        The Effects of Counseling about Death and Dying on Perceptions, Preparedness, and Anxiety Regarding Death among Family Caregivers Caring for Hospice Patients: A Pilot Study

        Purpose: This pilot study aimed to examine the influence of death counseling on perceptions, preparedness, and anxiety regarding death and dying among family caregivers of hospice patients. Methods: Death counseling developed based on the SPIKES model was provided to 37 family caregivers in a hospice and palliative care unit. Perceptions, preparedness, and anxiety regarding death were assessed with a self-administered structured questionnaire, and participants' scores before and after counseling were compared using the paired t-test. Results: Significant changes were found in perceptions, preparedness, and anxiety regarding death after counseling. Compared to before counseling, the scores for perceptions of death (t=-4.90, P<0.001) and preparedness for death and dying (t=-16.23, P<0.001) improved, while anxiety (t=3.72, P=0.001) decreased after counseling. Some changes were also found in the types of support that family caregivers needed to prepare for the death of their family members in the hospice care unit. Conclusion: The findings demonstrate that death counseling could help family caregivers prepare for the death of their loved ones. Hospice and palliative care providers should play a key role in supporting family caregivers of hospice patients by developing strategies for counseling.

        질 내 유산균의 Candida albicans에 대한 항진균 효과 연구

        Vulvovaginal candidiasis (VVC) is one of the urogenital infections occurring in women worldwide. Candida albicans is generally observed among various types of microorganisms causing VVC. Antibiotic therapy is typical, and the use of Lactobacilli probiotics is to be recognized as a promising alternative. The aim of this study was to select vaginal lactobacilli with probiotic properties against C. albicans. In a previous study, we isolated 38 lactobacilli from vagina of Korean women and 20 isolates were shown to inhibit C. albicans. We further selected 10 isolates which were able to inhibit C. albicans less than 10<SUP>5</SUP> CFU/mL. Among these selected strains, Lactobacillus salivarius MG242 (identified by 16s rRNA sequencing) was finally selected based on its strong anti-candidal activity, acid/bile salt resistance and adhesion property. Indirect adhesion activity of MG242 measured by auto-aggregation assay showed more than 60% auto-aggregation after 5 h standing. Taken these results together, the selected strain MG242 may have potential for application in vagina health related products.

        흰쥐 관절염 모델에서 족삼리와 대릉 배오 전침의 상호억제 효과

        Objectives: We examined mutual inhibitory effects of combined acupoints in arthritic pain induce by carrageenan (CA). Electroacupuncture (EA) is considered a potentially useful treatment for arthritis. Although the analgesic effect of acupuncture is well documented, little is understood about its biological basis. There are many previous studies of positive effect of combined acupoint, this study was conducted to see the mutual inhibitory effects produced by combined acupoint (ST36 and PC7) on arthritic rats. Methods: For the induction of inflammatory pain rat model, CA was injected into the knee joint cavity. There are four groups; EA was applied to bilateral PC7 acupoints(PC7 group), ST36 acupoints (ST36 group), and both PC7 and ST36 acupoints (ST36+PC7 group) except for the control group. The pain level were assessed to determine the change in weight bearing force. We also examined the COX-2 expression in dorsal horn using immunohistochemistry and western blot analysis. Results: The ST36+PC7 group data showed the significant reduction of weight bearing force and the induction of COX-2 protein expression compared with the ST36 group. Conclusions: Simultaneous EA applied to the ST36 and PC7 acupoints reduced the analgesic effect of the ST36 group on knee inflammatory pain.

