Objectives : The purpose of the present study is to observe the neuroprotective effect of the $NeuBo153^{\circledR}$ on transient focal cerebral ischemia in rats. Methods : $NeuBo153^{\circledR}$ was made by mixing the herbs, mainly the root of Panax ginseng, the root of Rehmannia glutinosa and Poria cocos, the stem bark of Acanthopanax senticosus, the root of Scutellaria baicalensis and Mel, and heating for 96 hours. Transient Focal cerebral ischemia (2 h of ischemia, 22 h of reperfusion) was induced by intraluminal suture method with SD rats. Sensory motor function was tested by rotarod test, prehensile traction test, beam balance test and foot fault test at 24 h after ischemia. The brain slices were stained by 2% 2, 3, 5-triphenyltetrazolium chloride and the infarct volume was measured by graphic analyzer at 24 h after ischemia. Results : $NeuBo153^{\circledR}$ treated group did not show significant differences compared with vehicle treated group in body temperature. Oral administration of $NeuBo153^{\circledR}$ reduced brain infarct volume by 29.7% compared with vehicle treated group. $NeuBo153^{\circledR}$ also showed protective effects on sensory motor functional deficits. Conclusion : $NeuBo153^{\circledR}$ treatment reduced brain damage and improved functional deficits induced by MCAo. It showed neuroprotective effects even when treatment was relayed 2 h after injury. Further research is required to evaluating long term functional recovery am accurate therapeutic range and mechanisms.

N/A Objectives: The root of Rhodiola sachalinensis A. bor (RS), which has been used to strengthen qi(補氣), reduce heat of lung (淸縮), nourish heart (養心), stop bleeding (止血), reduce edema(消腫) for the treatment of fatigue, amnesia, bleeding and edema. It`s flavour and channel tropism is sweet (甘), acting on the lung (肺). The purpose of the study we report here was to determine the neuroprotective effect of the roots of RS on transient focal cerebral ischemia in SD rats. Methods: The root of RS was lyophilized after extraction with 85% methanol. We occluded middle cerebral artery for 90 minutes and reperfused for 24 hours. The brain slices were stained by 2 % 2, 3, 5-triphenyltetrazolium chloride and the infarct volume was measured by graphic analyzer. Results: The root extracts of RS showed significant neuroprotective effects (100 mg/kg, p〈0.05) compared with control group. The neuroprotective efficacy was about 35%. Immunohistochemical data explained the protective effects of RS via attenuation of COX-2 induction in neocortex and inhibition of microglia activation in the penumbral region. Conclusions: The root of RS has neuroprotective effects on transient focal cerebral ischemia in Spargue-Dawley rats and the mechanism might be the attenuation of COX-2 induction and microglial activation.

N/A Recent reports have proposed that Ephedrae Herba may modulate the immune response on allergy or asthma. Chemokines are important for the recruitment of leukocytes to sites of infection, which is essential in host defense. The thymus and activation-regulated chemokine (TARC) is a CC chemokine which potentially plays a role via a paracrine mechanism in the development of allergic respiratory diseases. Objectives : The objective of this study is to investigate the effect of Ephedrae Herba on the secretion of TARC of human bronchial epithelial cell Methods : Enzyme-Linked Immunosorbent Assay (ELISA) was performed to detect the secretion of TARC . The cytotoxicity was measured by MTT assay. Results : Ephedrae Herba significantly inhibited the secretion of TARC with a dose-dependant manner. The effective dosage did not have the cytotoxicity on human bronchial epithelial cell. Conclusion : Results of our study show that Ephedrae Herba would play an important role in modulation of TARC in Human bronchial epithelial cells.
Myristica fragrans Seed Extract Protects Against Dextran Sulfate Sodium–Induced Colitis in Mice

Nutmeg (seed of Myristica fragrans [MF]) is one of the most commonly used spices in the world and also a well-known herb for the treatment of various intestinal diseases, including colitis in traditional Korean medicine. The purpose of the current study was to investigate whether water extract of MF (MFE) can protect against dextran sulfate sodium (DSS) induced colitis in a mouse model. Colitis was induced by 5% DSS in balb/c mice. MFE (100, 300 or 1000 mg/kg) was orally administered to the mice twice a day for 7 days. Body weight, colon length, clinical score, and histological score were assessed to determine the effects on colitis. Proinflammatory cytokines (interferon-c, tumor necrosis factor-a, interleukin [IL]- 1b, and IL-6) were measured to investigate the mechanisms of action. MFE dose dependently inhibited the colon shortening and histological damage to the colon. However, it did not prevent weight loss. MFE also inhibited proinflammatory cytokines. The current results suggest that MFE ameliorates DSS-induced colitis in mice by inhibiting inflammatory cytokines. Further investigation, including the exact mechanisms is needed.
Telomerase induction in astrocytes of Sprague-Dawley rat after ischemic brain injury

Telomerase, a reverse transcriptase, consists of an RNA template and protein polymerase. This ribonucleoprotein protects the linearized chromosomal end region and elongates the telomere during chromosomal replication. Telomerase is not expressed in adult somatic cells but it shows high activity in most cells during embryonic development. We report, by RT-PCR and immunohistochemical results, that the induction of telomerase protein catalytic subunit (TERT) in transient middle cerebral artery occlusion induced brain injury. TERT mRNA emerged 24 h after ischemia. We examined which brain cell expressed TERT in the penumbra region of injured brain. The expression of TERT began from 24 h and remained until 5 days after ischemia. We identified that TERT was co-localized with the astrocyte marker, GFAP, at 3 days after ischemia. This is strong evidence that TERT is induced in astrocytes when the brain is damaged by ischemia, and that this enzyme may play an important role in ischemic brain injury.
Effect of Cibotii Rhizoma on the Growth Hormone Secretion of Adolescent Male Rats

뇌하수체전엽에서 분비된 성장호르몬의 작용으로 성장이 촉진된다는 것이 지금까지의 정설이고, 최근 그 외 부갑상선 호르몬, 성호르몬, 국소호르몬 등이 관련되어 있다는 연구보고가 있다. 목적 : 성장촉진 효능이 있는 구척추출물의 작용기전 중 성장호르몬 분비에 대한 영향을 밝히기 위해 본 실험이 구상되었다. 방법 : 성장기 흰쥐에게 구척추출물을 투여하고 성장촉진 효과를 나타낸 농도에서, 투여시간에 따른 혈중 성장호르몬의 변화를 측정하였다. 결과 : 구척 투여전 20분에서 투여후 60분까지 총 80분간의 혈중 성장호르몬 변화를 측정한 결과, 투여후 5분에서 혈중 성장호르몬이 약간 증가하는 경향을 나타내었다. 5분후의 혈중 성장호르몬의 양을 구척추출물 10 mg/kg과 100 mg/kg의 농도별로 투여한 후 측정하였으나 성장호르몬의 증가를 관찰할 수 없었다. 결론 : 구척은 유의한 성장촉진작용을 나타내지만, 혈중 성장호르몬의 양을 증가시키지는 않았다. 따라서 구척의 성장촉진 효능에 대한 작용기전에 성장호르몬 분비촉진기전은 없는 것으로 사료된다.
