        Multi‑tissue lipotoxicity caused by high‑fat diet feeding is attenuated by the supplementation of Korean red ginseng in mice

        Background Excessive intake of fat, one of the causes of obesity, is associated with low-grade infammation in various susceptible organs and eventually causes tissue toxicity. This study examines the multifaceted suppressive efects of Korean red ginseng extract (KRG) on high-fat diet (HFD)-induced lipotoxicity and infammatory responses in the aorta, liver, and brain. Methods Male C57BL/6 mice were fed HFD with or without KRG for 12 weeks. The improvement efect in KRG on lipotoxicity and infammatory potential was determined in the blood and the aorta, liver, and brain tissues. Results KRG signifcantly inhibited 3-hydroxy-3-methylglutaryl coenzyme A (HMG-CoA) reductase activity by >20% in vitro. KRG supplementation suppressed HFD-associated body weight gain, lipid profle changes, and excessive fat deposition in the liver and increased leptin, insulin, and ALT levels in the blood. Infammatory markers in the aorta, liver, and brain were also signifcantly reduced by KRG treatment. In microvascular endothelial cells, the 15% cyclic stretch-mediated upregulation of ICAM-1 and vascular cell adhesion protein-1 (VCAM-1) expression was signifcantly attenuated in the presence of KRG. Conclusion KRG supplementation attenuates HFD-mediated body weight gain, lipid profle changes, and multi-tissue infammatory responses.

      • 박실지의 대형수생식물상과 그 분포에 관한 연구

        The flora(macro-hydrophyte) Vegetation of Baksil reservoir and surrounding area was investigated from March, 2005 to October, 2005. The Macro-hydrophyte of this area consist of 33 taxa : 3 variety, 30 species, 30 genera and 22 family. The Maro-hydrophyte of this area consist of Emergent Plants : 16 species, 14 genera, 11 family, Submerged plants : 4 species, 4 genera, 3 family, Free-floating Plants : 3 species, 3 genera, 2 family, Floating leaved Plants : 2 species, 2 genera, 2 family, Hygrophytes Plants : 5species, 5 genera, 4 family. The communites of Jeongyang lake : Miscanthus Sacchariflorus community, Zizania latifolia community, Phragmites communis community, Tyhpa orientalis community and Trapa japonica community. The life form spectrum : Emergent Plants 49%, Submerged Plants 12%, Free-floating Plants 9%, Floating leaved plants 6%, Hygrophytes plants 15%.

      • 핵전달 방법에 의한 Aspergillus 속 종간잡종 형성에 관하여

        핵전달 방법에 의해 A. nidulans FGSC 514와 A. niger의 종간잡종을 얻었고 그들 특성을 관찰한 결과는 다음과 같다. 1. A. nidulans FGSC 514의 원형질체 형성은 삼투안정제인 0.6M KCI과 0.5% Novozym 234를 2시간 처리하여 2x10^7 원형질체를 얻었다. 2. A. niger의 원형질체 형성은 삼투안정제로 0.6M KCI, 0.5% Novozym 234, 0.3% β-glucuronidase와 0.4% BSA를 2시간 처리하여 6x10^7 원형질체를 얻었다. 3. A. niger의핵을 추출하여 A. nidulans FGSC 514 원형질체를 형질전환시켜 3.5%의 형질전환률을 얻었다. 4. 형질전환체의 섬유소 분해효소 활성은 CMCase, avicelase와 pectinase에서 대체적으로 증가하였고 β-glucosidase에서는 모균주보다 약간 감소하는 경향을 보였다. 5. 형질전환체의 건조균체량은 모균주에 비해 대체적으로 높게 나타났고 배양 10시간부터 25시간까지 급격한 증가를 보였으나 40시간 후에는 거의 증가하지 않았다. 6. 형질전환체의 DNA 함량은 1.5∼2.23㎍/10^7pts로 모균주에 비해 약 1.4∼2.0배 증가하였다. 7. 세포외 수용성 단백질은 전기영동 양상은 형질전환체들은 유사하나 모균주의 A. nidulans FGSC 514와 A. niger과는 양상이 다른 중간형태를 나타내었다. The properties of interspecific hybrids obtained by nuclear transfer method between A. nidulans FGSC 514 and A. niger wild type were analyzed by enzyme activity, protein determination, dry weight, DNA content, and total solube protein. The protoplast formation from A. niger was 6x10^7 pts with the treatment of 0.6M KCI, osmotic stabilizer, 0.5% novozym 234, 0.3% β-glucuronidase and 0.4% BSA for 2 hrs. When the isolated nuclei of A. niger wild type were transformed the protoplasts of A. nidulans FGSC 514, transformation frequency was 3.5%. It was found that the enzyme activities in degradation of cellulose and polysaccharide were increased in CMCase, avicelas and pectinase but decreased in β-glucosidase. The strains in which enzyme acttivity in general had a tendency of increasing apecific activity and protein content. The dry weight of transformants were increased more than the parent and increased fast from 10 to 25 hrs. The DNA content of transformants were 1.5∼2.23㎍ and 1.42∼2.04 times more than that of both A. nidulans FGSC 514 and A. niger wild type. Electrophoretic patterns of extracellular solube protein were different between parental and transformant strains.

