        항생제 다제내성균 Staphylococcus aureus SA2로부터 분리한 테트라사이클린 내성 플라스미드 pKH6의 염기서열

        한국에서 임산균주로부터 분리한 테트라사이클린(Tc) 내성 플라스미드 pKH6의 전체 염기서열을 결정하여 가장 대표적인 Tc 내성 플라스미드인 pT181과 비교하였다. pKH6의 전체 길이는 4439bp로 pT181과 동일하게 나타났으며 염기서열은 7개의 염기에서 차이를 보였다. 전부 다 염기쌍이 치환된 형태로 나타났으며 그 중 3개는 coding 부위에서 일어났다. coding 부위에서 일어난 경우에도 1개는 동일한 아미노산을 coding 하였기 때문에 pT181과 차이가 없었으며 RepC와 Pre 단백질에 있어 각각 1개의 아미노산에서 차이를 나타내었다. The complete nucleotide sequence of pKH6, a tetracycline-resistance (Tc^r) plasmid isolated from multi-drug resistant Staphlococcus aureus SA2, has been determined and compared with that of the staphylococcal Tc^r plasmid pT181. The nucleotide sequences of the two plasmids are in agreement except for 7 nucleotides. All differences are caused by base pair substitutions. Among 6 substitutions, 3 occurred in coding regions. However, only tow base substitutions in coding regions resulted in changes of amino acid sequences in two different ORFs of repC and Pre proteins.

      • 암석 슬러지를 이용한 건자재 개발

        본 연구는 1993년부터 춘천시 신동면 형돌리 산 62번지 내에서 흑운모 화강암 및 편마암 암석을 원자재로 사용하여 건설기초재료인 쇄설골재 및 인공모래를 생산하는 업체로부터 제품 생산 후 발생하는 토양 스러치 케익이 연간 약 10만 입방정도 발생한다는 사실에 기초하여 석산에서 채취된 페잔토(석분)의 화학적, 물리적, 역학적 특성을 조사하여 석분의 이용 가능성을 확인하기 위한 실험으로 단위 시멘트량을 400kg, 잔골재율을 45% 그리고 물-시멘트비를 65%로 고정하고 잔골재 중 석분을ㄹ 0%, 10%, 20%, 30%, 40%, 50% 및 60%로 치환하여 제조된 콘크리트의 기초적 성질을ㄹ 시험하였다. 그 결과 석분의 치환율이 높을 수록 공기량은 증가하고 슬럼프값이 감소하였는데 이는 미세한 입자인 석분을 잔골재의 일부로 치환함으로써 비표면적이 증가하였기 때문인 것으로 사료된다. 압축강도와 휨강도는 모두 석분의 치환율이 증가할수록 압축강도와 휨강도는 다소 감소하는 경향을 나타냈으나 큰 성능차이는 보이지 않는 다는 것을 알 수 있었다. 이상의 실험을 통해 석분을 시멘트 콘크리트에 사용했을 경우 약간의 성능저하는 있지만 잔골재로의 활용이 가능하다는 것을 알 수 있었다. 그러므로 앞으로 건조수축, 균열, 크리이프 특성 및 내구성에 과한 연구가 더욱 심도있게 이루어져야 폐자원인 석분의 이용이 가능하리라 판단된다. This study aims to make cement concrete using crushed stone fines which are produced at Chuncheon city, Hyuldongri and find it's applications. According to the experimental results, when ratio of crushed stone fines was increased, air contents were increased and slump values were decreased. Also, the compressive strength and flexural strength were decreased when ration of crushed stone fines was increased. Although these strengths are decreased, the crushed stone fines can be used to construction materials because the decreased amounts are not so much.

