        piRNA-1742 promotes renal cell carcinoma malignancy by regulating USP8 stability through binding to hnRNPU and thereby inhibiting MUC12 ubiquitination

        Accumulating studies have confirmed that PIWI-interacting RNAs (piRNAs) are considered epigenetic effectors in cancer. We performed piRNA microarray expression analysis on renal cell carcinoma (RCC) tumor tissues and paired normal tissues and performed a series of in vivo and in vitro experiments to explore piRNAs associated with RCC progression and investigate their functional mechanisms. We found that piR-1742 was highly expressed in RCC tumors and that patients with high piR-1742 expression had a poor prognosis. Inhibition of piR-1742 significantly reduced tumor growth in RCC xenograft and organoid models. Mechanistically, piRNA-1742 regulates the stability of USP8 mRNA by binding directly to hnRNPU, which acts as a deubiquitinating enzyme that inhibits the ubiquitination of MUC12 and promotes the development of malignant RCC. Subsequently, nanotherapeutic systems loaded with piRNA-1742 inhibitors were found to effectively inhibit the metastasis and growth of RCC in vivo. Therefore, this study highlights the functional importance of piRNA-related ubiquitination in RCC and demonstrates the development of a related nanotherapeutic system, possibly contributing to the development of therapeutic approaches for RCC.

        Comparison of Liver Transplantation and Liver Resection for Hepatocellular Carcinoma Patients with Portal Vein Tumor Thrombus Type I and Type II

        Purpose: The aim of this study was to compare the efficacy of liver transplantation (LT) and liver resection (LR) for hepatocellularcarcinoma (HCC) patients with portal vein tumor thrombus (PVTT) and to investigate risk factors affecting prognosis. Materials and Methods: A total of 94 HCC patients with PVTT type I (segmental PVTT) and PVTT type II (lobar PVTT) were involvedand divided into LR (n=47) and LT groups (n=47). Recurrence-free survival (RFS) and overall survival (OS) were comparedbefore and after inverse probability of treatment weighting (IPTW). Prognostic factors for RFS and OS were explored. Results: Two treatment groups were well-balanced using IPTW. In the entire cohort, LT provided a better prognosis than LR. Among patients with PVTT type I, RFS was better with LT (p=0.039); OS was not different significantly between LT and LR(p=0.093). In subgroup analysis of PVTT type I patients with α-fetoprotein (AFP) levels >200 ng/mL, LT elicited significantly longermedian RFS (18.0 months vs. 2.1 months, p=0.022) and relatively longer median OS time (23.6 months vs. 9.8 months, p=0.065). Among patients with PVTT type II, no significant differences in RFS and OS were found between LT and LR (p=0.115 and 0.335,respectively). Multivariate analyses showed treatment allocation (LR), tumor size (>5 cm), AFP and aspartate aminotransferase(AST) levels to be risk factors of RFS and treatment allocation (LR), AFP and AST as risk factors for OS. Conclusion: LT appeared to afford a better prognosis for HCC with PVTT type I than LR, especially in patients with AFP levels>200 ng/mL.

        Surface and foam property of perfluoroalkyl polyoxyethylene ether phosphate salt in aqueous-ethanol system

        Foam is used in manyfields with excellent performance. Moreover, disinfectants with ethanol as themain ingredient have increasingly been used in people's lives. However, the foam research of an ethanolwatermixed system is still in the initial stage. In this work, a perfluoroalkyl polyoxyethylene etherphosphate salt surfactant (FEP-Na) was synthesized through a new molecular design. The surfactant hasgood interfacial adsorption behavior in 50% ethanol-water mixing system, and the approximatelyspherical aggregates are observed in the volume phase of the solution. Moreover, FEP-Na showed goodfoaming property in 50% ethanol-water mixed solution. Interestingly, high concentration solutions withhigh surface activity have lower foam stability. And under the microscope, we see that the bubble burst isdominated by the phenomenon of coalescence. In this regard, we explain the mechanism of foamstability by electrostatic double layer theory and interfacial dilation rheology. It provides a new choice forthe synthesis of foaming agent in the ethanol-water mixed system at a concentration of 50%.

