        An extremely thermostable maltogenic amylase from <i>Staphylothermus marinus</i>: <i>Bacillus</i> expression of the gene and its application in genistin glycosylation

        <P><B>Abstract</B></P> <P>The most extremely thermostable maltogenic amylase (SMMA) from archaeon <I>Staphylothermus marinus</I> has many potential applications in food processing. To ensure safety of microbial origin, a recombinant plasmid containing the enzymic gene and a constitutive promoter AmyR2 was constructed, and then transformed into a GRAS microorganism <I>Bacillus subtilis</I>. The purified SMMA from the liquid cultures of <I>Bacillus</I> has a specific activity of 66.96U/mg, two times more than that from <I>Escherichia coli</I>. SMMA was further employed to catalyze the genistion glycosylation using γ-CD as both glucosyl donors and solubilizer. Glycosylated genistins with one to four additional α-glucosyls and a molar percentage of 69.87% in genistin reaction mixture were identified and quantified by HPLC–UV–MS. The glycosylated genistins at 0.2–1.2mM showed an enhanced DPPH free radical scavenging capacity. To our knowledge, this is the first report on the <I>Bacillus</I> expression of archaeal maltogenic amylase.</P> <P><B>Highlights</B></P> <P> <UL> <LI> A fused gene of SMMA and a constitutive promoter AmyR2 was transformed into <I>B. subtilis</I>. </LI> <LI> Specific activity of purified SMMA from <I>B. subtilis</I> was two times more than that from <I>E. coli</I>. </LI> <LI> SMMA of safe microbial origin attached different number of glucosyls from γ-CD to genistin at 90°C. </LI> <LI> Glycosylated genistins showed an enhanced DPPH free radical scavenging capacity than genistin. </LI> </UL> </P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

        Mechanistic target of rapamycin and an extracellular signalingregulated kinases 1 and 2 signaling participate in the process of acetate regulating lipid metabolism and hormone-sensitive lipase expression

        Objective: Acetate plays an important role in host lipid metabolism. However, the network of acetate-regulated lipid metabolism remains unclear. Previous studies show that mitogen-activated protein kinases (MAPKs) and mechanistic target of rapamycin (mTOR) play a crucial role in lipid metabolism. We hypothesize that acetate could affect MAPKs and/or mTOR signaling and then regulate lipid metabolism. The present study investigated whether any cross talk occurs among MAPKs, mTOR and acetate in regulating lipid metabolism.Methods: The ceramide C6 (an extracellular signaling-regulated kinases 1 and 2 [ERK1/2] activator) and MHY1485 (a mTOR activator) were used to treat rabbit adipose-derived stem cells (ADSCs) with or without acetate, respectively.Results: It indicated that acetate (9 mM) treatment for 48 h decreased the lipid deposition in rabbit ADSCs. Acetate treatment decreased significantly phosphorylated protein levels of ERK1/2 and mTOR but significantly increased mRNA level of hormone-sensitive lipase (HSL). Acetate treatment did not significantly alter the phosphorylated protein level of p38 MAPK and c-Jun aminoterminal kinase (JNK). Activation of ERK1/2 and mTOR by respective addition in media with ceramide C6 and MHY1485 significantly attenuated decreased lipid deposition and increased HSL expression caused by acetate.Conclusion: Our results suggest that ERK1/2 and mTOR signaling pathways are associated with acetate regulated HSL gene expression and lipid deposition.

