        Expression Profiles of Retinoblastoma Protein and p27Kip1 According to HER2 Amplification Status in Primary Breast Cancer

        Purpose: Human epidermal growth factor receptor-2 (HER-2)/neu amplification affects the cell proliferation through the modulation of multiple G1 cell cycle regulators in breast tumor cells. We performed this study to investigate whether retinoblastoma protein (pRB) and p27Kip1 were differently expressed according to the HER2 amplification status in human breast cancer. Methods: HER2 amplification was assayed by fluorescence in situ hybridization and the expression of cell cycle regulators were assayed by immunohistochemistry on 153 consecutive invasive breast cancers. The proliferative activity of breast cancer was analyzed according to the HER2 amplification and cell cycle protein expression status. Results: HER2 amplification was observed in 39 (25.5%) of 153 breast cancers. In the HER2 amplified breast cancers, the pRB expression was significantly increased (p=0.011) whereas there was no significant relationship between HER2 amplification and p27Kip1 expression. There was an inverse correlation between pRB expression and Ki-67 labeling index in the HER2 amplified breast cancers (p=0.036). In contrast, Ki67 labeling index was significantly decreased as p27Kip1 expression increased in HER2 non-amplified breast cancers (p=0.028). In HER2 non-amplified breast cancers, we could not observe any association between the pRB expression and Ki67 labeling index. Conclusion: The proliferation of the breast cancers was associated with pRB expression in HER2 amplified tumors whereas it was associated with p27Kip1 expression in HER2 non-amplified tumors. The results of the current study indicate that the cell proliferative activity of the breast cancer is under different growth signal pathways according to HER2 amplification status.

      • Everolimus-Eluting Stent Implantation for Unprotected Left Main Coronary Artery Stenosis

        Objectives: This study sought to evaluate the safety and efficacy of second-generation drug-eluting stents (DES) for patients with unprotected left main coronary artery (ULMCA) stenosis. Background: The clinical benefit of second-generation DES for ULMCA stenosis has not been determined. Methods: The authors assessed 334 consecutive patients who received everolimus-eluting stents (EES) for ULMCA stenosis between 2009 and 2010. The 18-month incidence rates of major adverse cardiac or cerebrovascular events (MACCE), including death, myocardial infarction (MI), stroke, or ischemia-driven target vessel revascularization (TVR), were compared with those of a randomized study comparing patients who received sirolimus-eluting stents (SES) (n = 327) or coronary artery bypass grafts (CABG) (n = 272). Results: EES (8.9%) showed a comparable incidence of MACCE as SES (10.8%; adjusted hazard ratio [aHR] of EES: 0.84; 95% confidence interval [CI]: 0.51 to 1.40; p = 0.51) and CABG (6.7%, aHR of EES: 1.40; 95% CI: 0.78 to 2.54; p = 0.26). The composite incidence of death, MI, or stroke also did not differ among patients receiving EES (3.3%), SES (3.7%; aHR of EES: 0.63; 95% CI: 0.27 to 1.47; p = 0.29), and CABG (4.8%; aHR of EES: 0.67; 95% CI: 0.29 to 1.54; p = 0.34). However, the incidence of ischemia-driven TVR in the EES group (6.5%) was higher than in the CABG group (2.6%, aHR of EES: 2.77; 95% CI: 1.17 to 6.58; p = 0.02), but comparable to SES (8.2%, aHR of EES: 1.14; 95% CI: 0.64 to 2.06; p = 0.65). Angiographic restenosis rates were similar in the SES and EES groups (13.8% vs. 9.2%, p = 0.16). Conclusions: Second-generation EES had a similar 18-month risk of MACCE for ULMCA stenosis as first-generation SES or CABG.         Hypoallergenic and Physicochemical Properties of the A2 β-Casein Fraction of Goat Milk

        Goat milk has a protein composition similar to that of breast milk and contains abundant nutrients, but its use in functional foods is rather limited in comparison to milk from other sources. The aim of this study was to prepare a goat A2 β-casein fraction with improved digestibility and hypoallergenic properties. We investigated the optimal conditions for the separation of A2 β-casein fraction from goat milk by pH adjustment to pH 4.4 and treating the casein suspension with calcium chloride (0.05 M for 1 h at 25°C). Selective reduction of β- lactoglobulin and αs-casein was confirmed using sodium dodecyl sulphate-polyacrylamide gel electrophoresis and reverse-phase high-performance liquid chromatography. The hypoallergenic property of A2 β-casein fraction was examined by measuring the release of histamine and tumor necrosis factor alpha from HMC-1 human mast cells exposed to different proteins, including A2 β-casein fraction. There was no significant difference in levels of both indicators between A2 β-casein treatment and the control (no protein treatment). The A2 β-casein fraction is abundant in essential amino acids, especially, branched-chain amino acids (leucine, valine, and isoleucine). The physicochemical properties of A2 β-casein fraction, including protein solubility and viscosity, are similar to those of bovine whole casein which is widely used as a protein source in various foods. Therefore, the goat A2 β-casein fraction may be useful as a food material with good digestibility and hypoallergenic properties for infants, the elderly, and people with metabolic disorders.

