      • T 세포활성항원 CTLA-4의 기능에 관한 연구 : T 세포에서 표적세포 살해능과 CTLA-4 발현과의 연관성 Target Cytotoxicity of T Cell Correlates with CTLA-4 Production

        CTLA-4, a T cell activation antigen and a homologue of CD28, was originally identified as a gene by a series of reverse genetic approaches. While CD28 molecule has been characterized well as a stimulator of T-cell function via enhanced lymphokines production and stablilization of those mRNA, most of the functions of CTLA-4 remain unknown. It has been widely accepted that CTLA-4 functions as an immune suppressor which is down-regualting the function of CD28. We previously showed that 34 KD form of this antigen mainly expressed CD8+ subset, a cytotoxic or suppressor, of activated peripheral blood lymphocyte. Based on our previous finding this study was conducted to further characterize immunological function of CTLA-4 especially in terms of involvement of this molecule in T-cell effector function mediating target cell cytotoxicity. 4 human T cell clones with different target cytotoxicities were employed in this study. NBL46 (CD4+) and NBL77 (CD8+) were cytotoxic and NBL32 (CD4+) and NBL58 (CD 8+) were non-cytotoxic to target LBF cell in target cell chromium release assay. And in Western blot assay 34 kD antigen was detected in NBL46 (CD4+) and NBL77 (CD8+) clones but not in NBL32 (CD4+) and NBL58 (CD8+) clones. It was summarized that expression of the CTLA-4 antigen was associated with cytotoxicity but not with subset phenotypes of T cells. In conclusion CD8+ T subset of PHA-stimulated PBL was major and only CTLA-4 producer and this molecule was induced during mid to late stage of T cell activation. The cytotoxicity of T cell clones to its target cell was directly correlated with its CTLA-4 production and vice versa. And it was highly suggested that primary function of CTLA-4 may involved in T cell effector function which mediates target cell killing.

        비유피-4 정(염산프로피베린 20㎎)에 대한 건일염산프로피베린 정의 생물학적동등성

        The purpose of the present study was to evaluate the bioequivalence of two propiverine hydrochloride tablets. BUP-4 (Jeil Pharm. Co., Ltd.) and Kuhnil Propiverine Hydrochloride (Kuhnil Pharm. Co., Ltd.), according to the guidelines of the Korea Food and Drug Administration (KFDA). The propiverine release from the two propiverine hydrochloride formulations in vitro was tested using KP Ⅷ Apparatus Ⅱ method with a variety of dissolution media (pH 1.2, 4.0, 6.8 buffer solutions, water and blend of polysorbate 80 into pH 6.8). Twenty six healthy male subjects, 23.73 ± 2.79 years in age and 67.04 ± 7.93 kg in body weight, were divided into two groups and a randomized 2 x 2 cross-over study was employed. After one tablet containing 20 mg as propiverine hydrochloride was orally administered, blood was taken at predetermined time intervals and the concentrations of propiverine in serum were determined using HPLC method with UV detector. The dis-solution profiles of two formulations were similar at all dissolution media. Besides, the pharmacokinetic parameters such as AUC" C _(max) and T _(max) were calculated and ANOVA test was utilized for the statistical analysis of the parameters using logarithmically transformed AUC, C_(max), and untransformed T_(max). The results showed that the differences between two formulations based on the BUP-4 were 0.17%, 7.98% and 4.55% for AUC,, C_(max), and respectively. There were no sequence effects between two formulations in these parameters. The 90% confidence intervals using logarithmically trans-formed data were within the acceptance range of log(0.8) to log(1.25) (e.g., log(0.88)-log(l .12) and log(0.90)-log(l.15) for AUC, and _(max), respectively). Thus, the criteria of the KFDA bioequivalence guideline were satisfied, indicating Kuhnil Propiverine Hydrochloride tablet was bioequivalent to BUP-4 tablet.

        The purpose of this study was to explore the developmental trends in social skills of the normal, mental retardation, and learning disabilities children at primary schools. A total of 314 children(100 normal children, 100 mental retardation children, and 114 learning disabilities children) participated in this study. Social skills(Cooperation, Assertion, and Self-control skill) of children measured by Gresham and Elliott's Social Skills Rating System(SSRS). The results of the study were as followings: First, social skill of the normal children was found to be linear trend. However, social skill of the mental retardation and learning disabilities children were not found consistent trend. Second, cooperative skill within social skills of the normal, mental retardation, and learning disabilities children were found to be linear trend. Third, assertive skill within social skills of the normal children was found to be linear trend. However, assertive skill of the mental retardation and learning disabilities children were not found to be consistent trend. Forth, self-control skill within social skill of the normal and mental retardation children were found to be linear trend. However, self-control skill of the learning disabilities children was not found to be consistent trend.

