        Melatonin protects endothelial progenitor L255cells against AGE-induced apoptosis via autophagy flux stimulation and promotes wound healing in diabetic mice

        Wound healing is delayed in diabetic patients. Increased apoptosis and endothelial progenitor cell (EPC) dysfunction are implicated in delayed diabetic wound healing. Melatonin, a major secretory product of the pineal gland, promotes diabetic wound healing; however, its mechanism of action remains unclear. Here, EPCs were isolated from the bone marrow of mice. Treatment of EPCs with melatonin alleviated advanced glycation end product (AGE)-induced apoptosis and cellular dysfunction. We further examined autophagy flux after melatonin treatment and found increased light chain 3 (LC3) and p62 protein levels in AGE-treated EPCs. However, lysosome-associated membrane protein 2 expression was decreased, indicating that autophagy flux was impaired in EPCs treated with AGEs. We then evaluated autophagy flux after melatonin treatment and found that melatonin increased the LC3 levels, but attenuated the accumulation of p62, suggesting a stimulatory effect of melatonin on autophagy flux. Blockage of autophagy flux by chloroquine partially abolished the protective effects of melatonin, indicating that autophagy flux is involved in the protective effects of melatonin. Furthermore, we found that the AMPK/mTOR signaling pathway is involved in autophagy flux stimulation by melatonin. An in vivo study also illustrated that melatonin treatment ameliorated impaired wound healing in a streptozotocin-induced diabetic wound healing model. Thus, our study shows that melatonin protects EPCs against apoptosis and dysfunction via autophagy flux stimulation and ameliorates impaired wound healing in vivo, providing insight into its mechanism of action in diabetic wound healing.

        Intravitreal implantable magnetic micropump for on-demand VEGFR-targeted drug delivery

        <P><B>Abstract</B></P> <P>In this paper, we propose an intravitreal implantable magnetic micropump integrated with micro check valve capable of on-demand vascular endothelial growth factor receptor (VEGFR)-targeted drug delivery for the treatment of age-related macular degeneration, diabetic retinopathy and other eye pathologies characterized by ocular neoangiogenesis. Precise on-demand drug release is realized by the deflection of the magnetic membrane assembly according to the external magnetic field, and the membrane assembly consists of a thin elastic polydimethylsiloxane (PDMS) membrane and a cylindrical magnetic nanoparticle-PDMS composite block. Additionally, a micro check valve composed of two PDMS layers was integrated into the micropump to realize a diode-like one-directional drug delivery and prevent undesired drug diffusion. For specifically targeting VEGFR and suppression of VEGF-induced proliferation of microvascular endothelial cells, anti-Flt1 gold nanocomplexes are synthesized. <I>In vitro</I> and <I>in vivo</I> experiments and quantitative analysis are carried out in order to verify our proposed concept: precise drug release control according to the external magnetic field, targeting to microvascular endothelial cells, and efficient and on-demand drug delivery from the proposed micropump to the macular area of rabbit's eye.</P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

        Adaptive robust target tracking control of marching tank under high-speed maneuvering condition

        This paper focuses on the problem of target tracking control of marching tanks under the condition of high-speed maneuver based on sliding mode disturbance observer. First, the dynamic model of marching tank is established by using multi-body dynamics software. The nonlinear factors such as barrel flexibility, clearance between barrel and bushing, and uncertainty sources such as road excitation are considered. Second, the mechatronics model of tank bidirectional stabilization system considering transmission clearance and friction is established in numerical simulation software. Aiming at the tank bidirectional stabilization system, an adaptive robust controller based on sliding mode disturbance observer is designed. Finally, the real-time control and target tracking of the tank bidirectional stabilization system are realized by co-simulation. The results show that the sliding mode disturbance observer improves the stability accuracy of the system by 43.22 %. Comparing the simulation results with the PID control, the controller designed in this paper has the characteristics of rapid tracking speed and high robustness, and the advantage is more obvious under the condition of highspeed maneuver.

        Principle Issues and Future Prospect on Sliding Arc Ablation of Metal Rail

        Arc ablation occurs when the armature passes over the surface of metal rail. Ablation not only seriously afects the service life, but also may cause the failure to launch. In this paper, the research status of sliding arc ablation technology is summarized from the mechanism of sliding arc ablation, the characterization method of sliding arc ablation degree, and the enhanced design of materials or structures. Three key issues that need to be further solved are listed: the infuence mechanism of sliding arc ablation on microstructure and properties of metal materials, the service properties evaluation and quantitative characterization methods of metal material, as well as the modifed and enhanced design of materials or structures. In response to those problems, eforts are needed to improve the awareness of sliding arc ablation with the theoretical basis and characterization methods, as well as guide the improvement and strengthening of rail design.

