MgAl hydrotalcite synthesized via co-precipitation method from magnesium and aluminum nitrateswas calcined to produce Mg4AlO5.5 mixed oxides. The oxides were modified by introducing LiNO3 andapplied to the synthesis of glycerol carbonate from glycerol and dimethyl carbonate. The research resultsindicated that LiNO3/Mg4AlO5.5 was an efficient catalyst for the synthesis of glycerol carbonate. The fullconversion of glycerol and 96.28% yield of glycerol carbonate were obtained after reacting at 80 8C for1.5 h in the presence of LiNO3/Mg4AlO5.5. The structure and properties of the catalysts were studied bymeans of XRD, TGA, BET, CO2-TPD and Hammett indicator method. It was found that the basic strengthand the basicity of the catalysts were enhanced after addition of LiNO3. Over Mg4AlO5.5 oxides, LiNO3 waseasily converted to LiAlO2 which constituted new strong basic sites. Too high basic strength and basicityof catalysts may improve the conversion of glycerol at the cost of GC selectivity reduction.

Vesicular stomatitis virus G glycoprotein (VSV-G) has been widely used for pseudotyping retroviral, lentiviral, and artificial viral vectors. The objective of this study was to establish a potential approach for large-scale production of VSV-G. To this end, VSV-G was cloned with an N-terminal His-tag into Pichia pastoris expression vector pPIC3.5K. Three clones (Muts) containing the VSV-G expression cassette were identified by PCR. All clones proliferated normally in expansion medium, whereas the proliferation was reduced significantly under induction conditions. VSV-G protein was detected in cell lysates by western blot analysis, and the highest expression level was observed at 96 h post induction. VSV-G could also be obtained from the condition medium of yeast protoplasts. Furthermore, VSV-G could be incorporated into Ad293 cells and was able to induce cell fusion, leading to the transfer of cytoplasmic protein. Finally, VSV-G-mediated DNA transfection was assayed by flow cytometry and luciferase measurement. Incubation of VSV-G lysate with the pGL3-control DNA complex increased the luciferase activity in Ad293 and HeLa cells by about 3-fold. Likewise, incubation of VSV-G lysate with the pCMV-DsRed DNA complex improved the transfection efficiency into Ad293 by 10% and into HeLa cells by about 1-fold. In conclusion, these results demonstrate that VSV-G could be produced from P. pastoris with biofunctionalities, demonstrating that large-scale production of the viral glycoprotein is feasible.

A novel and convenient route for the synthesis of a series of thalidomide derivatives is described. Compound 2 was cyclized with different amines under alkaline condition to obtain 4-nitro substituted phthalimidines 3a-d. Hydroxylactams 4a-d were produced via bromination and hydroxylation. Different acyl chlorides were reacted with hydroxylactams to provide the desired esters 5a-d. All compounds were evaluated by MTT assay for their inhibitory activities against HCT-116, MG-63, MCF-7, HUVEC and HMVEC cell lines in vitro. Most of them showed no obvious cytotoxic effect on normal human cells, compounds 4a-d, $5a_2$, $5a_4$, $5a_5$, $5b_2$, $5c_2$ and $5d_2$ exhibited potent antitumor activities, among which compounds $5a_2$ and $5b_2$ were more effective than 5-FU.

Two kinds of semi-hard cheeses, with Monascus purpureus and without M. purpureus, were manufactured, and effects of M. purpureus on physicochemical properties and proteolysis were evaluated during 36 days of ripening. Addition of M. purpureus changed the microbial survival and showed no significant effect on physicochemical properties of the cheeses, including dry matter and pH. Regardless on the rind or in the core, the indices of proteolysis had no significant difference (p>0.05), whereas there were significant differences of total free amino acid (FAA) and individual FAA between cheeses; this indicated that M. purpureus had no significant effect on the primary proteolysis, but affected the content and ratio of individual FAAs during maturation. Electrophoretic analysis showed strong degradation of αs1-casein in the core and on the rind of cheeses, while β-casein was highly degraded on the rind but less in the core. Thus, Monascus spp. might have a potential application in the manufacture of cheeses.

Fusarium oxysporum trypsin (FOT) is a fungal serine protease similar to mammal trypsin. The FOT could be successfully expressed in Pichiapastoris by engineering the natural propeptide APQEIPN. In this study, we constructed two recombinant enzymes with engineered amino acid sequences added to the N-terminus of FOT and expressed in P. pastoris. The N-terminal residues had various effects on the structural and functional properties of trypsin. The FOT, and the recombinants TE (with peptide YVEF) and TS (with peptide YV) displayed the same optimum temperature (40°C) and pH (8.0). However, the combinants TE and TS showed significantly increased thermal stability at 40°C and 50°C. Moreover, the combinants TE and TS also showed enhanced tolerance of alkaline pH conditions. Compared with those of wildtype FOT, the intramolecular hydrogen bonds and the cation π-interactions of the recombinants TE and TS were significantly increased. The recombinants TE and TS also had significantly increased catalytic efficiencies (referring to the specificity constant, kcat/Km), 1.75- fold and 1.23-fold than wild-type FOT. In silico modeling analysis uncovered that the introduction of the peptides YVEF and YV resulted in shorter distances between the substrate binding pocket (D174, G198, and G208) and catalytic triad (His42, Asp102, and Ser180), which would improve the electron transfer rate and catalytic efficiency. In addition, N-terminal residues modification described here may be a useful approach for improving the catalytic efficiencies and characteristics of other target enzymes.