      • 레진접착제를 도포한 상아질에 대한 합착용 시멘트의 전단결합강도

        The purpose of this study was to confirm the formation of hybrid layer and resin tags in dentin tissue and the possibility of bonding between luting cements used for the prosthesis and the resinous surface coated with resin bonding agents to prevent the dentin hypersensitivity after abutment preparation. Some resin bonding agents, which may have the possibility of bonding with polyacrylic acid as a liquid ingredient of polycarboxylate and glass ionomer cements, were selected. All-Bond desensitizer containing NTG-GMA and BPDM, Scotch-Bond Multipurpose plus containing HEMA, and XR-bond containing organophosphate were selected as a coating agent. Dental cements were zinc phosphate, polycarboxylate, and glass ionomer cement. After the exposed dentin surface of premolars was ethced with 10% phosphoric acid and coated with resin bonding agents, the morphology of treated surfaces and the resin tags and hybrid layers on sectioned surfaces were observed by SEM. Shear bond strength between the resin bonding agents and 3 kinds of cements was measured 24 hours after bonding. On the debonded surfaces of the shear bond strength tested specimens, the cement tags and the bonding sites between the resin materials and cements were examined by SEM. Following conclusions were drawn 1. Coating of dentin with resin bonding agents had no effect on the shear bond strength of zinc phosphate cement. 2. Both of polycarboxylate and glass ionomer cements showed the increased shear bond strength by the dentinal coating with Scotch-Bond Multipurpose plus containing HEMA. However, in the case of dentinal coating with some agents containing NTG-GMA and BPDM or organophosphate, polycarboxylate cement exhibited the lowered shear bond strength, and glass ionomer cement showed the unchanged shear bond strength. 3. Complete obstructions of dentinal tubules were observed on the dentin coated with All-Bond desensitizer or XR-bond, but distinct shape of the orifices of dentinal tubules was observed consistently on the dentin coated with Scotch-Bond Multipurpose plus. 4. The hybrid layer was thickest on the dentin coated with Al1'Bond desensitizer, and the length of resin tags was longest on the dentin coated with Scotch-Bond Multipurpose plus. 5. On the debonded specimens which had been bonded with polycarboxylate cement or glass ionomer cement after coating with Scotch-Bond Multipurpose plus, the cement tags and the bonding sites between the resinous surface and the cements could be examined.

        고지방식이가 백서 골격근 및 지방조직의 지방분해에 미치는 영향

        연구배경:지방산화의 증가가 골격근에서 인슐린 저항성을 유발한다는 주장이 꾸준히 제기되어 나왔으나, 최근 제2형 당뇨병 환자나 인슐린저항성 상태에서 골격근의 지방산화능이 저하되어 있다는 보고도 있어 논란이 있는 상태이다. 본 연구에서는 고지방식이를 투여하여 인슐린저항성을 유발한 백서에서 microdialysis 기법을 이용하여 조직내의 지방분해 결과로 간질액내로 방출되는 글리세롤 농도를 골격근 및 지방조직에서 직접 측정하여 지방분해의 변화를 알아보고자 하였다. 방법:정상 Sprague­Dawley 백서를 두 군으로 나누어 4주간 고지방식이 및 저지방식이를 투여하였다. 4주간 기저상태 및 고인슐린 정상혈당 클램프 중의 골격근 및 지방조직에 microdialysis probe를 삽입하고 일정한 농도의 글리세롤을 포함하는 dialysate를 통과시켜 주입액과 유출액의 글리세롤 농도 차이로부터 조직 간질의 글리세롤 농도를 산출하엿다. 결과:기저상태에서 골격근 및 지방조직 간질의 글리세롤 농도는 저지방식이군에 비하여 고지방식이군에서 유의하게 낮았다. 고인슐린 정상혈당 클램프 중에는 골격근 및 지방조직 간질의 글리세롤 농도가 양군에서 모두 억제되었는데, 기저 상태와 마찬가지로 골격근 및 지방조직 모두에서 저지방식이군보다 고지방식이 군에서 더 낮았다. 기저 상태에 대한 고인슐린 클램프중의 글리세롤 방출 억제 정도는 저지방식이군에 비하여 고지방식이군에서 덜 억제되는 경향을 보였는데, 골격근조직에서만 통계적으로 유의하였다. 결론:본 연구 결과는 고지방식이에 의한 인슐린저항성 모델에서 골격근 및 지방조직내 지방분해가 억제되어 있음을 보여주며, 기존의 포도당­지방산 cycle 이론과는 달리 조직내 지방산화의 증가보다는 지방산화능의 감소에 따른 세포내 지방축척이 인슐린저항성과 관련될 가능성을 시사하였다. Background : It has been hypothesized that increased fat oxidation reduces glucose utilization in skeletal muscle, and is responsible for the insulin resistance associated with obesity or high-fat feeding. In contrast, there have been reports that fat oxidation capacity was decreased in skeletal muscles from insulin resistant subjects. This study was undertaken to examine whether insulin resistance in high-fat fed rats is associated with increased lipolysis in skeletal muscle and adipose tissue. Methods : Two groups of Sprague-Dawley rats were fed either high-fat or low-fat diets for 4 weeks. Lipolysis in skeletal muscle and adipose tissue was determined by measurement of interstitial 1 glycerol concentrations by a microdialysis method in basal and hyperinsuline mic-euglycemic clamp conditions. Results : In basal state, plasma glycerol levels and interstitial glycerol concentrations of skeletal muscle, and adipose tissue were lower in high-fat fed than in low-fat fed rats. The degree of suppression of glycerol release by the hyperinsuline mia was smaller in the high-fat diet than in the low-fat diet group. However, plasma and interstitial glycerol concentrations during the hyperinsuline mic euglycemic clamps were also lower in the high-fat diet group. Conclusion : Lipolysis was decreased in skeletal muscle and adipose tissue of insulin resistant, high-fat fed rats. These results support the idea that limited fat oxidation capacity resulting in lipid accumulation in tissues, rather than increased fat oxidation perse, is responsible for the insulin resistance associated with high-fat feeding (J Kor Diabetes Asso 24:641~651, 2000).

