        SOD2와 NDPK2 유전자 집적에 의한 페튜니아의 아황산가스 저항성 증진

        유전자 집적에 의해 아황산가스 저항성 증진 페튜니아를 개발하고자 비생물적스트레스 저항성 유전자로 널리 알려진 SOD2 와 NDPK2유전자가 각각 도입된 SOD2 형질전환 계통 SOD2-2-1-1-35(T₄)[S(T₄)]와 NDPK2 형질전환 계통 NDPK2-7-1(T₂)[N7-1(T₂)]간 상호교잡을 실시하여 교잡 제1세대를 획득하였다. 교잡 제1세대 중 SOD2와 NDPK2유전자가 모두 집적된 개체 획득율은 32.1-73.0%이었고, SOD2와 NDPK2유전자가 모두 집적된 개체는 SOD2 또는 NDPK2유전자가 단독 도입된 개체에 비해 아황산가스 30ppm처리 피해율이 2.6-5.1배 낮아 아황산가스 저항성이 증진되었음을 확인하였다. 또한 아황산가스 저항성이 증진된 교잡 제1세대를 자가수분에 의해 세대진전 시켜 획득한 교잡 제2세대의 아황산가스 저항성 검정을 통하여 교잡 제1세대에서의 아황산가스 저항성이 후대에서도 안정적으로 발현함을 확인하였다. 또한, 광합성 효율 증진 조사를 통해 아황산가스 저항성이 증진된 SOD2와 NDPK2유전자 집적 후대가 아황산 가스에 대한 스트레스를 덜 받고 있음을 확인하였고, RT-qPCR분석을 통해 SOD2와 NDPK2유전자 집적 후대의 아황산가스 저항성 증진이 SOD2와 NDPK2 유전자 집적에 의한 효과임을 확인하였다. The aim of this study was to develop a transgenic petunia with enhanced resistance to sulfur dioxide (SO₂) gas by stacking two genes, SOD2 and NDPK2, which are both known to confer resistance to abiotic stresses. The first-generation hybrids (TF1) were obtained through reciprocal crosses between an SOD2-transgenic line SOD2-2-1-1-35(T₄)[S(T₄)] and an NDPK2- transgenic line NDPK2-7-1(T₂)[N7-1(T₂)]. Approximately 32.1-73.0% of the first-generation hybrids (TF₁) carried both SOD2 and NDPK2 genes. These hybrids showed 2.6 and 5.1 times less damage than hybrids carrying only SOD2 or NDPK2 genes, respectively, when they were treated with SO₂ gas at 30 ppm. This confirmed that the heterozygous hybrids were more resistant to SO₂ than the hybrids carrying either one of the resistance genes. Second-generation hybrids (TF₂) were obtained by selfing the TF₁ individuals. We confirmed the expression of the stacked genes in the TF₂ hybrids by phenotypic observation of their response to SO₂ gas at 30 ppm as well as using RT-qPCR and photosynthetic efficiency.

