고온.내산성 Bacillus sp. SJ-15를 이용한 음식물 쓰레기의 호기적 퇴비화

고온 내산성 미생물을 퇴비에서 분리하고, 생리 및 생화학적 특성을 조사하여 Bacillus sp. SJ-15로 명명하였다. 생장을 위한 최적 온도와 pH는 각각 $55^{\circ}C$와 5.0 이었다. 분리된 SJ-15 균주는 음식물쓰레기의 고온 고속 퇴비화 공정에 적용하였다. 퇴비화 과정에서 16시간 만에 최대 생균수인 $9.2{\times}10^9/ml$를 나타내었다. 본 퇴비화 공정은 개시 100일 후에 약 88%의 감량율을 나타내었고, 퇴비성분의 농도를 분석한 결과 본 공정을 통해 생산된 퇴비는 염분을 제외하면 유기질 비료의 기준을 모두 만족하는 것으로 나타났다. A thermoacidophilic bacterium was isolated from the compost and designated as Bacillus sp. SJ-15 by physiological and biochemical characteristics. The optimum temperature and pH for growth were at $55^{\circ}C$ and pH 5.0, respectively. The strain SJ-15 was adapted in process of accelerated high-temperature composting of garbage. The highest viable cell count of composting process reached to $9.2{\times}10^9/ml$ in 16 hours. After running times of 100 days, the composting process showed a reduction rate of approximately 88%, and the concentrations of components were sufficiently high or low to satisfied the standard of organic compost except for salinity.

Paenibacillus macerans 유래의 cycloinulooligosaccharide fructanotransferase (CFTase) 유전자(cft)를 Saccharomyces cerevisiae SEY2102에 발현시키기 위해 대장균과 효모의 shuttle vector인 pYES2.0에 subcloning 하였다. 구축된 pYGECFTN (8.6 kb) plasmid를 S. cerevisiae SEY2102에 형질전환하였고, uracil이 결핍된 SD 배지에서 선별하였다. cft 유전자는 선별된 형질전환체(S. cerevisiae SEY2102/pYCECFTN)에서 GAL1 promoter 조절하에 성공적으로 발현되어 cyclofructan(CF)을 생성함을 TLC로 확인하였다. 그러나, 균체 외로의 효소 분비는 이루어지지 않았고 cytoplasm보다 periplasmic space에 많이 존재하였다 S. cerevisiae에서 발현된 P. polymyxa유래 CFTase보다 P. macerans 유래 CFTase의 CF 생성이 image analyzer로 확인한 결과, 더 많음을 알 수 있었다. 효소반응 5분째부터 CF가 생성됨을 확인하였고, 최적온도와 최적 pH는 각각 $45^{\circ}C$와 pH 8.0로 나타났으며, $55^{\circ}C$까지 효소활성이 안정적으로 유지되었다. Dahlia tubers, chicory root, Jerusalem artichoke 등의 inulin 기질에 따른 반응산물 분석 결과, 모든 기질로부터 CF가 생산되었으며, dahlia tubers와 Jerusalem artichoke로부터 가장 효과적으로 생성되었다. The cycloinulooligosaccharide fructanotransferase (CFTase) gene (cft) from Paenibacillus macerans was subcloned into an E. coli-yeast shuttle vector, pYES2.0, resulting in pYGECFTN. The plasmid pYGECFTN (8.6 kb) was introduced into Saccharomyces cerevisiae SEY2102 cells and then the transformants were selected on the synthetic defined media lacking uracil. The cft gene expression in yeast transformant was demonstrated by the analyses cyclofructan (CF) spots on thin-layer chromatogram. The recombinant CFTase was not secreted into the medium and localized in the periplasmic space. The production of CF was observed after 5 min of the enzymatic reaction with inulin. The optimun pH and temperature for CF production were found to be at pH 8.0 and $45^{\circ}C$, respectively. Enzyme activity was stably maintained up to $55^{\circ}C$. The CF was produced from all inulin sources and was most efficiently produced from dahlia tubers and Jerusalem artichokes.