        자색고구마가 사염화탄소 투여에 의한 흰쥐의 간손상 보호에 미치는 영향

        The purpose of this study was to investigate the effects of dietary purple sweet potato(lpomoea batatas) powder on serum lipid levels and antioxidative enzymes in normal and pretective effect on hepatotoxicity rats induced by carbon tetrachlolide. Four groups of rats (3-week-old inbred Sprague-Dawley male rats) were normal rats fed control diet(C)_(1) induced hepatotoxicity rats fed control diet(EC)_(1) normal rats fed purple sweet potato diet(P), and induced hepatotoxicity rats fed purple potato sweet diet(EP). Rats were induced by single injection of 50% carbon tetrachloide(0.1 mL/100 g B.W., i.p.). The rats were fed ad libitum each of the experimental diet for 5 weeks. After 5 weeks the rats were sacrificed and activities of antioxidant enzymes and lipid peroxidation products were determined in their liver homogenates. But serum concentrations of lipid was not significant in all groups. Serum alanine aminotransferase(ALT/GPT) and aspartate aminotransferase(ALT/GPT)of the EC and EP groups were heigher than the C and P groups. The hepatic glucose 6-phosphatase(G9Pase) activity of the group fed purple potato diet(P) was lower than the other groups(p<0.05). However, The glutathione peroxidase(GPx) activities was not statistically different between the groups. Renal glutathione S-transferase(GST) activity of the EC and EP groups were lower than the C and P groups(p<0.05). In conclusion, these results suggest that purple sweet potato is believed to be possible protective effect on hepatotoxicity rats induced by carbon tetrachlolide.

        Using acclimated sludge with inorganic nitrogen medium for three months and non-acclimated sludge, raw sludge, under aerobic condition we investigated characteristics biological nitrogen removal. As results, e acclimated sludge under aerobic condition accomplished nitrification efficiently. But the samples from the reactor operated raw sludge had higher ammonium concentration rather than the concentration which was loaded by the medium. This means when we are trying to reorganize existing facilities, we should take high initial ammonium concentration into account.

      • 1980年度 痢疾樣 大便을 가진 患兒들에 對한 臨床的 細菌學的 觀察

        1980年 1月부터 1980年 8月까지 滿 8個月間 痢疾樣 大便을 主訴로 本 小兒科에서 入院및 來院한 患兒 75名을 對象으로 臨床的, 細菌學的 觀察을 하여 다음과 같은 咸績을 얻었다. 年齡別 分布는 1歲에서 5歲 사이가 35例(47%)로 가장 높았으며 이들中 Shigella는 23例(30%), Entameba histolytica는 13例(17%)였고 Vibrio parahaemolyticus는 한 例도 檢出되지 않았다. 檢出된 Shigella 23菌株는 모두 Shigella flexneri 였다. 月別分布는 6月 16例(20%), 7月 30例(40%)로서 最多數를 占하였으나 細菌性 痢疾의 경우 月別에 관계없이 均等하게 分布하고 있었다. 臨床症狀및 理學的 所見으로는 細菌性痢疾의 경우 38℃ 以上의 發熱 19例(83%), 嘔吐 6例(26%), 腹痛 8例(35%), 痙攣 5例(22%)였으며, 아메바性 痢疾에서는 發熱 4例(31%), 痙攣 1例(8%)로서 細菌性 痢疾에서 아메바性 痢疾보다 發熱과 痙攣이 빈번히 나타났다. 泄瀉의 性狀에서는 細菌性 痢疾 23例中 血粘液性이 19例(83%)였는데 비해 아메바性 痢疾 13例에서는 粘液性이 8例(62%)였다. 泄瀉의 回數에서도 細菌性 痢疾에서는 대부분이 11回以上이었는데 比해 아메바性 痢疾에서는 대부분이 10回未滿이었다. 末梢血液에서의 白血球數는 全例의 35%에서 白血球 增多症을 보였다. 分離된 Shigella 23菌株에 對한 抗生劑 感受性 檢査를 平板混合포稀釋法에 依해 調査해 본 結果 kanamycin, gentamicin, amikacin, cephaloridine, rifampin이 100%의 感受性을 나타내어 가장 높았고, nalidixic acid에서도 87%의 感愛性을 나타냈으며, sulfonamide, chloramphenicol, streptomycin, tetracycline, ampicillin, bactrim 에서는 한 菌株를 제외한 모든 菌株에서 耐性을 나타냈다. 그리고 이들 Shigella 23菌株中 1菌株를 제외한 나머지 모든 菌株에서 6種 以上의 抗生物質에 對해 多抗生物質耐性을 보여 주었다. A clinical and laboratory study was conducted on 75 children with dysentery-like stool, who were admitted or visited to our pediatric department during 8 months period from January to August 1980. The following results were obtained: One to five years of age group was affected most frequently (47%). Shigella flexneri was identified by stool culture in 23 cases (30%) and Entameba hitolytica in 13 cases (17%). In bacillary dysentery group, fever was the most common symptom occuring in 83%, followed by abdominal pain in 35%, vomiting in 26% and convulsion in 22%. In amoebic dysentery group, fever was noted in 31% and convulsion in 8%, showing some contrast to the frequency of fever and CNS manifestation. About the character of diarrheal stools, 83% of bacillary dysentery group showed bloody, mucoid stool and 62% of amoebic dysentery group mucoid ones. Frequency of diarrhea was 11 times per day or more in 73% of bacillary dysentery patients, and 10 times per day or less in 92% of amoebic dysentery patients. The result of sensitivity test of isolated Shigella to various antibiotics were as follow: 100% sensitive to kanamicin, gentamicin, amikacin, cephaloridine and rifampin. 87% to nalidixic acid, and 4.3% to sulfonamide, chloramphenicol, streptomycin, tetracyclin, ampicillin and bactrim. And 22 out of 23 isolated Shigella strains revealed multiplydrug-resistance patterns to 6 or more antibiotics.

