        Ginsenoside Rb1 exerts neuroprotective effects through regulation of Lactobacillus helveticus abundance and GABA<sub>A</sub> receptor expression

        Background: Ginsenoside Rb1 (Rb1), one of the most abundant protopanaxadiol-type ginsenosides, exerts excellent neuroprotective effects even though it has low intracephalic exposure. Purpose: The present study aimed to elucidate the apparent contradiction between the pharmacokinetics and pharmacodynamics of Rb1 by studying the mechanisms underlying neuroprotective effects of Rb1 based on regulation of microflora. Methods: A pseudo germ-free (PGF) rat model was established, and neuroprotective effects of Rb1 were compared between conventional and PGF rats. The relative abundances of common probiotics were quantified to reveal the authentic probiotics that dominate in the neuroprotection of Rb1. The expressions of the gamma-aminobutyric acid (GABA) receptors, including GABAA receptors (α2, β2, and γ2) and GABAB receptors (1b and 2), in the normal, ischemia/reperfusion (I/R), and I/R+Rb1 rat hippocampus and striatum were assessed to reveal the neuroprotective mechanism of Rb1. Results: The results showed that microbiota plays a key role in neuroprotection of Rb1. The relative abundance of Lactobacillus helveticus (Lac.H) increased 15.26 fold after pretreatment with Rb1. I/R surgery induced effects on infarct size, neurological deficit score, and proinflammatory cytokines (IL-1β, IL-6, and TNF-α) were prevented by colonizing the rat gastrointestinal tract with Lac.H (1 × 10<sup>9</sup> CFU) by gavage 15 d before I/R surgery. Both Rb1 and Lac.H upregulated expression of GABA receptors in I/R rats. Coadministration of a GABA<sub>A</sub> receptor antagonist significantly attenuated neuroprotective effects of Rb1 and Lac.H. Conclusion: In sum, Rb1 exerts neuroprotective effects by regulating Lac.H and GABA receptors rather than through direct distribution to the target sites.

        System Reliability Analysis of Rock Slope Stability Involving Correlated Failure Modes

        This paper aims to propose a systematic quantitative method for system reliability evaluation of rock slope with plane failure involving multiple correlated failure modes. A probabilistic fault tree approach is presented to model system reliability of rock slope. An n-dimensional equivalent reliability method is employed to perform the system reliability analysis of the slope involving multiple correlated failure modes. Reliability sensitivity analyses at three different levels, namely, the single limit state function level, single failure mode level, and system reliability level, are carried out to study the effect of variables on reliability. An example is presented to demonstrate the validity and capability of the proposed approach. The results indicate that the system reliability of rock slope involving multiple correlated failure modes can be evaluated efficiently using the proposed approach. The system probability of failure is overestimated if the correlations between different failure modes are ignored. The relative importance of different failure modes to the system reliability can differ considerably. The sensitivity coefficients of basic random variables strongly depend on the selected sensitivity analysis level. The system reliability is sensitive to the location of the tension crack and the percentage of the tension crack filled with water.

        This research takes Chinese overseas students of W university in Jeollabuk-do in 2018 as subjects, and attempts to understand the influence of acculturation stress on health behaviors. Firstly, the average acculturation stress of Chinese overseas students is 1.88±.40, and there is no significant stress. For general characteristics, differences (p<.01) occurred in TOPIK, underlying factor “designated factor” (p<.01)“fear”, “perceived hostility”, benefitted from scholarship in“others” is “cultural shock”(p<.05), differences of “designated hostility”(p<.001), “cultural shock”, “sense of guilt” (p<.0-05) under healthy condition. Secondly, the average of Chinese overseas students’health-enhancing behaviors is 3.6±.5 (5 as full scores), which is a high score in general. For general characteristics, based on“health risk”(p<.05), a series of differences“ physical activities”(p<.01),“nutrition”(pm.001), a significant difference occurred in“pressure management”from (p<.05). Difference occurred in “physical activity” (p<.05) of living status, and there is a significant difference in“physical activity”(p<.01) and “stress management”(p<.001) in health condition. Based on above research findings, all Chinese overseas students live in Jeolla buk-do should improve health responsibility awareness in terms of health-enhancing behaviors, and special health management is needed for physical activity and cultural adaptation conditions.