N/A Objectives: To determine accurate diagnosis and proper treatment, we reviewed colonoscopic and histologic findings of flat adenoma of colon, Methods: We studied retrospectively 2148 cases of colonoscopic findings performed in our hospital from March of 1993 to September of 1995. Results: 1) The incidence of adenoma is 9.5%(203 cases), and that of flat adenoma is 1.6%(34 cases). 2) The location of flat adenoma is 3 cases in rectum(8.8%), 15 cases in sigmoid colon(44.1%), 9 cases in descending colon(26.5%), 3 cases in transverse colon(8.8%) and 4 cases in ascending colon and cecum(11.8%). 3) The diameter of flat adenoma was smaller than 5mm in 17 cases(50.0%), between 5-10mm in 8 cases(23.5%) and larger than 10mm in 9 cases (26.5%). Mean size was 9.2mm. 4) In colonoscopic features, type IIa was 25 cases (73.5%), type IIa + IIc was 5 cases(14.7%) and lateral spreading tumor was 4 cases(11.8%). 5) In histologic findings, tubular adenoma was 26 cases(76.5%), tubulovillous adenoma was 6 cases (17.6%) and serrated adenoma was 2 cases(5.9%). 6) The incidence of severe dysplasia or carcinoma was zero in smaller than 5mm, 25.0%(2 case) in 5-10mm and 55.6%(5 cases) in larger than 10mm. 7) The incidence of severe dysplasia or carcinoma was 11.5%(3 cases) in tubular adenoma and 66.7%(6 cases) in tubulovillous adenoma. 8) It was confirmed by abdominal CT scan or operation that lesions were limited to intraepithelium in 4 cases, mucosa in 2 cases submucosa in 1 case and no lymph node metastasis in any case. Conclusion: Even though flat adenoma of colon was smaller than polypoid adenoma, the incidence of malignant change was higher. When it was smaller than 10mm, the incidence of submucosal invasion or lymph node invasion was rare. Therefore endoscopic mucosal resection(EMR) can be the treatment of choice in flat adenoma smaller than 10mm, and after EMR, it is desirable to decide the treatment modality depending on the histologic findings.

Lymphangioma is a benign tumor composed of numerous small or large thin-walled lymphatic spaces. It shows a predilection for head, neck, and axilla. Jejunal lymphangioma in children is extremely rare. We present a case of 13-year-old girl with jejuno-jejunal intussusception secondary to lymphangioma in jejunum. To our knowledge, this is the second report of jejunal lymphangioma causing intussusception in a child. (Korean J Gastroenterol 2007;49:256-259)
양강 에탄올 추출물이 마우스에서 면역조절작용에 미치는 영향

