        Evaluation and validation of reference genes for SYBR Green qRT-PCR normalization in Sesamia inferens (Lepidoptera: Noctuidae)

        The pink stemborer, Sesamia inferens (Walker) (Lepidoptera: Noctuidae), has gradually become amajor threat to rice production in Asian rice growth area. Using reverse-transcription quantitative real-time PCR (qRT-PCR) to investigate gene expression of S. inferens under different conditions is a popular technique. However, numerous studies demonstrate that the experimental conditions affect the stability of reference genes. Thus, it is necessary to normalize qRT-PCR data under a specific condition using a stable reference gene as an internal control. In the present study, five algorithms (Delta Ctmethod, Bestkeeper, geNorm,Normfinder, and RefFinder) and seven candidate reference genes were evaluated to find out the most suitable reference genes under different biotic and abiotic conditions including different tissues and developmental stages, exposure to insecticides, different rice varieties, and different diets. Our results showed that the more stable series of reference genes were as follows: GAPDH, RPL10, and RPS3 for different tissues;GAPDH and RPS3 for developmental stages; RPS3 and EF1 for larval insecticide exposure; RPS3 and RPL10 for larvae reared on different rice varieties; RPS3 and ACTB for larvae fed on different diets. Hence, a combination of the best sets of reference genes for normalization of expression analysis will increase the accuracy and reliability of qRT-PCR data in S. inferens. In addition, these results establish the basis for selection of reference genes in other lepidopterous insect species.

        Bi@H-TiO2/B-C3N4 heterostructure for enhanced photocatalytic hydrogen generation activity under visible light

        Herein, a ternary Bi@H-TiO2/B-C3N4 type II heterostructure is successfully designed and prepared viametal Bi as cocatalyst for photocatalytic H2 generation. As expected, under k > 300 nm light, the highestphotocatalytic hydrogen production rate (HPR) of Bi@H-TiO2/B-C3N4 is up to 223.08 lmol g1 h1. Excitingly, under visible light (k > 400 nm), the highest HPR of Bi@H-TiO2/B-C3N4 is increased to18.84 lmol g1 h1, which is about 67.3 times, 37.7 times and 6.8 times as high as that of H-TiO2, BC3N4and H-TiO2/B-C3N4 under the same conditions, respectively. Moreover, the electrochemical impedancespectroscopy and transient photocurrent tests manifest that the interface resistance of Bi@HTiO2/B-C3N4 composite decreases and the photocurrent density increases after loading metal Bi. Theseresults indicate that loading metal Bi as cocatalyst (providing more electron trapping sites) is beneficialto improve photocatalytic activity in the type II heterostructure composite. And the electron transferdirections (B-C3N4 ? H-TiO2 ? Bi or B-C3N4 ? Bi) are proved by ultraviolet photoelectron spectroscopytest. Therefore, the Bi@H-TiO2/B-C3N4 photocatalyst with strong visible light absorption is expected to bea promising candidate in photocatalytic fields under visible light.

        Short-term Power Load Forecasting Based on Phase Space Reconstruction and EMD-ELM

        With the increasing complexity of the world energy structure, the uncertainty of the power system increases significantly, and the accuracy of the short-term power load forecasting is of great significance to the safe, economical and reliable operation of the power system. In order to further improve the accuracy of short-term power load forecasting, this paper innovatively combines the theories such as phase space reconstruction, empirical mode decomposition, and extreme learning machine to establish a new short-term power load forecasting model based on phase space reconstruction and EMD-ELM. The prediction results show that the root means square error, average relative error and global maximum relative error of the short-term power load forecasting model based on phase space reconstruction and EMD-ELM are much smaller than other forecasting models, which verifies the correctness and effectiveness of the short-term load forecasting method.

