      • 미술치료 석사과정생의 소아암 환아를 대상으로 한 병원학교 미술치료 실습 체험연구

        본 연구의 목적은 미술치료전공 석사과정생들이 소아암 환아를 대상으로 진행하는 병원학교의 미술치료 실습을 통해 어떤 체험을 하며, 그 체험의 의미와 본질이 무엇인지를 이해하는데 두었다. 이를 위해 Max van Manen의 해석학적 현상학 연구방법을 적용하여, 체험을 더욱 생생하고 깊이 있게 이해하고자 하였다. 2010년 9월부터 2010년 11월까지 심층 면담에 자발적으로 참여 의사를 밝힌 9명의 연구 참여자를 통해 자료를 수집하였으며, 이를 토대로 9개의 본질적 주제와 25개의 하위주제들을 도출하였다. 현상학적 반성과 글쓰기 등의 과정을 통하여, 연구 참여자들의 소아암 환아를 대상으로 한 병원학교 미술치료 실습 체험은 다음과 같은 결론을 내리고 있다. 첫째, 면역력이 약한 소아암 환아를 대상으로 하는 병원학교의 미술치료 실습에서 실습 초기나 실습 종료기 이후에도 환아에게 지나치게 몰입되어 있는 자신을 발견한다. 둘째, 실습현장에서는 미술치료 적 접근이외에, 소아암 환아의 특성을 잘 알고 융통성 있게 대처할 수 있는 철저한 준비와 경험이 요구됨을 자각한다. 셋째, 집단 미술치료는 소아암 환아에게 내면 표출, 또래와의 소통, 공감의 기회 등을 통해 자기개방과 치유로 연결시킨다. 넷째, 소아암 환아를 위해서는 병원학교 의료진, 환아의 가족, 실습생 및 자원 봉사자, 이들 모두의 원활한 소통이 요망되며, 특히 소아암 환아의 심리적 돌봄을 위해 전문적이고 역량있는 미술치료사가 필요하다. Purpose of study will understand that meaning and substance that art therapy major students through practice for childhood cancer patient in hospital school. I applied a hermeneutic phenomenology study way of 'Max van Manen', and the nine people who showed spontaneously in a participation opinion to an interview were selected for this. I collected data through intense interviews until November 2010 from September 2010, and it was derived from 25 subordinate subjects of 9 main subjects. Based on the research result, this study about experience of group art therapy practice with pediatric cancer patients in the hospital school made the following conclusions. First, art therapy practice in hospital schools who it to the immune weak childhood cancer patients in hospital schools and I find oneself who is immersed in excessively to patients after the practice early days or practice end. Second, besides art therapy approaches in practice, I am conscious of required preparation and experience that know about childhood cancer patients characteristic and cope flexibility. Third, group art therapy lets childhood cancer patients connect by own open healing through inside expression, communication with an age, an opportunity of sympathy. Fourth, hospital medical staffs, patient's family, trainees, and volunteers are demanded communication without a hitch for pediatric cancer patients. Especially, need to professional and capable art therapist for pediatric cancer patient's psychological care.

        Application of Response Surface Methodology to Optimize the Extraction of Acetylcholinesterase Inhibitors from Rhodiola sachalinensis

        Response surface methodology (RSM) was utilized to optimize extraction conditions for the acetylcholinesterase inhibitor from Rhodiola sachalinensis. Optimized values for AChE inhibition and extraction yield were 19.41 and 13.35%, respectively, and these were in good agreement with the experimental values from validation, suggesting that RSM is a useful tool.

