Mastoparan B (MPB)는 벌독으로부터 정제된 양친매성α-helical 구조를 취하면서 14개의 아미노산 잔기로 구성된 염기성 항균성 펩타이드로서 여러 가지 생물막과 작용한다. 본 연구에서는 우리 나라 연안의 적조 (HABs, harmful algal blooms)를 일으키는 4종의 적조생물 (Alexandrium tamarense, Chattonella catenatum)에 대한 MPB의 살조효과를 조사하였다. MPB의 4종의 적조생물에 대한 살조효과는 31.3㎍/㎖에서 세포의 lysis또는 ecdysis와 같은 형태로 현미경으로 관찰할 수 있었다. 또한 MPB는 C. marina 및 C. polykridorides에 대해서 A. tamarense와 G. catenatum보다 더욱 강한 살조효과를 나타내었다. 이러한 MPB의 HABs에 대한 살조효과연구는 새로운 살조물질을 개발하기 위한 자료가 될 것으로 생각된다. Mastoparan B (MPB), an antimicrobial cationic peptide isolated from the venom of the hornet Vespa basalis, is a basic amphipathic a-helical peptide composed of fourteen amino acid residues. In this study, we have investigated the algicidal effect of MPB against hatmful algae blooms (HABs) casative Alexandrium tamarense, Chattonella marina, Cochlodinium polykrikoides and Gymnodinium catenatum. The algicidal effect of MPB showed in the concentration of 31.3 ㎍/㎖ to 500 ㎍/㎖ against 4 HAB species and observed cell lysis or cell ecdysis by microscopy. MPB reacted more sensitive to C. marina and C.polykrikoides than A. tamarense and G. catenatum. The algicidal study of MPB against HABs will provides much insight into development of new algicidal substances.

생리활성을 지닌 신경펩타이드의 구조와 활성간의 관계를 규명하기 위하여 고상법으로 합성한 세종류의 neuropeptide γ(mammalian-,trout- 그리고 goldfish-neuropeptide γ)를 사용하여 연구하였다. Circular dichroism spectra에 의하면 mammalian-,trout- 와 goldfish-neuropeptide γ는 완충액 조건하에 모두 random한 구조를 나타내었다. 중성 및 산성 지질 존재 하에서, mammalian-과 trout- neuropeptide γ는 여전히 random한 구조를 취하였다. 그러나, goldfish-neuropeptide γ는 중성 및 산성지질하에서 부분적으로 α-helix구조를 나타내었다. 장관 수축활성에 있어서는 carp 장관, guinea-pig 회장 그리고 rat십이지장에 대하여 비교하였다. Carp에 대해서는 goldfish-neuropeptide γ= trout- neuropeptide γ>mammalian-neuropeptide γ순으로 활서이 나타났다. 그러나, guinea-pig 회장과 rat십이지장에 대해서 mammalian-neuropeptide γ는 어류 유래성 neuropeptide g들 보다 높은 수축활성을 나타내었다. 이러한 결과들은 neuropeptide γ들이 종-특이적인 활성을 나타낸다는 것을 제시한다. The relationship between structure and biological activity was studied on the three neuropeptides(mammalian-,trout- and goldfish-neuropeptide γ)that were syntheized by the solid-phase method. Circular dichroism spectra showed that mammalian-,trout- and goldfish-neuropeptide γadopted an undered structure in buffer solution. In the presence od neutral and acidic liposomes, mammalian-and trout- neuropeptide γ also took a random structure. However, goldfish-neuropeptide γ took an α-helical structure in acidic liposomes. The intestinal motility response was investigated with carp interstines, guinea-pig ileums and rat duodenums. In case of carp intestine, contractine activity was as follows:goldfish-neuropeptide γ= trout- neuropeptide γ> mammalian-neuropeptide γ. On the other hand, the contracile activity of mammalian-neuropeptide γ was more potent than trout- and goldfish-neuropeptide γ in the guinea-pig ileums and rat duodenums. these results suggest that neuropeptide γs showed the species-specific activity.
Genetic diversity and structure analysis of wild soybean (Glycine soja) in Korea

Undomesticated soybeans (Glycine soja) are an important source of genetic variation for introducing useful traits to domesticated soybeans (Glycine max). Although Korea is known as the origin of the soybean, a little is known about genetic diversity and structure analysis of G.soja. The objectives of this study were to investigate the genetic diversity and the structure analysis of wild soybeans, and to construct a core collection of G. soja accessions in Korea. To evaluate the genetic diversity and structure analysis of G.soja, we analyzed allelic profiles at 21 SSR loci of 1028 accessions using POWERMARKER V3.25. These markers generated a total of 581 alleles over all loci. The number of alleles per locus ranged from 21 to 40, with a mean of 28 alleles per locus and a mean gene diversity of 0.886 in this accessions tested. Polymorphic information content value ranged from 0.737 to 0.946, with an average of 0.877. Using STRUCTURE V2.34, wild soybean originated from Korea was divided into two distinct populations, largely corresponding to two geographic regions. Population 1 consisted of eight sub-groups corresponds to mountains; population 2 to entire regions in Korea. Based on theses 21 SSR markers, a core collection development was performed by POWERCORE V1.0. A G. soja core collection consisted of 148 accessions which were established from 1028 accessions in Korea. Most accessions of the core collection were belonged to population 2 and only four were belonged to population 1. These results of this study would provide valuable information for future breeding programs using the G. soja core collection.

