Glycosylation on Urinary Exosome for Biomarker Discovery

IgA Nephropathy (IgAN) and thin basement membrane nephropathy (TBMN) have identical hematuria symptom but different seriousness in renal function. In case of TBMN, renal function is usually normal but IgAN is the most common cause of end-stage renal failure. In general, the biopsy of the kidney is performed to distinguish between IgAN and TBMN. There are a lot of needs for clinicians to have a sensitive and specific biomarker for differentiation IgAN and TBMN. Exosomes which are small cell-derived vesicles have been suggested as potential biomarkers in diagnosis of disease because these are secreted from original cell with intracellular fluids and apical membrane. Urinary exosomes, 40-100nm vesicles secreted to urine by renal cells, may provide non-invasive diagnosis manner of kidney disease. Glycosylation is highly sensitive to the biological environment changes and considerably affected by disease states. Glycan is known as playing key roles in cancer metastasis and intracellular recognition. Therefore, the examination of glycosylation change in urinary exosome may be a new way for potential biomarkers to differentiate TBMN and IgAN. In this study, we have analyzed 18 individuals exosomes secreted into urine from renal epithelial cells. Samples were composed of three different groups, mainly TBMN, IgAN patients, and healthy volunteers. Briefly, N-glycans from urinary exosomes were released by PNGase F digestion and then enriched by solid phase extraction using a porous graphitized carbon cartridge. N-glycans were eluted with three different solution (10% ACN, 20% ACN, and 40% ACN with 0.05% TFA in H2O) based on the glycan size and polarity. Enriched glycans were analyzed for qualitative and quantitative profiling using high resolution MALDI-TOF/TOF mass spectrometry and performed tandem MS to obtain extensive structure information. We also separated isomer-specific glycans by nano-LC chip/Q-TOF mass spectrometry. We found that high mannose glycans are high in abundance in normal group. Complex, non-sialylated fucosylated tri-, and tetraantennary glycans are significantly different between normal and patients, which suggest the potential biomarker.

In this study, it is reviewed that histamine-induced wheal and flare responses are potential sur-rogate endpoints for predicting the clinical effects of antihistamines in patients with allergic skin diseases. Histamine plays an important role in allergic response by inducing degranulation of mast cells due to allergen exposure and mediating the inflammatory reaction. Thus, suppression of histamine-induced wheal and flare has been noted as surrogate markers for efficacy of Hl receptor antagonists. In addition, allergy skin prick test and intradermal test using histamine have been used to diagnose the histamin-induced allergic reaction. However, it has been well known that allergic diseases are not only mediated by histamine. but also by var-ious immunological inflammatory responses. Previous studies reported that there is a lack of evidence about the correlation between antihistamines to predict clinical efficacy and antihistamine efficacy, although hista-mine-induced wheal and flare responses may be useful indicators of the dose-response relationship. In con-clusion, the evaluation for the suppression of wheal and flare after histamine injection is reasonable for de-termining the treatment of allergic simple skin diseases, but there is a limit to evaluate the efficacy in com-plex inflammatory diseases mediated IgE or T cells, or other immune complex. Therefore, further studies are needed to identify additional surrogate endpoints to predict the therapeutic effect of antihistamines in other inflammatory diseases such as allergic rhinitis, atopic conjunctivitis, allergic asthma, and so on.
Customer Service Assist System based on Natural Language Processing

This paper proposes a novel assist system for customer service representatives based on natural language processing (NLP). In the proposed system, an NLP model calculates the relationships between a question from a customer and all the questions in a given FAQ list. Based on the model’s calculation, the system will recommend several FAQs that are more similar to the customer’s question than the others in the FAQ list and then the representative responses, whether the recommended questions are actually similar to the customer’s question or not. Since these responses become the data for the NLP model’s next training, the NLP model’s accuracy can be incrementally enhanced by repetitive fine-tuning with the accumulated data. The experimental result shows that the proposed system can effectively help customer service representatives as well as incrementally improve via the automatically accumulated data.

The site-selective and metal-free C–H nitrationreaction of quinoxalinones and pyrazinones as biologicallyimportant N -heterocycles with t -butyl nitrite is described. Awide range of quinoxalinones were effi ciently applied in thistransformation, providing C7-nitrated quinoxalinones withoutundergoing C3-nitration. From the view of mechanisticpoint, the radical addition reaction exclusively occurred atthe electron-rich aromatic region beyond electron-defi cientN -heterocycle ring. This is a fi rst report on the C7–H functionalizationof quinoxalinones under metal-free conditions. In contrast, the nitration reaction readily takes placeat the C3-position of pyrazinones. This transformation ischaracterized by the scale-up compatibility, mild reactionconditions, and excellent functional group tolerance. Theapplicability of the developed method is showcased by theselective reduction of NO 2 functionality on the C7-nitratedquinoxalinone product, providing aniline derivatives. Combinedmechanistic investigations aided the elucidation of aplausible reaction mechanism.

R을 이용한 광역시의 대기오염물질에 대한 데이터 시각화

1995년부터 2014년까지 약 20년간의 기상청의 대기오염 자료를 바탕으로 이를 3가지의 다른 방법으로 시각화 하였다. 첫 번째는 2014년 주요 광역시의 연중 월별로 대기 오염 물질들을 시각화 하였고, 두 번째는 1995년부터 2014년까지 주요 광역시의 연도별로 대기 오염 물질들을 시각화하였다. 마지막으로 대한민국 지도 위에 2014년 주요 광역시의 대기 오염 물질들을 점 모양으로 시각화하였다. 자료 시각화에는 묘의 기본 내장 시각화 함수 plot()과 시각화 패키지 ggplot2, ggmap를 사용하였다