        Tumor-suppressor Protein p53 Sensitizes Human Colorectal Carcinoma HCT116 Cells to 17α-estradiol-induced Apoptosis via Augmentation of Bak/Bax Activation

        17α-estradiol (17α-E₂)의 에폽토시스 유도활성에 미치는 종양억제단백질 p53의 조절효과를 조사하고자, 17α-E₂에 의해 유도되는 에폽토시스 현상들을 인체 대장암 세포주 유래 클론인 HCT116 (p53+/+) 및 HCT116 (p53-/-) 세포에서 비교하였다. HCT116 (p53+/+) 및 HCT116 (p53-/-) 세포를 17α-E₂ (2.5~10 μM)로 처리하거나 혹은 HCT116 (p53+/+) 및 HCT116 (p53-/-) 세포를 10 μM 17α-E₂로 시간 별로 처리한 결과, HCT116 (p53+/+)에 있어서는 세포독성과 에폽토시스-관련 sub-G₁ peak의 비율은 처리농도와 시간에 의존적으로 나타났다. 그러나 HCT116 (p53-/-) 세포의 경우는 이러한 현상이 미약하게 나타났다. 17α-E₂에 의해 유도되는 비정상적 유사분열방추사 형성, 중기판 염색체 배열의 미완성, 이에 따른 유사분열정지(G₂/M arrest) 등의 현상은 HCT116 (p53+/+) 및 HCT116 (p53-/-) 세포에서 유사한 수준으로 나타났다. 이에 반해, 17α-E₂에 의해 유도되는 Bak과 Bax의 활성화, 미토콘드리아의 막전위 상실(Δψm loss), 그리고 PARP 분해 등의 현상은 HCT116 (p53-/-) 세포에 비해 HCT116 (p53+/+) 세포에서 훨씬 높은 수준으로 확인되었다. 아울러 17α-E₂로 처리된 HCT116 (p53+/+) 세포에서 확인되는 p53 (Ser-15)의 인산화 및 p53 수준의 증가와 일치하여, 세포 내의 p21및 Bax 수준도 현저히 증가하였다. 이때 17α-E2로 처리된 HCT116 (p53-/-) 세포에서는 p21 및 Bax의 발현수준이 매우 낮았다. 한편, 에폽토시스 억제단백질인 Bcl-2 단백질수준은 HCT116 (p53-/-) 세포에 비해 HCT116 (p53+/+) 세포에서 다소 낮았으나, 이러한 Bcl-2 단백질 수준은 17α-E₂ 처리 후에도 크게 변화하지 않는 것으로 나타났다. 이러한 결과들은 17α-E₂ 처리에 의해 유도되는 에폽토시스 유도 경로의 구성원들의 변화, 즉 비정상적 유사분열방추사 형성 및 이에 따른 유사분열정지(G2/M arrest), 뒤이은 Bak 및 Bax의 활성화, 미토콘드리아의 막전위 상실, 그리고 이에 수반되는 caspase cascade 활성화 및 PARP 분해로 진행되는 에폽토시스 현상들 중에서, Bak 및 Bax의 활성화 단계가 종양억제단백질 p53의 에폽토시스 증진 활성에 의해 양성적으로 조절되는 작용 타켓임을 보여준다. The regulatory effect of the tumor-suppressor protein p53 on the apoptogenic activity of 17α-estradiol (17α-E₂) was compared between HCT116 (p53+/+) and HCT116 (p53-/-) cells. When the HCT116 (p53+/+) and HCT116 (p53-/-) cells were treated with 2.5~10 μM 17α-E₂ for 48 h or with 10 μM for various time periods, cytotoxicity and an apoptotic sub-G₁ peak were induced in the HCT116 (p53+/+) cells in a dose- and time-dependent manner. However, the HCT116 (p53-/-) cells were much less sensitive to the apoptotic effect of 17α-E₂. Although 17α-E₂ induced aberrant mitotic spindle organization and incomplete chromosome congregation at the equatorial plate, G₂/M arrest was induced to a similar extent in both cell types. In addition, 17α-E₂-induced activation of Bak and Bax, Δψm loss, and PARP degradation were more dominant in the HCT116 (p53+/+) than in the HCT116 (p53-/-) cells. In accordance with enhancement of p53 phosphorylation (Ser-15) and p53 levels, p21 and Bax levels were elevated in the HCT116 (p53+/+) cells treated with 17α-E₂. The HCT116 (p53-/-) cells exhibited barely or undetectable levels of p21 and Bax, regardless of 17α-E₂ treatment. On the other hand, although the level of Bcl-2 was slightly lower in the HCT116 (p53+/+) than in the HCT116 (p53-/-) cells, it remained relatively constant after the 17α-E₂ treatment. Together, these results show that among the components of the 17α-E₂-induced apoptotic-signaling pathway, which proceeds through mitotic spindle defects causing mitotic arrest, subsequent activation of Bak and Bax and the mitochondria-dependent caspase cascade, leading to PARP degradation, 17α-E₂-induced activation of Bak and Bax is the upstream target of proapoptotic action of p53.

