      • Detection of Individuals Restoring Fertility by DNA Fragment Converted into STS (sequence tagged site) in Red pepper

        Co-segregation of male fertility with DNA markers selected by RAPD analysis as being potentially linked to the restorer gene (Rf) for Cytoplasmic male sterility (CMS) was analyzed using segregating F2 population. One RAPD marker directly linked to the Rf locus was identified. Amplification of OPT-02/570 using the STS primers generated a monomorphic band of each fertile plants randomly selected F2 progenies. From these results, this specific marker would be strongly linked to be restoring gene. The use of STS marker is effective in overcoming the reliability of the RAPD phenotype and improving their utility for MAS, co-dominant STS markers are especially very useful.

      • 유동층형 생물반응기를 이용한 구연산 생산

        유동층형 생물반응기를 이용하여 Aspergillus niger에 의한 구연산 생산에 관해 연구하였다. 실험결과 균체의 형상은 기질의 물리적 특성 특히 발효액의 점도에 영향을 미쳐 최종적으로는 구연산의 생산 효율에 영향을 미치는 중요한 인자의 하나인을 알 수 있었다. 균체의 크기를 비롯한 형상은 배양기간중 발효액의 교반속도를 변화시켜 조절하였다. 또 질소원이 균체의 성장과 구연산의 생산에 중요한 역할을 하고 있음을 확인 할 수 있었으며, 최적의 질소농도가 존재함을 관찰하였다. 구연산의 생산효율을 높이기 위한 시도로 구연산 발효액으로 부터 균체를 분리하여 다시 이용하는 2단계에 걸친 유가식 배양을 실시한 결과 기존의 회분식 배양에서보다 높은 생성효율을 얻을 수 있었다. A Study on citric acid production by Aspergillus niger was carried out in a fluidized bed bioreactor. Morphology is one od the important factors affecting the physical properties of the medium, especially viscosity, and consequently on the citric acid productivity. During the cultivation, the pellet size was controlled by changing the agitation speed. It was also observed that nitrogen source was found to be important on the cell growth and citric acid production and had an optimum concentration. 2-stage fed batch system was examined and it showed higher productivity than the conventional batch system. The cells after separation from the broth containing citric acid was reused for the next batch.

        Background : Mucins possess the unique function of protecting and lubricating the epithelial surface and other important functions such as call growth, direct implication in the fetal development, the epithelial renewal and differentiation, the epithelial integrity, carcinogenesis, immune regulation, cellular adhesion and metastasis. Purpose : This study was done to provide the significance of alteration of MUC1 and MUC2 expression in colorectal adenocarcinoma, A series of 131 colorectal adenocarcinomas including 11 mucinous carcinomas were screened immunohistochemically for their expression of MUCI and MUC2, Materials and mehtods : Of 131 carcinomas, 76 (58,5%) were MUCI positive and 91 (68, 9%) were MUC2 positive, In normal colonic goblet cells, MUCl was not expressed but MUC2 was expressed in cytoplasm, Conclusion There were up-regulation of MUCI and down-regulation of MUC2 in colorectal carcinomas, The frequency of MUC2 positivity according to differentiation was statistically reliable. (p=0.0001)

      • 화학공학 전공 학생의 취업을 위한 복합 STC 운영

        우리 영동대학교에서 1998년 이후 지금까지 진행하여 온 특별 주제 학습과정인 STC(Special Topics COurse)프로그램을 기존의 운영틀에서 탈피하여 한층 더 발전시킨 복합STC로 활용하고자 하였으며, 특히 이를 졸업생의 취업 및 진학의 방법으로 최대한 활용하고자 계획·운영하였다. 그 결과 취업 및 진학과 같은 진로 확립은 물론 전공지식의 확대, 자격시험 대비능력의 향상, 학생과 교수간의 유대강화 등과 같은 괄목할 만한 성과를 얻을 수 있었으며, 교수들의 산업체 방문을 통한 산학협동의 계기 마련에도 커다란 성과를 얻었다. We have upgraded the STC(Special Topics Course) program, which has been carried out in our university since 1995, to more structured program so that we could train our chemical engineering students better. This program was geared to help students find better jobs and enter well-recognized graduate schools. Consequently, we had most students successful in job searches as well as entrance exams for graduate school; besides, those who actively participated in our STC program were able to attain better professional knowledge and acquire various professional engineers certificates. On top of that, this program allowed us to build up closer relationships with our students. We also made university-industry cooperation possible through our visits to industries nearby.