      • 당뇨병과 동반된 화농성 간농양의 임상상 및 예후인자

        연구배경: 감염질환은 혈관질환과 함께 당뇨병의 중요한 사망원인의 하나이며, 혈당조절이 불량할수록 감염 빈도가 증가됨은 잘 알려져 있다. 간농양의 발생이 있어서도 당뇨병은 중요한 기저질환 중의 하나이다. 일반적으로 간농양의 가장 흔한 원인균은 Escherichi coli라고 알려져 있으나, 최근 당뇨병을 가진 간농양 환자에서 Klesiella pneumoniae의 검출이 증가되고 있다. 또 최근 연구에 의하면 K.pneumonie가 간농양의 가장 흔한 원인균이며, K. pneumoniae에 의한 간농양은 K.pneumoniae 이외의 균주에 의한 간농양보다 당뇨병과 밀접히 관련되어 있다고 보고된 바 있다. 이에 저자들은 간농양 환자를 대상으로 당뇨병환자의 빈도를 알아보고, 그 원인균 및 특징을 비당뇨병 환자들의 경우와 비교해 보고자 하였다. 방법: 1992년 1월부터 1999년 6월까지 방사선학적 검사 혹은 수술에 의해 간농양이 확인되거나, 경피적 천자나 수술을 통한 배농액 배양, 혹은 혈액 검사에서 원인균이 증명된 182명의 환자를 대상으로 후향적 임상고찰을 실시하였다. 결과: 화농성 농양을 가진 167명의 환자(남:94명, 여:73명)중 당뇨병을 가진 환자는 54명 (남:30명, 여:24명)으로 32.3%를 차지하였다. 환자의 평균나이는 당뇨군에서 62.1 ± 13.4세, 비당뇨군에서는 55.1 ±15.5세로 당뇨군에서 높았으며(p=0.0021), 남녀간의 평균나이 비교 시 두 군 모두 여성의 나이가 의미있게 높았다. 비당뇨군에 비하여 당뇨군의 경우 저알부민혈증의 빈도와 aspartate transaminase가 증가되어있었다. 원인균주는 K. pneumoniae (당뇨군54%, 비당뇨군 39.1%), E. coli(당뇨군 17.5%, 비당뇨군 18.8%), Pseudomonas aeruginosa, Streptococcus viridans, Enterococcus 순이었으며 두 군간의 의미는 차이는 없었으나 K. pneumoniae의 경우 다른 균종에 비해 단독감염의 빈도가 높았다(82.8%). 사망률은 당뇨군에서 16.7%, 비당뇨군에서 7.1%로 당뇨군에서 의미있게 높았으며(p=0.019), 당뇨군에서의 사망환자는 모두 패혈증이 동반되어 있었다. 당뇨환자의 사망군은 생존군에 비해 당뇨병 이환기간이 길고 혼합감염의 빈도가 높았다(p=0.046). 검사실 소견에서는 총 빌리루빈 및 알카리성 포스파타제의 증가가 관찰되어 있었다. 결론: E. coli가 간농양의 주된 원인균이라는 기존 보고와는 달리 당뇨군과 비당뇨군 모두 K. pneumoniae가 간농양의 주된 원인균이었다. 또한 당뇨 유병기간이 길고 당뇨조절이 불량한 환자에서 혼합감염이며 총 빌리루빈 및 알카리성 포스파타제의 증가등과 같은 위험인자가 존재할 경우 사망률이 증가될 수 있으므로 이들 군에서는 적극적인 치료가 필요할 것으로 사료된다. Background: Liver abscesses are commonly associated with underlying disease, particularly diabete mellitus. The number of the liver abscesses caused by Klebslella pneumoniae in diabetic patient has been increased in Korea nowadays. This study was conducted to clarify the clinical presentation and prognostic factors of pyogenic liver abscesses, especially in diabetic patients and to determine the proportion of K. pneunomiae as a pathogen in liver abscess in Korea. Methods: Medical records of 167 patients treated for pyogenic liver abscess from January, 1992 through June, 1999 were reviewed retrospectively in detail. Major Pathogenic organism, clinical manifestations, prognostic factors, the importance of diabetes mellilus as an underlying disease and its effect on clinical features and prognosis were analyzed. Results: Among 167 cases of pyogenic liver abscess, underlying diabetes mellitus was present in 32.3%. The mean age of patients was 62.7 13.4 years in diabetic liver abscess group and 55.1 15.5 years in non-diabetic liver abscess group. Most liver abscesses were cryptogenic in origin or secondary to the billary tree diseases. The clinical presentations among the two groups were not significantly different. When compared to patients without diabetes, patients with diabetes had significantly higher proportions of hypoalbuminemia and elevated aspartate transaminase level. The most common organism of the pyogenic abscess was K. pneumoniae in both groups. Percutaneous drainage of the abscess with appropriate antibiotics was the most commonly used therapeutic modality in liver abscess. The mortality rate of diabetic liver abscess is 16.1 % and 7.1% in nondiabetic liver abscess. Complications, especially septicemla, were found more frequently in patients with diabetes than in patients without diabetes (64.8% vs 28.3%, septicemia : 31.5% vs 9.7%). The factors influencing mortality in the diabetic liver abscess were disease duration, mixed infection, presence of septicemia, elevated bilirubin and alkaline phosphatase. Conclusions: In contrast to prior report that the E. coli was the most common pathogen in liver abscess, we found that K. pneumoniae was the most common organism cultured in liver abscess. Diabetic patients have more complications and higher mortality than patients without diabetes. Early detection and proper treatment are needed to improve the outcome for diabetic patient with liver abscess.

