        HIV-negative plasmablastic lymphoma: report of 8 cases and a comprehensive review of 394 published cases

        BackgroundHuman immunodeficiency virus (HIV)-negative plasmablastic lymphoma (PBL) is a rare entity of diffuse large B-cell lymphoma (DLBCL). The clinicopathological features of and optimal treatment for HIV-negative PBL remain largely unknown.MethodsTo gain insight into this distinct lymphoma, we summarized the clinicopathologic charac-teristics of 8 unpublished HIV-negative PBLs and performed a comprehensive review of 394 published cases.ResultsOf the 8 unpublished PBLs, the median patient age was 53.0 years. Four patients pre-sented with stage IV disease. All 8 patients showed a plasma cell-like immunophenotype. Of the six patients who received anthracycline-based chemotherapy, including two who received bortezomib, three patients achieved a continuous complete response, two pa-tients died due to disease progression, and one patient was lost to follow-up. The other two patients achieved continuous complete response after receiving chemotherapy com-bined with radiotherapy and surgery. Of the 402 patients, the majority were male, with a mean age of 58.0 years. EBV infection was detected in 55.7% of the patients. The median survival times of the patients who received CHOP or CHOP-like regimens and intensive regimens were not reached and 23.0 months, respectively, and the intensive regimen did not improve the survival outcome (P=0.981). Multivariate analysis showed that EBER remained the only independent factor affecting overall survival (OS).ConclusionHIV-negative PBL is a distinct entity with a predilection for elderly and immunosup-pressed individuals. Intensive chemotherapy had no apparent survival benefits over the CHOP regimen in terms of OS; the prognosis of this disease is poor with current chemo-therapy methods, and treatment remains a challenge.

        Molecular identification and expression patterns of carboxylesterase genes based on transcriptome analysis of the common cutworm, Spodoptera litura (Lepidoptera: Noctuidae)

        Carboxylesterases (CXEs) belong to a family of metabolic enzymes that are widely distributed in insects and other organisms and can rapidly degrade the components of sex pheromones and plant volatiles with an acetate functional group. The common cutworm, Spodoptera litura, is an important agricultural pest around the world, causing vast economic losses every year. The female sex pheromones of S. litura comprise four acetates, Z9, E11-14:OAc; Z9, E12-14:OAc; Z9-14:OAc; and E11-14:OAc, but the degradation mechanisms of these components are not well understood. By analysing previously obtained transcriptomic data of the sex pheromone glands,we identified a total of 24 putative CXE genes in S. litura. Gene expression patterns and phylogenetic analysis revealed 5 genes with antennae-specific or biased expression, and clusteredwith genes showed involvement in the degradation of sex pheromones or other detoxification in other insects. SlitCXE10was expressed specifically in the antennae of both sexes, and SlitCXE14, 17, 19, and 21 had high antenna biased expression. Interestingly, RT-PCR and qPCR tests indicated that SlitCXE24 had significantly higher expression in PG than in other tissue, and that it could be a potential candidate gene for sex pheromone degradation in PG. This study is the first to provide solid background information for the further elucidation of sex pheromone degradation, and ultimately provides potential targets for the disruption of sexual communication in S. litura for new pest management.

        Anatomical Study of the Accessory Tendon of the Extensor Hallucis Longus Muscle and Its Clinical Application

        Background: The accessory tendon of the extensor hallucis longus (ATEHL) muscle is a common abnormal structure, and its clinical significance remains debatable. In this study, we provide the incidence of the ATEHL and characterize its morphological types in Asian cadavers and investigate its clinical applications. Methods: The tendons from 50 adult cadaveric feet, fixed in 10% formalin, were analyzed. We measured the length and width of both the ATEHL and the extensor hallucis brevis (EHB). Results: All dissected specimens had an ATEHL. The first metatarsophalangeal joint was surrounded by an accessory tendon that inserted onto the joint capsule and the dorsal base of the proximal phalanx. We classified the ATEHL into 3 types based on their directions. Differences in ATEHL type based on sex were not statistically significant. Conclusions: We found an ATEHL in all cadaveric specimens in this study. We surmise that the ATEHL acts as an antagonist with the EHB when the toe is extending, which might help prevent the occurrence of hallux valgus deformity.

