        A study for development of natural red pigment in food and drug industry was proceeded to anthocyanin from prunus yedoensis Matsum, isolated through silica gel column and further subjected to TLC. The color effect and stability from several factors was detected by spectrophotometry in 520 nm and was compared to amaranth and then 1/200 times of it. The content of the color of the fruits of Prunus yedoensis Matsum was 0.6%. As this pigment was more safe about 7.4 times in ??,but was influenced by pH(3.0-8.0) and by some metal ions (??, ??, ??, ??),and respectively stable to temperature, sun light, ultra violet ray and organic acids than amaranth, and was not given a bad infouences by sugars, recovery of another characteristics of this pigment or co-factors to compensate and promote the color effect, for example Cu(), is expected.

      • Larson-Miller 법을 이용한 보일러튜브의 크립 잔존수명에 관한 연구

        The boiler components are operated at high temperature, and their materials are subject to the time dependent creep deformation. The materials eventually reach the fracture stage due to the creep damage, and thus the designer should pay attention to the creep damage, and thus the designer should pay attention to the creep lifetime to prevent the catastrophic fracture of those components. In this study, test specimens were prepared from the primary superheater in Yousu power plant which have been used for 11 years, and the creep tests were performed to measure the remaining lifetime. To predict the remaining lifetime of used materials, an equation was proposed by using the Larson-Miller parameter which can estimate the lifetime of unused materials. The primary superheater was turned out to be risky, and the remaining lifetime was predicted to be about 2 years. The fractography captured by the scanning electron microscope showed that the fracture was transgranular with dimples for all the specimens.

        Image-based three dimensional radiation treatment planning(3D RTP) has a potential of generating superior treatment plans. Advances in computer technology and software developments quickly make 3D RTP a feasible choice for routine clinical use. However, it has become clear that evaluation of a 3D plan is more difficult than a 2D plan. A number of tools has been developed to facilitate evaluation of 3D RTP both qualitatively and quantitatively. For example, beam's eye view(BEV) is one of the most powerful and time-saving method as a qualitative tool. Dose-volume histogram(DVH) has been proven to be one of the most valuable method for a quantitative tool. But it has a limitation to evaluate several different plans for biological effects of the tissue and critical organ. Therefore. there is a strong interest in developing quantitative models which would predict the likely biological response of irradiated organs and tissues. such as tumor control probability(TCP) and normal tissue complication probability(NTCP). DVH and NTCP of hepatoma were evaluated for three dimensional conformal radiotherapy(3D CRT). Also, 3D RTP was analysed as a dose optimization based on beam arrangement and beam modulation. The value of NTCP in patients with hepatitis was higher than that in patients without hepatitis. Also the value of n =0.32(volume factor) was highly correlated to prediction of radiation hepatitis. Therefore, we concluded that the possibility of radiation hepatitis was highly suggested if NTCP was over 40%.

        In current design, the strength formula for a Gusset-Tube connections stiffened with rib-plate is proposed only in the limited case because of its complexity of the behavior. So in this study, experimental study and finite element analysis were performed to investigate the behavior and strength of a Gusset-Tube connections stiffened with rib-plate. Because of limitations in loading capacity, the specimens were fabricated with 1/3 scale of full size. Tensile coupon tests and stub column tests were also carried out. Of primary interest here are the stiffened effect of Gusset-Tube connections considering lateral force ratio, gusset plate length, eccentricity, stiffened length. Consequently, we intend to obtain accumulated database to propose practical strength equation and suggest rational reinforcing method of Gusset-tube connection.

      • 고온·알칼리성 Bacillus sp.의 Catabolite Repression 저항성 변이주로부터 Cellulase 및 Xylanase의 생산

        고온·알칼리성 cellulase 및 hemicellulase 생성 미생물을 분리할 목적으로 pH 10.3의 PY-CMC배지에서 55℃로 3회 연속 직접배양하여 효소 생성능이 우수한 20여균주를 분리하였고, 그 중에서 2개균주를 최종 선정하여 균학적 성상을 조사한 결과 고온·알칼리성 Bacillus속과 거의 일치하였다. 선정, 동정된 고온·알칼리성 Bacillus sp. F204와 F1020은 α-1, 4-glucose 등에서는 효소 생합성이 왕성하였으나, glucose와 같은 단당류에서는 효소가 전혀 유도되지 않았는데, 이 두 균주는 carbon catabolote repression을 받고 있었다. 2-deoxy glucose는 야생주에서는 강력한 repressor로 작용하였고, 야생주의 catabolite repression을 해제 시키는데 antimetabolite로 작용한다는 것이 밝혀졌다. NTG(500㎍/㎖, 50℃, 20∼60분)와 UV(2분) 처리후 2-DG(20mM)에 직접배양하여 강력한 catabolite repression resistant strain인 M-211과 M-367을 분리하였다. M-211과 M-367은 2-DG를 energy원으로 이용하였으며, glucose에 대하여 CMCase는 1.5%, avicelase 및 xylanase는 약 1.0%까지 저항성을 보였다. 또한 두개의 변이주의 효소 활성은 CMCase 2배, avicelase 1.5배 및 xylanase3배의 증가를 보였으며, 각 효소생합성을 위한 gene expression은 cAMP-independent transcription을 하는 것으로 나타났다. Cellulase and xylanase synthesis of thermophilic alkalophilic Bacillus sp. F204 and F1020 isolated in our lab, were repressed by glucose and induced by compounds structurally related to α-1,4 linkage (avicel cellulose and xylan et. al.). Catabolite repression-resistant mutants were obtained through treatment of NTG(500㎍./㎖), UV(2 min) and enrichment culture in 2-deoxy glucose (20 mM) containing PY-glucose on the wild type strain. T재 catabolite repression resistant mutants were not repressed by glucose and produced cellulase complex and xylansase when in grown in the presence of 1.0% glucose and 30mM 2-DC, which were repressors in wild type. Enzyme production was greater with the mutants (CMCase of 2 times, avicelase of 1.5 times, xylanase of 3 times) than with the wild type strains under various test conditions. T재 mutants M-211 and M-367 were potential catabolite repression resistant mutants and their gene expression was cAMP independent transcription.