      • 약물의 폐 전달을 위한 염화 벤제토늄 기반 리포좀의 제조 및 평가

        Benzethonium chloride-containing egg phosphatidylcholine-based liposomes (BC/EPC-LIPOs) were developed for lung-targeted drug delivery. BC/EPC-LIPOs were fabricated via thin-film hydration method with various BC-to-EPC molar ratios of 1:4 (F1), 1:1 (F2), and 4:1 (F3). Cyanine5.5 (Cy5.5) was loaded as a fluorescent probe for the biodistribution study during the film-forming step. BC/EPC-LIPOs were evaluated by using dynamic light scattering analysis, where the mean diameter and zeta-potential values of F1, F2, and F3 were observed to be 69.5 ± 3.1 nm and 50.06 ± 1.00 mV, 59.0 ± 2.3 nm and 51.30 ± 2.98 mV, and 694.2 ± 243.3 nm and 55.01 ± 3.16 mV, respectively. To evaluate the lung distribution efficiency of the developed liposomes, F1 and F2, which possess suitable particle sizes for the intravenous administration, were injected into the tail vein of ICR mice. At 6 h post-injection, the lung, liver, kidney, heart, spleen, and brain were dissected, and the radiant efficiency (RE) values of the or- gans were measured using the near-infrared fluorescence imaging method. The lung RE value of F2 was 1.69-fold higher than that of F1, indicating a more efficient lung targetability of F2. This phenomenon can be attributed to the higher content of BC in F2 than F1. Considering the lung-targetability, as well as the biocompatible natures of BC and EPC, the developed BC/EPC-LIPOs can be used as an efficient lung-targeted drug delivery system.

      • 濟州道 菜蔬·花卉園藝의 生産實態, 育成方案 및 流通構造改善에 관한 硏究

        濟州道의 施設을 理用한 菜蔬類와 球根切花類 및 洋蘭類의 재배실태를 調査한 結果는 다음과 같이 要約할 수 있다. 1. 作目이 토마토와 오이에 編重되어 있어 딸기等을 첨가하여 作目이 多樣化하여야 할 것으로 思料되었다. 2. 施設內의 土壤管理에 대한 認識이 不足하며, 均衡施肥가 施行되지 않고 있었고 土壤酸度도 矯正해야 할 농가가 많았다. 3. Mg이 缺乏된 施設이 대부분이므로 이의 是正이 時急한 課題였다. 4. 施設內 土壤에서 鹽類集積의 被害가 우려되는 곳은 거의 없었으나 裁培年限이 5年以上된 施設에서는 土壤改良이 이뤄저야 할 것으로 思料되었다. 5. 作付體係를 改善해서 年 2期作 體係가 確立되어야 겠으며 施設管利技術이 普及, 育苗技術의 改善 및 共同育苗를 施行해서 健全苗의 養成이 重要課題였다. 6. 濟州道內에서 産業的으로 栽培되고 있는 蘭類 中에서 熱帶産 Cymbidium類가 全體 洋蘭 栽培量의 67.8%로 가장 많았는데 그 中에서도 Eiko, sharnell-5, Kenny가 數的으로 많았다. 7. 溫帶産蘭(東洋蘭)類 中에서는 새우난초類가 數的으로 가장 많았고, 나도풍난, Dendrobrum moniliforme, Cymbidium 風蘭의 順位이었으며, 自生寒蘭은 대 40,396촉이 栽培되고 있었다. 8. 供試 5種의 球根類들은 virus 감염이 甚하여 全量이 virus 無病株로 대체되어야 할 것으로 판단되었다. 9. 서귀포지역의 土壤은 害蟲이 많이 分布하여 球根切花栽培에 큰 위협조건으로 나타났으며 앞으로 도양소독에 대하여 보다 집약적이고 효율적인 方法이 개발되어야 하겠다. 10. 한라산의 고냉지를 利用한 球根의 低溫處理 效果에 對하여 많은 적응시험이 요망되며 道當局은 고냉지의 개발과 利用에 적극적인 지원을 투여하여야 할 것이다. 11. 植栽되고 있는 5種의 球根類 모두 50% 以上이 外國수입에 의존하고 있으므로 國內의 組織培養技術을 利用한 virus 無病良質球根의 生産이 절실히 요망된다. Studies on various cultural practices of vegetable crops, and orchids have been conducted in Chejudo. Followings are the results of such studies. 1. Too much importance has been given to Tomato and Cucumber crops. An additional crop such as Strawberry should be included to broaden the studies. 2. In general, lack of informations on soil managements appear to be prevailing; scheduled fertilization, for instance, has not been performed and pH level must be adjusted on many farm lands. 3. Mg deficiency has been very much in common, therefore, is corection are matter of utmost importance. 4. Problems arising from salt accumulation were nearly unimportant, however, soil improvements through various cultural practices are recommended for those soils which have been continuous cropping for more than 5 years. 5. Planting system should be improved so as to establish 2-crops/year system. In addition, proper extention of various soil management techniques and improvement on nursing techiques are of inportant matters. Commonly sharing community nursing practices must be accompanied in order to provide healthy-seedlings. 6. Among commercially growing orchid species in Chejudo, the tropical Cymbidium species found to be the most widely grown, nearly 67.8% 'Kiko', 'Sharnell-5', 'Kenny' appear to be most in common. 7. Among oriental orchids, Calanthe spp are found to be most prevalent followed in the order of Aerides japonica, Dendrobium moniliforme, Cymbidiums, respectively. 8. The total number of the shoots of Cymbidium kanran being cultivated in Chejudo were found to be 40,396. 9. Virus infections appear to be universal in all 5 species of bulbs studied therefore, virus-free stocks must be essential. 10. Sogwipo-soil, in general, heavily infected by soil-born insects of various kinds are badly affecting bulb productions. More efficient methods of soilsanitations must be studied and introduced in the future. 11. The studies of low temperature treatment on bulb crops by utilizing the high altitude growing in the area of mountain Halla are in great need. An extensive assistance from the provincial government are very much in need in order for the development and utilization of the high-altitude cultivation. 12. More than 50% of the bulbs of the 5 species grown are being imported. Domestic production utilizing 'meristem culture' which enable the virus free stocks are in great demand.

