      • Diatomaceous earth를 이용한 경제적이고 효율높은 homemade PCR purification column의 제조 및 평가

        In recent years the amount of sequencing has increased tremendously, especially due to the introduction of automated DNA sequencing. PCR is principal method for the synthesis of templates for DNA sequencing. Consequently, the need for PCR cleanup for template preparation has increased. Already, many PCR purification kits are marketed. This DNA purification kits uses most glass membrane or silica gel, So, can do rapidly efficient purification by simple miniprep. But, efficiency of purification kits is different according to manufacturer and is very expensive. Therefore, we compare the homemade Diatomaceous earth column with major manufacturer's spin column methods for DNA purification. As a result, we found that the homemade Diatomaceous earth column method resulted in equal or better yields relative to the commercial purification kits.

      • 합성 Zeolite A 의 Detergent Builder 로써의 이용

        세제 builder로써의 합성 zeolite A를 하동 고령토로부터 합성하였다. 경도, 염화나트륨의 농도, linear alkyl benzene sulfonate와 합성 zeolite A와 sodium triphosphate의 비율을 달리 하는 세제 용액 중에서 non-finished cotton, resin finished cotton, polyester-cotton blend cloths를 세탁하여 세탁율과 칼슘이온농도를 측정하였다. zeolite A와 sodium triphosphate의 중량비가 1인의 경우의 실험결과는 다음과 같다. 세제의 양이 많을수록 세탁율은 증가하였으나 16˚d에서 3g/1 이상의 세제농도에서는 세탁율은 거의 증가하지 않았다. 세제농도는 경도 8˚d에서는 1.5g/1, 16˚d에서는 3g/1임을 알았으며 세탁율과 제거된 칼슘농도는 염화나트륨의 농도에는 무관하였음을 알았다. Synthetic zeolite A as a laundry derergent builder was prepared from domestic Hadong Kaolin. Non-Finished cotton, resin finished cotton and polyester-cotton blend cloths were washed in the detergent solutions which were containing various hardness, sodium chloride concentration and ratio of linear alkyl benzene sulfonate to zeolite A or to sodium triphosphate, and then cleaning performances and calcium ion concentrations were measured. The followings were the results of experiment in which the weight ratio of zeolite A to sodium triphosphate was 1. The more amount of detergent the more cleaning performance increased, but the performance was almost not inclreased with the concentration of detergent over 3g/1 at 16˚d. It was observed that the concentration of detergent was needed 1.5g/1 at hardness 8˚d and 3g/1 at hardness 16˚d. It was obtained that the cleaning performance and the calcium binding capacity were independent on the concentration of sodium chloride.

      • 암 생쥐에서 Testosterone으로 인한 유해감각예민도 둔화와 이에 대한 Naloxone의 차단효과

        In order to examine the effect of testosterone on antinociception and the role of opioid system as an underlying mechanism, thirty-seven adult female mice(Institute of Cancer Research) were randomized to Naloxone(n=18) and Control group(n=19). Testosterone propionate(1mg/kg, ip) with naloxone HCL(2mg/kg, ip) or with normal saline(5ml/kg, ip) was administered on the first and the third experimental days. Nociceptive sensitivity was measured by Tail flick test(TFL, sec) before and after injection on the first and the third experimental days. The same dose of testosterone was injected on the second experimental day. The results were as follows; 1.TFL of control group was significantly increased by testosterone. TFL of control group measured on the first experimental day before and after injection were 5.5±1.1 sec(mean±sd) and 6.3±1.2 sec, respectively, and the change was significant(t=4.06, p=0.001). That measured on the third experimental day before injection was 5.9±1.0, showing a significant increase compared to that of the first experimental day(t=2.37, p=0.029). 2.Testosterone-induced antinociception was blocked by naloxone. In naloxone group, change in TFLs measured before(5.6±0.8) and after(5.3±0.9) injection was not significant on the first experimental day. On the third day, TFL measured before injection(6.6±1.2) was increased significantly than that of the first experimental day(t=2.87, p=0.011), but it decreased to baseline level after injection of naloxone(5.3±1.3; t=4.73, p=0.000). From these results, it is suggested that testosterone has antinociceptive effect, which is mediated by endogenous opioid system.

