        3D Numerical Simulation of Ice Accretion on a Rotating Surface

        A novel 3D mathematical model for water film runback and icing on a rotating surface is established in this work, where both inertial forces caused by the rotation and shear forces due to the air flow are taken into account. The mathematical model of the water film runback and energy conservation of phase transition process is established, with a cyclical average method applied to simulate the unsteady parameters variation at angles of attack. Ice accretion on a conical spinner surface is simulated and the results are compared with the experimental data to validate the presented model. Then Ice accretion on a cowling surface is numerically investigated. Results show that a higher temperature would correspond to a larger runback ice area and thinner ice layer for glaze ice. Rotation would enhance the icing process, while it would not significantly affect the droplet collection efficiency for an axi-symmetric surface. In the case at angle of attack, the effect of rotation on ice shape is appreciable, ice would present a symmetric shape, while in a stationary case the shape is asymmetric.

        Anti-inflammation compounds from the seedpods of Pongamia pinnata (L.) Pierre guided by the bioactivity and UPLCHRESIMS

        Pongamia pinnata (Linn.) Pierre has anti-inflammatory activity and could significantly decrease serum tumor necrosis factor-a and IL-10 in arthritic rats. Previous research indicated the typical chemical constituent in P. pinnata is furanoflavone. Guided by anti-inflammatory active assay and UPLC-HRESIMS chromatography, 22 compounds were isolated from the ethanol extract of P. pinnata seedpods. One novel furanoflavone, 40-hydroxypinnatin, was elucidated by HRESIMS, 1D- and 2D-NMR spectra. The 21 known compounds, including 9 furanoflavone, were identified by comparing their NMR data with the previous data in reference. In the known compounds, 5 were isolated for the first time from the species. The anti-inflammatory activities were assayed by assessing LPS-induced NO production in BV-2 cells. 12 compounds can inhibit the production of NO without cytotoxicity at concentration of 50 lM. Among them, compounds 4 can significantly inhibit the production of NO, with the IC50 value of 31.36 lM.

        Melatonin protects endothelial progenitor L255cells against AGE-induced apoptosis via autophagy flux stimulation and promotes wound healing in diabetic mice

        Wound healing is delayed in diabetic patients. Increased apoptosis and endothelial progenitor cell (EPC) dysfunction are implicated in delayed diabetic wound healing. Melatonin, a major secretory product of the pineal gland, promotes diabetic wound healing; however, its mechanism of action remains unclear. Here, EPCs were isolated from the bone marrow of mice. Treatment of EPCs with melatonin alleviated advanced glycation end product (AGE)-induced apoptosis and cellular dysfunction. We further examined autophagy flux after melatonin treatment and found increased light chain 3 (LC3) and p62 protein levels in AGE-treated EPCs. However, lysosome-associated membrane protein 2 expression was decreased, indicating that autophagy flux was impaired in EPCs treated with AGEs. We then evaluated autophagy flux after melatonin treatment and found that melatonin increased the LC3 levels, but attenuated the accumulation of p62, suggesting a stimulatory effect of melatonin on autophagy flux. Blockage of autophagy flux by chloroquine partially abolished the protective effects of melatonin, indicating that autophagy flux is involved in the protective effects of melatonin. Furthermore, we found that the AMPK/mTOR signaling pathway is involved in autophagy flux stimulation by melatonin. An in vivo study also illustrated that melatonin treatment ameliorated impaired wound healing in a streptozotocin-induced diabetic wound healing model. Thus, our study shows that melatonin protects EPCs against apoptosis and dysfunction via autophagy flux stimulation and ameliorates impaired wound healing in vivo, providing insight into its mechanism of action in diabetic wound healing.
