        2-Substitiuted Thio- and Amino-5,8-dimethoxy-1,4-naphthoquinones as a Novel Class of Acyl-CoA: Cholestrol Acyltransferase Inhibitors

        A series of 2-alkyl or 2-arylthio-5,8-dimethoxy-1,4-naphthoquinones (2-Thio-DMNQ, 5a-s) and 2-alkylamino-5,8- dimethoxy-1,4-naphthoquinones (2-Amino-DMNQ, 6a-k) was synthesized and evaluated for their ACAT inhibitory activities. Among them, the 2-dodecylthio-DMNQ 5l and 2-isobutylamidoundodecylthio-DMNQ 5r showed the most potent ACAT inhibitory activities with $IC_{50}$ value of 22.8 and 24.4 ${\mu}M$, respectively. In a structure-activity relationship study, 2-thio-DMNQs with side chains of carbon number 11 $\sim$ 15 exhibited significant ACAT inhibitory activities.

        MicroRNA-196b enhances the radiosensitivity of SNU-638 gastric cancer cells by targeting RAD23B

        <P><B>Abstract</B></P> <P>Gastric cancer is characterized by resistance to ionizing radiation. The development of resistance to radiotherapy in gastric cancer patients is one of the obstacles to effective radiotherapy. MicroRNAs are small well-conserved non-coding RNA species that regulate post-transcriptional activation. Our study aimed to investigate the role of miR-196b in radiation-induced gastric cancer. In the present study, we found that miR-196b expression was significantly reduced following radiation. The ectopic miR-196b expression sensitized SNU-638 gastric cancer cells and increased γ-H2AX foci upon radiation treatment. Bioinformatics analysis suggested that the DNA repair protein RAD23B was a putative target gene of miR-196b. Overexpression of miR-196b suppressed RAD23B expression in SNU-638 cells. Reporter assays further showed that miR-196b inhibited RAD23B 3′-UTR luciferase activity. Knockdown of RAD23B by small interfering RNA transfection closely mimicked the outcomes of miR-196b transfection, leading to impaired DNA damage repair in gastric cancer cells. Our results show that miR-196b improved radiosensitivity of SNU-638 cells by targeting RAD23B. Our data indicate that miR-196b is a potential target to enhance the effect of radiation treatment on gastric cancer cells. These findings will provide evidence for a new therapeutic target in radiotherapy.</P>

        春秋战国至西汉初期的“三礼”,蕴含了儒家伦理道德和社会政治思想,能为“具身德育”研究提供滋养,主要体现有三。其一,每个人都应重视内心道德的修养,即便闲居独处也应自律慎独。其二,为人弟子者对待师长和尊者应注重言行细节,在求教、同行、洒扫等情境中务必诚敬谦和。其三,为官者理应对民众施行因地制宜、因时而动的教化,“修六礼、明七教、齐八政”,重视礼对民众身体的规范,促进民风淳善朴实、社会安宁祥和。“三礼”充分考虑了 “身体塑造道德”的事实,其间的德育理念穿越千载。如今我们摒弃迂拘、承袭中正,见证先贤智慧启迪当下、指引未来。 The “Three Rites” from the Spring and Autumn Period and the Warring States Period to the early Western Han Dynasty contain Confucian ethics and social and political thoughts, which can provide nourishment for the study of “embodied moral education”. There are three main manifestations. Firstly, everyone should pay attention to the cultivation of inner morality, even if they live alone, they should be self-disciplined and cautious. Secondly, ,as disciples, they should pay attention to the details of their words and actions when dealing with their teachers and masters, and be sincere and humble in situations such as seeking advice, traveling together, and sweeping. Thirdly, as officials, they should educate the people according to local conditions and the times, “Practice the six rites, clarify the seven teachings, and Doing eight things”, pay attention to the regulation of the people’s bodies by etiquette, and promote the integrity and simplicity of the people and the peace and harmony of the society. The “Three Rites” fully considered the fact that “the body shapes morality”, and the concept of moral education in them has gone through thousands of years. Today, we abandon formality and inherit Zhongzheng, witnessing the wisdom of the sages to enlighten the present and guide the future.

        RNA sequencing reveals that Prx II gene knockout can down-regulate the allograft rejection of dermal mesenchymal stem cells

        In this study, we used RNA sequencing (RNA-seq) to analyze and compare bulk cell samples from wild-type (WT) dermal mesenchymal stem cells (DMSCs) (n = 3) and Prx II knockout DMSCs (n = 3). The purpose of the study was to elucidate the role of Prx II on allogeneic immune rejection of transplanted DMSCs. The results revealed differential expression of 472 genes (176 up-regulated and 296 down-regulated; p ≤ 0.05) between the PrxII+/+ (WT) and PrxII−/− sample groups. When highly regulated genes were categorized according to the Gene Ontology (GO) molecular function classification and the Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis, the PrxII−/− samples showed a robust downward trend in allograft rejection. The study identified 43 all immunologically rejected differentially expressed genes, of which 41 showed lower expression in the PrxII−/− vs. PrxII+/+ (WT) samples. These findings suggest that Prx II gene knockout may down-regulate the allograft rejection that occurs during DMSCs transplantation and improve the survival rate of DMSCs in the host. This study provides a new perspective on the clinical treatment of stem cell transplantation.

