      • 농업 부산물 바이오매스 특성에 따른 반탄화 공정 최적화: 1. 질량감소 예측 모델 개발

        본 연구에서는 국내에서 버려지고 있는 농업부산물의 에너지로서 이용 가능성이 분석되었다. 농업부산물 바이오매스는 화석연료와 동일하게 활용가능하며 지속적으로 생산되는 에너지원이다. 하지만 높은 수분 함량에 따른 보관문제, 화석연료와 비교하여 낮은 발열량등의 단점을 지니고 있어 전처리공정이 요구된다. 따라서 효율적인 활용을 위한 전처리 공정으로 반탄화가 선정되었다. 반탄화공정은 수분제거를 통해 에너지가 증가되어 저장·운송에 이점으로 인하여 최종적인 에너지 가치가 증대된다. 실험은 문헌탐색을 통한 반탄화공정 적용 가능 농업부산물(고추대)과 실험조건(200, 230, 270C, 10, 20, 30, 40 min)이 선정되었으며, 1차원 시뮬레이션을 통한 분석이 이루어졌다. 이때 반탄화 실험을 통하여 발생되는 질량감소량과 발열량 변화량은 높은 상관관계(r<sup>2</sup>=0.9528)를 가지고 있으며 본 연구에서는 반탄화공정의 질량감소량 예측 모델개발이 수행되었다. 질량감소량 분석을 위하여 열 중량 분석(Thermo Gravimetric Analyzer, TGA)이 수행되었으며 승온속도(5, 7.5, 15, 22.5, 30, 37.5°C min<sup>-1</sup>)에 따른 반응속도상수가 도출되었다. 1차원 시뮬레이션 분석을 통하여 바이오매스 내부 온도를 예측하였으며 앞서 도출된 반응속도 상수를 이용하여 질량감소량이 예측되었다. 질량감소량 실험 및 시뮬레이션 결과 평균 r<sup>2</sup>=0.97로 상관관계를 나타내고 있으며 공정온도별 평균제곱근오차(Root Mean Square Error, RMSE)가 200°C-0.0165, 230°C-0.0191, 270°C-0.0207로 나타났다. 공정온도 증가에 따라 정확도가 감소되었으며, 이는 고온의 실험조건에서 발생되는 비응축가스와 온도변화에 의한 열분해 특성에 기인한 것으로 판단된다. 최종적으로 더 나아가서 질량감소 모델을 통한 발열량 예측을 위한 분석이 이루어져야한다.

        This study was conducted to optimize the processing of high quality surimi gel from unmarketable cultured bastard halibut Paralichthys olivaceus. According to endogenous enzyme activity and processing optimization, high quality surimi gel from unmarketable cultured bastard halibut was prepared by mixing 3.0% (w/w) salt, 2.4% (w/w) starch, 5.0% (w/w) egg white and 4.8% (w/w) ice water in a Stephan mixer, set at 5˚C for 24 h, followed by boiling for 30 min, and finally cooling for 30 min. The strength of the surimi gel from unmarketable cultured bastard halibut prepared by the above processing method was 1,257 g×cm, which was 33% higher than that of a commercial surimi gel from Alaska pollock, grade SA.

        원산지별 감초추출물의 항산화활성 증가를 위한 효율적인 추출조건 탐색

        본 연구에서는 한국 및 중국산 감초(Glycyrrhiza uralensis), 그리고 우즈베키스탄산 감초(Glycyrrhiza glabra)를 대상으로 추출용매, 추출온도, 추출시간 등 추출조건별 추출물을 제조하고 이들 추출물들의 추출 수율과 항산화 활성을 비교하여 최적의 추출조건을 선정하였다. 항산화 활성 중 자유 라디칼(1,1-phenyl-2-pic-rylhydrazyl, DPPH) 소거활성은 85% 에탄올로 60 ℃에서 6 h 동안 추출한 조건에서 한국 감초로부터 얻은 추출물이 가장 높은 활성(46.05%)을 나타내었다. 루미놀 발광법을 이용한 추출물들의 총 항산화능 측정 실험과 피부 광노화에 중요한 <sup>1</sup>O<sub>2</sub>으로 유도된 세포막 손상에 대한 추출물들의 세포 보호 효과를 측정한 실험 모두에서도 위의 조건에서 가장 높은 항산화 활성을 나타내었다. 특히, 한국 감초는 τ<sub>50</sub>이 116.4 min으로 비교 물질인(+)-α-tocopherol (28.5 min)보다 약 4 배나 높은 세포 보호 효과를 나타내었으며, 추출 수율은 18.75%로 우즈베키스탄 및 중국 감초보다 각각 1.2 배 및 2.5 배의 추출 수율을 나타내었다. 따라서, 본 연구 결과는 항산화 소재로 화장품에 응용하기 위하여 감초로부터 추출물을 얻기 위한 최적의 조건은 85% 에탄올로 60 ℃에서 6시간 동안 추출하는 것임을 보여주었다. In this work, licorice extracts were prepared using various extraction conditions such as extraction solvent, temperature, and time from Glycyrrhiza uralensis (G. uralensis) produced in Korea and China and Glycyrrhiza glabra (G. glabra) in Uzbekistan. The optimum extraction condition was selected from the extraction yields and antioxidative activities of extracts. Korea licorice extracts showed the highest free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scav-enging activity (46.05%) under the extraction condition of 85% ethanol at 60 ℃ for 6 hours. The prominent ROS (reactive oxygen species) scavenging activity using luminol-dependent chemiluminescence assay and the cellular protective effect against <sup>1</sup>O<sub>2</sub> induced cellular membrane damage were also shown from the extracts obtained from the same condition. Especially, Korea G. uralensis extracts exhibited the higher prominent protective effect (τ<sub>50</sub> = 116.4 min) than (+)-α-tocopherol (τ<sub>50</sub> = 28.5 min) and the extraction yield of Korea licorice extract was 18.75%, which is 1.2 times and 2.5 times higher than that of Uzbekistan and China, respectively. These results indicate that the extraction condition of 85% ethanol at 60 ℃ for 6 hours is optimal to prepare licorice extracts, which can be applicable as antioxidative cosmetic materials.