      • 경연 대회를 통한 초ㆍ중고생의 창의력 신장 효과에 관한 연구

        미래 사회를 대비하기 위한 개인의 개성을 존중과 창의적인 능력을 지닌 인간을 양성을 목적으로 충청남도 지역의 초 ㆍ 중고등학생들을 대상으로 한 창의력 경연대회가 공주대학교 과학교육연구소에서 개최되었다. 충청남도 초 ㆍ중 ㆍ 고등학생들을 대상으로 실시한 창의력 경연대회는 창조적 아이디어를 내어 스스로 문제를 해결해 나갈 수 있는 교육 프로그램을 제공하는 좋은 기회였음이 확인되었으나, 일부의 팀에서만 참신하고 재미있는 아이디어들이 제안되었다. 여학생의 참가자 수가 상당히 증가하였으며, 대체적으로 입상권의 학생들의 상당 부분이 여학생이라는 것이 특이할만하다. 학년이 올라감에 따라서 창의적인 아이디어를 내는 양이나 질에 있어서 뒤떨어지는 것으로 분석되었으며, 토론 학습의 적응력이 미흡하고 발표력 및 청취력에 있어서도 떨어지는 것으로 나타났다. 따라서 중 ㆍ고등학교의 학교교육에서도 학생들에게 프로젝트형 탐구학습 프로그램의 적용은 물론 토론식 수습을 통한 학생들의 사고의 전환의 기회를 제공해야 할 것으로 사료된다. The creativity competition was held to foster the creativity of elementary and secondary school students in the Institute of Science Education, Kongju national University. It was found that the creativity competition effected on the improvement of inventing creative ideas and problem solving activities. But, only some competitors participated in semifinal and final contests, presented original and interesting ideas. More female students participated in competition than last year. More female participants were awarded a prize than males. Totally, participants were defective in discussion and communication, and presentation and listening ideas. Increasing grades correlated inversely with creativity and originality. There is need of inquiry project teaming programs and presenting opportunities of conversion of thinking by discussing instructions in school

        21세기의 정보화 산업사회와 지식기반 사회에서 우리의 아이들이 자신의 목소리로 세계 속에서 살아가게 하기 위해서는 다른 사람과 다른 아이디어를 낼 수 있도록 교육의 틀을 바꾸지 않으면 안된다. 다라서 21세기의 미래 사회를 살아갈 학생들을 가르치는 교육의 현장 역시 창의적인 인간을 길러낼 수 있도록 변해야 한다. 즉, 미래 사회를 대비하기 위해서는 각 개인의 개성을 존중하는 분위기에서 창의적인 능력을 지닌 인간을 양성해야 한다는 것을 의미한다. 따라서, 학교 안에서의 교육뿐만이 아니라 학교 밖에서도 다양한 학습 경험을 통해서 교육 과정이 추구하는 인간상을 구현할 수 있도록 활동할 수 있는 기회가 학생들에게 제공되어야 한다. 창의력을 발휘하여 문제를 해결하는 능력을 기르기 위해서는 개별적이 아닌 여러 명의 동료가 주어진 과제를 해결할 수 있는 기회를 갖고 짧은 시간이 아닌 장시간 동안 고민하고 그 해결책을 찾는 활동을 가질 기회가 필요하다. In order to educate our students with their special talents for the informational and knowledge based age expected in the 21st century, we must improve our educational programs such that students are able to create their own fresh ideas. Hence the present educational environment should be changed such that they are willing to respect others' characters and personality with their own original ideas. Therefore, we must make them have the opportunity that they can acquire many experiences through the school education and the social education program, so that they may grow up to the person with the humanity and creativity. In this study we have suggested new educational program where students engage themselves in long term research projects with collaboration of the team and creativity of individuals.

        Effects of Ethanol on Na<SUP>⁢</SUP>-dependent Solute Uptake in Rabbit Renal Brush-Border Membrane Vesicles

        <P> This study was undertaken to examine the effect of ethanol on Na<SUP>⁢</SUP>-dependent transport systems (glucose, phosphate, and dicarboxylate) in renal brush-border membrane vesicles (BBMV). Ethanol inhibited Na<SUP>⁢</SUP>- dependent uptakes of glucose, phosphate, and succinate in a dose-dependent manner, but not the uptakes of Na<SUP>⁢</SUP>-independent. The H<SUP>⁢</SUP>/TEA antiport was reduced by 8% ethanol. Kinetic analysis showed that ethanol caused a decrease in V<SUB>max</SUB> of three transport systems, leaving K<SUB>m</SUB> values unchanged. Ethanol decreased phlorizin binding, which was closely correlated with the decrease in V<SUB>max</SUB> of Na<SUP>⁢</SUP>-glucose uptake. These results indicate that ethanol inhibits Na<SUP>⁢</SUP>-dependent uptakes of glucose, phosphate, and dicaboxylate and that the reduction in V<SUB>max </SUB>of Na<SUP>⁢</SUP>-glucose uptake is caused by a decrease in the number of active carrier proteins in the membrane.