      • A Novel Method for Thermal Analysis of MF Transformers with Hollow Windings

        One of the key problems in the research and development of high-power electronic transformer is the thermal analysis of high-frequency or medium frequency transformer under non-sinusoidal load. At present, there is a lack of a convenient and effective method. Taking the thermal problem of an medium frequency transformer with a hollow conductor structure as an example, this paper presents an analysis method suitable for researchers. Firstly, based on the finite element model, the loss of hollow winding is studied by numerical analysis. Then, based on the similarity principle, the equivalent reduction method for calculating the core loss of medium frequency transformer is presented. Finally, the measurement data of the magnetic properties of the transformer core and the measurement data of the transformer loss under the medium frequency excitation are given. The complete research work can provide a method for the thermal analysis of high frequency or medium frequency transformer.

        Experimental and numerical analysis of unsteady pressure pulsation in a centrifugal pump with slope volute

        This study experimentally and numerically investigates the unsteady flow in a centrifugal pump with special slope volute under variousconditions to illustrate the detailed flow structures and pressure pulsation within the model pump. Whole flow passage is consideredduring the numerical simulation; pressure pulsation signals are extracted using nine fast-response pressure transducers. The Root meansquare (RMS) method is introduced to deal with the discrete components at fBPF of the different monitoring points along the volute casing,which is an effective attempt to evaluate the overall pulsating level of the model pump. Results show that numerical method canpredict the components at fBPF effectively; however, it has limited ability in capturing noise frequencies motivated by unsteady separateflow and non-linear interaction effect. Around the nominal flow rate, the predicted amplitudes at fBPF agree well with the experimentalresults, showing larger difference at the off-design conditions. To predict the pulsating level of the components at fBPF, two fitted equationsof the RMS values versus the flow rate and specific speed are carried out, which would be very helpful in evaluating the pressurepulsation level in the centrifugal pump.

        Particle image velocimetry measurement of complex flow structures in the diffuser and spherical casing of a reactor coolant pump

        Understanding of turbulent flow in the reactor coolant pump (RCP) is a premise of the optimal design ofthe RCP. Flow structures in the RCP, in view of the specially devised spherical casing, are more complicatedthan those associated with conventional pumps. Hitherto, knowledge of the flow characteristics ofthe RCP has been far from sufficient. Research into the nonintrusive measurement of the internal flow ofthe RCP has rarely been reported. In the present study, flow measurement using particle imagevelocimetry is implemented to reveal flow features of the RCP model. Velocity and vorticity distributionsin the diffuser and spherical casing are obtained. The results illuminate the complexity of the flows in theRCP. Near the lower end of the discharge nozzle, three-dimensional swirling flows and flow separationare evident. In the diffuser, the imparity of the velocity profile with respect to different axial crosssections is verified, and the velocity increases gradually from the shroud to the hub. In the casing, velocitydistribution is nonuniform over the circumferential direction. Vortices shed consistently from thediffuser blade trailing edge. The experimental results lend sound support for the optimal design of theRCP and provide validation of relevant numerical algorithms.

        Exploring the Heat Treatment Parameters, Microstructural Evolution, and Mechanical Properties of Ti–6Al–4V Alloy Fabricated by a Selective Laser Melting Process

        The effects of heat treatment parameters on the mechanical properties of a selective laser melting Ti–6Al–4V alloy werestudied using an L9 (34) type orthogonal test. Results showed that the solution temperature and aging temperature were themost influential factors relative to the mechanical properties. The optimum heat treatment was then obtained as 920 °C/2 h/water quenching + 550 °C/3 h/air cooling and the corresponding tensile strength was 1045.2 MPa and the elongation was13.6%. Compared to a solution temperature of 850 °C, more fine secondary α phase (αS) and higher geometrically necessarydislocations (GND) density were obtained when solution temperature was 920 °C, which promoted the increased strengthby increasing the interface strengthening effect of αS/β and the dislocation strengthening. However, the microstructure of thecontinuously coarsening αGB resulted in decreased elongation when the solution was at 920 °C. As the aging temperatureincreased from 550 to 650 °C, the GND density was reduced and the microstructures of primary α (αP), αS with variouscrystallographic orientations, lamellar α, and equiaxial αP were coarsened. Therefore, the elongation of the specimen agingat a higher aging temperature increased while the strength decreased.

