      • Neuroprotective Effect of Human Adipose Stem Cell-Derived Extract in Amyotrophic Lateral Sclerosis

        <P>Amyotrophic lateral sclerosis (ALS) is a devastating human neurodegenerative disease. The precise pathogenic mechanisms of the disease remain uncertain, and as of yet, there is no effective cure. Human adipose stem cells (hASC) can be easily obtained during operative procedures. hASC have a clinically feasible potential to treat neurodegenerative disorders, since cytosolic extract of hASC contain a number of essential neurotrophic factors. In this study, we investigated effects of hASC extract on the SOD1 G93A mouse model of ALS and in vitro test. Administration of hASC extract improved motor function and prolonged the time until symptom onset, rotarod failure, and death in ALS mice. In the hASC extracts group, choline acetyltransferase immunostaining in the ventral horn of the lumbar spinal cord showed a large number of motor neurons, suggesting normal morphology. The neuroprotective effect of hASC extract in ALS mice was also suggested by western blot analysis of spinal cord extract from ALS mice and in vitro test. hASC extract treatment significantly increased expression of p-Akt, p-CREB, and PGC-1 alpha in SOD1 G93A mouse model and in vitro test. Our results indicated that hASC extract reduced apoptotic cell death and recovered mutant SOD1-induced mitochondrial dysfunction. Moreover, hASC extract reduced mitochondrial membrane potential. In conclusion, we have demonstrated, for the first time, that hASC extract exert a potential therapeutic action in the SOD1 G93A mouse model of ALS and in vitro test. These findings suggest that hASC hold promise as a novel therapeutic strategy for treating ALS.</P>

      • Decitabine, a DNA methyltransferases inhibitor, induces cell cycle arrest at G2/M phase through p53-independent pathway in human cancer cells

        Decitabine (5-aza-2'-deoxycytidine), an inhibitor of DNA methyltransferases, has a wide range of anti-metabolic and anti-cancer activities. Decitabine also induces cell cycle arrest at G2/M phase and apoptosis in human cancer cells. However, the cellular and molecular mechanisms of this cell cycle arrest are poorly understood. In the present study, we investigated the roles of the tumor suppressor p53 and the cyclin-dependent kinase (Cdk) inhibitor p21 following decitabine-induced G2/M arrest in human cancer cells. DNA flow cytometric analyses indicated that decitabine induced a G2/M arrest in AGS gastric and A549 lung carcinoma cell lines, which have wild type p53. Western blot analyses using whole cell lysates from AGS cells demonstrated that decitabine treatment did not change the steady-state level of Cdks and Cdk inhibitor p27, but it partially inhibited expression of cyclin A, cyclin B1, and Cdc25C proteins. However, similar results were found using the A549 cell line, where decitabine induced a dramatic up-regulation of both p53 and p21 expression, and the increased levels of p21 were associated with increased binding of p21 with Cdks, cyclin A, and cyclin B1. Knockdown of p53 by small interfering RNA (siRNA) markedly abolished p53 induction by decitabine in AGS cells, yet p53 siRNA had no attenuating effect on p21 induction. In addition, depletion of p21 expression with siRNA, but not p53, significantly attenuated decitabine-induced G2/M arrest. We also observed that decitabine strongly induced G2/M arrest associated with p21 induction in both p53 allele-null (-/-) HCT116 and wild type p53 (+/+) HCT116 cell lines. Therefore, our data indicated that p21 plays a crucial role in decitabine-induced G2/M arrest and operates in a p53-independent manner.

        고인슐린증이 없었던 인슐린종 1 예

        Insulinoma is a disorder characterized by hypoglycemia due to endogenous hyperinsulinemia. The elevated plasma insulin level and hypoglycemia should be established for the diagnosis of an insulinoma. Other laboratory methods such as immunoreactive insulin to glucose(IRI/G) ratio, C-peptide, proinsulin, and some provocation tests are also employed to detect an insulinoma because hyperinsulinemia is often not found. Especially, IRI/G ratio has been considered as very sensitive and specific test to confirm the insulinoma without peripheral hyperinsulinemia. We report a case of insulinoma without hyperinsulinemia and elevation of IRI/G ratio, which was diagnosed by endoscopic ultrasonography and THPVS(Transhepatic portal venous sampling). It is suggested that more confirmatory tests will be necessary for the diagnosis of insulinoma especially when it is highly suspected although IRI/G ratio is not increased(J Kor Soc Endocrinol 10: 439-444, 1995).

        Identification of Novel SNPs in Bovine Insulin-like Growth Factor Binding Protein-3 (IGFBP3) Gene

        The insulin-like growth factors (IGFs), their receptors, and their binding proteins play key roles in regulating cell proliferation and apoptosis. Insulin-like growth factor binding protein-3 (IGFBP3, OMIM #146732) is one of the proteins that bind to the IGFs. IGFBP3 is a modulator of IGF bioactivity, and direct growth inhibitor in the extravascular tissue compartment. We identified twenty-two novel single nucleotide polymorphisms (SNPs) in IGFBP3 gene in Korean cattle (Hanwoo, Bos taurus coreanae) by direct sequencing of full gene including -1,500 bp promoter region. Among the identified SNPs, five common SNPs were screened in 650 Korean cattle; one SNP in promoter (IGFBP3 G-854C), one in 5'UTR region (IGFBP3 G-100A), two in intron 1 (IGFBP3 G+421T, IGFBP3 T+1636A), and one in intron 2 (IGFBP3 C+3863A). The frequencies of each SNP were 0.357 (IGFBP3 G-854C), 0.472 (IGFBP3 G-100A), 0.418 (IGFBP3 G+421T), 0.363 (IGFBP3 T+1636A) and 0.226 (IGFBP3 C+3863A), respectively. Haplotypes and their frequencies were estimated by EM algorithm. Six haplotypes were constructed with five SNPs and linkage disequilibrium coefficients (|D'|) between SNP pairs were also calculated. The information on SNPs and haplotypes in IGFBP3 gene could be useful for genetic studies of this gene.