        진로미결정 대학생의 심리상담 경험에 관한 현상학적 연구

        본 연구는 진로미결정 대학생의 심리상담 경험에 대한 심리적 본질과 경험적 구조를 파악하는 것을 목적으로 한다. 이를 위해 A, B 대학교에 재학 중인 여학생 4명, 남학생 2명, 총 6명을 대상으로 심층 면담을 실시하였다. 수집된 자료는 현상학적 연구 방법을 통해 경험의 공통된 의미 단위를 기초로 하여 8개의 구성 요소와 23개의 하위 구성 요소를 도출하였다. 분석 결과, 진로미결정 대학생은 진로를 생각하면 머리가 복잡하고 가슴은 답답하여, 숨통을 틔우고 싶어 상담실을 두드린다. 한줄기 빛에 마음이 녹아 말문이 열리고, 곪은 상처를 입 밖으로 터트린다. 내 존재가 정성껏 수용되는 경험을 하여, 내면에 정서적으로 어떤 불편함이 있었는지 깨닫는다. 정서적으로 안정을 찾으며 진로 고민의 실마리를 얻는다는 변화를 보였는데, 이것을 역동적이고 지속적으로 일어난 의미 있는 경험들의 연속 속에서 하나의 현상으로 파악할 수 있었다. 연구 결과를 토대로 진로미결정 대학생의 심리상담 경험을 ‘곪은 상처가 아물어 정서적 안정감을 찾고 진로를 결정함’으로 그 본질적 의미를 규명하였고, 이에 대한 학문적・실천적 함의를 논하였으며, 연구의 의의와 제한점 및 제언을 제시하였다. The purpose of this study was to shed light on the meaning and the nature of their experiences on psychology counseling experience of the career undecided undergraduate clients. It s specifically meant to determine the nature of their experiences by investigating why the undergraduate clients came to the university advisement center, how the undergraduate clients solve a problem, what their experiences meant and what influence the experiences exerted on them in-depth interviews. Giorgi s phenomenological method, which is one of qualitative research methods, was utilized to analyze the experiences of the undergraduate clients. The subjects in this study were six selected the career undecided undergraduate clients. They were interviewed in depth, and the collected interview data were analyzed and interpreted by Giorgi s method. This study used the vivid stories obtained from six participants as purposive sampling and these interviews were done for two or three times after research agreement. As a result, 1343 meaning unit were derived and summarized into 149 meaning units, and back again tied to 23 sub-components and finally divided into 8 categories. The findings of the study on the nature of their experiences were as follows: First, participants in the psychology counseling application recognized their difficulties as career concerns. Second, participants had problems with attachment. Third, the core of the interventions in participants psychology counseling was the counselor s sincerity. So far, the career undecided undergraduate clients were solved it by seeking a sense of security based on the sincerity of the counselor. Yet the findings of the study might not be able to generalize since just a small number of the undergraduate clients with type based on the career decision by emotional factor were investigated. Based on the these results, the implications and limitations of this study were discussed, and the future research directions were suggested.

        Induction of steroid sulfatase expression by tumor necrosis factor-α through phosphatidylinositol 3-kinase/Akt signaling pathway in PC-3 human prostate cancer cells

        Steroid sulfatase (STS) is responsible for the hydrolysis of aryl and alkyl steroid sulfates and has a pivotal role in regulating the formation of biologically active estrogens. STS may be considered a new promising drug target for treating estrogen-mediated carcinogenesis. However,the molecular mechanism of STS expression is not well-known. To investigate whether tumor necrosis factor (TNF)-α is able to regulate gene transcription of STS, we studied the effect of TNF-α on STS expression in PC-3 human prostate cancer cells. RT-PCR and Western blot analysis showed that TNF-α significantly induced the expression of STS mRNA and protein in a concentration- and time-dependent manner. Treatment with TNF-α resulted in a strong increase in the phosphorylation of Akt on Ser-473 and when cells were treated with phosphatidylinositol (PI) 3-kinase inhibitors such as LY294002 or wortmannin, or Akt inhibitor (Akt inhibitor IV), induction of STS mRNA expression by TNF-α was significantly prevented. Moreover, activation of Akt1 by expressing the constitutively active form of Akt1 increased STS expression whereas dominant-negative Akt suppressed TNF-α-mediated STS induction. We also found that TNF-α is able to increase STS mRNA expression in other human cancer cells such as LNCaP, MDA-MB-231,and MCF-7 as well as PC-3 cells. Taken together, our results strongly suggest that PI 3-kinase/Akt activation mediates induction of human STS gene expression by TNF-α in human cancer cells.

