Background/Aims: SKI306X, a mixed extract of three herbs, Clematis mandshurica(CM), Prunella vulgaris (PV), and Trichosanthes kirilowii (TK), is chondroprotective inanimal models of osteoarthritis (OA). The objectives of this study were to investigateits effect on interleukin (IL)-1β-induced degradation of glycosaminoglycan(GAG) and the basis of its action in human OA cartilage, as well as to screen forthe presence of inhibitors of matrix metalloproteinase (MMP)-13 and a disintegrinand metalloprotease with thrombospondin motifs (ADAMTS)-4 in SKI306Xand its component herbs, as well as in fractions from SKI306X. Methods: Human OA chondrocytes and cartilage explants were obtained duringtotal knee replacements and incubated with IL-1β ± oncostatin M with or withoutSKI306X or its component herb extracts. GAG degradation was assayed in cartilageexplants using a commercial kit. Expression of genes involved in cartilagedestruction was measured by real-time polymerase chain reaction using chondrocyteRNA. SKI306X was fractionated by preparative liquid chromatography totest for the presence of inhibitors of MMP-13 and ADAMTS-4. Results: SKI306X and PV inhibited IL-1β-induced GAG release from cartilage explants,and SKI306X, CM, PV, and TK inhibited IL-1β-induced MMP gene expression. Unexpectedly, SKI306X greatly stimulated IL-1β + oncostatin M-inducedADAMTS-4 gene expression, probably due to its TK component. Some fractionsof SKI306X also inhibited ADAMTS-4 activity. Conclusions: SKI306X and its herbal components inhibit GAG degradation andcatabolic gene expression in human OA chondrocytes and cartilage explants. SKI306X likely also contains one or more ADAMTS-4 inhibitor.

Background/Aims: Ampullary adenomyoma is a benign lesion whose malignant potential has yet to be confirmed. Despite its benign nature, adenomyoma is frequently misdiagnosed as a carcinoma or adenoma and is overtreated by extensive surgery. This study was performed to analyze the clinical, pathological, and immunohistochemical features of adenomyomas in the ampulla of Vater. Methods: Nine cases of adenomyoma in the ampulla of Vater, diagnosed in Chungbuk National University Hospital between 2008 and 2011, were enrolled in this study. We reviewed the clinical data on the symptoms, laboratory data, and radiologic findings of the abdominal computed tomography and endoscopic retrograde cholangiopancreatography. For pathological analysis, all the slides were reviewed by one pathologist, and immunohistochemical stainings with antibodies against cytokeratin 7 (CK7), cytokeratin 20 (CK20), α-smooth muscle actin (α-SMA), and Ki-67 antigen were performed. Results: All the cases were CK7 positive and CK20 negative. A strong cytoplasmic expression of α-SMA was confirmed in all cases. The Ki-67 index was less than 1% in eight cases and 5% in one case. Four cases underwent endoscopic papillectomy, and one case received surgical ampullectomy during colorectal cancer surgery. Five cases that underwent endoscopic or surgical treatment remained symptom-free for three years. Four cases that were closely observed with repeated endoscopic examinations exhibited no interval changes in the papillary lesions. Conclusions: Endoscopic biopsy and immunohistochemistry can aid in the diagnosis of ampullary adenomyomas. Endoscopic papillectomy or surgical ampullectomy is adequate for the treatment of symptomatic ampullary adenomyomas. (Korean J Gastroenterol 2013;62:352-358)

In this paper the experiments of breakdown characteristics by temperature change of $SF_6$ gas($GSF_6$), and $SF_6$ liquid ($LSF_6$) in model GIS (Gas Insulated Switchgear) are described. From the experiment's results, the breakdown characteristics classify the vapor stage of $SF_6$ according to Paschen's law, in which the gas & liquid coexisted stage of voltage value increases, resulting in much deviation and the breakdown of voltage ($V_B$) low stage as the interior of the chamber gets filled with a mixture of $SF_6$ that is not liquefacted and remaining air that cannot be ventilated. The ability of $LSF_6$ insulation is higher than the high-pressurized $SF_6$ gas. The breakdown characteristics of $LSF_6$ were produced by bubble formed evaporation of $LSF_6$ and bubbles caused by high electric emission. It is considered in this paper that the results are fundamental data for electric insulation design of superconductor and cryogenic equipments machinery that will be studied and developed in the future.