        Current-Lead Design for Cryocooled HTS Fault Current Limiters

        <P>A comprehensive design procedure is presented for the current leads in HTS fault current limiters continuously refrigerated by a cryocooler. Two specific aspects are focused here in addition to the standard lead-design. The first is an optimization with taking into account the heat conduction through electrical insulator needed for high-voltage applications. Through a rigorous analysis, it is verified that the standard design should be modified in order to minimize the overall heat leak. The second is the transient thermal behavior that may be caused by excessive current. A numerical analysis is performed to estimate the peak temperature after a short current-limiting period, including the effect of fins as enhanced local heat capacity. Some of the design details are demonstrated for the specifications of FCL's under development in Korean R&D programs.</P>

        항구포자충제제의 사용 여부에 따른 구포자충 감염실태 조사

        The present study was conducted to investigate the prevalence of Eimeria infection according to anti-coccidiosis drugs treatment in 1,300 slaughtered chickens from 130 farms in Gyeongnam, Jeonnam, Jeonbuk, Gyeongbuk and Chungnam during the period from October 2002 to August 2003. The prevalence of Eimeria infection by drug administration for treatment and prevention or non-administration was shown 71.4%, 82.6% and 72.3%, respectively, and so infection rate of chicken farms was similar independent on drug administration or not. The prevalence of Oocysts infection level of chicken farms by administrated drug for treatment was shown lower (25.3%) than for prevention (35.4%) and non-treatment (36.2%). The prevalence of Oocysts infection level by drug administration under 2 days for treatment was shown higher infection rate (32.2%) than over 3 days (20%) and also under 2 days for prevention has similar tendency of that of treatment and so, it was conclusion that administration of drug over 3 days has the effect for treatment and prevention of Eimeria infection. The prevalence of Oocysts infection level by age for prevention has various level from 48.8% to 22.9% but on 22~25 days was shown the most higher ratio (48.8%) and on 19∼21 days was shown the lowest rate (22.9%) among the all age groups.

      • Comparison of prasugrel versus clopidogrel in Korean patients with acute myocardial infarction undergoing successful revascularization

        <P><B>Abstract</B></P> <P><B>Background</B></P> <P>Although there have been several reports that prasugrel can improve clinical outcomes, the efficacy and safety of prasugrel is unknown in Korean patients with acute myocardial infarction (AMI) undergoing successful revascularization.</P> <P><B>Methods</B></P> <P>A total of 4421 patients [637 patients were prescribed prasugrel (60/10 or 5mg, loading/maintenance dose) and 3784 patients clopidogrel (600 or 300/75mg)] with AMI undergoing successful revascularization were enrolled from the core clinical cohort of Korea Acute Myocardial Infarction Registry-National Institute of Health.</P> <P><B>Results</B></P> <P>After propensity score matching (637 pairs), there were no significant differences in baseline clinical and procedural characteristics and in-hospital medications between the two groups. The primary efficacy endpoint, defined as the composite of cardiac death, MI, stroke, or target vessel revascularization at 6 months showed no significant difference between prasugrel and clopidogrel (2.4% vs. 2.9%, <I>p</I> =0.593). Also, no difference was observed in the composite of cardiac death, MI, or stroke during hospitalization between two groups (0.8% vs. 0.9%, <I>p</I> =0.762). However, the incidence of in-hospital Thrombolysis in Myocardial Infarction (TIMI) major or minor bleeding was significantly higher in prasugrel compared with clopidogrel (5.3% vs. 2.7%, <I>p</I> =0.015). In multivariate linear regression analysis, trans-femoral intervention, use of glycoprotein IIb/IIIa inhibitors, use of calcium channel blocker, and use of prasugrel were independent predictors of in-hospital TIMI major or minor bleeding [odds ratio (OR)=6.918; 95% confidence interval (CI)=2.453–19.510, OR=2.577; 95% CI=1.406–4.724, OR=4.016; 95% CI=1.382–11.668, OR=2.022; 95% CI=1.101–3.714].</P> <P><B>Conclusions</B></P> <P>Our study shows that the recommended dose of prasugrel had significantly higher in-hospital bleeding complications without reducing ischemic events compared with clopidogrel. However, further large-scale, long-term, randomized clinical trials are required to accurately assess the efficacy and safety of prasgurel and to find out the optimal dose for Korean AMI patients.</P>