        Genome-wide locus-specific DNA methylation repatterning may facilitate rapid evolution of mercury resistance in rice

        Background: Albeit a relatively stable epigenetic modification, DNA methylation in plants can be repatterned and play important roles in response to biotic and abiotic stresses. However, whether DNA methylation dynamics may contribute to cope with mercury (Hg) stress in plants remains to be fully investigated. Objective: To probe the potential roles of DNA methylation dynamics in coping with Hg stress in rice. Methods: Whole-genome bisulfite sequencing was used to profile the DNA methylation patterns of a rice Hg-resistant line (RHg) selected from a heterozygous mutant of the DNA methyltransferase 1 gene (OsMET1+/-), together with its Hg-sensitive wild-type plants of cv. Nipponbare (Nip) under both normal and Hg stress conditions. Results: Genome-wide locus-specific differential methylation regions (DMRs) were detected between RHg and Nip under normal condition, the predominant DMR patterns were CG hypo-DMRs, CHG hypo-DMRs and CHH hyper-DMRs. In both lines, more hyper- than hypo-DMRs were detected at all three sequence contexts (CG, CHG and CHH) under Hg stress relative to normal condition. Comparison of DNA methylation changes in the two lines under Hg stress indicates that RHg had a more dynamic methylome than the control (Nip). Original DMRs in RHg trended to transform to opposite status (from hyper- to hypo- or vice versa) under Hg stress condition. Gene ontology analysis revealed that Hg-resistance-related DMGs were enriched in diverse biological processes. Conclusions: Our results suggest genome-wide locus-specific DNA methylation repatterning can facilitate rapid acquisition of Hg resistance in rice.

      • Comparative analysis of nano-scale structural and electrical properties in AlGaN/GaN high electron mobility transistors on SiC and sapphire substrates.

        <P>A comparison of AIGaN/GaN HEMTs fabricated on both 4-in SiC and sapphire substrates was performed. Due to the high crystalline quality with one order lower dislocation density of GaN on a SiC substrate, a better two-dimensional electron gas (2DEG) mobility with high values of drain current density (780 mA/mm) and a better extrinsic transconductance (240 mS/mm) were observed. We demonstrate GaN-on-SiC HEMTs with a periphery gate width of 200 microm, exhibiting a unity-gain cut-off frequency (f(T)) = 29.6 GHz, a maximum frequency of oscillation (f(MAX)) = 63.2 GHz, and an output power density of 6.4 W/mm with a 55% power added efficiency (PAE) at 10 GHz. A surface roughness of 0.828 nm and 1.025 nm and an X-ray diffraction (XRD) GaN (0002) full-width at half-maximum (FWHM) of 120 s and 919 s were measured for the SiC and sapphire-based AIGaN/GaN HEMTs, respectively. The SiC substrate has been shown to be an optimal solution for fabricating HEMTs for X-band high-power applications, which require excellent performances.</P>

      • Association of the XRCC1 c.1178G>A Genetic Polymorphism with Lung Cancer Risk in Chinese

        The X-ray repair cross-complementing group 1 protein (XRCC1) plays important roles in the DNA base excision repair pathway which may influence the development of lung cancer. This study aimed to evaluate the potential association of the XRCC1 c.1178G>A genetic polymorphism with lung cancer risk. The created restriction site-polymerase chain reaction (CRS-PCR) and DNA sequencing methods were utilized to evaluate the XRCC1 c.1178G>A genetic polymorphism among 376 lung cancer patients and 379 controls. Associations between the genetic polymorphism and lung cancer risk were determined with an unconditional logistic regression model. Our data suggested that the distribution of allele and genotype in lung cancer patients was significantly different from that of controls. The XRCC1 c.1178G>A genetic polymorphism was associated with an increased risk of lung cancer (AA vs GG: OR=2.91, 95%CI 1.70-4.98, p<0.001; A vs G: OR=1.52, 95%CI 1.22-1.90, p<0.001). The allele A and genotype AA may contribute to risk of lung cancer. These preliminary results suggested that the XRCC1 c.1178G>A genetic polymorphism is statistically associated with lung cancer risk in the Chinese population.