Apidaecin Ib had strong antimicrobial activity against several tested Gram-negative bacteria including Escherichia coli, Enterobacter cloacae, and Shigella flexneri (MECs; $0.3-1.5{\mu}g/mL$), but showed no activity against all the tested Gram-positive bacteria including Bacillus subtilis, Micrococcus luteus, Staphylococcus aureus and one yeast, Candida albicans (MECs; > $125{\mu}g/mL$). Interestingly, this peptide showed potent antibacterial activity only against Edwardsiella species (MECs; $0.6-3.6{\mu}g/mL$) among the tested fish pathogenic bacteria through a bacteriostatic process and showed no significant hemolytic activity. Apidaecin Ib took an unordered structure in all environments and also had very weak membrane perturbation activity even at $25{\mu}M$. Anti-Edwardsiella activity of apidaecin Ib is stronger than those of other antimicrobial polypeptides or antibiotics, but its activity is salt-sensitive. These results suggest that apidaecin Ib has Edwardsiella speciesspecific antibacterial activity and could be applied as new preventive or control additives for Edwardsiella species infection in freshwater fish aquaculture.
Distinct roles of E3-paralogue genes promote early flowering in late flowering soybean cultivars

Soybean (Glycine max (L.) Merr) is a short day plant and has been adapted to various climates and environments during cultivation. However, the cultivation area is restricted to a very narrow range of latitudes. To date, nine major genes (E1 to E8 and J) have been reported to control the flowering time and maturity. Here, we evaluated the role of E2, E3, E4, and their paralogue genes in late flowering soybean cultivars under long day (LD) conditions using Soybean yellow common mosaic virus (SYCMV)-based virus-induced gene silencing (VIGS) system. A total of nine VIGS constructs were infiltrated into two fully expanded cotyledons and primary leaves. After inoculation with these VIGS constructs on Jangyeobkong, which is a late-flowering cultivar, phenotypic traits were evaluated for the first flowering dates (FFDs) and pod maturities under LD conditions. The FFDs of the silenced plants occurred 50-56 days after sowing (das), while the non-silenced plants bloomed on 60-61 days. We found that the E3 paralogue-silenced plants flowered the fastest and responsive genes were identified to be associated with the promotion of flowering time. As the knock-down of E3 paralogue, expression of E1 was up-regulated, E2 was no difference, E3 and E4 genes were down-regulated in the silenced plants. Expression of GmFT2a and GmFT5a is known to be controlled by E3 and E4. Interestingly, GmFT5a were highly expressed in SYCMV:E3 paralogue-silenced plants, whereas the expression of GmFT2a was not significant. These results support that GmFT5a is able to independently promote flowering under LD conditions.
A New Six-rowed Barley Cultivar, "Hwanggeumchal" with Waxy Cover and Yellow Aleurone

“Hwanggeumchal”, a new covered waxy cultivar derived from the cross between “Chalssalbori” and “Milyang 65” with waxy endosperm and early maturing cultivars, respectively was developed at Honam Agricultural Research Institute (HARI), NICS, RDA, 2006. The origin of “Hwanggeumchal” is “Suwon 403” (SB942090-B-B-B-42-1). The initial cross was made in 1994 and the selected line showed a high yield and good quality characteristics under yield trial test in 2003. “Suwon 403” consistently performed well for three years (2004-2006) from the four locations of regional yield trial (RYT) in Korea and released as “Hwanggeumchal”. The characteristics of “Hwanggeumchal” were the following: rate I growth habit, green leaf and stem, compact spike and with long rough awns. The heading date was April 30 in upland and April 28 in paddy field, which was 1 and 4 days later than that with check cultivar, “Seodunchalbori”. The culm length was 81 cm which was 3 cm shorter than those with check cultivar. It showed spike length of 4.5 cm and 633 spikes per m2, 64 grains per spike, 27.6 g of 1,000-grain weight and 632 g of test weight. It showed stronger winter hardiness and higher resistance to barley yellow mosaic virus (BaYMV) than those with check cultivar. It had yellow aleurone and showed higher water absorption, expansion rate and total phenol content than those of check cultivar, but lower protein, β-glucan content and pearling rate. The average yield of the pearled grain in the RYT was 4.04 ton ha-1 in upland and 4.12 ton ha-1 in paddy field, which was 10% higher and 4% lower than that of the check cultivar, respectively. This cultivar would be suitable for the area above the daily minimum mean temperature of -8℃ in January in Korean peninsula.