        First Principles Study of Structural and Electronic Properties of Pentagonal and Hexagonal Noble Metal Nanowires

        The equilibrium structure and electronic properties of four ultrathin free-standing pentagonal and hexagonal noble metal nanowires, that is, copper nanowires (CuNWs), silver nanowires (AgNWs), gold nanowires (AuNWs) and platinum nanowires (PtNWs), have been studied comprehensively by adopting a first-principles simulation based on the density-functional theory. The staggered topologies are more stable than the eclipsed ones by analyzing the bonding energy. The staggered ones with a linear atom chain in the center of the pentagonal or hexagons topologies are the preferred structures for CuNWs and AgNWs, but the staggered ones without a linear atom chain in the center of the pentagon or hexagon are the preferred structures for AuNWs and PtNWs due to the increasing core–core repulsions. The calculated electronic band structures and density of states present that all the noble metal nanowires are metallic. The projected densities of states (PDOS) of dominant d-states and the charge density show that the narrower d-state moved to the Fermi energy and metallic bonding character for all the noble metal nanowires.

        Efficient Analysis of the Deformation and Fracture Properties of Nano-SiO2 Reinforced ABS/PMMA Composites

        This study investigated the damage mechanism of acrylonitrile-butadiene styrene (ABS)/poly(methyl methacrylate) (PMMA)/nano-SiO2 composites. The distortion and fracture behavior of ABS/PMMA/nano-SiO2 compositeswere predicted by developing the homogenization theory and the surface-based cohesive method. This model simulatedthe elementary action of elastoplastic damage in the matrix, the fracture for the nano-SiO2 reinforcement, and traction separation for interfaces. The experiment and morphology of the fracture section were validated by the modeling results. Thedamage mechanisms of ABS/PMMA/nano-SiO2 composites were researched with the numerical results, and the roles ofnano-SiO2 strength in the mechanical properties of ABS/PMMA blends were discussed in detail

      • Expression and Prognostic Significance of lncRNA MALAT1 in Pancreatic Cancer Tissues

        Background: Long non-coding RNAs (lncRNAs) have been recently observed in various human cancers. However, the role of lncRNAs in pancreatic duct adenocarcinoma (PDAC) remains unclarified. The aim of this study was to detect the expression of lncRNA MALAT1 in PDAC formalin-fixed, paraffin embedded (FFPE) tissues and to investigate the clinical significance of the MALAT1 level. Methods: The expression of MALAT1 was examined in 45 PDAC and 25 adjacent non-cancerous FFPE tissues, as well as in five PDAC cell lines and a normal pancreatic epithelium cell line HPDE6c-7, using qRT-PCR. The relationship between MALAT1 level and clinicopathological parameters of PDAC was analyzed with the Kaplan-Meier method and Cox proportional hazards model. Results: The relative level of MALAT1 was significantly higher in PDAC compared to the adjacent normal pancreatic tissues (p=0.009). When comparing the MALAT1 level in the cultured cell lines, remarkably higher expression of MALAT1 was found in aspc-1 PDAC cells compared with the immortal pancreatic duct epithelial cell line HPDE6c-7 (q=7.573, p<0.05). Furthermore, MALAT1 expression level showed significant correlation with tumor size (r=0.35, p=0.018), tumor stage (r=0.439, p=0.003) and depth of invasion (r=0.334, p=0.025). Kaplan-Meier analysis revealed that patients with higher MALAT1 expression had a poorer disease free survival (p=0.043). Additionally, multivariate analysis indicated that overexpression of MALAT1, as well as the tumor location and nerve invasion, was an independent predictor of disease-specific survival of PDAC. Conclusion: MALAT1 might be considered as a potential prognostic indicator and may be a target for diagnosis and gene therapy for PDAC.

        A Case of Simultaneously Diagnosed Lung Adenocarcinoma and Endobronchial Inflammatory Myofibroblastic Tumor with Two Distinct Types of ALK Translocation

        A 61-year-old male patient was simultaneously diagnosed with lung adenocarcinoma and inflammatory myofibroblastic tumor (IMT). The lung adenocarcinoma and IMT harbored two distinct types of ALK translocation, LOC101927285-ALK, and TPM3-ALK, respectively. The ALK Ventana showed strong positivity on both lesions. The patient was therefore given an endobronchial cryotherapy and ALK inhibitor crizotinib. The tumors showed durable response however the left lung adenocarcinoma relapsed at 17th month post-crizotinib treatment. Tissue re-biopsy on the resistant tumor revealed an ALK exon 23 C1156Y missense mutation in addition to LOC101927285-ALK mutation. Further RNA-based sequence uncovered that the noncoding region rearrangement is the fusion mutation of EML4-ALK. The patient was therefore received alectinib, and the tumor exhibited partly response. Overall, it is very rare that two types of pulmonary tumors exist in one patient driven by two distinct ALK fusions, which emphasizes the necessity of gene sequencing in clinical decision-making and individualized therapy.
