        Roseovarius tibetensis sp. nov., a halophilic bacterium isolated from Lake LongmuCo on Tibetan Plateau

        Two Gram-stain negative halophilic strains, designated as LM2T and LM4, were isolated from Lake LongmuCo on Tibetan Plateau. These two strains were aerobic, catalaseand oxidase- positive, nonmotile and rod-shaped organisms. Phylogenetic analysis based on 16S rRNA gene sequences indicated that LM2T and LM4 belong to the genus Roseovarius, with Roseovarius tolerans EL-172T (97.3% and 97.4% 16S rRNA gene sequence similarity, respectively) and Roseovarius azorensis SSW084T (95.5% and 95.6% 16S rRNA gene sequence similarity, respectively) as their closest neighbors. Q-10 was the sole respiratory quinone of these two strains. The major fatty acids were C18:1 ω7c/C18:1 ω6c, C16:0, C19:0 cyclo ω8c, and 11-methyl C18:1 ω7c. The polar lipids included phosphatidylglycerol, diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylcholine, phospholipid of unknown structure containing glucosamine, and unidentified aminolipid. The DNA G + C content was between 64.2 and 64.5 mol%. DNA-DNA hybridization showed 96.7% relatedness between LM2T and LM4, 24.9% relatedness between LM2T and R. tolerans EL-172T, and 36.3% relatedness between LM4 and R. tolerans EL-172T. Based on phylogenetic analysis, DNA-DNA hybridization, a range of physiological and biochemical characteristics, LM2T and LM4 belong to the same species and were clearly distinguished from the type strains of the genus Roseovarius. It was evident that LM2T and LM4 could be classified as a novel species of the genus Roseovarius, for which the name Roseovarius tibetensis sp. nov. is proposed. The type strain is LM2T (= CGMCC 1.16230T = KCTC 62028T).

        Green’s-function-based-finite element analysis of fully plane anisotropic elastic bodies

        In the paper, an anisotropic Green’s function based hybrid finite element was developed for solving fully plane anisotropic elastic materials. In the present hybrid element, the interior displacement and stress fields were approximated by the linear combination of anisotropic Green’s functions derived by Lekhnitskii formulation, the element frame fields were constructed by the interpolation of general shape functions widely used in the conventional finite element, and then they are linked by a new double-variable hybrid functional. Because the approximated interior fields exactly satisfied the governing equations related to anisotropic elasticity, all integrals in the present hybrid functional were performed along the element boundary and theoretically arbitrary hybrid polygonal element can be constructed. Finally, the present hybrid polygonal element with four edges was verified by making comparison of numerical results and exact solutions in a cantilever composite beam made with angled lamina.

        Halomonas tibetensis sp. nov., isolated from saline lakes on Tibetan Plateau

        Strains pyc13T and ZGT13 were isolated from Lake Pengyan and Lake Zigetang on Tibetan Plateau, respectively. Both strains were Gram-negative, catalase- and oxidase-positive, aerobic, rod-shaped, nonmotile, and nonflagellated bacteria. Phylogenetic analysis based on 16S rRNA gene sequences showed that strains pyc13T and ZGT13 belong to the genus Halomonas, with Halomonas alkalicola 56-L4-10aEnT as their closest neighbor, showing 97.4% 16S rRNA gene sequence similarity. The predominant respiratory quinone of both strains was Q-9, with Q-8 as a minor component. The major fatty acids of both strains were C18:1 ω6c/C18:1 ω7c, C16:1 ω6c/C16:1 ω7c, C16:0, and C12:0 3OH. The polar lipids of both strains consisted of phosphatidylethanolamine, phosphatidylglycerol, diphosphatidylglycerol, glycolipid, phospholipids of unknown structure containing glucosamine, and unidentified phospholipids. The DNA G + C content of pyc13T and ZGT13 were 62.6 and 63.4 mol%, respectively. The DNA-DNA hybridization values of strain pyc13T were 34, 41, 61, 35, and 35% with the reference strains H. alkalicola 56-L4-10aEnT, H. sediminicola CPS11T, H. mongoliensis Z-7009T, H. ventosae Al12T, and H. fontilapidosi 5CRT, respectively. Phenotypic, biochemical, genotypic, and DNA-DNA hybridization data showed that strains pyc13T and ZGT13 represent a new species within the genus Halomonas, for which the name H. tibetensis sp. nov. is proposed. The type strain is pyc13T (= CGMCC 1.15949T = KCTC 52660T).