        GC/NPD를 이용한 다성분 잔류농약의 동시분석

        Pesticides were extracted from samples with 70% acetone and methylene chloride in order, and then cleaned up via open-column chromatography apparatus packed with fiorisil, and finally analyzed simultaneously the organophosphorus pesticides using GC/NPD, Ultra-2 and Ultra-1 fused silica capillary columns were used to separate and identify the products. Recovery of most analytes from soybean sample, taken from pesticide residues well, was greater than(80%) for all except(6) analytes. This method can simultaneously determine multiple pesticides with a high degree of accuracy and precision.

      • Retinoic acid에 의해 유발된 흰쥐의 구개열에서 Transforming Growth Factor-beta 1, 2, 3의 발현의 변화

        Objective : Craniofacial malformations are among the most frequent congenital birth defects in humans : cleft palate, that is inadequate fusion of the palatal shelves. Retinoic acid is teratogenic in many species, producing multiple malformations, including cleft palate. Also it plays an important role in embryogenesis, by regulating morphogenesis, cell proliferation, differentiation, and deposition of the extracellular matrix. The effects of retinoic acid which leads to cleft palate vary depending on the stage of development exposed. After exposure of embryonic mice to retinoic acid on gestational day(GD) 10, abnormally small palatal shelves form. And the elevation of palatal shelves is delayed. The growth and differentiation factor, transforming growth factor-beta(TGFβ) family is thought to play important roles in multiple developmental processes, especially, the development of the embryonic palate. The expression of the TGFβ family in embryonic palate suggests a functional role for these molecules. In this study, author has determined the tissue distributions of TGFβ family in murine embryonic palate, and compared them in terms of developmental stage and locations in normal and retinoic acid-induced cleft palate. Methods and Material : Retinoic acid was administered orally at gestational day(GD) 10 to ICR mice. The pregnant mice were sacrifeced on GD 12, 13. 14, 15 to obtain the fetuses and immunohistochemistry was performed. And using the scanning electron microscope, morphology of normal palate and cleft palate was studied. Results : In the retinoic acid-treated fetuses, palatal shelves did not elevate and cleft palate was induced. On GD 13 in the palates of the retinoic acid-treated fetuses, expression of the TGFβ1, 3 decreased dramatically, but that of TGFβ2, did not change. The expression of the TGFβ1, 3 in the palates of the retinoic acid-treated fetuses, increased on GD 14, 15, Significant differences in TGFβ family expression were observed with areas of the palate. Conclusion : These results suggest that retinoic acid inhibits the expression of TGFβ 1, 3, which is critical in the developing palate, and elevation of palatal shelves is delayed and impaired, therefore cleft palate is induced. These data demonstrate a role for retinoic acid and retinoic aicd-induced TGFβ family in the regulation of palatal development and suggest a role for TGFβ in retinoic acid-induced cleft palate.

      • 와룡산의 식물상과 곤충상

        The flora and insect fauna of Mt. Waryong was surveyed. The flora was surveyed from 1998 to 2001 and insect fauna was done from spring to autumn of 2000. The vascular plants collected from this region are 815 taxa: 41 orders, 133 families, 476 genera, 702 species, 1 subspecies, 96 varieties, and 16 forma. In the area, Korean endemic plants were 18 taxa(2.2% among total 815 taxa); cultivated plants were 154 taxa (18.9% among total 815 taxa). Also the naturalized plants were 48 taxa(5.9% among total 815 taxa and 26.7% among the total naturalized plants in Korea): 19 families, 40 genera, and 8 species. The total insect specimens were classified into 6 orders, 53 families, and 239 species. In hemiptera, 11 families and 25 species were classified, and 8 families and 28 species were contained in homoptera. diptera held 6 families and 11 species, hymenoptera had 4 families and 9 species, and coleoptera had 4 families 35 species. And we could classified 20 families and 131 species in lepidoptera.

      • 정상인과 혈액질환 환자에서의 혈소판 평균용적치에 관한 연구

        Molphologic study of platelet have been done by many authors for the past years, they investigated that the heterogeneity of platelet size was shown even in normal person and various hematologic disorder. In the past the platelet size was not utilized in clinical medicine because of its technical difficulties of measuring. Recently, and automated blood cell counter, Coulter counter Model Splus Ⅱ®, was introduced and platelet size could be easily gained, which expressed by mean platelet volume(MPV). Therefore, this study was planned to evaluated the clinical significance of MPV in health person and various hematologic disease. The following results were obtained: 1. In healthy person, MPV was decreased along with increasing the platelet number. 2. In thrombocytopenic patients, MPV was decreased more than normal. 3. In various hematologic disease, MPV was decreased in acute leukemia, marrow failure and aplastic anemia, normal in iron deficiency. anemia, increased in reactive marrow, MPV was varied by platelet number in various hematologic disease. This disclosed that MPV was related with marrow function and production of megakaryocytes. From these results, MPV could be used as a valuable parameter in the evaluation of platelet together with measuring the platelet number. Particular relation-ship between MPV and various hematologic disease were not observed. We should be more study for MPV.
