        Selection of High Laccase-Producing Coriolopsis gallica Strain T906: Mutation Breeding, Strain Characterization, and Features of the Extracellular Laccases

        Commercial application of laccase is often hampered by insufficient enzyme stocks, with very low yields obtained from natural sources. This study aimed to improve laccase production by mutation of a Coriolopsis gallica strain and to determine the biological properties of the mutant. The high-yield laccase strain C. gallica TCK was treated with N-methyl-N-nitro Nnitrosoguanidine and ultraviolet light. Among the mutants isolated, T906 was found to be a high-production strain of laccases. The mutant strain T906 was stabilized via dozens of passages, and the selected ones were further processed for optimization of metallic ion, inducers, and nutritional requirements, which resulted in the optimized liquid fermentation medium MF9. The incubation temperature and pH were optimized to be 30°C and 4.5, respectively. The mutant strain T906 showed 3-times higher laccase activity than the original strain TCK under optimized conditions, and the maximum laccase production (303 U/ml) was accomplished after 13 days. The extracellular laccase isoenzyme 1 was purified and characterized from the two strains, respectively, and their cDNA sequence was determined. Of note, the laccase isoenzyme 1 transcription levels were overtly increased in T906 mycelia compared with values obtained for strain TCK. These findings provide a basis for C. gallica modification for the production of high laccase amounts.

        Identification of Volatile Components in Phyllanthus emblica L. and Their Antimicrobial Activity

        The volatile components and in vitro antimicrobial activities of Emblica (Phyllanthus emblica L.) essential oils (EOs) obtained by hydrodistillation (HD-EO) and supercritical fluid extraction (SFE-EO) were investigated. The compositions of volatile compounds in these oils were tentatively determined by gas chromatography-mass spectrometry. The antimicrobial activites of these two extracts were investigated with microbiological tests against Gram-positive and Gram-negative bacteria and three pathogenic fungi. The main components of both oils were β-caryophyllene, β-bourbonene, 1-octen-3-ol, thymol, and methyleugenol. Both essential oils showed a broad spectrum of antimicrobial activity against all the tested microorganisms. Gram-positive bacteria were more sensitive to the investigated oils than Gram-negative bacteria. SFE-EO exhibited a higher antifungal activity compared to HD-EO.

        A Comparative Study on the Offshore Intensification of Supertyphoon Rammasun (2014) and Typhoon Rumbia (2013): the Role of Summer Monsoon

        The offshore intensificationmechanisms of Supertyphoon Rammasun (2014) and Typhoon Rumbia (2013) are analyzed and compared based on the best track typhoon data provided by the Shanghai Typhoon Institute of China Meteorological Administration (STI/CMA), ECMWF (CMA and JMA) objective (subjective) predicted typhoon track and intensity, and reanalysis data of the National Centers for Environmental Prediction/National Center for Atmospheric Research (NCEP/NCAR). Results show that the offshore sea surface temperature over 28 °C was conducive to the intensification of the two typhoons in the coastal area prior to their landfall in South China. Rammasun was embedded in an extremely active phase of a 10–20-day oscillation in the summer monsoon over the South China Sea and western North Pacific prior to its landfall. The low-level jet transported warm and moist air to the inner core of Rammasun and the deepening of the southwesterly monsoon flow up to 200 hPa reduced vertical wind shear, both favorable for the offshore rapid intensification of Rammasun. In 2013, the summer monsoon over the South China Sea and western North Pacific experienced quasi-20-day and quasi-10-day oscillations. The out of phase of the two oscillations resulted in relatively weak tropical and subtropical summer monsoon flow prior to the landfall of Rumbia. As a result, the low-level monsoon flow transported relatively less moist air into Rumbia, and the shallow monsoon flow was not conducive to the weakening of vertical wind shear, resulting in a less rapid offshore intensification of Rumbia than Rammasun.