        경기·인천 지역 주부의 특수란의 구매와 계란의 보관 및 조리에 관한 연구

        The purpose of this study was to investigate the storage and cooking of eggs in addition to the purchase of special eggs. This survey was carried out through questionnaire and subjects were 429 housewives whose children were middle or high school students in Kyunggi-do and Inchon. Most housewives began to purchase special eggs one year ago. When housewives purchased special eggs, the first consideration was in order of nutrition, freshness, price and preference. The higher housewives' education level was, the higher they took nutrition into consideration. More than half of housewives replied that the proper package of special eggs was 10 eggs or more, and perceived that the period between producers of eggs and consumers was less than 5 days. Most housewives did the freshness test by checking roughness of shell surface or the interior quality of egg yolk and white after purchase. Also most housewives stored eggs in the inside rack of refrigerator door. Most housewives served dishes made from eggs more than 2 or 3 times per week and perceived that soft-boiled eggs are foods easy of digestion. These results suggested that it is necessary to supply fresh and nutritious eggs and to develop various dishes made from eggs for quickly-prepared meals.

        아마릴 정(글리메피리드 2㎎)에 대한 글리메드 정의 생물학적 동등성

        The purpose of the present study was to evaluate the bioequivalence of two glimepiride tables, Amaryl^(?)(Handok/Aventis Pharm. Co., Ltd.) and Glimed (Kuhn Ⅱ Pharm. Co., Ltd.), according to the guidelines of Korea Food and Drug Administration (KFDA). The glimepiride release from the two glimepiride formulations in vitro was tested using KP Ⅷ Apparatus Ⅱ method with a variety of dissolution media (pH 1.2, 4.0, 6.8 buffer solution, water and blend of PSB 80 into each dissolution medium). Twenty six healthy male subjects, 22.65±2.19 years in age and 66.55±8.85 kg in body weight, were divided into two groups and randomized 2×2 cross-over study was employed. After one tablet containing 2 ㎎ as glimepiride was orally administered, blood was taken at predetermined time intervals and the concentrations of glimepiride in serum were determined using HPLC method with UV detctor. The dissolution profiles of two formulations were similar at all dissolution media. Besides, the pharmacokinetic parameters such as AUC_(t), C_(max) and untransformed T_(max). The results showed that the differences between two formulations based on the Amaryl were -3.70, -8.28 and 0.61% for AUC_(t), C_(max) and T_(max), respectively. There were no sequence effects between two formulations in these parameters. The 90% confidence intervals using logarithmically transformed data were within the acceptance range of log(0.8) to log(1.25)(e.g., log(0.84)∼log(1.04) and log(0.82)∼log(1.03) for AUC_(t) and C_(max), respectively). Thus, the criteria of the KFDA guideline for the bioequivalence were satisfied, indicating Glimed tablet and Amaryl tablet were bioequivalent.