        Theoretical Analysis and Optimization of Extrinsic Fabry-Perot Interferometer Optical-fiber Humidity-sensor Structures

        The theoretical analysis and optimization of extrinsic Fabry-Perot interferometer (EFPI) opticalfiber humidity sensors are deeply investigated. For a typical dual-cavity structure composed of an optical fiber and a humidity-sensitive membrane (HSM), the changes in refractive index (RI) and initial length are discussed for polymer materials and porous oxide materials when relative humidity (RH) increases. The typical interference spectrum is simulated at different RH using MATLAB. The spectral change caused by changing HSM RI and initial length are simulated simutineously, showing different influences on humidity response. To deeply investigate the influence on RH sensitivity, the typical response sensitivity curves for different HSM lengths and air-cavity lengths are simulated. The results show that the HSM is the vital factor. Short HSM length can improve the sensitivity, but for HSM RI and length the influences on sensitivity are opposite, because of the opposite spectral-shift trend. Deep discussion and an optimization method are provided to solve this problem. According to analysis, an opaque HSM is helpful to improve sensitivity. Furthermore, if using an opaque HSM, a short air cavity and long HSM length can improve the sensor’s sensitivity These results provide deep understanding and some ideas for designing and optimizing highly sensitive EFPI fiber humidity sensors.

        Manufacture and antibacterial characteristics of Eucommia ulmoides leaves vinegar

        In this work, the fermentation conditions and theantibacterial characteristics of Eucommia ulmoides leavesvinegar (EV) were studied. By single factor orthogonaltest, it was found that under optimal fermentation conditions(bran addition 10%, sugar addition 8%, leaven addition0.3% and acetic acid bacteria solution 12%), the aceticacid content and CA content of EV were 45.5 ± 2.8 mg/mL and 0.98 ± 0.08 mg/mL, respectively. Then, by thedisc diffusion method, it was concluded that the antibacterialeffect of EV was significantly higher than that ofEucommia ulmoides leaves enzymatic hydrolysate andZhenjiang aromatic vinegar (P\0.05). An investigationinto action mode of EV against Bacillus subtilis indicatedthat, under the combined action of CA and acetic acid, EVexerted its antibacterial effect by damaging bacterial cellwall and cell membrane, increasing the cell permeabilitywhich resulted in the structural lesions and release of cellcomponents, thus led to cell death.

        The Fingerprinting of Huangjinju Powder for Injection on Chinese Patent Medicine by XRD Fourier

        The purpose is to study the identification method of Huangjinju powder for injection and the medicinal materials by the fingerprint off-ray Diffraction Fourier (XRDF). We used the same method on both the studying of Huangjinju and the medicinal materials. Then we selected a few components alignment to compare. We analyzed the data by setting up the deviation $d({\AA})$ as ${\pm}0.05$ to calculate the rate of special mark on the sample (Px) and on the patent (P). The special XRDF of Huangjinju$[d({\AA})/(I/I_0)]$ have 5 peaks that have not expressed in medicinal materials. Therefore Px is 22.73%. Flos Trollii Chinensis has 3 special marks and Px is 17.65%. Flos Chrysanthemi Indici has 1 special mark and Px is 3.57%. Its coincided interplanar spacing with the patent is $2.907{\AA}$. Flos Lonicerae Japonicae has 6 special marks and Px is 23.08%. Its special mark in the patent are 4.95/14 and 4.50/15, respectively. The P is 9.09%. Its coincided interplanar spacing with the patent is $2.910{\AA}\;and\;3.05{\AA}$, respectively. The number of special XRDF mark peaks of baicalin is 9 and Px is 18.37%. Its coincided interplanar spacing with the patent is $2.910{\AA}$. It has visible mark and specificity adopting XRDF fingerprint to identify Huangjinju and medicinal materials. Establishing the quality standard is a synthetic index that depends both on special marks in the medicinal materials of the patent and on the coincidence peak data.