      • Gene Silencing of β-catenin by RNAi Inhibits Proliferation of Human Esophageal Cancer Cells by Inducing G0/G1 Cell Cycle Arrest

        Objectives: The aim of the present study was to explore mechanisms underlying the effects of down-regulating ${\beta}$-catenin expression on esophageal carcinoma (EC) cells. Methods: Cell cycle distribution and apoptosis were determined using flow cytometry and annexin V apoptosis assay, respectively. Transmission electron microscopy (TEM) was used to examine changes in ultrastructure, while expression of cyclin D1 protein and mRNA was detected by western blot and real-time PCR. Proliferating cell nuclear antigen (PCNA) and extracellular signal-regulated kinase (ERK) 1-2 were evaluated by Western blot analysis. PCNA labeling index (LI) was determined by immunocytochemistry. Results: Compared with pGen-3-con transfected and Eca-109 cells, the percentage of G0/G1-phase pGen-3-CTNNB1 transfected cells was obviously increased (P<0.05), with no significant difference among the three groups with regard to apoptosis (P>0.05). pGen-3-CTNNB1 transfected cells exhibited obvious decrease in cyclin D1 mRNA and protein expression (P<0.05) and the ultrastructure of Eca-109 cells underwent a significant change after being transfected with pGen-3-CTNNB1, suggesting that down-regulating ${\beta}$-catenin expression can promote the differentiation and maturation. The expression of PCNA and the ERKI/2 phosphorylation state were also down-regulated in pGen-3-CTNNB1 transfected cells (P<0.05). At the same time, the PCNA labeling index was decreased accordingly (P<0.05). Conclusion: Inhibition of EC Eca-109 cellproliferation by down-regulating ${\beta}$-catenin expression could improve cell ultrastructure by mediating blockade in G0/G1 through inhibiting cyclin D1, PCNA and the MAPK pathway (p-ERK1/2).

        Value of contrast-enhanced ultrasonography in microwave ablation treatment of symptomatic focal uterine adenomyosis

        Purpose: This study evaluated the value of contrast-enhanced ultrasonography (CEUS) in the ultrasound-guided microwave ablation (MWA) treatment of symptomatic focal uterine adenomyosis.Methods: This retrospective study was conducted between March 2020 and January 2023, enrolling 52 patients with symptomatic focal uterine adenomyosis who had undergone MWA. All patients were examined with CEUS before and after MWA. The non-perfused volume (NPV) was compared between CEUS and dynamic contrast-enhanced magnetic resonance imaging (DCEMRI) following ablation. Therapeutic efficacy and safety were evaluated at 3-, 6-, and 12-month follow-ups. Additionally, this study explored the correlations between pre-treatment CEUS features and a volume reduction ratio indicating sufficient ablation, defined as 50% or more at the 3-month follow-up.Results: No significant differences in NPV were noted between CEUS and DCE-MRI immediately after MWA and during follow-up (all P>0.05). At the 3-month follow-up, the median VRRs for the uterus and adenomyosis were 33.2% and 63.9%, respectively. Sufficient ablation was achieved in 69.2% (36/52) of adenomyosis cases, while partial ablation was observed in the remaining 30.8% (16/52). The identification of non-enhancing areas on pre-treatment CEUS was associated with sufficient ablation (P=0.016). At the 12-month follow-up, significant decreases were observed in both the uterine and adenomyosis volumes (all P<0.001). Dysmenorrhea and menorrhagia were significantly alleviated at 12 months, and no major complications were encountered.Conclusion: CEUS can be used to evaluate the ablation zone of focal adenomyosis that has been treated with MWA, similarly to DCE-MRI. The identification of non-enhancing areas on pretreatment CEUS indicates satisfactory treatment outcomes.         Molecular identification and sex distribution of two chemosensory receptor families in Athetis lepigone by antennal transcriptome analysis

        Chemosensory receptors play central roles in insect behavior and can potentially be used as new targets for pest control. Athetis lepigoneMöschler (Lepidoptera: Noctuidae) is a polyphagous insect pest found in many countries in Europe and Asia. However, systemic identification of the chemosensory receptors in the antennae has not been reported. In the present study, we obtained the antennal transcriptome of A. lepigone using Illumina sequencing technology and identified 80 candidate chemosensory receptors, including 61 transcripts encoding for odorant receptors (ORs) and 19 for ionotropic receptors (IRs). This result is similar to that reported for other lepidopteran species, suggesting that our approach successfully identified ORs and IRs of A. lepigone. RT-PCR was used to determine the sex distribution patterns of all OR and IR genes. Some OR and IR genes showed male- or femalespecific/ predominant expression. Our results help understand the functions of chemosensory receptor genes in A. lepigone as well as other insects, and suggest that these genes could be potential targets for developing environment-friendly behavioral antagonists and pesticides against A. lepigone.