      • 複合微生物을 利用한 간장開發에 관한 硏究

        韓國 고유의 간장을 開發코자 Aspergillus oryzae, Aspergillus niger 및 Rhizopus delemar와 Bacillus natto 菌으로 製麴하고 kogi의 配合比를 달리하여 간장을 담구고 熟成(7週) 過程 중에 化學成分의 調査와 관능검사를 실시한 結果는 다음과 같다. 1) 간장시료 중의 아미노태 질소함량은 혼합미생물시험구(0.57∼0.79%)가 Asp.oryzae만의 대조구(0.48%)보다 많았고 그중 Asp. oryzae+Rhiz. delemar 시험구(0.79%)가 가장 많았으며 Asp. niger의 혼합은 Rhiz. delemar보다 아미노태 질소의 유리량이 적었다. 2) 혼합미생물처리구(4.37∼7.96%)의 환원당량은 대조구(3.53%)보다 많았으며 그중 Asp. niger+Rhiz. delemar시험구가 제일 많았다. Asp. oryzae에 Rhiz. delemar의 복합은 Asp.niger보다 환원당량이 많았다. 3) 혼합미생물처리구의 총산량(0.38∼0.82%)은 대조구(0.33%)보다 많았고 그중 Asp. oryzae+Rhiz. delemar구(0.81%)와 Asp. oryzae+Rhiz. delemar+Bacillus nato구(0.82%)가 제일 많았으며 Asp. oryzae+Rhiz. delemar구는 Asp. oryzae+Asp. niger구보다 더 生酸量이 많았다. 4) 관능검사의 총점수에서 대조구(25점)보다 많은 시험구는 3구인데 그중 Asp. oryzae+Rhiz. delemar+Bacillus nato구(30점)가 가장 良好하고 다음 Asp. oryzae+Asp. niger, Asp. oryzae+Asp. niger+Rhiz. delemar구(29점)이다. In order to develop better soy-sauce, the research made different kinds of Koji, using microorganism such as Aspergillus oryzae, Aspergillus niger, Rhizopus delemar. and Bacillus natto each. Several kinds of soy-sauce, in which Kojis were mixed by different rate, were compared each other for their changes of chemical components during fermentation period (Seven weeks) and their quality examined by panel test. The results were summarized as follows; 1. The soy-sauce using the mixed koji, Asp. oryzae+Rhiz, delemar., showed the the highest NH₂-N content by 0.79% and the soy-sauce using the Koji made by composite microorganism showed higher NH₂-N content (0.57∼0.79%) than that of standard soy-sauce using Asp. oryzae Koji (0.48%). The content of NH₂-N in soy-sauce of Asp. oryzae+Asp. niger Koji was less than in soy-sauce of Asp. oryzae+Rhiz. delemar Koji. 2. The content of reducing sugar in soy-sauce was highest when the mixed Koji, Asp. niger+Rhiz, delemar was used, the nest was the composite microorganism Koji by 4.37∼7.96%, and the lowest was the standard commercial Koji by 3.53%. The mixed Koji using Asp. oryzae and Rhiz. delemar showed higher reducing sugar content than that of Asp. oryzae+Asp. niger Koji. 3. Total acid content in soy-sauce was highest when the mixed Kojis, Asp. oryzae+Rhiz. delemar and Asp. oryzae+Rhiz. delemar+Baillus natto., both by 0.81% and 0.82%, respectively. Total acid content in soy-sauce using composite microorganism Koji showed higher(0.38∼0.82%)than that of standard Koji(0.33%), and Asp. oryzae+Rhiz. delemar showed higher value than Asp. oryzae.+Asp.niger in total acidcontent. 4. Panel test score was highest for the soy-sauce used the mixed Koji, Asp. oryzae+Rhiz. delemar+Bacillus natto and the two kinds of soy-sauce used the mixed Koji, Asp. oryzae+Asp. niger and Asp. oryzae+Asp. niger+Rhiz, delemar., were judged as better than standard soy-sauce.

