      • Fashionable styles and information sources: an exploratory study of Hangzhou, China

        The exploratory study aims to examine Hangzhou fashion in the context of globalization and localization through in-depth interviews. Sixteen young females from Hangzhou were interviewed. In the 1990s young females from Hangzhou had a weak concept of fashionable style and they just had a preference for the feminine style, which is derived from a regional culture. In the 2010s, young females’ concept regarding fashion have changed from fashionable (时髦) to stylish (有型), leading to a variety of fashionable styles. The popular Korean, European and minimalist styles mostly originate from foreign dramas and fashion advertisements from the Internet. This qualitative research verified that there was a diminishing sense of locality and cultural identity in young females’ fashion in Hangzhou. However, the impact of globalcultural eclecticism and mass media imagery appears larger for young females’ fashion in the city, especially through the Internet, especially in the spread of SNS, online shopping malls, and online video sites. Therefore, fashion brands should expand the brand culture influence through online media. Presently, the mobile apps in Hangzhou such as Wechat and Instagram may be the most helpful tool for brand culture spreading.

        Molecular cloning and expression analysis of the highly conserved eukaryotic translation initiation factor 5A (eIF-5A) from Antheraea pernyi

        Eukaryotic initiation factor 5A (eIF‐5A) is a highly conserved protein found in all eukaryotic organisms that plays a key role in the regulation of many cellular processes including translation elongation, cell proliferation, programmed cell death, mRNA turnover and decay, and abiotic stress responses. In this study, the eIF‐5A gene from the Chinese oak silkworm Antheraea pernyi (Lepidoptera: Saturniidae) was characterized. The full‐length ApeIF‐5A cDNA of 1056 bp includes a 5′‐untranslated region (UTR) of 138 bp, a 3′‐UTR of 435 bp, and an open reading frame of 483 bp encoding a polypeptide of 160 amino acids. The deduced ApeIF‐5A protein shares 99 %, 82 %, and 72 % sequence identity with orthologs in Bombyx mori, Drosophila melanogaster and Homo sapiens, indicating high conservation during animal evolution. Real‐time quantitative reverse transcription PCR revealed expression in all four developmental stages and in all nine tissues tested, consistent with an important role in development. After challenge with lipopolysaccharide, the expression levels of ApeIF‐5A were markedly upregulated. Phylogenetic analysis of amino acid sequences revealed A. pernyi eIF‐5A was closely related to B. mori eIF‐5A, consistent with traditional classification and other molecular data. The results indicate the potential value of eIF‐5A in phylogenetic analysis.

        Biomimetic apatite formed on cobalt-chromium alloy: A polymer-free carrier for drug eluting stent

        <P><B>Abstract</B></P> <P>In this study, sirolimus (SRL) was loaded within biomimetic apatite formed on cobalt-chromium (Co-Cr) alloy, which has been reported for the first time, to inhibit the in-stent restenosis. Two different groups of loading SRL within biomimetic apatite were prepared: Group A (mono-layer of apatite/SRL) and Group B (bi-layer of apatite/SRL). Group A and Group B showed the biphasic pattern of SRL release up to 40 and 90days, respectively. The attachment of human artery smooth muscle cell (HASMC) for both Group A and Group B was significantly inhibited, and proliferation dramatically decreased with the release of SRL. Noteworthily, biomimetic apatite alone also suppressed the SMC proliferation. The porous biomimetic apatite uniformly covered Co-Cr stent without crack or webbings. After balloon expansion, the integrity of biomimetic apatite was sufficient to resist delamination or destruction.         Enhancement of α-ketoglutarate Production in Torulopsis glabrata: Redistribution of Carbon Flux from Pyruvate to α-ketoglutarate

        This manuscript aimed at increasing the production of α-ketoglutarate by the multi-vitamin auxotrophic yeast Torulpsis glabrata CCTCC M202019. The carbon flux was redistributed from pyruvate to α-ketoglutarate node by manipulating the specific activity of pyruvate dehydrogenase complex (PDH), pyruvate carboxylase (PC), pyruvate decarboxylase (PDC), and α-ketoglutarate dehydrogenase complex (KGDH). By proper increase of PDH, PC, and PDC activities, α-ketoglutarate in fermentation broth could be accumulated to the levels of 17.1 g/L, 21.6 g/L, and 31.2 g/L, respectively. In addition, decrease in the specific activity of KGDH also resulted in an enhanced α-ketoglutarate synthesis.         Downregulation of FoxM1 sensitizes nasopharyngeal carcinoma cells to cisplatin via inhibition of MRN-ATM-mediated DNA repair