        Evidence for the Presence of Long-Lived Plasma Cells in Nasal Polyps

        Purpose: Plasma cells and immunoglobulins (Igs) play a pivotal role in the induction and maintenance of chronic inflammation in nasal polyps. During secondary immune responses, plasma cell survival and Ig production are regulated by the local environment. The purpose of the present study was to investigate the presence of long-lived plasma cells (LLPCs) and specific survival niches for LLPCs in human nasal polyps. Methods: Nasal mucosal samples were cultured with an air-liquid interface system and the Ig levels in culture supernatants were analyzed by enzyme-linked immunosorbent assay. The characteristics of LLPCs in nasal polyps were determined by immunohistochemistry and immunofluorescence. The expression of neurotrophins as well as their receptors was detected by quantitative real-time polymerase chain reaction, immunohistochemistry, immunofluorescence, and Western blotting. Results: The numbers of CD138+ total plasma cells and BCL2+ plasma cells were increased in both eosinophilic and non-eosinophilic nasal polyps compared with those in normal tissues. The production of IgG, IgA, and IgE was detected in culture supernatants even after a 32-day culture of nasal polyps. Although the total numbers of plasma cells were decreased in nasal polyps after culture, the numbers of BCL2+ plasma cells remained stable. The expression of nerve growth factor (NGF) as well as tropomyosin receptor kinase (Trk) A, a high-affinity receptor for NGF, was upregulated in both eosinophilic and non-eosinophilic nasal polyps. In addition, BCL2+ plasma cell numbers were positively correlated with NGF and TrkA mRNA expression in nasal mucosal tissues. Polyp plasma cells had the expression of TrkA. Conclusions: Human nasal polyps harbor a population of LLPCs and NGF may be involved in their prolonged survival. LLPCs may be a novel therapeutic target for suppressing the local Ig production in nasal polyps.

        Efficient Analysis of the Deformation and Fracture Properties of Nano-SiO2 Reinforced ABS/PMMA Composites

        This study investigated the damage mechanism of acrylonitrile-butadiene styrene (ABS)/poly(methyl methacrylate) (PMMA)/nano-SiO2 composites. The distortion and fracture behavior of ABS/PMMA/nano-SiO2 compositeswere predicted by developing the homogenization theory and the surface-based cohesive method. This model simulatedthe elementary action of elastoplastic damage in the matrix, the fracture for the nano-SiO2 reinforcement, and traction separation for interfaces. The experiment and morphology of the fracture section were validated by the modeling results. Thedamage mechanisms of ABS/PMMA/nano-SiO2 composites were researched with the numerical results, and the roles ofnano-SiO2 strength in the mechanical properties of ABS/PMMA blends were discussed in detail

        Parkinson’s Disease with Fatigue: Clinical Characteristics and Potential Mechanisms Relevant to α-Synuclein Oligomer

        Background and Purpose The aim of this study was to identify the clinical characteristics and potential mechanisms relevant to pathological proteins in Parkinson’s disease (PD) patients who experience fatigue. Methods PD patients (n=102) were evaluated using a fatigue severity scale and scales for motor and nonmotor symptoms. The levels of three pathological proteins—α-synuclein oligomer, β-amyloid (Aβ)1-42, and tau—were measured in 102 cerebrospinal fluid (CSF) samples from these PD patients. Linear regression analyses were performed between fatigue score and the CSF levels of the above-listed pathological proteins in PD patients. Results The frequency of fatigue in the PD patients was 62.75%. The fatigue group had worse motor symptoms and anxiety, depression, and autonomic dysfunction. The CSF level of α-synuclein oligomer was higher and that of Aβ1-42 was lower in the fatigue group than in the non-fatigue group. In multiple linear regression analyses, fatigue severity was significantly and positively correlated with the α-synuclein oligomer level in the CSF of PD patients, after adjusting for confounders. Conclusions PD patients experience a high frequency of fatigue. PD patients with fatigue have worse motor and part nonmotor symptoms. Fatigue in PD patients is associated with an increased α-synuclein oligomer level in the CSF

        Two new tirucallane triterpenoids from the leaves of Aquilaria sinensis

        Two new tirucallane triterpenoids, aquilacallanesA–B (1–2), together with 15 known compounds(3–17) were isolated from the leaves of Aquilaria sinensis. The structures of these new compounds were elucidated onthe basis of extensive spectroscopic analyses. All compoundswere evaluated for their cytotoxic activity againstfive human cancer cell lines. The known compounds,ursolic acid (7) and 5,7,40-trimethoxyflavone (14), exhibitedweak cytotoxic activity against some cells.