        Laccase-Mediated In Situ Oxidation of Dopamine for Dyeing of Human Hair

        Due to the damage of scalp because of chemical dyes, increasing attention had been paid to the ecological dyeingof human hair. To address the need for ecological restoration and dyeing of human hair, herein, a novel approach has beenproposed via laccase-mediated in situ oxidation of dihydroxy phenylalanine (dopamine). As laccase can catalyze dopamineoxidation and then polymerization to produce strongly colored polymers, different biological coloration processes of humanhair based on this process were investigated: (i) Human hair was treated with laccase alone; (ii) The absorption of human hairwith dopamine, followed by further catalysis of air; (iii) The absorption of human hair with dopamine, followed by furthercatalysis of laccase; (iv) Simultaneous laccase-mediated polymerization and dyeing of human hair; and (v) Polymerization ofDopamine with laccase and then dyeing human hair. The surface properties of dyed hair were evaluated in terms of ultraviolet(UV)-protection, color fastness, Fourier transforming infrared spectrum (FT-IR), optical microscope, and scanning electronmicroscopy (SEM). The results show that best hair dyeing effect was achieved via simultaneous laccase-mediatedpolymerization and dyeing. The dyed hair obtained K/S values of 28, light fastness of level 5, and UPF of 100+. The opticalmicroscope and SEM observations show that the surface of colored hair fibers was covered with thin burrs, which can beattributed to polydopamine attached with physical adhesive force and covalent reaction. FT-IR analysis shows that functionalgroups were present on the surface of the dyed hair which can be considered as introduction of hydroxyl radical groups. Thisbiological stain technique may be widely used in dyeing of hair with black color.

        ssc-miR-185 targets cell division cycle 42 and promotes the proliferation of intestinal porcine epithelial cell

        Objective: microRNAs (miRNAs) can play a role in a variety of physiological and pathological processes, and their role is achieved by regulating the expression of target genes. Our previous high-throughput sequencing found that ssc-miR-185 plays an important regulatory role in piglet diarrhea, but its specific target genes and functions in intestinal porcine epithelial cell (IPEC-J2) are still unclear. We intended to verify the target relationship between porcine miR-185 and cell division cycle 42 (CDC42) gene in IPEC-J2 and to explore the effect of miR-185 on the proliferation of IPEC-J2 cells. Methods: The TargetScan, miRDB, and miRanda software were used to predict the target genes of porcine miR-185, and CDC42 was selected as a candidate target gene. The CDC42-3' UTR-wild type (WT) and CDC42-3'UTR-mutant type (MUT) segments were successfully cloned into pmirGLO luciferase vector, and the luciferase activity was detected after co-transfection with miR-185 mimics and pmirGLO-CDC42-3'UTR. The expression level of CDC42 was analyzed using quantitative polymerase chain reaction and Western blot. The proliferation of IPEC-J2 was detected using cell counting kit-8 (CCK-8), methylthiazolyldiphenyl-tetrazolium bromide (MTT), and 5-ethynyl-2'-deoxyuridine (EdU) assays. Results: Double enzyme digestion and sequencing confirmed that CDC42-3'UTR-WT and CDC42-3'UTR-MUT were successfully cloned into pmirGLO luciferase reporter vector, and the luciferase activity was significantly reduced after co-transfection with miR-185 mimics and CDC42-3'UTR-WT. Further we found that the mRNA and protein expression level of CDC42 were down-regulated after transfection with miR-185 mimics, while the opposite trend was observed after transfection with miR-185 inhibitor (p<0.01). In addition, the CCK-8, MTT, and EdU results demonstrated that miR-185 promotes IPEC-J2 cells proliferation by targeting CDC42. Conclusion: These findings indicate that porcine miR-185 can directly target CDC42 and promote the proliferation of IPEC-J2 cells. However, the detailed regulatory mechanism of miR-185/CDC42 axis in piglets' resistance to diarrhea is yet to be elucidated in further investigation.