      • 보존적 치료로 호전된 요로감염에 의한 자발성 신파열

        자발성 신파열은 외상 혹은 수술 등의 병력 없이 신장 피막 혹은 신장 주위 혈종이 자발적으로 발생하는 드문 질환으로, 원인으로는 요로 결석[1], 결핵, 신우신염, 신농양 같은 감염성 질환이나[2] 신장의 종양 및 신장 이식 수술 후[3], 후천성 낭성 신환[4] 등이 관련이 있다. 그 중 복막 투석 환자에서 낭성 신질환에 의한 자발성 신파열은 국내 보고가 있었으나 요로감염에 의한 자발성 피막하 혈종을 동반한 신파열은 문헌 보고가 없었다. 저자들은 요로감염으로 진단된 환자에서 피막하 혈종을 동반한 신파열 1례를 경험하였기에 문헌 고찰과 함께 보고하는 바이다.

        양이온교환수지에 대한 Co(Ⅱ), Ni(Ⅱ), Cr(Ⅲ) 및 Fe(Ⅲ) 이온의 흡착 특성 : 원자로 일차 냉각재 계통내 탈염공정에의 적용 Application to the Demineralizing Process in a Primary Coolant System of PWR

        가압경수로 일차냉각수내 탈염공정의 효과적 운용을 위하여 Amberlite IRN-77 양이온 교환수지의 특성 및 Co(II), Ni(II), Cr(III), Fe(III) 이온의 흡착거동을 연구하였다. 용존 금속이온 농도가 약 200 mgL^-1 인 용액 100 mL에 대하여 이온교환수지 투입량은 약 0.6g이 가장 적절하였으며, 이온교환 반응 속도 측정 결과 대부분의 흡착은 반응 1 시간 이내에 신속히 발생하였다. 수지에서의 양이온 교환은 Langmuir 흡착등온선을 잘 따르는 것으로 나타났으나, Fe(III)의 경우, 다른 금속과는 달리 쉽게 평형에 이르지 못하였으며 이는 공기와 오래 접촉한 결과 철산화물 또는 수산화물을 형성하며 용액 중으로부터 침전되기 때문인 것으로 관찰되었다. 전자가가 동일한 Co(II)와 Ni(II)가 동시에 포함된 모의 조제용액에 대하여 흡착실험을 한 결과, 각 이온은 거의 동일한 흡착 거동을 보였다. 그러나 전자가가 큰 Cr(III)이 첨가된 경우, 이들은 기존에 수지에 흡착되어 있던 전자가가 낮은 금속들을 탈착시키고 대신 흡착하는 경향을 보였다. 따라서 이온교환수지를 이용한 효과적인 탈염 공정을 위해서는 용존 금속 상호간의 경쟁적 흡착관계의 규명이 필요한 것으로 여겨진다. Characteristics of Amberlite IRN-77, a cation exchange resin, and the mechanisms of its adsorption equilibria with Co(II), Ni(II), Cr(III) and Fe(III) and Fe(III) ions were investigated for the application of the demineralizing process in the primary coolant system of a pressurized water reactor (PWR). The optimum dosage of the resin for removal of the dissolved metal ions at 200 mgL^-1 was 0.6 g for 100 mL solution. Most of each metal ion was absorbed onto the resin in an hour from the start of the reaction. Each metal adsorption onto the resin could be well represented by Langmuir isotherms. However, in the case of Fe(III) adsorption, continuous formation of Fe-oxide or -hydroxide and its subsequent precipitation inhibited the completion of the equilibrium between the metal and the adsorbent. Cobalt(II) and Ni(II), which have an equivalent electrovalence, were adsorbed to the resin with a similar adsorption amount when they coexisted in the solution. However, Cr(III) added to the solution competitively replaced Co(II) and Ni(II) which were already adsorbed onto the resin, resulting in desorption of these metals into the solution. The result was likely due to a higher adsorption affinity of Cr(III) than Co(II) and Ni(II). This implies that the interactively competitive adsorption of multi-cations onto the resin should be fully considered for an efficient operation of the demineralizing ion exchange process in the primary coolant system.