        Experimental Study on Existing RC Circular Members Under Unequal Lateral Impact Train Collision

        With the fast growth of high-speed rail in recent years, derailment has become the first hidden danger of high-speed rail transportation. The high-speed train passes near the station building. So the train may derail and hit the station building. Building a high-speed railway station usually uses a reinforced concrete structure. As a result of high impact energy on the impact body, the reinforced concrete (RC) member may fail; the impact point is near the member's foot; the structural member's constraint can be considered fixed support. This paper investigates the dynamic behavior of four types of circular reinforced concrete members under unequal lateral impact loads. The RC member's failure mechanism and dynamic response addressed the significance of unequal lateral impact load. The usual circular reinforced concrete members are used as the model to perform the drop-weight impact test. The specimens' crack pattern, failure mechanism, impact, deflection, and strain time–history curves are obtained. Findings show that between the impact point and the adjacent support, shear fractures occur that fail in shear mode. Shear cracks are based on impact velocity, longitudinal reinforcement ratio, and stirrup ratio. One type is more destructive to members and nodes. A shear fracture occurs when a longitudinal reinforcement fractures towards the closer support. The effects of impact velocity, longitudinal reinforcement ratio, and stirrup ratio on the dynamic impact response are studied. The experimental results may help improve structural member impact resistance. The critical section (right side) computed the static shear resistance using shear force, whereas the maximum external load resistance determines static bending moment resistance. Understanding how circular members fail to be subjected to unequal lateral impact loads provides insight into circular RC members' impact design and damage evaluation.

        Whole Exome Sequencing in the Accurate Diagnosis of Bilateral Breast Cancer: a Case Study

        When faced with a case of bilateral breast cancer (BBC), understanding how to differentiate bilateral primary breast cancer from contralateral metastatic breast cancer is essential for treatment, but clear identification criteria have not been established to date. Diverse events play different roles in the therapy and prognosis of BBC; hence, it is of great significance to detect a more comprehensive and convincing technique to make an accurate differential diagnosis. We report a rare case of synchronous BBC in a 61-year-old Chinese woman. Based on her clinical and pathological features and the use of whole exome sequencing and cancer genome analysis, we concluded that the patient developed contralateral metastatic breast cancer which metastasized from left to right. Therefore, together with clinical, pathological and cancer genomics information, we could precisely define the origin and evolution of BBC.

        Effect of GZO thickness and annealing temperature on the structural, electrical and optical properties of GZO/Ag/GZO sandwich films

        The GZO/Ag/GZO sandwich films were deposited on glass substrates by RF magnetron sputtering of Gadoped ZnO (GZO) and ion-beam sputtering of Ag at room temperature. The effect of GZO thickness and annealing temperature on the structural, electrical and optical properties of these sandwich films was investigated. The microstructures of the films were studied by X-ray diffraction (XRD). X-ray diffraction measurements indicate that the GZO layers in the sandwich films are polycrystalline with the ZnO hexagonal structure and have a preferred orientation with the c-axis perpendicular to the substrates. For the sandwich film with upper and under GZO thickness of 40 and 30 nm, respectively, it owns the maximum figure of merit of 5.3 × 10-2 Ω-1 with a resistivity of 5.6 × 10-5 Ω cm and an average transmittance of 90.7%. The electrical property of the sandwich films is improved by post annealing in vacuum. Comparing with the as-deposited sandwich film, the film annealed in vacuum has a remarkable 42.8% decrease in resistivity. The sandwich film annealed at the temperature of 350 ℃ in vacuum shows a sheet resistance of 5 Ω/sq and a transmittance of 92.7%, and the figure of merit achieved is 9.3 × 10-2 Ω-1.

        Energy Efficiency Resource Allocation for MIMO Cognitive Radio with Multiple Antenna Spectrum Sensing

        The energy-efficient design of sensing-based spectrum sharing of a multi-input and multi-output (MIMO) cognitive radio (CR) system with imperfect multiple antenna spectrum sensing is investigated in this study. Optimal resource allocation strategies, including sensing time and power allocation schemes, are studied to maximize the energy efficiency (EE) of the secondary base station under the transmit power and interference power constraints. EE problem is formulated as a nonlinear stochastic fractional programming of a nonconvex optimal problem. The EE problem is transformed into its equivalent nonlinear parametric programming and solved by one-dimension search algorithm. To reduce searching complexity, the search range was founded by demonstration. Furthermore, simulation results confirms that an optimal sensing time exists to maximize EE, and shows that EE is affected by the spectrum detection factors and corresponding constraints.