        Inkjet Printing and In-Situ Crystallization of Biopigments for Eco-Friendly and Energy-Efficient Fabric Coloration

        Current methods of fabric dyeing have a significant negative effect on the environment via toxic effluent emissions, high water usage, and high energy consumption. We demonstrate, for the first time, low-energy inkjet printing of an ecofriendly and biodegradable pigment derived from widely available spalting fungi with synthesis by-products that are fully biodegradable. Vivid and stable coloration is achieved on polyester and cotton using benign acetone-based and ethanol-based biopigment inks, with no liquid effluent or runoff, and without any synthetic dyes or mordants. We investigate the influence of fabric type, solvent type, and the number of printing passes on the color type, intensity, and uniformity, pigment crystallization, and environmental stability. Greater spatial definition of the printed pigment in inkjet printing effects greater crystallization and more vivid and uniform coloration as compared to drop-casting, the primary method used till date to color fabrics with such fungal biopigments. pH-driven interactions between the fabric, solvent, and biopigment cause solvent-driven changes in color type (from bluish to reddish) and fabric-governed changes in color intensity. The impact of our findings on minimizing the environmental impact of fabric dyeing is discussed.

        STAT3, a Poor Survival Predicator, Is Associated with Lymph Node Metastasis from Breast Cancer

        Purpose: The aim of this study is to explore signal transducer and activator of transcription 3 (STAT3) expression in breast cancer and to analyze the detailed mechanism that STAT3 contributes to the progression of breast cancer. Methods: We retrospectively analyzed the clinicopathologic characteristics and overall survival (OS) of 140 breast cancer patients after curative surgery, and detected STAT3 expression, phosphorylated STAT3 (pSTAT3) expression, Ki-67 expression, vascular endothelial growth factor (VEGF)-C and -D expression in breast cancer tissues, and adjacent nontumor tissues. Survival analysis and relationship analysis were adopted for demonstrated the important mechanism of STAT3 contribution to progression of breast cancer. Results: STAT3 expression, pSTAT3 expression, Ki-67 expression, VEGF-C expression, and VEGF-D expression in breast cancer tissues were significantly higher than those in adjacent nontumor tissues, respectively. With survival analysis, only number of lymph node metastasis (N stage) was identified as the independent predictors of the OS of breast cancer patients. Besides, we demonstrated there was the most prominent correlation between STAT3 expression and lymph node metastasis in breast cancer tissues by using the multinominal regression method. Conclusion: STAT3, a poor survival biomarker potential association with lymph node metastasis, was suitable for predication the OS of breast cancer patients after curative resection.

        Moving Rate of Positive Patient Results as a Quality Control Tool for High-Sensitivity Cardiac Troponin T Assays

        Background: A small shift in high-sensitivity cardiac troponin T (hs-cTnT) assays can lead to different result interpretation and consequent patient management. We explored whether a small bias could be detected using conventional internal quality control (QC) procedures, evaluated the performance of moving average (MA)-based QC procedures, and proposed a new QC procedure based on the moving rate (MR) of positive patient results of hs-cTnT assays. Methods:The ability of conventional QC to detect a 5 ng/L bias was examined using the13s/ 22s/R4s multi-rule procedure as deviation rules.We developed MA and MR procedures for the hs-cTnT assay using eight months of patient data. The performance of different MA or MR procedures was investigated by calculating the median number of patient samples affected until a bias introduced into the dataset was detected (MNPed). After comparing the MNPed across different procedures, we selected an optimal MA or MR procedure for validation. Validation graphs were plotted using the minimum, median, and maximum number of results affected until bias detection. Results: Our conventional QC procedures could not detect a positive bias of 5 ng/L. When a positive bias was introduced, MNPed was much higher using MA than using MR, with cut-off values of 5 ng/L and 14 ng/L, respectively. MR validation charts for optimal procedures provided insight into the MR performance. Conclusions: The MR procedure could detect different errors with few false alarms. In the hs-cTnT assay, the MR procedure with a smaller cut-off value outperformed MA and conventional QC procedures for small bias detection.