        Three new diphenylpropanes from Celastrus hindsii

        Three new diphenylpropanes, HindsiipropaneA–C (1–3), together with one known arylpropyl quinoneGriffithane D (4), were isolated from Celastrus hindsii. Their structures were established by 1D and 2D nuclearmagnetic resonance spectroscopic analysis, and massspectroscopy. Compound 4 was firstly obtained in thisgenus. All the isolated compounds were evaluated in vitrofor cytotoxicity against four human tumor cell lines (A549,HCT116, MDA-MB-231, BEL7404) by the MTT assay.

      • Bioinformatic Prediction of SNPs within miRNA Binding Sites of Inflammatory Genes Associated with Gastric Cancer

        Polymorphisms in miRNA binding sites have been shown to affect miRNA binding to target genes, resulting in differential mRNA and protein expression and susceptibility to common diseases. Our purpose was to predict SNPs (single nucleotide polymorphisms) within miRNA binding sites of inflammatory genes in relation to gastric cancer. A complete list of SNPs in the 3'UTR regions of all inflammatory genes associated with gastric cancer was obtained from Pubmed. miRNA target prediction databases (MirSNP, Targetscan Human 6.2, PolymiRTS 3.0, miRNASNP 2.0, and Patrocles) were used to predict miRNA target sites. There were 99 SNPs with MAF>0.05 within the miRNA binding sites of 41 genes among 72 inflammation-related genes associated with gastric cancer. NF-${\kappa}B$ and JAK-STAT are the two most important signaling pathways. 47 SNPs of 25 genes with 95 miRNAs were predicted. CCL2 and IL1F5 were found to be the shared target genes of hsa-miRNA-624-3p. Bioinformatic methods could identify a set of SNPs within miRNA binding sites of inflammatory genes, and provide data and direction for subsequent functional verification research.

        Genetic variation and phylogenetic relationships of the ectomycorrhizal Floccularia luteovirens on the Qinghai-Tibet Plateau

        Floccularia luteovirens, as an ectomycorrhizal fungus, is widely distributed in the Qinghai-Tibet Plateau. As an edible fungus, it is famous for its unique flavor. Former studies mainly focus on the chemical composition and genetic structure of this species. However, the phylogenetic relationship between genotypes remains unknown. In this study, the genetic variation and phylogenetic relationship between the genotypes of F. luteovirens in Qinghai-Tibet Plateau was estimated through the analysis on two protein-coding genes (rpb1 and ef-1α) from 398 individuals collected from 24 wild populations. The sample covered the entire range of this species during all the growth seasons from 2011 to 2015. 13 genotypes were detected and moderate genetic diversity was revealed. Based on the results of network analysis, the maximum likelihood (ML), maximum parsimony (MP), and Bayesian inference (BI) analyses, the genotypes H-1, H-4, H-6, H-8, H-10, and H-11 were grouped into one clade. Additionally, a relatively higher genotype diversity (average h value is 0.722) and unique genotypes in the northeast edge of Qinghai- Tibet plateau have been found, combined with the results of mismatch analysis and neutrality tests indicated that Southeast Qinghai-Tibet plateau was a refuge for F. luteovirens during the historical geological or climatic events (uplifting of the Qinghai-Tibet Plateau or Last Glacial Maximum). Furthermore, the present distribution of the species on the Qinghai-Tibet plateau has resulted from the recent population expansion. Our findings provide a foundation for the future study of the evolutionary history and the speciation of this species.

        Age-induced Changes in Ginsenoside Accumulation and Primary Metabolic Characteristics of Panax Ginseng in Transplantation Mode

        Background: Ginseng (Panax ginseng Mayer) is an important natural medicine. However, a long culture period andchallenging quality control requirements limit its further use. Although artificial cultivation can yield a sustainablemedicinal supply, research on the association between the transplantation and chaining of metabolicnetworks, especially the regulation of ginsenoside biosynthetic pathways, is limited. Methods: Herein, we performed Liquid chromatography tandem mass spectrometry based metabolomic measurementsto evaluate ginsenoside accumulation and categorise differentially abundant metabolites (DAMs). Transcriptome measurements using an Illumina Platform were then conducted to probe the landscape of geneticalterations in ginseng at various ages in transplantation mode. Using pathway data and crosstalk DAMs obtainedby MapMan, we constructed a metabolic profile of transplantation Ginseng. Results: Accumulation of active ingredients was not obvious during the first 4 years (in the field), but followingtransplantation, the ginsenoside content increased significantly from 6 8 years (in the wild). Glycerolipidmetabolism and Glycerophospholipid metabolism were the most significant metabolic pathways, as Lipids andlipid-like molecule affected the yield of ginsenosides. Starch and sucrose were the most active metabolic pathwaysduring transplantation Ginseng growth. Conclusion: This study expands our understanding of metabolic network features and the accumulation of specificcompounds during different growth stages of this perennial herbaceous plant when growing in transplantationmode. The findings provide a basis for selecting the optimal transplanting time.

