피칸(Carya illinoensis C. KocH)으로부터 지질대사물질의 분리 및 동정

피칸(Carya illinoensis C. Koch) 열매를 80% MeOH로 추출하고, 추출물을 극성에 따라 n-hexane, CHCl_(3), EtOAc, n-BuOH 및 물로 분배, 추출하였다. 이 중 EtOAc분획으로부터 silica gel column chromatography를 반복하여 4종의 지질화합물을 분리하였다. 분리된 화합물 화학구조는, NMR 및 MS 등의 스펙트럼을 해석하여 oleic acid methyl ester, linoleic acid methyl ester, β-sitosterol, daucosterol(3-O-β-D-glucopyaranosyl β-sitosterol)으로 동정하였다. Carya illinoensis C. Koch was extracted with 80% aqueous MeOH, and the concentrated extract was partitioned with n-hexane, CHCl_(3), EtOAc, n-BuOH and H_(2)O, successively. From the EtOAc fraction, four compounds were isolated through the repeated silica gel column chromatographies. From the results of physico-chemical data including NMR, MS and IR, the chemical structures of the compounds were determined as oleic aicd methyl ester(1), linoleic aicd methyl ester(2), β-sitosterol(3), and daucosterol(4). They were the first to be isolated from C. illinoensis.

<P>Recently, adenovirus (Ad) has been utilized as a viral vector for efficient gene delivery. However, substantial immunogenicity and toxicity have obstructed oncolytic Ad’s transition into clinical studies. The goal of this study is to generate an adenoviral vector complexed with multidegradable bioreducible core-cross-linked polyethylenimine (rPEI) polymer that has low immunogenicity and toxicity while having higher transduction efficacy and stability. We have synthesized different molecular weight rPEIs and complexed with Ad at varying molar ratios to optimize delivery of the Ad/polymer complex. The size and surface charge of Ad/rPEIs were characterized. Of note, Ad/rPEIs showed significantly enhanced transduction efficiency compared to either naked Ad or Ad/25 kDa PEI in both coxsackievirus and adenovirus receptor (CAR) positive and negative cancer cells. The cellular uptake result demonstrated that the relatively small size of Ad/16 kDa rPEIs (below 200 nm) was more critical to the complex’s internalization than its surface charge. Cancer cell killing effect and viral production were significantly increased when oncolytic Ad (RdB/shMet, or oAd) was complexed with 16 kDa rPEI in comparison to naked oAd-, oAd/25 kDa PEI-, or oAd/32 kDa rPEI-treated cells. This increased anticancer cytotoxicity was more readily apparent in CAR-negative MCF7 cells, implying that it can be used to treat a broad range of cancer cells. Furthermore, A549 and HT1080 cancer cells treated with oAd/16 kDa rPEI had significantly decreased Met and VEGF expression compared to either naked oAd or oAd/25 kDa PEI. Overall, these results demonstrate that shMet expressing oncolytic Ad complexed with multidegradable bioreducible core-cross-linked PEI could be used as efficient and safe cancer gene therapy.</P><P><B>Graphic Abstract</B> <IMG SRC='http://pubs.acs.org/appl/literatum/publisher/achs/journals/content/bomaf6/2015/bomaf6.2015.16.issue-7/acs.biomac.5b00538/production/images/medium/bm-2015-00538v_0010.gif'></P>

We have established a new National Reference Standard(NRS) of filgrastim to be used for the quality management of domestic genetic recombinant drugs. Filgrastim is a recombinant human granulocyte colony stimulating factor( G-CSF) and one of the first-generation biosimilars used as the hematopoietic agent. At first, the candidate substance for the NRS was manufactured on consignment in accordance with the manufacturing process of filgrastim. The identity, purity, potency and protein contents of the candidate substance were assessed using electrophoresis, HPLC systems and a biological assay, which showed it was comparable to filgrastim in quality. In addition the stability test showed it was stable for 12 months. We performed a collaborative study with other institutes to endow it with the labelled protein content and potency values. From the data, it has been assigned with the protein content of 0.240 mg/mL(95% confidence intervals: 0.238~0.242 mg/mL) and the potency value of 2.94 × 107 IU per vial. We had finally enrolled it as the first Korean NRS of filgrastim as approved by Working Committee on Development of Testing and Inspection in Korea Ministry of Food and Drug Safety.

The flowers of Hemisteptia lyrata B. afforded two known acylglycerylgalactosides, 2',3'-di-O-(9Z,12Z,15Zoctadecatrienoyl) glyceryl β-D-galactopyranoside (1) and 2'-O-(9Z,12Z,15Z-octadecatrienoyl)glyceryl β- D-galactopyranoside (2), and a new sesquiterpene galactopyranoside, 7-eudesmene-1β,4β-diol-1-O-β-Dgalactopyranoside (3). This is the first time that galactopyranosides (1-3) have been isolated from the genus Hemisteptia. Their structures and stereochemistry were elucidated by 1D and 2D NMR data, including COSY, NOESY and HMBC experiments

<P><B>Abstract</B></P> <P>Canine oocytes ovulated at prophase complete meiosis and continue to develop in presence of a high progesterone concentration in the oviduct. Considering that meiotic competence of canine oocyte is accomplished in the oviductal environment, we postulate that hormonal milieu resulting from the circulating progesterone concentration may affect oocyte maturation and early development of embryos. From 237 oocyte donors, 2620 oocytes were collected and their meiotic status and morphology were determined. To determine optimal characteristics of the mature oocytes subjected to nuclear transfer, a proportion of the meiotic status of the oocytes were classified in reference to time post-ovulation as well as progesterone (P4) level. A high proportion of matured oocytes were collected from >126h (55.5%) post-ovulation or 40–50ngmL<SUP>−1</SUP> (46.4%) group compared to the other groups. Of the oocyte donors that provided mature oocytes <I>in vivo</I>, there was no correlation between serum progesterone of donors and time post ovulation, however, time post-ovulation were significantly shorter for <30ng/mL group (P<0.05). Using mature oocytes, 1161 cloned embryos were reconstructed and transferred into 77 surrogates. In order to determine the relationship between pregnancy performance and serum progesterone level, embryos were transferred into surrogates showing various P4 serum levels. The highest pregnancy (31.8%) and live birth cloning efficacy (2.2%) rates were observed when the embryos were transferred into surrogates with circulating P4 levels were from 40 to 50ngmL<SUP>−1</SUP>. In conclusion, measurement of circulating progesterone of female dog could be a suitable an indicator of the optimal time to collect quality oocyte and to select surrogates for cloning.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Observation of the distribution of meiotic maturation state in canids after ovulation. </LI> <LI> Highest number of mature oocytes collected from oocyte donors with serum progesterone concentrations of 40–50ngmL<SUP>−1</SUP>. </LI> <LI> Serum progesterone levels of oocyte donors and surrogates may be used to estimate oocyte quality and determine pregnancy rate potential at the time of oocyte collection and embryo transfer. </LI> </UL> </P>