        옥수수 대로부터 생리활성물질 생산 증대를 위한 마이크로파 추출 공정 최적화

        옥수수 대는 셀룰로오스와 헤미셀룰로오스 이외에 높은 함량의 리그닌을 포함하고 있어 리그닌 분해를 통해 폴리페놀 생산이 가능하여 천연 항산화물 생산이 가능한 후보이다. 옥수수 대로부터 마이크로파 전처리를 이용해 폴리페놀과 플라보노이드 추출 증대를 위해 반응표면분석법(RSM)을 이용하여 공정조건을 최적화하였다. 폴리페놀과 플라보노이드의 함량은 마이크로파 출력과 추출 시간 증가에 따라 유의하게 증가하는 경향을 보였다(p<0.05). 조건 최적화에 있어 698.6 W, 240 sec, 0 mol 조건에서 최대 TPC 82.4 mg GAE/g DM과 플라보노이드 18.1 mg QE/g DM이 예측되었다. 기존 추출법인 속실렛과 마이크로파 추출을 비교하였을 때, 마이크로파 추출이 폴리페놀과 플라보노이드 생산에 있어 13.5와 8.0배가 각각 높고 짧은 추출 시간과 낮은 에너지 소비로 기존 추출 대비 효과적인 공정임을 확인하였다. 본 연구는 옥수수 대로부터 유용물질 생산 가능성과 마이크로파 추출법이 상업화 공정 적용이 가능한 효과적인 추출법임을 확인하고 옥수수 대를 이용한 폴리페놀과 플라보노이드 생산을 통해 바이오당 생산과 더불어 부산물 크레딧을 확보하여 바이오 에탄올 가격 경쟁력을 높일 수 있는 추출공정을 제안하였다는 데 의의가 있다고 하겠다. Corn stover is known as a good candidate for a functional food ingredient when the main lignocellulosic material, lignin, is used as bioactive materials as form of polyphenolic compounds. The purpose of this study was to determine the microwave extraction conditions under which total phenolic compounds (TPC) and flavonoid contents of corn stover were maximized. Microwave-assisted extracts using sulfuric acid ranging from 0 to 1.0 mol with extraction time between 40 and 240 sec were conducted by using response surface methodology (RSM). Microwave power showed significant effects (p<0.05) and the concentrations of TPC and flavonoids increased with increased level of microwave power and extraction time. The optimum conditions for corn stover extraction were determined as 698.6 W, 240 sec, and 0 mol sulfuric acid, and the predicted value of TPC and flavonoid is 82.4 mg GAE/g DM and 18.1 mg/g DM, respectively. Microwave extraction was evaluated as an economic process with low energy consumption, short extraction and high extraction yield of bioactive including TPC and flavonoids compared to conventional extractions.

        커피박으로부터 생리활성물질 생산 증대를 위한 열수추출 공정 개발

        커피침출 공정의 부산물인 커피박을 이용한 고부가가치 식품 및 화장품 소재 생산을 위해 열수추출 공정의 폴리페놀 추출인자(용매, 온도, 시간, 농도) 최적화를 수행하였다. 실험에 적용 된 모든 추출인자가 폴리페놀 생산에 유의한 효과가 있음을 확인할 수 있었고 NaOH 농도가 폴리페놀 생산에 미치는 효과가 가장 큰 것으로 평가되었다. 특히, 열수추출 보다는 산 또는 염기를 이용한 열수추출이 폴리페놀 추출 효과가 높았으며 NaOH 0.1 mol 첨가 시 증류수를 이용한 열수추출에 비해 1.5배 추출 효과가 증가하여 열수를 이용한 추출에 비해 염기-열수 추출이 보다 효과적임을 알 수 있었다. 폴리페놀 추출의 최적 조건인 100 ℃, 2 mol NaOH와 30 min 추출 조건에서 36.5 mg GAE/g DM를 얻을 수 있었다. 이는 최적화 이전의 열수추출에서 얻은 12.5 mg GAE/g DM 대비 2.9배 증가된 결과로 커피박은 항산화활성 등 기능성 강화 소재로 식품 및 화장품 산업에 활용이 가능할 것으로 판단된다. In this study, the optimization of extraction parameters (solvent, temperature, time, solvent concentration) for the maximization of polyphenol extraction was performed to produce value-added food and cosmetic additives using a byproduct of coffee extraction process (coffee residue). All of the extraction parameters evaluated in this experiment had significant effects on polyphenol extraction and the results showed the effect of NaOH concentration on the polyphenol production was most significant among tested parameters. Especially, hot water extraction using acid or base was effective rather than hot-water extraction and the addition of 0.1 mol of NaOH increased 1.5 times extraction concentration compared with hot-water extraction using distilled water. It was found that hot-water extraction with NaOH was more effective than hot-water extraction, and 36.5 mg GAE/g DM was obtained under optimum condition of 100 ºC, 2 mol of NaOH and 30 min. This result was 2.9 times higher than that of 12.5 mg GAE/g DM obtained from the hot-water extraction before optimization. Thus, coffee residue could be used for food and cosmetic industry as a high-value additive such as antioxidant.
