        Ginsenosides Promote Proliferation of Cultured Ovarian Germ Cells Involving Protein Kinase C-mediated System in Embryonic Chickens

        The effect of ginsenosides (GS) on germ cell proliferation was evaluated with a chicken ovarian germ-somatic cell coculture model and the mechanism involving protein kinase C (PKC) pathway was investigated. Ovarian cells were cultured in serum-free McCoy's 5A medium and challenged with GS alone or in combinations with PKC activator (phorbol 12-myristate 13-acetate, PMA) or inhibitor ($H_7$) for 48 h. The number of germ cells was counted and the proliferating cells were identified by immunocytochemistry of proliferating cell nuclear antigen (PCNA). Results showed that GS significantly increased germ cell proliferation and this stimulating effect was further increased by PMA, but inhibited by H7, in a dose-dependent manner. Moreover, GS-elevated PCNA expression and the PCNA -labeling index of germ cells displayed similar changes with the increased numbers of germ cells. These results indicated that GS stimulated proliferation of ovarian germ cells with involvement of the PKC-mediated system.

        Effects of Daidzein on mRNA Expression of Gonadotropin Receptors and P450 Aromatase in Ovarian Follicles of White Silky Fowls

        Effects of daidzein on expression of mRNAs of gonadotropin receptors (FSHR, LHR) and P450 aromatase (P450arom) were evaluated in ovarian follicles of white silky fowls. The hens were 13 months old in the post-peak period of egg laying and were randomly allocated as control and daidzein-treated groups, with daidzein supplemented to the basal diet at 10 mg/kg for 7 consecutive weeks. The mRNA expression of related genes was measured by semi-quantitative RT-PCR in the granulosa layers of the preovulatory follicle (PRF: F1, F2 ...) and follicular layers of the small yellow follicle (SYF), large white follicle (LWF) and atretic follicle (ATF). Results showed that daidzein supplementation significantly increased the number of SYF and LWF (p<0.05). The relative abundance of the FSHR mRNA decreased in the granulosa layers from F3 to F1, but LHR mRNA displayed opposite developmental changes. P450arom mRNA was highest in the SYF, but was very low in the granulosa layers after follicles finished selection. Treatment with daidzein resulted in increased mRNA expression of FSHR in F3 granulosa layer, LHR in granulosa layers of F3 to F1 and P450arom in LWF (p<0.05). These results indicated that dietary supplementation of daidzein up-regulated mRNA expression of gonadotropin receptors and P450arom to improve the development of preovulatory follicles in white silky fowls after the peak-laying period.

        Effects of dietary protein level on growth performance and nitrogen excretion of dairy heifers

        Objective: Protein supplementation is costly and can result in excess nitrogen (N) excretion. The objective of this study was to evaluate the effects of feeding different levels of dietary protein on average daily gain, body size, rumen fermentation, and nitrogen excretion of 8 to 10 month-old Holstein heifers. Methods: Thirty-six Holstein heifers were divided into 12 blocks according to age (273±6.2 d) and were randomly assigned to diets containing a low (10.2% dry matter [DM]), medium (11.9% DM), or high (13.5% DM) level of dietary crude protein (CP). All diets contained approximately 70% roughage and 30% concentrate with similar dietary metabolizable energy (ME) content (2.47 Mcal/kg). Results: Dry matter intake did not differ among the treatments, and average daily gain increased with the increasing dietary protein, 0.79, 0.95, 0.97 kg/d for low, medium, and high group, respectively. Body height increased linearly with increasing dietary CP but no other significant differences in body dimensions were found among the treatments. The increased ratios of dietary CP improved the rate of rear teat length growth remarkably (p<0.05). There was no difference in rumen pH or ruminal major volatile fatty acid (acetate, propionate, and butyrate) concentration among the 3 diets, but rumen ammonia-N concentration increased with the higher dietary CP (p<0.05). Increasing N intake led to increased total N excretion; urinary N excretion was significantly increased (p<0.05) but fecal N excretion was similar among the treatments. Conclusion: These data suggest that the diet containing 11.9% CP (ME 2.47 Mcal/kg) could meet the maintenance and growth requirements of 9 to 11 month-old Holstein heifers gaining approximately 0.9 kg/d.