        프로스타글란딘 E₁에칠에스테르의 외용 리오겔 제제 설계

        External lyogels containing prostaglandin E₁ethyl ester(PGE₁-EE), a produrg of prostaglandin E₁(PGE₁) as a therapeutic agent for erectile dysfunction, were formulated to overcome the aqueous instability and enhance the percutaneous absorption. Lyogels of PGE₁-EE were prepared with ethanol (EtOH)/proplyene glycol (PG) cosolvent system as a vehicle, cineol as an enhancer, and hydroxypropylcellusose as a gelling agent. In vitro percutaneous absorption studies were performed to determine the rate of PGE₁ absorption through rat or hairless mouse skin. The permeability of PGE₁-EE lyogel with enhancer was 16-fold greater than that of lyogel without enhancer. Cosolvent produced 9-fold increase in percutaneous absorption. Pharmacodynamic effects of lyogels were evaluated in mature male cats in terms of intracavernosal pressure (ICP). Lyogles containing 0.01% of PGE₁-EE showed higher ICP compared to intraurethral preparation of PGE₁(1%) and enhancer-free control lyogel. The shelf-life (t_(10%) of lyogel at refrigerated condition (4℃) was calculated as 928 days, which is 4.2 times longer than that of control hydrogel. As a result, PGE₁-EE was formulated successfully to a lyogel system with a selective enhancer and cosolvent system for the topical delivery of PGE₁.

      • 비정상 질확대경검사 소견을 보이는 환자에서 환상투열요법의 직접적 적용에 관한 연구

        Objective: Large Loop Excision of the Transformation Zone (LLETZ) has been widely used for evaluating cervical dysplasia/cancer. We performed this study to evaluate if colposcopically suspected cervical high-grade dysplasia or cancer could be directly evaluated and treated with LLETZ. Methods: 93 women who were scheduled for colposcopic evaluation because of an abnormal cervical cytology report were included in the study in Sanggye Paik Hospital from Jan to Dec 2001. Mean age of the study population was 38.6±9.0. colposcopy directed punch biopsy was not performed and histologic diagnosis was made by LLETZ. Sensitivity and specificity of colposcopic evaluation was calculated and compared with those of cervical cytology. Results: On histopathological examination by LLETZ, 11(11.8%) patients had invasive cancers including 3(3.2%) adenocarcinomas. The sensitivity and specificity of colposcopy for the detection of cervical dysplasia or cancer was 71.7% and 72.3% respectively, wile those of cytology was 56.5% and 74.5% respectively. Conclusion: LLETZ may be adequately performed for the diagnosis and treatment of cervical dysplasia/cancer based on the results of pap smear and colposcopy without punch biopsy. Cost benefit might be anticipated while cost-effectiveness analysis is necessary in the future.