      • 황색 포도상구균의 vancomycin 내성 원인 유전자의 동정

        임상 분리되는 Gram 양성균인 황색 포도상구균(Staphyhcoccus aureus)에 대한 각종 학생물질의 최소 발육저지농도 측정(Minimal Inhibitory Concentration: MIC test)과 내성 pattern을 조사하였으며, 이들 균주 중 특히 vancomycin에 대하여 높은 내성을 나타내는 균주(Vancomycin resistant Staphylococcus aureus: VRSA)에 대한 vancomycin 내성원인 유전자의 탐색 및 cloning을 실시하여 vancomycm 내성기구의 연구에 필요한 조사를 수행하였다. Vancomycm에 내성인 균주(VRSA)를 고온(43.5℃) 처리하여 계대 배양을 한 다음, replica plating method에 의 해 vancomycin 감수성 균주(Vancomycin-sensitive Staphylococcus aureus: VSSA)를 얻었으며, VRSA 균수와 VSSA 균주로부터 membrane protein을 준비한 다음 SDS-poly-acrylamide gel electrophoresis(SDS-PAGE)로 전기 영동하여 그 단백질 pattern을 비교한 결과, VRSA 균주에서는 분자량(Mr: relative molecular weight)이 약 127Kadl 그리고 30Kdal 정도 되는 부위에서 vancomycin 내성에 관계되는 듯한 특이적인 단백질을 관찰할 수 있었다. 그러나 VSSA 균주에서는 이러한 단백질을 관찰할 수 없었다. 한편 이들 균주로부터 plasmid DNA를 분리한 다음 agarose gel electrophoresis하여 그 pattern을 분석한 결과, VRSA 균주에서는 약50Kb부위에서 plasmid DNA밴드가 관찰되었으나 VSSA 균주에서는 아무런 밴드도 관찰되지 않았다. 이상의 결과로 볼때 vancomycin 내성유전자는 plasmid DNA상에 존재한다고 생각되어진다. This study was carried out to study the characterization of the gene that is responsible for the vancomycin-resistant Staphylococcus aureus (VRSA) In order to obtain vancomycin-sensitive S. aureus(VSSA)from VRSA stain, vancomycin-resistant S. aureus(VRSA) was subcultured twice at the high temp.(43.5℃)and then VSSA was obtained by replicaplating method after MIC test with vancomycin in high temp. treted VRSA strain. Membrane proteins were prepared from VRSA and VSSA strain and protein patterns of those were compared bny 7.5% SDS-polyacrylamide gel electrophoresis. Only in VRSA strain appeared novel protein bands at a position about 120 Kdal, 80 Kdal and 30Kda1, but it did not appear in VSSA strain. The patterns of plasmid DNA in VRSA and VSSA strain were clearly classified. Plasmid DNA band in VSSA strain appeared at a position about 50Kb, but it did not appear any band in VSSA strain. These results suggested that plasmid DNA may be encode the gene that is responsible for vancomycin-resistance.

        일차성 Streptococcus pneumoniae 균혈증에 동반된 흉부 하행 대동맥염 1예

        페렴구균에 의한 대동맥염은 국내에서 1예만 보고될 정도로 매우 드문 감염증이다. 저자들은 원발성의 폐렴구균혈증에 합병된 페렴구균 대동맥염 환자 1례를 경험하였기에 문헌고찰과 함께 보고하는 바이다. Bacterial aortitis is rare and without surgical intervention, it usually leads to uncontrolled sepsis or catastrophic hemorrhage, ultimately, resulting in death. In the postantibiotic era, the dominant infecting organisms are Staphylococcus aureus and Salmonella spp.. In spite of frequent pneumococcal bacteremia, vascular seeding is rare and there is only one published report in Korea. We describe a 77-year-old female with primary pneumococcal bacteremia followed by aortitis of the descending aorta, which was successfully treated by aneurysmectomy, wide excision of periaortic tissue, aortobiiliac bypass via prosthetic graft, and antibiotic therapy.