      • 전단보강근량에 따른 초고강도 콘크리트를 사용한 고층형 내력벽의 이력거동

        Three one fourth scale models using ultra high-strength concrete(f' =704 kg/㎠) are tested under the combined action of a constant axial and a horizontal load cyclically to failure. Such specimens are considered to represent the critical 3-story of low part in 60-story tall building of a structural wall system in area of high seismicity. The amount of vertical reinforcement and the level of applied axial stress are identical for the three wall tested. The cross-section of all walls is barbell shape. The aspect ratio(h /l ) of test specimen is 1.8. The primary objectives of this paper are to investigate the influence of the amount of horizontal reinforcement on the lateral resistance, failure mechanism, ductility and energy-dissipation capability of walls with ultra high-strength concrete. In contrast to what is widely believed, the horizontal web reinforcement does not appear to have a significant effect on shear capacity. Certainly, since the reduction of the web horizontal reinforcement to almost half the value specified by building codes doesn't affect the failure load, this effect in not accounted for by the truss analogy concept. The results obtained have helped to identify the causes of wall failure and have demonstrated that the concepts underlying current ACI Building Code provisions for the design of walls conflict with the observed structural behavior. It has been found that shear resistance is associated with triaxial compressive stress conditions that develop in the compressive zone of the section at the base of the wall.

        흉수에 발생한 상의신경세포종 1예

        We have recently experienced a case of intramedullary ependymoma in the upper thoracic spinal cord. A 46 years old man was admitted to our Department of Neurosurgery because of spastic paraplegia and disturbance of .urination since 6 years ago when the disability developed rather abruptly. Myelogram showed a subtotal block of the dye column at the level of T2-3 intervertebral space about where a fusiform enlargement of the cord was shadowed. Opening the duramater after total laminectomy performed through C7 to T3, a markedly swallen and enlarged spinal cord was under a great pressure. Spinal cord was paper thin and fully filled with a massive tumor inside of it. The intrameduallary tumor was mushroomed out on dorsal myelotomy. Histological study was verified to be ependymoma. The patient has not shown any improvement of neurological deficits in postoperative period.

        17α-hydroxylase 결핍증에 의한 원발성 무월경 1예

        The enzyme, 17α-hydroxylase, is necessary for both cortisol and estrogen synthesis. Deficiency of the hormone results in increased adrenocorticotrophic hormone (ACTH), follicle-stimulating hormone (FSH). Synthesis of progesterone, 11-deoxycorticosterone (DOC), corticosterone and aldosterone don't require the enzyme. The lack of estrogen results in primary amenorrhea and absent sexual maturation. The replacement of dexamethasone and estrogens has lowered the blood pressure and produced feminization. A 19-year-old female had 46,XX genotype and presented amonorrhea, absence of sexual characteristics, hypertension and hypokalemia. Endocrinologic studies demonstrated increased plasma progesterone, ACTH levels and low production of 17α-hydroxyprogesterone and testosterone. We report a rare case of 17α-hydroxylase deficency with a brief history and review of the literature.

        현미경적 폐전이를 동반한 갑상선 유두암 1예

        Although the papillary thyroid carcinoma generally follows an indolent course characterized by slow growth and abscence of distant metastasis, distant metastases to extrathyroid organ may occur in the lung, bone and brain in about 5% of patients. The lung metastases are usually diagnosed by plain X-ray, chest CT and 131I whole body scan. However, we present a case of papillary thyroid carcinoma with microscopic pulmonary metastasis that could not be detected by conventional method, such as chest X-ray, chest CT and 131I whole body scan. A 62-year-old female visited due to traumatic hemopneumothorax. In the pathology of resected tissues from lacerated lung parenchyme, metastatic papillary carcinoma was diagnosed without evidence of abnormality in radiologic examination and we diagnosed and treated an asymptomatic papillary thyroid carcinoma.