        Advanced Stability Analysis of the Tunnels in Jointed Rock Mass Based on TSP and DEM

        The discrete element method (DEM) can analyze the large deformation and large displacement of rock mass effectively, and it is widely used in underground engineering, slope engineering and other fields. However, due to the low accuracy of rock mass structural surface information acquisition, the application of discrete element method in the analysis of jointed rock mass stability is still deviated. In this paper, combined with the advantages of the tunnel seismic prediction (TSP) in obtaining discontinuous geological interface information and the discrete element method in the calculation and analysis of jointed rock mass stability, this paper proposes an advanced analysis method for jointed rock mass stability based on TSP and DEM. Compared to the traditional methods, the analysis results of the jointed rock mass stability are more reliable. Firstly, relying on the advanced detection system – Tunnel Seismic Prediction 203Plus, the unstructured rock mass structure information of the tunnel is obtained, and the spatial attitude of the discontinuous geological interface is further determined. Secondly, based on the Fish programming language, the non-continuous geological interface information can be expressed in the discrete unit software – 3D Distinct Element Code (3DEC). In this way, the excavation calculation model of the tunnels in jointed rock mass can be constructed. Finally, based on the DEM, the excavation of the tunnels in jointed rock mass can be simulated, analyze the stability of surrounding rock during the tunnel excavation process, and realize the stability analysis of surrounding rock stability of jointed rock mass. Based on the Huangjiazhuang Tunnel Project, this paper uses the above method to carry out on-site application. The results show that the location of the dangerous block is predicted to be consistent with the actual exposure of the tunnel surrounding rock based on TSP and DEM, which verify the accuracy and feasibility of this method, and the research results have practical guiding significance for the safe construction of the tunnels in jointed rock mass.

        Separation of valuable metals from mixed cathode materials of spent lithium-ion batteries by single-stage extraction

        With the intensity of resource scarcities and environmental problems, the disposal and recovery of spent lithium-ion batteries, especially recovery of valuable metals, becomes vital. In this work, a method of co-extracting nickel, cobalt, manganese and being separated from lithium by single-stage solvent extraction is proposed. The extraction and separation process of D2EHPA was studied. The effects of extractant concentration, saponification percentage, extraction time and O:A on the extraction efficiency of D2EHPA were systematically studied. Nearly 100% manganese, 94% cobalt and about 90% nickel were co-extracted and separated from lithium using D2EHPA in kerosene by single-stage extraction. The maximum value of separation factors (Ni/Li, Co/Li and Mn/Li) was 13.03, 23.42 and 1904.24. The mathematical model of extraction of four ions was developed by combination of Levenberg-Marquardt method and Universal Global Optimization method. The proposed extraction model accurately fits the experimental data and helps to predict the extraction efficiency of each metal under the corresponding conditions.

        Upregulation of Fas in epithelial ovarian cancer reverses the development of resistance to Cisplatin

        This study was to investigate the role of Fas in the developmentof Cisplatin-resistant ovarian cancer. On the cellularlevel, Fas expression was significantly reduced in Cisplatinresistant A2780 (A2780/CP) cells compared with A2780 cells. Fas silence with siRNA would promote tumor cell linesproliferation, facilitate tumor cell cycle transition of G1/S,prevent cell apoptosis, and promote cell migration. Expressionof drug resistance gene was negatively correlated to Fas. Innude mice metastasis model of human ovarian carcinoma bysubcutaneous transplantation, after Ad-Fas injected intratumorly,we found that upregulation of Fas could inhibit transplantationtumor tissue growth and reduce the expression of drugresistance gene. Our results indicated that upregulation of Fasin epithelial ovarian cancer reversed the development ofresistance to Cisplatin. In conclusion, our findings suggestedthat Fas might act as a promising therapeutic target for improvementof the sensibility to Cisplatin in ovarian cancer.

