Shigella spp. constitutes some of the key pathogens responsible for the global burden of diarrhoeal disease. With over 164 million reported cases per annum, shigellosis accounts for 1.1 million deaths each year. Majority of these cases occur among the children of the developing nations and the emergence of multi-drug resistance Shigella strains in clinical isolates demands the development of better/new drugs against this pathogen. The genome of Shigella flexneri was extensively analyzed and found 4,362 proteins among which the functions of 674 proteins, termed as hypothetical proteins (HPs) had not been previously elucidated. Amino acid sequences of all these 674 HPs were studied and the functions of a total of 39 HPs have been assigned with high level of confidence. Here we have utilized a combination of the latest versions of databases to assign the precise function of HPs for which no experimental information is available. These HPs were found to belong to various classes of proteins such as enzymes, binding proteins, signal transducers, lipoprotein, transporters, virulence and other proteins. Evaluation of the performance of the various computational tools conducted using receiver operating characteristic curve analysis and a resoundingly high average accuracy of 93.6% were obtained. Our comprehensive analysis will help to gain greater understanding for the development of many novel potential therapeutic interventions to defeat Shigella infection.

Shopping Behavior, Country Attitudes, and Evaluation of Countries of Origin in China

This article presents the results of a survey of 209 Chinese male consumers. In this study, consumer evaluations and attitude towards products made in industrialized and newly industrialized countries were obtained along with measures of consumer shopping behavior. The results indicate that industrialized countries were perceived as manufacturing products that are more reliable, technologically advanced, stylistic, and costly than newly industrialized countries. A duster analysis using moderating variables related to shopping for refrigerators, cameras, and t shirts revealed that the Chinese respondents could be grouped into four segments identified as Durables Enthusiasts, Duraoles Uninvolved, Inexperienced Shoppers, and Apparels Involved. Country attitudes and evaluations were fairly consistent across the newly industrialized countries but varied for industrialized countries across the four segments. Similar results also emerged from a correlation analysis of made in attitude scales (based on moderating shopping variables) with country evaluations. It was found that Chinese consumers' involvement with durable products was positively related to the evaluation of industrialized countries. Consumers belonging to the Durable Enthusiasts and Apparels Involved segments evaluated industrialized countries more favorably and believed to a greater extent that products made in these countries are reliable, technologically advanced, and stylistic than consumers belonging to the Durables Uninvolved and Inexperienced Shopper segments. Managerial and research implications are derived from these results.

Oxidative stress plays a pivotal role in the development of diabetes and hyperglycaemia. The protective effects of natural extracts against diabetes are mainly dependent on their antioxidant and hypoglycaemic properties. Broccoli (Brassica oleracea) exerts beneficial health effects in several diseases including diabetes; however, the mechanism has not been elucidated yet. The present study was carried out to evaluate the potential hypoglycaemic and antioxidant properties of aqueous broccoli extracts (BEs) in diabetic rats. Streptozotocin (STZ) drug was used as a diabetogenic agent in a single intraperitoneal injection dose of 50 mg/kg body weight. The blood glucose level for each rat was measured twice a week. After 8 weeks, all animals were fasted overnight and sacrificed; pancreatic tissues were homogenized and used for measuring oxidative DNA damage, biochemical assessment of glutathione (GSH), and total antioxidant capacity (TAC) as well as histopathological examination for pancreatic tissues was examined. Diabetic rats showed significantly higher levels of DNA damage, GSH depletion, and impaired TAC levels in comparison to non-diabetics (P<0.05). The treatment of diabetic rats with BE significantly reduced DNA damage and conserved GSH and TAC values (P<0.01). BE attenuated pancreatic histopathological changes in diabetic rats. The results of this study indicated that BE reduced the STZ mediated hyperglycaemia and the STZ-induced oxidative injury to pancreas tissue. The used in vivo model confirmed the efficacy of BE as an anti-diabetic herbal medicine and provided insights into the capacity of BE to be used for phytoremediation purposes for human type 2 diabetes.
Sixty‐five gene‐based risk score classifier predicts overall survival in hepatocellular carcinoma

<P><B>Abstract</B></P><P>Clinical application of the prognostic gene expression signature has been delayed due to the large number of genes and complexity of prediction algorithms. In the current study we aimed to develop an easy‐to‐use risk score with a limited number of genes that can robustly predict prognosis of patients with hepatocellular carcinoma (HCC). The risk score was developed using Cox coefficient values of 65 genes in the training set (n = 139) and its robustness was validated in test sets (n = 292). The risk score was a highly significant predictor of overall survival (OS) in the first test cohort (<I>P</I> = 5.6 × 10<SUP>−5</SUP>, n = 100) and the second test cohort (<I>P</I> = 5.0 × 10<SUP>−5</SUP>, n = 192). In multivariate analysis, the risk score was a significant risk factor among clinical variables examined together (hazard ratio [HR], 1.36; 95% confidence interval [CI], 1.13‐1.64; <I>P</I> = 0.001 for OS). <I>Conclusion:</I> The risk score classifier we have developed can identify two clinically distinct HCC subtypes at early and late stages of the disease in a simple and highly reproducible manner across multiple datasets.