        Floristic characteristics of vascular plants and first distributional report of Pseudostellaria baekdusanensis M. Kim in Yongneup wetland protected area

        Yongneup wetland protected area, the only high moor in Korea, is a core area to conserve biodiversity. Even though the Yongneup wetland protected area is relatively small, various plant species are distributed in the Yongneup wetland protected area because it includes various habitats showing different environmental gradients. Vascular plants distributed in the Yongneup wetland protected area were identified as a total of 376 taxa that is composed of 73 families, 217 genera, 322 species, 3 subspecies, 44 varieties and 7 forms. For endangered plants designated by the Ministry of Environment, 5 species including Trientalis europaea var. arctica, Lilium dauricum, Halenia corniculata, Lychnis wilfordii and Menyanthes trifoliata were found and 34 taxa were confirmed to be distributed only in the mountainous wetland habitats. Regarding naturalized plants, a total of 11 taxa were distributed, but most of them were distributed in the areas where artificial interference has occurred. And in areas inside the wetlands that are relatively well preserved, 2 species of Bidens frondosa and Erigeron annuus were observed. In this study, the occurrence and distribution of Pseudostellaria baekdusanensis M. Kim, which was recently found in Mt. Baekdu and reported as a new species, were identified in the Yongneup wetland protected area. A wetland is a very vulnerable area to drastic environmental changes and damages to its ecosystem could cause the extinction of rare plant species which are distributed only in the wetlands. Therefore, it is mandatory that current status of the Yongneup wetland protected area is evaluated and actions to prevent rapid environmental changes are taken. Fourteen separate investigations were conducted in 2013 and another four in 2014, to evaluate current status of the Yongneup wetland protected area. These investigations have provided us the basic information for future actions of conservation and restoration.

        대성동 주민의 삶의 질 개선을 위한 공공 커뮤니티 서비스 디자인 -통일맞이 첫마을 대성동 프로젝트 사례를 중심으로-

        본 연구의 목적은 DMZ내 유일한 민간 마을인 대성동 주민들의 삶의 질을 개선함과 동시에 남북 평화통일의 미래비전을 지향하는 아이디어를 창안하는 것이다. 대성동 주민의 삶의 질 개선은 2015년 행자부에서 마을의 노후화된 주택을 개량하는 과제로 시작되었다. 본 과제는 ‘정부3.0 국민디자인단’ 활동을 통해 서비스 디자인 관점에서 단순한 주택개량을 너머선 서비스 아이디어를 발굴하는 것을 목표로 한다. 전체적인 디자인 프로세스는 서비스 디자인 방법론에 기반 하였으며, ‘마을 삶의 질 개선’이란 측면에서 커뮤니티 디자인을 접목하여 진행하였다. 다양한 이해관계자와의 인터뷰를 통해본 연구의 큰 이해관계자를 ‘내부 주민’과 ‘외부 방문객’으로 정한 후 그들의 니즈와 핵심사항을 도출하였다. ‘남과 북’처럼 대립되는 요소를 연결시키고 소통하게 해주는 다리와 같은 개념을 본 연구의 디자인 프레임워크로 설정하였고, 문제해결을 위한 디자인 방향 키워드로 ‘역발상’, ‘조화’ 및 ‘미래지향’을 선정하였다. 이를 토대로 ‘내부 주민’에게는 부족한점을 채워주고 삶의 활력소가 되는 서비스 아이디어를, ‘외부 방문객’에게는 통일된 미래를 미리 만나봄과 동시에 이색적인 체험을 할 수 있는 관광 서비스 모델을 창안하였다. 결론적으로 커뮤니티 디자인이 접목된 서비스 디자인 특유의 생활밀착형 디자인 아이디어 발상을 통해 단순한 주택 개량을 넘어 삶의 질 향상에 기여하는 문화.사회.경제 등 다양한 측면에서의 요소를 발굴하였고, 동시에 분단과 통일이라는 대성동 마을 특유의 역사성과도 조화를 이루어내었다. The purpose of this study is to create service ideas regarding the vision for peaceful reunification in Korea and the improvement of life quality in the special community ‘Daesongdong’ - the only civilian habitation in DMZ. The effort from the Korean government to improve the quality of life in Daesongdong began by the Ministry of the Interior from 2015. Following that effort, this study aimed to expand ideas in a view of service design to generate solutions beyond just renovating current residences. The design process of the study was based on the service design process in mixture with community design. The two core stakeholders selected were ‘residents in Daesongdong’ representing ‘improving the quality of community life’ and ‘visitors to Daesongdong’ representing ‘towards peaceful unification of Korea.’ The design challenge was to harmonize these two rather separated themes. The concept behind the design framework was ‘bridge’ implying the connection of two opponents (South Korea and North Korea, separated past and reunited future, for instances.). Ideas were created with three design keywords in mind - harmony, towards future and reverse-thinking. As a result, a number of ideas in cultural, social, economical aspects beyond simple renovation of residences were produced and supported both themes in harmony.

        Formation and Related-Behavior of Micro-bowl Morphology Consisting of Ionic Palladium(II) Complexes

        Reaction of [(bpy)Pd](PF6)2 (bpy = 2,2'-bipyridine) with racemic bis(isonicotinoyl)-1,1'-bi-2-naphtholate (L) in acetone,and followed by addition of chloroform and solvent evaporation allows to form amorphous micro-bowl morphology consisting of [(bpy)PdL]2(PF6)4 without any template or additive. In contrast, the reaction and recrystallization in acetone for 1 week produce parallel-piped single crystals consisting of [(bpy)3Pd3(μ3-HPO4)2](PF6)2. The formations of micro-bowl and parallel-piped single crystal morphologies appear to be primarily associated with the kinetic and thermodynamic control, respectively. The formation of micro-bowls may be attributed to eruption of organic solvents. Cosolvent effects and chemical properties on the formation of micro-bowl morphology have been observed.