        Development of the vapor film thickness correlation in porous corrosion deposits on the cladding in PWR

        The porous corrosion deposits (known as CRUD) adhered to the cladding have an important effect on the heat transfer from fuel rods to coolant in PWRs. The vapor film is the main constituent in the two-phase film boiling model. This paper presents a vapor film thickness correlation, associated with CRUD porosity, CRUD chimney density, CRUD particle size, CRUD thickness and heat flux. The dependences of the vapor film thickness on the various influential factors can be intuitively reflected from this vapor film thickness correlation. The temperature, pressure, and boric acid concentration distributions in CRUD can be well predicted using the two-phase film boiling model coupled with the vapor film thickness correlation. It suggests that the vapor thickness correlation can estimate the vapor film thickness more conveniently than the previously reported vapor thickness calculation methods.

        Synergistic removal of arsanilic acid using adsorption and magnetic separation technique based on Fe3O4@ graphene nanocomposite

        A self-assemble nanocomposite of the nano-Fe3O4 and the reduced graphene oxide with D-glucose as thereductant and FeCl3 and graphene oxide as the pristine materials was prepared by an one-pothydrothermal method. The adsorption capacity of arsanilic acid by the nanocomposite was found to be313.7 11.2 mg g 1, reach the equilibrium within 15 min, and was independent of pH (3–6) and theconcentrations of ammonia nitrogen up to 1000 mg L 1. The adsorption of arsanilic acid followed pseudosecond-order kinetic model and Langmuir isotherm and was an endothermic and spontaneous process.

        Genomic and Transcriptomic Characterization Revealed the High Sensitivity of Targeted Therapy and Immunotherapy in a Subset of Endometrial Stromal Sarcoma

        Purpose The unique chromosomal rearrangements of endometrial stromal sarcoma (ESS) make it possible to distinguish high-grade ESS (HGESS) and low-grade ESS (LGESS) from the molecular perspective. Analysis of ESS at the genomic and transcriptomic levels can help us achieve accurate diagnosis of ESS and provide potential therapy options for ESS patients.Materials and Methods A total of 36 ESS patients who conducted DNA- and/or RNA-based next-generation sequencing were retrospectively enrolled in this study. The molecular characteristics of ESS at genomic and transcriptomic levels, including mutational spectrum, fusion profiles, gene expression and pathway enrichment analysis and features about immune microenvironment were comprehensively explored.Results <i>TP53</i> and <i>DNMT3A</i> mutations were the most frequent mutations. The classical fusions frequently found in <i>HGESS</i> (<i>ZC3H7B-BCOR</i> and <i>NUTM2B-YWHAE</i>) and LGESS (<i>JAZF1-SUZ12</i>) were detected in our cohort. <i>CCND1</i> was significantly up-regulated in HGESS, while the expression of <i>GPER1</i> and <i>PGR</i> encoding estrogen receptor (ER) and progesterone receptor (PR) did not differ significantly between HGESS and LGESS. Actionable mutations enriched in homologous recombination repair, cell cycle, and phosphoinositide 3-kinase/AKT/mammalian target of rapamycin pathways were detected in 60% of HGESS patients. Genes with up-regulated expression in HGESS were significantly enriched in five immune-related pathways. Most HGESS patients (85.7%) had positive predictors of immunotherapy efficacy. Moreover, immune microenvironment analysis showed that HGESS had relatively high immune infiltration. The degree of immune infiltration in HGESS patients with <i>ZC3H7B-BCOR</i> fusion was relatively higher than that of those with <i>NUTM2B-YWHAE</i> fusion.Conclusion This study investigated the molecular characteristics of ESS patients at the genomic and transcriptomic levels and revealed the potentially high sensitivity of targeted therapy and immunotherapy in a subset of HGESS with specific molecular features, providing a basis for guiding decision-making of treatment and the design of future clinical trials on precision therapy.

        본 연구의 목적은 먼저 중국 경제체제 개혁을 단계별로 그의 특징을 살펴보고 중국 경제체제 개혁에 따라 해운의 역할 변화과정을 고찰하며 그 변화 가운데 COSCO의 발전전략의 변화를 연구하는데 있다. 중국의 개방정책과 사회주의 시장경제 도입에 따라 중국의 외항해운 정책구조도 크게 변화되어 왔다. 이러한 환경변화 속에서 중국의 외항해운도 순수한 운송서비스를 제공하는 산업으로 변화해왔으며 해운기업도 정부의 하나의 기구로서 역할하기 보다는 독립적으로 책임을 부담하고 이익창출을 기업목표로 설정하는 독립기업으로 전환되었다. COSCO는 1984년 독립적 기업으로 전환하였고 1993년 기업집단으로 발전하였다. 특히 1993년 이후 COSCO는 선대경영체제의 개혁, 육상산업의 확장, 해외업무의 확대 등 개혁조치를 채택이후 기업은 계획경제체제의 管理方式에서 탈출하여 市場經濟의 관리경영체제로 전환하였다. COSCO는 중국 경제체제 변화에 따라 적절히 발전전략을 조정하여 기업의 경쟁력을 향상하고 있다.