The effect of adjunct culture Monascus purpureusBD-M-4 on the physicochemical, proteolytic, andlipolytic properties of surface mold-ripened cheese wereevaluated. During the maturation of Monascus-fermentedcheese, the total microbiota count and the content of solublenitrogen increased steadily, whereas the total proteincontent showed no significant difference. Moreover, a17-fold increase in total free amino acids was observed inMonascus-fermented cheese. The use of adjunct culture M. purpureus BD-M-4 in the production of surface-ripenedcheeses did not show a significant effect on the total fatcontent in the ripening period, nor did it change thelipolysis of cheese during ripening. Compared to 52 volatilecompounds of the control cheese, a total of 62 compoundswere detected in M. purpureus-fermented cheese, including 16 acids, 16 ketones, 11 alcohols, 5 aldehydes,11 esters, and 3 unclassified compounds.

Patch based Semi-supervised Linear Regression for Face Recognition
( Yuhua Ding ),( Fan Liu ),( Ting Rui ),( Zhenmin Tang ) 한국인터넷정보학회 2019 KSII Transactions on Internet and Information Syst Vol.13 No.8
To deal with single sample face recognition, this paper presents a patch based semi-supervised linear regression (PSLR) algorithm, which draws facial variation information from unlabeled samples. Each facial image is divided into overlapped patches, and a regression model with mapping matrix will be constructed on each patch. Then, we adjust these matrices by mapping unlabeled patches to [1,1,···1]T. The solutions of all the mapping matrices are integrated into an overall objective function, which uses ℓ2,1-norm minimization constraints to improve discrimination ability of mapping matrices and reduce the impact of noise. After mapping matrices are computed, we adopt majority-voting strategy to classify the probe samples. To further learn the discrimination information between probe samples and obtain more robust mapping matrices, we also propose a multistage PSLR (MPSLR) algorithm, which iteratively updates the training dataset by adding those reliably labeled probe samples into it. The effectiveness of our approaches is evaluated using three public facial databases. Experimental results prove that our approaches are robust to illumination, expression and occlusion.

The antihypertensive activity of few-flower wildrice was studied in spontaneously hypertensive rats (SHRs)with evaluation of blood pressure lowering effects andtranscriptional levels of the sarcoplasmic reticulum Ca2+-ATPase (SERCA2a) gene that is regulated by AngiotensinII (Ang II). SHRs were randomly divided into 5 groupswith 6 rats each. The systolic blood pressure (SBP) reachedthe lowest point 3 h after administration of a single dose ofpaste made from few-flower wild rice stem powder. TheSBP of SHR in the relatively high amount of RSP (HRSP)administrated group was reduced by approximately 30mmHg, compared to the negative control group, and wasnot significantly different from the positive control IPPcontrol group at a dose of 1.5 mg/kg body weight (p>0.05). RSP administrated SHRs showed a significantly higherSERCA2a transcription level than negative control SHRs(p<0.05). RSP administration had no negative effects onglycometabolism of SHR.

A series of SAPO-11 zeolites with different Si contents were prepared by hydrothermally synthesized method. They were characterized by ICP, XRD, SEM, FT-IR, N2 adsorption-desorption, NH3-TPD and 29Si MAS NMR, and evaluated by the methylation of naphthalene with methanol to 2,6-dimethylnaphthalene (2,6-DMN). According to XRD and SEM results, the crystallinity of SAPO-11 sample increased with increase of the Si content until the SiO2/Al2O3 ratio was up to 0.2. However, a reduction in the crystallinity was observed with further increase of the Si content of the synthesis. N2 adsorption-desorption results showed that all the samples possessed micropores and secondary mesopores. SAPO-11 sample with SiO2/Al2O3 ratio of 0.2 exhibited the largest secondary mesopores size distributions. NH3-TPD and 29Si MAS NMR showed that the Si content was incorporated into the framework affecting not only the acid sites but also the acid strength of SAPO-11. SAPO-11 with SiO2/Al2O3 ratio of 0.2 presented the high catalytic performances for the methylation of naphthalene, which was mainly attributed to the amount of secondary mesopores in the SAPO-11 zeolite.