      • On the receding contact plane problem for bi-FGM-layers indented by a flat indenter

        The major objective of this paper is to study the receding contact problem between two functional graded layers under a flat indenter. The gravity is assumed negligible, and the shear moduli of both layers are assumed to vary exponentially along the thickness direction. In the absence of body forces, the problem is reduced to a system of Fredholm singular integral equations with the contact pressure and contact size as unknowns via Fourier integral transform, which is transformed into an algebraic one by the Gauss–Chebyshev quadratures and polynomials of both the first and second kinds. Then, an iterative speediest descending algorithm is proposed to numerically solve the system of algebraic equations. Both semi-analytical and finite element method, FEM solutions for the presented problem validate each other. To improve the accuracy of the numerical result of FEM, a graded FEM solution is performed to simulate the FGM mechanical characteristics. The results reveal the potential links between the contact stress/size and the indenter size, the thickness, as well as some other material properties of FGM.

      • Application and Simulation of Distribution Function in Grape Drying Process

        In order to investigate the influence factors of Weibull distribution function and its application in drying, the drying process of grape slices in different microwave vacuum drying power (330, 460, 590W) and vacuum degree (-50, -70, -90kPa) was taken as the research object, and the Weibull distribution function was used to simulate and analyze its drying kinetic curves, the results showed that the Weibull distribution function was able to simulate microwave vacuum drying process of grape slice well: scaling parameterαwas related with microwave power and vacuum degree, and decreased with the rise of them; the influence of drying variable on the form parameterβwas relatively low. The estimated effective moisture diffusivity coefficient in the dry process was obtained by calculating, its value increased with the rise of microwave power and vacuum degree in the scope from 3.45078×10-7 to 6.74613×10-7m2/s; through Arrhenius equation, the activation energy of drying was calculated respectively 1.34701, 1.49099 and 1.57108 W/g under the vacuum degree of -50, -70 and -90kPa. This study has provided the technical basis for the application of Weibull distribution function in microwave vacuum drying technology of grape slices.

      • Study of the color features of the traditional clothing of the Tibet Autonomous Region of China

        China is a multiracial nation consisting of the Han majority and 55 ethnic minorities, each of which possesses unique cultural characteristics. Among them, the Tibetan Plateau, Sichuan province, Yunnan Province and the current Chinese Tibet Autonomous Region have a long history, traditional culture and a wide range of natural environments. As an example, the Hollywood blockbuster <2012> that shows "the fall of mankind” has received a lot of criticism as the United States acknowledges the domination of the Tibet. However, people are reminded of the Tibet at the end when it becomes the last area with survivors. Due to these characteristics, Tibetan politics hold an important place in China's politics, society, economics and culture. The study uses empirical research methods by analyzing the literature of the Tibetan people. The study of their literature helps to understand the culture of Tibet and to interpret the meaning behind the uses of different colors. The empirical study uses pictures of Tibetan traditional clothes. A total of 85 photos were used for the study of the color palette, which belonged to the Google Art & Design Museum, the Museum of Photography, and the Photo-Friendly Exhibition Hall. By analyzing the results, we can see the use of all the hue-tone colors. Among them, red, orange, yellow, green, indigo, and purple are more likely to be used than other colors. From all the hues, it is easily appreciated that the V hues were the most used. In addition, many color applications were used as well. These different senses of color represent different religions, whose different characteristics appear also on the traditional clothes. Red represents the color of fire and is regarded as a symbol of power. When thinking about red flags in Tibet, they think of the dragon as a symbolic color. Amber also represents the Buddhist temple of Buddhism. Dark blue symbolizes the eldest son, or Chang Cheong, being the most frequently worn color of the Tibetan region. Dark blue represents mystery and the plateau, with the color of blue skies and lakes. Green is the most common color, meaning vitality in the color of grass. Green color is closer to the public, and it is more frequent in a wide range of nomadic areas. Gold and silver represent wealth and dignity, and clothes with those colors represent the wealth and riches of the family. Since white symbolizes life and luck, it is one of the most frequently used colors. Black is one of the most sophisticated colors in people's lives. Most of the great men and women are inseparably related. There are black tents used in the homes of nomads, the black border of Tibet, and a pair of black cranes made by men and women in rural areas.