        Effects of Methoxsalen from <i>Poncirus trifoliata</i> on Acetylcholinesterase and Trimethyltin-Induced Learning and Memory Impairment

        <P>Previously, we identified methoxsalen (8-methoxy-2′,3′,6,7-furocoumarin) as the bioactive compound probably responsible for acetylcholinesterase (AchE) inhibition achieved by feeding crude extract of <I>Poncirus trifoliate.</I> To confirm the activity of methoxsalen, Institute of Cancer Research (ICR) mice were fed a control or a methoxsalen-supplemented diet for 4 weeks, and then learning and memory enhancing effects with respect to trimethyltin (TMT)-induced neurotoxicity were evaluated. The brain tissues of ICR mice were dissected after completion of the behavioral tests for biochemical analysis. Methoxsalen effectively reversed TMT-induced memory impairment on both Y-maze and passive avoidance tests. Brain AchE activity was inhibited by the oral consumption of all concentrations of methoxsalen. Moreover, the level of oxidative stress was significantly ameliorated in the groups on methodsalen containing diets. This is the first <I>in vivo</I> study conducted with methoxsalen in the field of AD research, and it indicates that further investigation of methoxsalen is warranted.</P>

        저장 기간 중 생마즙, 오미자 및 오디 첨가 샐러드드레싱의 품질 특성 및 항산화능의 변화

        본 연구는 항산화능이 우수한 오디, 오미자, 생마즙(0, 7, 18%)을 사용하여 개발한 고품질 샐러드드레싱의 저장(5℃, 4주) 기간 중 품질 특성 및 항산화능을 분석하였다 PH 맟 산도는 생마즙의 첨가량이 증가할수록 PH는 높아지고 산도는 낮아졌으며 저장기간에 따라 PH는 다소 증가하고 산도는 감소하였으나 그 변화 폭이 적어 품질 변화가 일어나지 않았다. 점도는 대조군은 83.2 cP로 낮았으나, 생마즙의 첨가량이 증가할수록 유의적으로 값이 증가하였으며(p<0.001) 저장기간에 따라 모든 군에서 점도가 증가하는 경향을 나타냈다. 유화안정성은 생마즙 첨가량과 상관없이 49~50의 수준으로 나타났으며 저장기간이 경과되어도 변화를 나타내지 않았다. 색도는 저장 초기 대조군에 비해 생마즙을 첨가한 실험군의 명도, 적색도, 황색도 수치가 높았으며 저장기간이 길어질수록 모든 군에서 명도, 적색도, 황색도 값이 감소하는 경향을 보였다. DPPH 라디칼소거능 IC(50) 값과 총 페놀함량을 통한 항산화성은 생마즙의 첨가량이 증가할수록 항산화성이 증가하였고, 저장기간에 따른 항산화성 저하는 관찰되지 않았다. 과산화물가는 저장 초기 1.51~1.12 meq/kg 이었으며, 생마즙 첨가량이 증가함에 따라 감소하였고, 4주 경과 후에도 큰 폭으로 증가하지 않았다. 이상의 결과로 본 실험에서 제조한 생마즙 오미자 및 오디 첨가 샐러드드레싱은 항산화성이 우수한 고품질 샐러드드레싱으로 저장(5℃, 4주)중 산패 및 품질에 큰 변화가 없어 저장성이 우수한 것으로 보여 진다. In this study, the quality characteristics and antioxidant activity of mulberry salad dressing prepared with yam juice during storage at 5℃ for four weeks were evaluated. The salad dressing was prepared with a salad base, mulberry, omija, and yam (4:4:6:1 or 4:4:6:3). Fresh yam juice was added to the salad dressing at 0, 7, and 18% levels. After four weeks storage, the pH increased whereas the acidity decreased. As the yam concentration increased, the pH increased to 3.84 and the acidity decreased to 0.14%. The Hunter color L (lightness), b (yellowness), and a (redness) values decreased over the storage period. After four weeks storage, the viscosity increased from 83.2 to 158.5 cp according to the amount of yam juice that was added. The antioxidant activity, such as the DPPH radical scavenging activity, and the total polyphenol content of the salad dressing improved as the yam concentration increased. Moreover, according to the storage time, the peroxide value did not increase. These results show that the mulberry salad dressing to which omija and fresh yam juice were added maintained its freshness with high antioxidative activity during storage.