Therefore, if the questions that confronts us are solved and the reasonable supporting method to floriculture rearing is established, the cultivation of flowers will be florished.         酸棗仁이 CT105에 의한 신경세포 상해 및 白鼠의 기억에 미치는 영향

        Objective: This study was designed to research whether demonstrate neuroprotective and memory enhancing effects of Z1BU, suggesting its beneficial actions for the treatment of AD. Methods: The present study investigated effects of the dichloromethane extract of Semen Zizyphi Spinosae(ZIBU) on neurotoxicity and reactive oxygen species(ROS) and nitric oxide(NO) formation in PC 12 cell line overexpressed with carboxyl-terminal 105 amino acid peptide fragment of amyloid precursor protein(CT105). In addition, the present study evaluated its radical scavenging activity and effects on acetylcholinesterase(AChE) activity. Furthermore, effects on neuronal loss in CAl area and cognitive deficits induced by scopolamine treatment were also evaluated in rats. Result: we found in this study that ZIBU significantly inhibited apoptotic neuronal death induced by CT105 overexpression in PC12 cells, with the IC_(50) value of 23 ㎍/ml. Based on phase-contrast and fluorescence microscopic examinations, ZIBU reversed morphological changes of CT105-expressed PC-12 cells. It was also found that Z1BU significantly induced neurite outgrowth and inhibited formation of ROS and NO.Z1BU scavenged DPPH radicals and inhibited AChE activity. Furthermore, it attenuated memory impairment induced by the injection of scopolamine in rats, based on passive avoidance test. Crystal violet staining of rat brain slices exhibited increase in the viability of pyramidal neurons in hippocampal CAI area. Conclusion: these results demonstrate neuroprotective and memory enhancing effects of ZIBU, suggesting its beneficial actions for the treatment of AD.

      • 고도비만 환자에서 복강경 위소매절제술 후 발생한 누공의 내시경 치료 1예

        Laparoscopic sleeve gastrectomy can reduce morbidity and mortality in patients with morbid obesity, but it can cause complications such as a gastrointestinal leak. A 30-year-old morbidly obese female who had type 2 diabetes mellitus and hypertension with estimated body mass index of 40.2 kg/m2 was admitted. Laparoscopic sleeve gastrectomy was performed. On postoperative day 19, a leak was suspicious on physical examination and radiologic findings. Conservative management was performed, but the patient was hemodynamically unstable and imminently septic. After laparoscopic drainage procedure, esophagogastroduodenoscopy was performed and revealed the fistula opening at staple line just below gastroesophageal junction. Fibrin tissue adhesive was injected around the fistula and the esophageal covered stent was inserted to cover the leak. At 14th days after stent insertion, the barium study confirmed no more leak.