        폐렴의 진단에서 정량적 기관지폐포 세척액 배양의 유용성

        N/A Background: The aim of this study is to evaluate the usefulness of quantitative culture of bronchoalveolar lavage (BAL) fluid for the diagnosis of bacterial pneumonia and identification of causative agents. Methods: Study group consisted of 30 epiaodes in 28 patients, enrolled from January 1995 through June 1996. Inclusion criteria were 1) presence of respiratory symptoms such as cough, sputum or dyspnea 2) increased peripheral blood leukocyte count (≥11,000/mm³) 3) Fever (≥38.3ºC) 4) purulent sputum 5) new or progressive infiltrate on chest radiography. For the diagnosis of pneumonia and its causative agents, sputum smear and culture, blood culture and BAL fluid studies were performed. BAL fluid studies included differential count of white blood cell, BAL fluid smear and culture, detection of elastin fibers and presence of intracellular organisms (ICO). Quantitative culture of BAL fluid was considered positive if colony forming units was more than 1.0×10(4)/ml. Positive criteria for ICO was presence of microorganism in more than five per 100 of phagocytes, Result: Recruited were 22 males and 6 females. The mean age was 57.5±13.5 years (range 25-84), Of 30 episodes underwent BAL fluid studies, 19 cases were diagnosed to be bacterial pneumonia, S. aureus (7 cases) was the most common causative agent and was followed by P. aeruginosa (4), E. cloacae (2), A baumanii (1), H. influenzae (1) and a-hemolytic Streptococcus (1). Sensitivity of quantitative culture of BAL fluid for the diagnosis of bacterial pneumonia was 68.4% and its specificity was 63.6%. Elastin fibers were detected in 5 cases (31%) and ICO over 5% in 3 cases (15.7%). When criteria of quantitative culture of BAL fluid, detection of ICO and elastin fibers were applied together, diagnostic rate of pneumonia was 84.2% (16/19). Conclusion: Quantitative culture of BAL fluid was sensive and specific compared to sputum and b1ood culture for the diagnosis of bacterial pneumonia, It was suggested that detection of ICO and elastic fibers in BAL fluid could raise the diagnostic rate of bacterial pneumonia.

        조각자(주엽) 나무의 생리활성물질 Gleditschia이 비육돈육의 이화학적 성상에 미치는 영향

        The effects of dietary Gleditschia on fatty acid composition, lipid oxidation, and pork quality were investigated. Pigs (n=40) were fed a diet containing 0, 0.1, 0.2, 0.4, and 1% Gleditschia for 14 weeks and slaughtered at 110 kg average. The longissimus throracis et lumborum muscle was collected at 24 hr postmortem. Pork loin chops (3 cm thick) were packaged aerobically and stored at $4^{\circ}C$ for 7 days. Samples were analyzed for fatty acid composition, ultimate pH, thiobarbituric acid-reactive substance (TBARS), color ($L^*$, $a^*$, $b^*$), drip loss and water-holding capacity. There was no significant difference of moisture and crude fat percent in between treatment and control group, and the color stability of pork loin better in all treat groups was more improved during cold storage. The change of pH was significantly increased (p<0.05) in 0.2, 0.4, 1% treatment groups compare to the control group. Water-holding capacity of pig loins was significantly higher (p<0.05) in all treat groups than in control group; 0.2% treat group was the highest in the water holding capacity followed by 0.4, 1, and 0.1% treat groups. Less drip loss of pig loin was observed with samples from Gleditschia-fed pigs except 1% treat group. Unsaturated fatty acid were tend to be decreased and saturated fatty acid were tend to be increased in Gleditschia-treated group. Thiobarbituric acid-reactive substance value of control was significantly higher than that of the Gleditschia-fed group (p<0.05).

        Controlled infiltration profile of SiC coating layer on graphite by Si vapor deposition reaction

        SiC-coated graphite was successfully fabricated at different temperatures (1300-1600 oC) through a silicon vapor depositionreaction (Si-VDR) process. Si powder was used for the Si source of the SiC coating layers. When Si powder was evaporatedat high temperature near the melting point of bulk Si, Si gases are moved and changed into Si liquid at the surface of thegraphite. The high-temperature process facilitated the formation of SiC coating layers on the graphite. The microstructural,mechanical, and thermal oxidation properties of the coated graphite were investigated.