        여수산단 대기 중 휘발성유기화합물질 농도 경향

        The concentrations of volatile organic compounds (VOCs) in the ambient air were measured at three sites (Samil-dong, SRO and EFMC) in Yeosu industrial complex during September 2000 to August 2001. Air samples were collected for 24 hours in Silicocan canister (6ℓ) with constant flow samplers every 6 days and analyzed using a cryogenic preconcentration system and a GC/MS. At each site, we identified 35 species known as on both the carcinogenic and mutagenicity by the EPA US (TO-14 manual). No relationship was found between VOCs concentration at three sampling points. Furthermore, the result shows that there appears to be a variety distribution of the concentration. BTX, vinyl chloride and high concentrations of I, 2-dichloroethane were observed at the sampling sites. Especially, high concentration of toxic VOCs, such as vinyl chloride, chloroform, I, 2-dichloroethane and benzene were shown at environmental facilities management cooperation (EMFC) site. They seem to be emitted from the facility of wastewater treatment in Yeosu industrial complex. It was difficult to tell the seasonal variation of total VOCs concentration. Nevertheless, the concentration in winter was typically higher than in summer. The concentrations of toxic VOCs contents in Yeosu industrial complex were generally lower than those in Ulsan complex, although those were similar or less than in Seoul and Daegu. Whereas, toluene and styrene emitted from Yeosu industrial complex were higher than those of Edmonton industrial complex in Canada. Especially, toluene was third times higher than those observed from Texas, USA.

        A 8-year-o1d castrated male Shit-tzu dog was presented with chronic, severe and recurrent bilateral otitis externa. Clinical signs were stenosis, swelling, severe discharges from external auditory canal and pruritus of the ear. A diagnosis of the patient was made based on history, Physical examination, otoscopic examination, cytological examination and radiography. The dog was diagnosed to end-stage otitis externa and followed by treatment with systemic cyclosporine administration and topical solution. Clinical symptoms were resolved after 4 weeks cyclosporine therapy.

        여수석유화학산단 내 VOCs에 대한 오염원 분류표의 개발 및 CMB 모델에 의한 기여도 산정

        The purposes of this study were to characterize the local levels of VOCs (volatile organic compounds), to develop source profiles of VOCs, and to quantify the source contribution of VOCs using the CMB (chemical mass balance) model. The concentration of VOCs had been measured every 6-day duration in the SRO monitoring site in the Yeosu Petrochemical Industrial Complex from September 2000 to August 2002. The total of 35 target VOCs, which were included in the TO-14 designated from the U.S. EPA, was selected to be monitored in the study area. During a 24-h period, the ambient VOCs were sampled by using canisters placing about 10~15 m above the ground level. The collected canisters were then analyzed by a GC-MS in the laboratory. Aside from ambient sampling at the SRO site, the VOCs had been intensively and massively measured from 8 direct sources and 4 general sources in the study area. The results obtained in the study were as follows; first, the annual mean concentrations of the target VOCs were widely distributed regardless of monitoring sites in the Yeosu Petrochemical Industrial Complex. In particular, the concentrations of BTX (Benzene, Toluene, Xylene), vinyl chloride were higher than other target compounds. Second, based on these source sample data, source profiles for VOCs were developed to apply a receptor model, the CMB model. Third, the results of source apportionment study for the VOCs in the SRO Site were as follows; The source of petrochemical plant was apportioned by 31.3% in terms of VOCs mass. The site was also affected by 16.7% from wastewater treatment plant, 14.0% from iron mills, 8.4% from refineries, 4.4% from oil storage, 3.8% from automobiles, 2.3% from fertilizer, 2.3% from painting, 2.2% from waste incinerator, 0.6% from graphic art, and 0.4% from gasoline vapor sources.

        소목 천연 염색에 관한 연구 Ⅰ : 정제 알루미늄 화합물들의 매염효과에 대하여 Mordanting Effect of Purified Aluminum Compounds

        Currently, as mordants, heavy metals are being used in dyeing process in order to improve dye uptake and dye fastness. These chemicals, however, are mostly harmful to human body and the environment along with the pollution by effluent. Traditionally, rice straw ash solution, lime, etc. have been used as mordants in order to improve the effects on dye uptake and color fastness of dyestuffs. The study of natural mordants would, therefore, be vital to the development and succession of our traditional dyeing methods. In this study, as a preliminary step toward the use of rice straw ash solution for the nautral dyeing using Caesalpinia sappan, several aluminum compounds were employed as mordants. When the purified aluminum compounds were used, on the basis of unmordanted cotton fabric, the WS values of the pre-mordanted and simultaneously mordanted fabrics increased by 2 or 3 times, while those of the post-mordanted fabrics decreased.

        If we are looking for something that can be found anywhere in public places, homes, and and even toilets, we will be able to choose a mirror that can easily reflect us. To satisfy the convenience needs of people, we can automatically introduce various useful information on mirrors that are often used by anyone, or add functions such as measuring human body temperature in line with the recent trend of public health due to the COVID-19 pandemic. The system presented in this paper aims to improve the convenience of users who frequently look in the mirror by displaying visual information on a translucent mirror as well as being able to reflect the image as explained above.