      • A phase II study of irinotecan plus cisplatin for patients with advanced stage IIIB or IV NSCLC previously treated with nonplatinum-based chemotherapy

        <B>BACKGROUND.</B><P>Irinotecan (I) and cisplatin (P) are active chemotherapy agents with clinical synergy in non–small-cell lung cancer (NSCLC). We evaluated the efficacy of IP regimen as a salvage treatment of patients with NSCLC that progressed after nonplatinum-containing regimen(s).</P><B>METHODS.</B><P>Eligibility required histologically confirmed NSCLC, bidimensionally measurable disease, ECOG PS 0-2, and progressive disease after nonplatinum-based chemotherapy. Treatment consisted of I (65 mg/m<SUP>2</SUP>) and P (30 mg/m<SUP>2</SUP>) i.v. on Days 1 and 8 of a 21-day cycle, for a maximum of 6 cycles. An informed consent was obtained from all patients.</P><B>RESULTS.</B><P>Between August 2002 and May 2004, 32 patients with median age of 56 years (range, 42–74) were enrolled. Twenty-four (75%) patients were men, and 28 (88%) had ECOG PS 0 or 1. Twenty-five patients had adenocarcinoma and 6 had squamous-cell carcinoma. All patients were evaluated for response and toxicity, and the response rate was 40.6%. After a median follow-up of 18.5 months, the median survival time was found to be 9.3 months, with a 1-year survival rate of 43.8%. Toxicities were moderate and manageable, with 47% G3 and 9% G4 neutropenia, 19% G3 diarrhea, and 22% G3 asthenia. There was no G4 nonhematologic toxicity.</P><B>CONCLUSIONS.</B><P>The irinotecan and cisplatin combination is an active and well-tolerated regimen for the patients with advanced NSCLC that progressed after nonplatinum-containing regimen(s).       • 위상형 CGH(computer generated hologram)를 이용한 비구면 파면 및 Bessel beam 제작

        We have designed and fabricated computer generated holograms(CGH) to generate ashepheric wavefront and Bessel beam. Designed CGH is recorded onto the photopolymer which is used as a volume hologram material mating CGHOE(computer generated holographic optical element). To investigate properties of fabricated CGHOE, intensities of Bessel beam are measured as a function of propagation(z-axis) and transverse (x-axis) distances.

        We fabricated photopolymer hologram and explained hologram formation processes. Dynamic self-diffraction oscillation of photopolymer is observed by two wave mixing experiment and it is well explained by the modified Kogelnik's coupled wave equation.

      • 알콕시기와 아릴옥시기가 치환된 폴리포스파젠의 합성 및 특성에 관한 연구

        Polyphosphazenes are represented as a general formula [(R_1R_2)p=N]_n, where R_1 and R_2 are representing organic substituent groups. Poly[(2,2,2-trifluoroethoxy)(p-chlorophenoxy)phosphazenes] were synthesixed by the 2,2,2-trifluoroethoxy and p-chlorophenoxy. The polymers were characterized by means of element analysis, 1^H-NMR,(31)^P-NMR, FT-IR and gel permeation chromatograpy. The weights of all the polymers are above 10^6, where their chromatogrames, in particular, exhibit a monomodal and narrow molecular weoght distribution with low (M ̄w/Mn) (2.7∼3.77) values in contrast to the bimodal and broad (M ̄w/Mn〉10) molecular weight distributions in known poly(organophosphazenes) systems. The thermal properties of the copolymers reveal appropriately median values between those of each homopolymers. The glass temperatures of the copolymers exhibit between -47℃ and 16℃. The thermal degradation starts at 334℃ and ends up to 460℃ with two step decompositions. The tensile strength of the copolymers is intermediate between those of each homopolymer. However, ultimate elongation values show higher valve than those of each homopolymers.

        This paper begin a new approach in the water-gate controller using radio communication. The dissertation is on the controllable device development of water-gate in the remote distance from water-gate trough the transceiver by radio communication. The proposed water-gate control device is simple in structure, an suitable for implementation of water-gate control in the remote distance. The remote controller water-gate device inspected the up and down motion of water-gate through the LCD displayer, so that it was very safety driving about the surroundings information, over loading and position of water-gate, and so on.

      • 레이저에 의한 폐활성슬러지의 전처리에 관한 기초연구

        The objective of this study have been carried out to investigate the solubilization and optimum operation of waste activated sludge by laser for sewage sludge reduction. The concentration of waste activated sludge was adjusted to 0.4%, 0.6%, 0.8%, 1.0%, 1.5% of total solids, respectively. Laser modules were used for pretreatment of waste activated sludge out to use laser module(wavelength : 635~650nm, powermax : <5mW, operating electric power : 6V) in the laser pretreatment. Waste activated sludge was evaluated the temperature, pH, SCOD, SCOD/TCOD rate for the 24 hours. The results showed that SCOD/TCOD rates were increased at the low concentration of total solids, because permeability characteristics of laser were largely interrupted by medium in the sludge. The SCOD/TCOD rates were gradually increased until 1hr of irradiation. At the 0.4% of TS concentration, SCOD/TCOD rates were most effective by increasing sludge temperature from 20.0℃ to 26.3℃, decreased pH from 7.1 to 6.5 and increased concentration of SCOD from 58mg/L to 85mg/L.