        LncRNA-GAS5/miR-382-3p axis inhibits pulmonary artery remodeling and promotes autophagy in chronic thromboembolic pulmonary hypertension

        Background: We have clarified the role of miR-382-3p in chronic thromboembolic pulmonary hypertension (CTEPH), but what is less clear lies in its upstream regulatory mechanism. Objective: To explore the regulation mechanism of GAS5/miR-382-3p axis on CTEPH. Methods: In vitro, we constructed cell models by treating Pulmonary Artery Smooth Muscle Cells (PASMCs) with platelet-derived growth factor-BB (PDGF-BB). The effects of different concentrations of PDGF-BB on the activity of PASMCs were tested by cell counting kit-8 (CCK-8). The upstream lncRNA of miR-382-3p was screened and confirmed through bioinformatics analysis, RNA pull-down, quantitative reverse transcription polymerase chain reaction (qRT-PCR), dual luciferase reporter gene and RNA immunoprecipitation assays. The effects of GAS5/miR-382-3p axis on the viability, migration, and expressions of autophagy- and angiogenesis-related proteins were confirmed by rescue experiments (CCK-8, wound healing and western blot). In vivo, animal models by perfusing autologous blood vessels, the effects of GAS5 overexpression or silencing on the expressions of miR-382-3p, angiogenesis- and autophagy-related genes, mean pulmonary arterial pressure (mPAP) and pulmonary artery wall were determined by biological signal acquisition system, hematoxylin-eosin staining, qRT-PCR and western blot. Results: PDGF-BB dose-dependently promoted PASMCs viability. XIST and GAS5 expressions in PASMCs were affected by the concentration of PDGF-BB, but only GAS5 can be pulled down by miR-382-3p probe. GAS5 targeted miR-382-3p to inhibit the viability and migration of PAMSCs, mPAP in CTEPH rats, pulmonary artery wall thickening and angiogenesis, and promote autophagy. Conclusions: GAS5/miR-382-3p axis is involved in the regulation of pulmonary artery remodeling and autophagy in CTEPH.

        Analysis of the effect of Rhodopseudomonas palustris on the lead exposure rat model using 1H-NMR-based metabolomics of urine and plasma

        Background Rhodopseudomonas palustris had the eff ect of adsorbing and removing lead ions. Metabolomics is a comprehensive analysis based on several metabolites or biomarkers. Objective The paper aimed to investigate the restorative eff ect and the potential mechanisms of R. palustris in rats with the lead exposure model using 1 H-NMR metabolomics. Results Nine endogenous metabolites from rat plasma and 13 endogenous metabolites from rat urine were identifi ed as potential biomarkers of the symptoms of lead poisoning. The changes of metabolites levels in the plasma included lower levels of valine, acetoacetate, and N-acetyl-glycoprotein and a higher concentration of lactate, acetate, creatine, threonine, glutamine, and asparagine in model group compared with control group. Furthermore, increased citrate, succinate, dimethylamine, pyruvate, acetoacetate, α -oxoglutarate and asparagine, accompanied by decreased LDL/VLDL, taurine, creatinine, glucose and hippurate levels were observed in the model group compared with the control group in the urine. Biochemical parameters and histopathology examination dovetailed well with the metabolomics data. Treatment with R. palustris at a certain degree returned the levels of these metabolites to normal levels. Plasma and urine metabolomic analysis showed changes associated with energy metabolism, amino acids and fatty acid metabolism of lead exposure rats. Conclusion The eff ects of removing lead ions of R. palustris in rats with lead exposure were confi rmed. This research demonstrated that 1 H-NMR-based metabolomics was a promising tool to identify potential biomarkers and unravel protective eff ect mechanisms of R. palustris in lead poisoning rats.