        Chemoresistance is the primary obstacle in the treatment of locally advanced and metastatic nasopharyngeal carcinoma (NPC). Recent evidence suggests that the transcription factor forkhead box M1 (FoxM1) is involved in chemoresistance. Our group previously confirmed that FoxM1 is overexpressed in NPC. In this study, we investigated the role of FoxM1 in cisplatin resistance of the cell lines 5-8F and HONE-1 and explored its possible mechanism. Our results showed that FoxM1 and NBS1 were both overexpressed in NPC tissues based on data from the GSE cohort (GSE12452). Then, we measured FoxM1 levels in NPC cells and found FoxM1 was overexpressed in NPC cell lines and could be stimulated by cisplatin. MTT and clonogenic assays, flow cytometry, γH2AX immunofluorescence, qRT-PCR, and western blotting revealed that downregulation of FoxM1 sensitized NPC cells to cisplatin and reduced the repair of cisplatin-induced DNA double-strand breaks via inhibition of the MRN (MRE11-RAD50-NBS1)-ATM axis, which might be related to the ability of FoxM1 to regulate NBS1. Subsequently, we demonstrated that enhanced sensitivity of FoxM1 knockdown cells could be reduced by overexpression of NBS1. Taken together, our data demonstrate that downregulation of FoxM1 could improve the sensitivity of NPC cells to cisplatin through inhibition of MRN-ATM-mediated DNA repair, which could be related to FoxM1-dependent regulation of NBS1.         Experimental Study and Analytical Modeling on Fatigue Properties of Pervious Concrete Made with Natural and Recycled Aggregates

        As a solid pollutant, the recycled aggregate can be reused to replace the natural aggregate to cast pervious concrete, promoting resource recycling and reducing environmental pollution. Pervious concrete is usually applied to transportation engineering as pavements and decks, which are often subjected to fatigue loads in service. Therefore, it is essential to investigate the fatigue mechanical properties of pervious concrete. In this study, four-point cyclic bending loading test of natural aggregate pervious concrete and recycled aggregate pervious concrete were conducted under four different stress levels. By analyzing the experimental results, the mechanical performances, including hysteresis curve characteristics, damping ratio, dynamic elastic modulus and cyclic strain, of two kinds of pervious concrete under cyclic loading were revealed. Based on the improved EPF model, the relationship between fracture parameters, plastic strain and unloading strain were obtained. Besides, the relationship between the loading cycles and the ratio of plastic strain to unloading strain was received according to fatigue testing data under different stress levels. Further, the simplified fatigue model of pervious concrete was proposed and the experimental data was fitted with the model results. The fitting result reached a good agreement.

      • Laminin functionalized biomimetic apatite to regulate the adhesion and proliferation behaviors of neural stem cells

        <P><B>Background</B></P><P>Functionalizing biomaterial substrates with biological signals shows promise in regulating neural stem cell (NSC) behaviors through mimicking cellular microenvironment. However, diverse methods for immobilizing biological molecules yields promising results but with many problems. Biomimetic apatite is an excellent carrier due to its non-toxicity, good biocompatibility, biodegradability, and favorable affinity to plenty of molecules. Therefore, it may provide a promising alternative in regulating NSC behaviors.</P><P><B>Methods</B></P><P>Biomimetic apatite immobilized with the extracellular protein – laminin (LN) was prepared through coprecipitation process in modified Dulbecco’s phosphate-buffered saline (DPBS) containing LN. The amount of coprecipitated LN and their release kinetics were examined. The adhesion and proliferation behaviors of NSC on biomimetic apatite immobilized with LN were investigated.</P><P><B>Results</B></P><P>The coprecipitation approach provided well retention of LN within biomimetic apatite up to 28 days, and supported the adhesion and proliferation of NSCs without cytotoxicity. For long-term cultivation, NSCs formed neurosphere-like aggregates on non-functionalized biomimetic apatite. A monolayer of proliferated NSCs on biomimetic apatite with coprecipitated LN was observed and even more stable than the positive control of LN coated tissue-culture treated polystyrene (TCP).</P><P><B>Conclusion</B></P><P>The simple and reproducible method of coprecipitation suggests that biomimetic apatite is an ideal carrier to functionalize materials with biological molecules for neural-related applications.</P>

        Antinociceptive effects of oleuropein in experimental models of neuropathic pain in male rats

        Background: The present investigation explored the therapeutic actions of oleuropein along with the possible signaling pathway involved in attenuating neuropathic pain in chronic constriction injury (CCI) and vincristine-induced neuropathic pain in male rats. Methods: Four loose ligatures were placed around the sciatic nerve to induce CCI, and vincristine (50 μg/kg) was injected for 10 days to develop neuropathic pain. The development of cold allodynia, mechanical allodynia, and mechanical hyperalgesia was assessed using different pain-related behavioral tests. The levels of H<sub>2</sub>S, cystathionine-γ-lyase (CSE), cystathionine-β-synthase (CBS), orexin, and nuclear factor erythroid-2-related factor 2 (Nrf2) were measured in the sciatic nerve. Results: Treatment with oleuropein for 14 days led to significant amelioration of behavioral manifestations of neuropathic pain in two pain models. Moreover, oleuropein restored both CCI and vincristine-induced decreases in H<sub>2</sub>S, CSE, CBS, orexin, and Nrf2 levels. Co-administration of suvorexant, an orexin receptor antagonist, significantly counteracted the pain-attenuating actions of oleuropein and Nrf2 levels without modulating H<sub>2</sub>S, CSE and CBS. Conclusions: Oleuropein has therapeutic potential to attenuate the pain manifestations in CCI and vincristine-induced neuropathic pain, possibly by restoring the CSE, CBS, and H<sub>2</sub>S, which may subsequently increase the expression of orexin and Nrf2 to ameliorate behavioral manifestations of pain.