        The Facile Synthesis of Composition-Tunable Pt—Pd Bimetallic Nanocatalysts and Their Electrocatalytic Properties in Formic Acid

        <P>In this work, Pt-Pd nanoalloys with various compositions were synthesized using a solution plasma process with pure Pt, pure Pd and Pt80Pd20 (wt.%) alloy electrodes. The elemental compositions, structures and morphologies of the Pt-Pd nanoalloys were characterized using an inductively coupled plasma-optical emission spectrometer, energy dispersive X-ray spectroscopy, X-ray diffraction and high resolution transmission electron microscopy. In order to examine the electrochemical activities for formic acid oxidation, cyclic voltammetry was conducted using a potentiostat in a nitrogen-purged 0.5 M H2SO4 and 0.5 M HCOOH electrolyte solution. The compositions of as-synthesized Pt-Pd nanoalloys were measured by following: Pt82Pd18, Pt69Pd31, Pt41Pd59 and Pt29Pd71. From the results of the electrochemical characterization, the Pt69Pd31 nanoalloys showed much higher catalytic activity than the other Pt-Pd nanoalloys. Furthermore, and the CO poisoning was strongly affected by the Pd amounts in the synthesized Pt-Pd nanoalloys.</P>

      • Responsive Surface Methodology Optimizes Extraction Conditions of Industrial by-products, <i>Camellia japonica</i> Seed Cake

        <P><B>Background:</B></P><P>The central nervous system is easily damaged by oxidative stress due to high oxygen consumption and poor defensive capacity. Hence, multiple studies have demonstrated that inhibiting oxidative stress-induced damage, through an antioxidant-rich diet, might be a reasonable approach to prevent neurodegenerative disease.</P><P><B>Objective:</B></P><P>In the present study, response surface methodology was utilized to optimize the extraction for neuro-protective constituents of <I>Camellia japonica</I> byproducts.</P><P><B>Materials and Methods:</B></P><P>Rat pheochromocytoma cells were used to evaluate protective potential of <I>Camellia japonica</I> byproducts.</P><P><B>Results:</B></P><P>Optimum conditions were 33.84 min, 75.24%, and 75.82°C for time, ethanol concentration and temperature. Further, we demonstrated that major organic acid contents were significantly impacted by the extraction conditions, which may explain varying magnitude of protective potential between fractions.</P><P><B>Conclusions:</B></P><P>Given the paucity of information in regards to defatted <I>C. japonica</I> seed cake and their health promoting potential, our results herein provide interesting preliminary data for utilization of this byproduct from oil processing in both academic and industrial applications.</P><P><B>SUMMARY</B></P><P><P>Neuro-protective potential of <I>C. japonica</I> seed cake on cell viability was affected by extraction conditions</P><P>Extraction conditions effectively influenced on active constituents of <I>C. japonica</I> seed cake</P><P>Biological activity of <I>C. japonica</I> seed cake was optimized by the responsive surface methodology.</P></P> >[FIG OMISSION]</BR><P><B>Abbreviations used:</B> GC-MS: Gas chromatography-mass spectrometer, MTT: 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide, PC12 cells: Pheochromocytoma, RSM: Response surface methodology.</P>