        Effect of sintering temperature on properties of lightweightporous ceramics prepared by foam impregnation method

        In this paper, cheap mineral materials were used as the base materials of lightweight porous ceramics prepared through foamimpregnation method. The effect of the sintering temperature on the properties of the prepared porous ceramics was studied. The porous ceramic was mainly composed of amorphous silicon oxide, crystalline cordierite and mullite phases, and a smallamount of alumina phase. As the sintering temperature increased, the porosity of porous ceramics gradually decreased from94% to 92%, and the bulk density increased from 0.173 gcm-3 at 1100 ℃ to 0.194 gcm-3 at 1200 ℃. The best sinteringtemperature was 1180 ℃. The porosity of the porous ceramics sintered at 1180 ℃ was 92.14%, the volume weight was 0.189gcm-3, the shrinkage rate was 15.80%, the compressive strength was 0.79 MPa, and the thermal conductivity was 0.295 Wm-1k-1. The lightweight porous ceramic has high porosity, low density and good thermal insulation, as well as low cost, having greatpotential for application in fields such as thermal insulation, adsorption, and environmental protection.

        MiR-214 inhibits apoptosis in thyroid epithelial follicular cells induced by amiodarone through the FASL/MAPK pathway

        Background Hashimoto's thyroiditis (HT), also known as chronic lymphocytic thyroiditis, is one of the most common autoimmune disease (AITD) in clinical practice. It is urgent to explore the mechanism of amiodarone-induced thyroid dysfunction. Objective This study aims to assess the expression levels of miR-214 and FasL in amiodarone contact type of HT, and the effect of miR-214 on cell viability and apoptosis and potential mechanism. Results We found that miR-214 was low expressed in the tissues of amiodarone-treated thyroiditis patients. MiR-214 increased the survival rate of amiodarone-induced thyroid epithelial follicular cells and inhibited apoptosis. Mechanically, we found that miR-214 could bind to FASL and regulate MAPK signaling pathway through FASL. Conclusions Our results suggested that miR-214 could be a potential therapeutic target for Hashimoto's thyroiditis.

        IRE1α protects against osteoarthritis by regulating progranulin-dependent XBP1 splicing and collagen homeostasis

        Osteoarthritis (OA) is a full-joint, multifactorial, degenerative and inflammatory disease that seriously affects the quality of life of patients due to its disabling and pain-causing properties. ER stress has been reported to be closely related to the progression of OA. The inositol-requiring enzyme 1α/X-box-binding protein-1 spliced (IRE1α/XBP1s) pathway, which is highly expressed in the chondrocytes of OA patients, promotes the degradation and refolding of abnormal proteins during ER stress and maintains the stability of the ER environment of chondrocytes, but its function and the underlying mechanisms of how it contributes to the progression of OA remain unclear. This study investigates the role of IRE1α/ERN1 in OA. Specific deficiency of ERN1 in chondrocytes spontaneously resulted in OA-like cartilage destruction and accelerated OA progression in a surgically induced arthritis model. Local delivery of AdERN1 relieved degradation of the cartilage matrix and prevented OA development in an ACLT-mediated model. Mechanistically, progranulin (PGRN), an intracellular chaperone, binds to IRE1α, promoting its phosphorylation and splicing of XBP1u to generate XBP1s. XBP1s protects articular cartilage through TNF-α/ERK1/2 signaling and further maintains collagen homeostasis by regulating type II collagen expression. The chondroprotective effect of IRE1α/ERN1 is dependent on PGRN and XBP1s splicing. ERN1 deficiency accelerated cartilage degeneration in OA by reducing PGRN expression and XBP1s splicing, subsequently decreasing collagen II expression and triggering collagen structural abnormalities and an imbalance in collagen homeostasis. This study provides new insights into OA pathogenesis and the UPR and suggests that IRE1α/ERN1 may serve as a potential target for the treatment of joint degenerative diseases, including OA.