        <P>Matricellular protein SPON2 acts as an HCC suppressor and utilizes distinct signaling events to perform dual functions in HCC microenvironment.</P><P>Tumor-associated macrophages (TAM) represent key regulators of the complex interplay between cancer and the immune microenvironment. Matricellular protein SPON2 is essential for recruiting lymphocytes and initiating immune responses. Recent studies have shown that SPON2 has complicated roles in cell migration and tumor progression. Here we report that, in the tumor microenvironment of hepatocellular carcinoma (HCC), SPON2 not only promotes infiltration of M1-like macrophages but also inhibits tumor metastasis. SPON2-α4β1 integrin signaling activated RhoA and Rac1, increased F-actin reorganization, and promoted M1-like macrophage recruitment. F-Actin accumulation also activated the Hippo pathway by suppressing LATS1 phosphorylation, promoting YAP nuclear translocation, and initiating downstream gene expression. However, SPON2-α5β1 integrin signaling inactivated RhoA and prevented F-actin assembly, thereby inhibiting HCC cell migration; the Hippo pathway was not noticeably involved in SPON2-mediated HCC cell migration. In HCC patients, SPON2 levels correlated positively with prognosis.         Gamma and X-ray irradiation as a phytosanitary treatment against various stages of Planococcus lilacinus (Hemiptera: Pseudococcidae)

        The cacao mealybug, Planococcus lilacinus Cock, is an important quarantine pest. Infested commodity should be subject to appropriate phytosanitary treatment, while irradiation is recommended for the cacao mealybug. Radio-tolerance comparison tests were conducted on the crawler, nymphs, and adult females of P. lilacinus at the X-ray radiation doses of 40, 80, and 120 Gy, respectively. The results showed that irradiation had a strong effect on preventing of development and reproduction; the adult female stage was identified as the most tolerant. During the following dose–response tests, among young and late females X-ray-irradiation (20–100 Gy), the late females were most tolerant when preventing F 1 generation 2nd instars emergence was used as the evaluation criterion. Minimum absorbed dose and its 95 % fiducial limits to provide probit 9 efficacy at 95 % confidence level (100 % mortality/inhibition in an estimated population of 93,616 individuals) were 131.5 Gy (122.5, 142.6 Gy) and 144.4 Gy (132.7, 159.4 Gy), estimating from the probit analysis on dose-mortality data of 1–30 and 1–10-day-old neonates laid by late females, respectively. In the large-scale confirmatory tests, a total of estimating 97,384 late females of P. lilacinus rearing on the pumpkins fruits were irradiated with gamma-ray at the target dose of 135 or 145 Gy (measured doses 126.1–163.0 Gy), which resulted in no F 1 generation 2nd nymphs developing during a 6-week post-treatment period. The treatment efficacy calculated is 99.9969 % at the 95 % confidence level. Therefore, a minimum absorbed dose of 163.0 Gy is recommended for phytosanitary treatment of P. lilacinus in infested commodity.

        Schisandrin B Improves the Hypothermic Preservation of Celsior Solution in Human Umbilical Cord Mesenchymal Stem Cells

        BACKGROUND: Human umbilical cord mesenchymal stem cells (hUCMSCs) have emerged as promising therapy for immune and inflammatory diseases. However, how to maintain the activity and unique properties during cold storage and transportation is one of the key factors affecting the therapeutic efficiency of hUCMSCs. Schisandrin B (SchB) has many functions in cell protection as a natural medicine. In this study, we investigated the protective effects of SchB on the hypothermic preservation of hUCMSCs. METHODS: hUCMSCs were isolated from Wharton’s jelly. Subsequently, hUCMSCs were exposed to cold storage (4 C) and 24-h re-warming. After that, cells viability, surface markers, immunomodulatory effects, reactive oxygen species (ROS), mitochondrial integrity, apoptosis-related and antioxidant proteins expression level were evaluated. RESULTS: SchB significantly alleviated the cells injury and maintained unique properties such as differentiation potential, level of surface markers and immunomodulatory effects of hUCMSCs. The protective effects of SchB on hUCMSCs after hypothermic storage seemed associated with its inhibition of apoptosis and the anti-oxidative stress effect mediated by nuclear factor erythroid 2–related factor 2 signaling. CONCLUSION: These results demonstrate SchB could be used as an agent for hypothermic preservation of hUCMSCs.