        Development of <sup>64</sup>Cu-NOTA-Trastuzumab for HER2 Targeting: A Radiopharmaceutical with Improved Pharmacokinetics for Human Studies

        <P>The purpose of this study was to develop <SUP>64</SUP>Cu-labeled trastuzumab with improved pharmacokinetics for human epidermal growth factor receptor 2 (HER2). <B>Methods:</B> Trastuzumab was conjugated with SCN-Bn-NOTA and radiolabeled with <SUP>64</SUP>Cu. Serum stability and immunoreactivity of <SUP>64</SUP>Cu-NOTA-trastuzumab were tested. Small-animal PET imaging and biodistribution studies were performed in a HER2-positive breast cancer xenograft model (BT-474). The internal dosimetry for experimental animals was determined using the image-based approach with the Monte Carlo N-particle code. <B>Results:</B> <SUP>64</SUP>Cu-NOTA-trastuzumab was prepared with high radiolabeling yield and radiochemical purity (>98%) and showed high stability in serum and good immunoreactivity. Uptake of <SUP>64</SUP>Cu-NOTA-trastuzumab was highest at 48 h after injection as determined by PET imaging and biodistribution results in BT-474 tumors. The blood radioactivity concentrations of <SUP>64</SUP>Cu-NOTA-trastuzumab decreased biexponentially with time in both mice with and mice without BT-474 tumor xenografts. The calculated absorbed dose of <SUP>64</SUP>Cu-NOTA-trastuzumab was 0.048 mGy/MBq for the heart, 0.079 mGy/MBq for the liver, and 0.047 mGy/MBq for the spleen. <B>Conclusion:</B> <SUP>64</SUP>Cu-NOTA-trastuzumab was effectively targeted to the HER2-expressing tumor in vitro and in vivo, and it exhibited a relatively low absorbed dose due to a short residence time. Therefore, <SUP>64</SUP>Cu-NOTA-trastuzumab could be applied to select the right patients and right timing for HER2 therapy, to monitor the treatment response after HER2-targeted therapy, and to detect distal or metastatic spread.</P>

        Development of a Theranostic Convergence Bioradiopharmaceutical for Immuno-PET Based Radioimmunotherapy of L1CAM in Cholangiocarcinoma Model

        <P><B>Purpose:</B></P><P>Cholangiocarcinoma is a malignancy of bile duct with a poor prognosis. Conventional chemotherapy and radiotherapy are generally ineffective, and surgical resection is the only curative treatment for cholangiocarcinoma. L1-cell adhesion molecule (L1CAM) has been known as a novel prognostic marker and therapeutic target for cholangiocarcinoma. This study aimed to evaluate the feasibility of immuno-PET imaging–based radioimmunotherapy using radiolabeled anti-L1CAM antibody in cholangiocarcinoma xenograft model.</P><P><B>Experimental Design:</B></P><P>We prepared a theranostic convergence bioradiopharmaceutical using chimeric anti-L1CAM antibody (cA10-A3) conjugated with 1,4,7-triazacyclononane-1,4,7-triacetic acid (NOTA) chelator and labeled with <SUP>64</SUP>Cu or <SUP>177</SUP>Lu and evaluated the immuno-PET or SPECT/CT imaging and biodistribution with <SUP>64</SUP>Cu-/<SUP>177</SUP>Lu-cA10-A3 in various cholangiocarcinoma xenograft models. Therapeutic efficacy and response monitoring were performed by <SUP>177</SUP>Lu-cA10-A3 and <SUP>18</SUP>F-FDG-PET, respectively, and immunohistochemistry was done by TUNEL and Ki-67.</P><P><B>Results:</B></P><P>Radiolabeled cA10-A3 antibodies specifically recognized L1CAM <I>in vitro</I>, clearly visualized cholangiocarcinoma tumors in immuno-PET and SPECT/CT imaging, and differentiated the L1CAM expression level in cholangiocarcinoma xenograft models. <SUP>177</SUP>Lu-cA10-A3 (12.95 MBq/100 μg) showed statistically significant reduction in tumor volumes (<I>P</I> < 0.05) and decreased glucose metabolism (<I>P</I> < 0.01). IHC analysis revealed <SUP>177</SUP>Lu-cA10-A3 treatment increased TUNEL-positive and decreased Ki-67-positive cells, compared with saline, cA10-A3, or <SUP>177</SUP>Lu-isotype.</P><P><B>Conclusions:</B></P><P>Anti-L1CAM immuno-PET imaging using <SUP>64</SUP>Cu-cA10-A3 could be translated into the clinic for characterizing the pharmacokinetics and selecting appropriate patients for radioimmunotherapy. Radioimmunotherapy using <SUP>177</SUP>Lu-cA10-A3 may provide survival benefit in L1CAM-expressing cholangiocarcinoma tumor. Theranostic convergence bioradiopharmaceutical strategy would be applied as imaging biomarker-based personalized medicine in L1CAM-expressing patients with cholangiocarcinoma.</P>