        Optimization of Submerged Fermentation Medium for Matrine Production by Aspergillus terreus, an Endophytic Fungus Harboring Seeds of Sophora flavescens, Using Response Surface Methodology

        Different endophytes isolated from the seeds of Sophora flavescens were tested for their ability to produce matrine production. Response surface methodology (RSM) was applied to optimize the medium components for the endophytic fungus. Results indicated that endophyte Aspergillus terreus had the ability to produce matrine. The single factor tests demonstrated that potato starch was the best carbon source and the combination of peptone and NH<sub>4</sub>NO<sub>3</sub> was the optimal nitrogen source for A. terreus. The model of RSM predicted to gain the maximal matrine production at 20.67 μg/L, when the potato starch was 160.68 g/L, peptone was 24.96 g/L and NH<sub>4</sub>NO<sub>3</sub> was 2.11 g/L. When cultured in the optimal medium, the matrine yield was an average of 20.63 ± 0.11 μg/L, which was consistent with the model prediction. This study offered an alternative source for the matrine production by endophytic fungus fermentation and may have far-reaching prospect and value.

        Protective Effects of Bacillus coagulans JA845 against D-Galactose/AlCl<sub>3</sub>-Induced Cognitive Decline, Oxidative Stress and Neuroinflammation

        Recently, the efficacy of probiotics in treatment of neurodegenerative disorders has been reported in animal and clinical studies. Here, we assessed the effects of Bacillus coagulans JA845 in counteracting the symptoms of D-galactose (D-gal)/AlCl<sub>3</sub>-induced Alzheimer’s disease (AD) in a mice model through behavioral test, histological assessment and biochemical analysis. Ten weeks of pretreatment with B. coagulans JA845 prevented cognitive decline, attenuated hippocampal lesion and protected neuronal integrity, which demonstrated the neuroprotective features of B. coagulans JA845 in vivo. We also found that supplementation of B. coagulans JA845 alleviated amyloid-beta deposits and hyperphosphorylated tau in hippocampus of D-gal/AlCl<sub>3</sub>-induced AD model mice. Furthermore, B. coagulans JA845 administration attenuated oxidative stress and decreased serum concentration of inflammatory cytokines by regulating the Nrf2/HO-1 and MyD88/TRAF6/NF-κB pathway. Our results demonstrated for the first time that B. coagulans has the potential to help prevent cognitive decline and might be a novel therapeutic approach for the treatment of neurodegenerative diseases.

        In the paper, a large customer satisfaction evaluation index system is constructed, which involves five broad categories including corporate image, service attitude service quality, and 21 individual indexes, for example popularity, Value for money and so on. By using fuzzy theory, the fuzzy evaluation model of large customer satisfaction is been established. And then, based on a questionnaire survey for the main districts and counties in Daqing, the paper completes large customer satisfaction evaluation of the China Netcom Corporation Daqing branch and gives the advantages and disadvantages in the customer service, in order to further improving the large customer satisfaction.

        Yellow pigment from gardenia fruit: structural identification and evaluation of cytotoxic activity in HepG2 cells by induction of apoptosis

        The preparation process of yellow pigment (YP) from gardenia (Gardenia jasminoides) fruit was investigated, and the main components of YP were characterized by liquid chromatography-time of flight-mass spectrometer/mass spectrometer (LC-TOF–MS/MS). Furthermore, cytotoxic activity in HepG2 cells by induction of apoptosis was also evaluated. The preparation results indicated that the color value of YP was 498.34, which was 8.6 times higher than crude YP. Fifteen compounds in YP were identified, and crocins were the predominant compounds. The cell experiment results showed that YP inhibited the proliferation of HepG2 cells in a time- and dose-dependent manner. Moreover, YP also inhibited HepG2 cells in G2/M stage, increased the level of intracellular reactive oxygen species (ROS), and enhanced cell apoptosis. Real-time quantitative polymerase chain reaction (RT-PCR) analysis revealed the up-regulation of caspase-3, 8, 9, and bax and down-regulation of bcl-2 in HepG2 cells. Overall, these findings suggested that YP had potential cytotoxic activity in HepG2 cells by induction of apoptosis, which might be beneficial to human health.