        Effects of dandelion (Taraxacum sp.,) supplements on lactation performance, antioxidative activity, and plasma metabolome in primiparous dairy cows

        Objective: This study evaluated the effects of dandelion supplements on lactation performance, circulating antioxidative activity and plasma metabolomics in primiparous dairy cows. Methods: A total of 60 mid-lactation dairy cows (milk yield = 34.29±0.34 kg/d; days in milk = 151.72±2.36 days) were divided into 4 treatment groups randomly, comprising the addition of dandelion at 0, 100, 200, 400 g/d per head. The experiment lasted for 8 weeks with an extra 10 days’ pre-feeding period. Milk and blood samples were collected, and plasma samples were selected to perform metabolomics analysis. Results: Supplementing 200 g/d of dandelion increased the yield of milk and lactose (p≤ 0.05). The milk somatic cell counts (p≤0.05) were lower in all dandelion groups than those in the control group. The activity of glutathione peroxidase (p≤0.05) and superoxide dismutase (p≤0.05) were increased and plasma malondialdehyde (p = 0.01) was decreased when cows were fed 200 g/d dandelion. Plasma metabolomics analysis showed that 23 hub differential metabolites were identified in the 200 g/d dandelion group. These metabolites such as ribose, glutamic acid, valine, and phenylalanine were enriched in D-glutamine and D-glutamate metabolism (p = 0.06, impact value = 1), phenylalanine, tyrosine, and tryptophan biosynthesis (p = 0.05, impact value = 0.5), and starch and sucrose metabolism (p = 0.21, impact value = 0.13). Moreover, correlation analysis showed that circulating ribose, mannose, and glutamic acid were positively related to milk yield. Conclusion: Dandelion supplementation could improve lactation performance and elevate the plasma carbohydrate and amino acids metabolism and antioxidative activity. Supplementation of 200 g/d dandelion is recommended for lactating dairy cows.

        Genome-wide Identification and Expression Analyses of RPP13-like Genes in Barley

        Plants have evolved a series of mechanisms to resist pathogens infection. The nucleotide-binding site and leucine-rich repeat (NBS-LRR) family contains the largest number of plant disease resistance genes in plants. Recognition of Peronospora Parasitica 13-like (RPP13-like) genes belong to this superfamily and play important roles in the resistance of various plant diseases including the downy mildew caused by Peronospora parasitica. In this study, 21 RPP13-like genes were identified in barley via bioinformatics. These genes all contained CC, NB-ARC and LRR domains. The physical and chemical properties, chromosome locations, gene structures, protein motifs, 3D protein structures, and microarray based expression dynamics of these genes, as well as their phylogenetic relationship with other plant species were analyzed. Non-expression of MLOC_19262.1 was detected without pathogen infection. When barley was inoculated with the powdery mildew pathogenic fungus, the expression of MLOC_ 19262.1 reached a very high level, suggesting that this gene is an important and promising candidate resistance gene for further study. The two RPP13-like genes, MLOC_57007.2 and MLOC_5059.1 may be involved in barley regular or abiotic stress induced physiological metabolism in specific tissues or at specific developmental stages; furthermore, these functions may be associated with specific domains. These findings provided evidence for the functional diversity of plant pathogen resistance genes and will be helpful for the future characterization of the PRR 13-like gene subfamily.

        The Mechanism of Rockbolt in the Jointed Rock Under Uniaxial Tension

        In this research, the rockbolt mechanism of a jointed rock mass under uniaxial tension was systematically revealed with a laboratory test and a numerical simulation. It was found that the rockbolt rock mass experienced five stages under uniaxial tension, the densification stage, elastic stage, plastic deformation stage, progressive debonding stage, and complete debonding stage. The stress-strain curve and ultimate tensile strength of a rockbolt rock mass were analyzed by taking the rockbolt spacing and rockbolt angle as variables. It was found that the improvement effect of the reduction of the rockbolt spacing on the ultimate tensile strength was limited. When the rockbolt spacing was reduced to a certain limit, the stress concentration area between adjacent rockbolts was connected and destroyed, resulting in the increase of the rockbolt rock strength becoming smaller, and even having a downward trend. The increase of the rockbolt angle led to the change of the stress mode and failure mode of the whole structure, and the ultimate tensile strength first increased and then decreased. The optimal rockbolt angle was between 60° and 70°. It is worth noting that there was an obvious mechanical occlusion between the thread on the rockbolt surface and the rock mass, resulting in the multi-stage step-down characteristic of the stress-strain curve in the complete debonding stage. The results of this study can provide a reference for the design and construction of similar projects.
