinforce the electrocatalytic properties of catalysts. Fe3O4@MnO2–rGO nanocomposites were characterized by scanning electron microscope, Fourier transform infrared spectroscopy and X-ray photoelectron spectroscopy. Using AFP as a model analyte, the proposed sandwich-type electrochemical immunosensor exhibited a wide linear range of 0.01–50 ng · mL-1 with a low detection limit of 5.8 pg · mL-1. Moreover, the Fe3O4@MnO2–rGO-based peroxidase mimetic system displayed an excellent analytical performance with low cost, satisfactory reproducibility and high selectivity, which could be further extended for detecting other disease-related biomarkers.

        Influence of Monascus purpureus BD-M-4 on the physicochemical properties, proteolysis and volatile compounds of surface mouldripened cheese

        The effect of adjunct culture Monascus purpureusBD-M-4 on the physicochemical, proteolytic, andlipolytic properties of surface mold-ripened cheese wereevaluated. During the maturation of Monascus-fermentedcheese, the total microbiota count and the content of solublenitrogen increased steadily, whereas the total proteincontent showed no significant difference. Moreover, a17-fold increase in total free amino acids was observed inMonascus-fermented cheese. The use of adjunct culture M. purpureus BD-M-4 in the production of surface-ripenedcheeses did not show a significant effect on the total fatcontent in the ripening period, nor did it change thelipolysis of cheese during ripening. Compared to 52 volatilecompounds of the control cheese, a total of 62 compoundswere detected in M. purpureus-fermented cheese,including 16 acids, 16 ketones, 11 alcohols, 5 aldehydes,11 esters, and 3 unclassified compounds.

        Transcriptome profiling identifies immune response genes against porcine reproductive and respiratory syndrome virus and Haemophilus parasuis co-infection in the lungs of piglets

        Background: Co-infections of the porcine reproductive and respiratory syndrome virus (PRRSV) and the Haemophilus parasuis (HPS) are severe in Chinese pigs, but the immune response genes against co-infected with 2 pathogens in the lungs have not been reported. Objectives: To understand the effect of PRRSV and/or HPS infection on the genes expression associated with lung immune function. Methods: The expression of the immune-related genes was analyzed using RNA-sequencing and bioinformatics. Differentially expressed genes (DEGs) were detected and identified by quantitative real-time polymerase chain reaction (qRT-PCR), immunohistochemistry (IHC) and western blotting assays. Results: All experimental pigs showed clinical symptoms and lung lesions. RNA-seq analysis showed that 922 DEGs in co-challenged pigs were more than in the HPS group (709 DEGs) and the PRRSV group (676 DEGs). Eleven DEGs validated by qRT-PCR were consistent with the RNA sequencing results. Eleven common Kyoto Encyclopedia of Genes and Genomes pathways related to infection and immune were found in single-infected and co-challenged pigs, including autophagy, cytokine-cytokine receptor interaction, and antigen processing and presentation, involving different DEGs. A model of immune response to infection with PRRSV and HPS was predicted among the DEGs in the co-challenged pigs. Dual oxidase 1 (DUOX1) and interleukin-21 (IL21) were detected by IHC and western blot and showed significant differences between the co-challenged pigs and the controls. Conclusions: These findings elucidated the transcriptome changes in the lungs after PRRSV and/or HPS infections, providing ideas for further study to inhibit ROS production and promote pulmonary fibrosis caused by co-challenging with PRRSV and HPS.

        Targeting treatment of bladder cancer using PTK7 aptamer-gemcitabine conjugate

        Gemcitabine (GEM) is one of the first-line chemotherapies for bladder cancer (BC), but the GEMs cannot recognize cancer cells and have a low long-term response rate and high recurrence rate with side effects during the treatment of BC. Targeted transport of GEMs to mediate cytotoxicity to tumor and avoid the systemic side effects remains a challenge in the treatment of BC.Based on a firstly confirmed biomarker in BC-protein tyrosine kinase 7 (PTK7), which is overexpressed on the cell membrane surface in BC cells, a novel targeting system protein tyrosine kinase 7 aptamer-Gemcitabine conjugate (PTK7-GEMs) was designed and synthesized using a specific PTK7 aptamer and GEM through auto-synthesis method to deliver GEM against BC. In addition, the antitumor effects and safety evaluation of PTK7-GEMs was assessed with a series of in vitro and in vivo assays.PTK7-GEMs can specifically bind and enter to BC cells dependent on the expression levels of PTK7 and via the macropinocytosis pathway, which induced cytotoxicity after GEM cleavage from PTK7-GEMs respond to the intracellular phosphatase. Moreover, PTK7-GEMs showed stronger anti-tumor efficacy and excellent biosafety in three types of tumor xenograft mice models.These results demonstrated that PTK7-GEMs is a successful targeted aptamer-drug conjugates strategy (APDCs) to treat BC, which will provide new directions for the precision treatment of BC in the field of biomarker-oriented tumor targeted therapy.