본 연구는 면역억제 마우스에서 양강 에탄올 추출물이 면역증강에 미치는 영향을 평가해보았다. 마우스에 cyclophosphamide를 2회 복강 주사한 후 양강 추출물을 30, 100, 300 mg/kg 용량으로 4주간 경구투여 한 후, 체중 및 면역장기 무게, 비장세포수, 혈청 사이토카인 농도 및 혈청 면역글로불린의 농도를 측정하였다. 실험 결과 체중과 비장세포수는 양강 추출물 투여 시 대조군과 비교하여 유의한 차이를 보이지 않았다. 혈청의 IL-2, TGF-β 및 IFN-γ 농도는 AO 100군에서 대조군에 비해 유의적인 증가를 보였고(p<0.05) IL-4 농도는 실험군에서 유의한 차이를 보이지 않았다. 혈청 내 IgM의 농도는 대조군에 비해 양강 추출물 투여군 모두에서 유의적으로 증가하였고 (p<0.05), IgA의 농도는 양강 추출물 투여군에서 증가하는 경향을 보였는데 특히 AO100군에서 유의성 있게 증가하였다(p<0.05). 본 연구 결과는 양강 에탄올 추출물은 혈청 내 사이토카인 농도와 면역글로불린 농도를 증가시켜 면역력 증강에 기여할 것으로 보이며 특히 100 ㎎/㎏을 투여하였을 때 효과가 가장 큰 것으로 나타났다. The purpose of this study was to investigate the immunoregulatory effect of Alpinia officinarum (AO) ethanol extract in immunocomprised mice. Mice were injected intraperitoneally with an immunosuppresive drug, cyclophosphamide, and then administrated orally with 30, 100 and 300 ㎎/㎏ of ethanol extract of Alpinia officinarum (AO 30, AO 100, AO 300, respectively). The concentrations of cytokine and immunoglobulin (IgM, IgA, IgG) of serum were measured. The body weight of mice and spleen cell number of AO fed group showed no significant difference compared to control group. The concentrations of several cytokines including IL-2, IFN-γ, TGF-β of serum showed a significant increase in the AO 100 group compared to the control and other groups (p<0.05). And, IL-4 level showed no significant difference in the experimental groups. The supplementation of AO (30, 100, 300 ㎎/㎏) increased the concentration of IgM significantly (p<0.05). And, the concentration of IgA was significantly increased in the AO 100 group (p<0.05) compared to control group. It can be concluded that Alpinia officinarum ethanol extract may enhance immune function by promoting cytokine production and immunoglobulin.

In this study, we analyzed the anti-oxidative and anti-inflammatory activities of seven medicinal herbs. All extracts of the tested herbs, Euryale ferox Salisbury, Echinops setifer Iljin, Amomum cardamomum Linne, Tetrapanax papyriferus, Illicium verum Hook. f., Typha orientalis Presl, and Piper longum Linne, exhibited potent anti-oxidative activity as confirmed by DPPH radical scavenging capacity. Lipopolysaccharide (LPS) induced nitric oxide (NO) production, in the RAW 264.7 cell line, was also ameliorated by all extracts` treatments in a dose dependent manner. NO suppressive activity originated from the inhibition of inducible nitric oxide synthase (iNOS) protein expression by the extracts. Three extracts, E. ferox S., I. verum Hook. f., and P. longum L., possessed suppressive activity against, not only iNOS, but also cycloxygenase 2 (COX-2) protein expression. These three extracts may then serve as potential candidates for non steroidal analgesic inflammation drugs (NSAIDs). Furthermore, all extracts induced anti-oxidative enzyme, heme oxygenase 1, protein expression. Taken together, these results provide an important new insight into the fact that various medicinal herbs possess potent anti-oxidative and anti inflammatory activities and might be utilized as promising agents in the field of health products. Further studies for the identification of the active compounds from medicinal herbs are clearly needed.
진행성 위암과 동반된 후복막 섬유화증 7예의 임상적 고찰

Background/Aims: Retroperitoneal fibrosis is a fibrosing disease process which frequently develops ureteral obstruction. Malignant retroperitoneal fibrosis has been reported that various types of cancer including cancer of the breast, stomach, prostate, lung, cervix, uteri, colon, pancreas, ovary, and even Hodgkin's disease. While the pathogenesis remains obscure, small foci of metastatic neoplasm in the retroperitoneal space can elicit a desmoplastic reaction in secondary form to malignancy. Methods: Recently we experienced 7 cases of retroperitoneal fibrosis with advanced gastric cancer. So, we report 7 cases of retroperitoneal fibrosis with clinical and radiologic characteristics. Results: The most common signs and symptoms were related irreversible renal dysfunction. The urologic finding noted medial deviation of the involved ureters and hydronephrosis Conclusions: Clinical manifestations are abdominal pain, back pain, hydronephrosis, and uremia. When sudden back pain and hydronephrosis develop in advanced malignancy, physicians should be considerd for retroperitoneal fibrosis.