      • 온실재배와 노지재배한 금산 깻잎의 품질 특성과 향기성분

        깻잎을 이용한 건강 식품을 개발하고자 먼저 깻잎의 주산지인 금산에서 5월 온실재배한 깻잎과 8월 노지재배한 깻잎들의 품질특성과 향기성분을 조사하였다. 시료깻잎 모두 4.0%의 조단백질과 0.8%의 조지방을 함유하고 있었고, 플라보노이드의 함량은 온실재배 깻잎 25.2%, 노지재배 깻잎 26.5%로 시료깻잎간에 큰 차이는 나타나지 않았으며 조사포닌 함량도 2,7%와 2,8%로 비슷하였다. 시료깻잎의 효소 활성은 오직 알칼리성 단백질 분해효소 활성만이 8월 노지재배 깻잎에서만 물 추출물 7.1 unit, 에탄올 추출불 11.8 unit을 보였고 기타 효소 활성은 없었다. 8월 노지재배 깻잎의 조직감 특성으로 먼저 견고성과 탄력성 등은 잎의 상층부와 중층부 보다 잎받침을 포함하는 하층부에서 높았고 응집성은 하층부가 약간 약하였으며 5월 온실재배 깻잎은 씹힘성이 다서 낮았을 뿐 노지재배 깻잎과 비슷하였다. 5월 온실재배와 8월 노지재배 깻잎에는 1-octen-3-ol, linalool, β-caryorhyllene, α-caryophyllene, α-famesene, perilla ketone, nerolidol, eugenol. α-cadinol 등 14종∼15종의 향기성분을 함유하고 있었고 주요한 향기성분은 perilla ketone 이었다. Quality and flavor compounds of perilla leaves cultivated in greenhouse(May) and field (August) in Geumsan province were investigated and compared. All perilla leaves contained 4.0% crude protein and 0.8% crude lipid. C겨de flavonoid contents of perilla leaves cultivated in greenhouse and field showed 25.2% and 26.5%, respectively and each crude saponin content was cultivated in field. Hardness and chewness of bottom parts of field-perilla leaves were higher than those of top and middle part, whereas the cohesiveness of top parts and middle parts of perilla leaves were higher than that of bottom part. Furthermore, texture properties of greenhouse-perilla leaves were similar with those of field-perilla leaves except chewness. Nine kind of flavor compounds such as 1-octen-3-ol, linalool, β-caryophyllene, α-caryophylene, α-franesene, perilla ketone, nerolidol, eugennl, α-cadinol were identified in greenhouse-perilla and field-perilla leaves, showing that main flavor compound was perilla ketone.

      • 항갑상선제로 치료한 그레이브스병 환자의 관해예측인자

        Background: Most cases of Graves' disease have been treated by antithyroid drugs in Korea. The major drawbacks of antithyroid drugs are the relative high rate of relapse and the inconvinience of long term therapy. Measurement of TBII and TRH stimulation test have been considered as the most reliable tests predicting relapse, but the predictive power is not high enough to use in clinical situation. This study was performed to find good prognostic indicators in Graves' disease patients after the discontinuation of antithyroid drug therapy. Subjects and methods : We restrospectively evaluated 96 patients with Graves' disease who showed normal TRH test at the end of antithyroid drug therapy and were followed for more than one year. Serum T3, T4, T3/T4 ratio, TBII, Anti-TPO Ab and TGAb levels were measured at the time of diagnosis and at the end of therapy. Two to three months after withdrawal of antithyroid medication, serum TSH and free T3 responses to TRH were examined. These parameters were compared between the relapse group and the remission group. Results : Among the 96 patients, 22 patients(22.9%) relapsed(Group Ⅱ) and 74 patients(77.1%) remained in remmision(Group Ⅰ). No significant difference was observed between the relapse and the remission group in clinical parameters, serum T3, T4, TSH, T3/T4 ratio, TBⅡ and Anti-TPO Ab determined before and after treatment. Serum TSH and free T3 levels measured after TRH stimulation were also similar in both groups. Serum TGAb levels determined at the end of treatment were significantly higher in the remission group(p<0.05). Relapse following the discontinuation of therapy occured within 24 months in 72.7% of the relapsed cases. After 24 months, relapse rate was reduced significantly. Conclusion : These results suggest that high serum TGAb levels could be a favorable prognostic indicator for the long term remission of Graves' disease treated with antithyroid drugs, and may suggest that Hashimoto's thyroiditis is combined with Graves' disease in those patients.
