        종 분포 모형을 이용한 구상나무림의 지속 및 쇠퇴에 관한 연구 - 전라남도 광양시 백운산을 중심으로 -

        The present study investigated the habitats of Korean fir trees (Abies koreana E. H. Wilson) on Mt. Baekwun (Baekwun-san), determined the current distribution, quantified the contribution of biological and nonbiological environmental factors affecting the distribution, derived actual and potential habitats, presented a plan for the establishment of protected areas, applied RCP 8.5 climate change scenario to analyze the effects of climate change on the future distribution of Korean fir trees, and predicted future potential habitats. According to the results of the study, 3,325 Korean fir trees (DBH >= 2.5 cm) inhabited Mt. Baekwun, and their distribution area was approximately 150 ha. Populations of Korean fir trees were confirmed to exist at an altitude of 900 m above sea level and were distributed up to 1,200 m. Based on potential distribution, areas appropriate for habitation by Korean fir trees were analyzed to be 450 ha, three times the current distribution area, with a focus on Sang Peak (Sang-bong), Eokbul Peak (Eokbul-bong), Ddari Peak (Ddari-bong), and Dosol Peak (Dosol-bong). The forest stands near Sang Peak, the main peak, were evaluated as those with the most appropriate potential for the habitation of Korean fir trees, and populations of the trees tended to prefer the northern slope rather than the southern slope. When climate change scenario RCP 8.5 was applied and future potential distribution was analyzed, the habitats were expected to decrease in area to 20 ha by 2050, with a focus on Sang Peak, and areas appropriate for habitation were predicted not to exist by 2080. Judging from such results, as global warming accelerates, the habitats of Korean fir trees are clearly expected to move from lowlands to highlands.

        경부도상 피판을 이용한 구강내 결손부의 재건

        Various cutaneous as well as myocutaneous flaps have been designed for the reconstruction of tissue defects caused by the excision of oral cancer. Among these flaps, cervical island skin flap have been introduced by Farr et al and more have developed by Tashiro et al. This flap has many advantages. The flap minimizes donor size by use of cervical operation wound, flap size available is adequate for most oral defects and the procedure is relatively simple and time saving. However, this flap is not applicable in patients where there are large tissue defects and metastasis is suspected. We used this flap for it's rapid simple, and effective, primary closure of oral defects after cancer ablation and we have found this flap very useful for the reconstruction of relatively small oral defects.

        사람 뇌척수액중 Transthyretin의 항산화작용

        Protective effects of human cerebrospinal fluid antioxidants against enzyme inactivation caused by metal-catalyzed oxidation systems were investigated. When purified glutamine synthetase(GS) was incubated with human cerebrospinal fluid(CSF), the enzyme was progressively inactivated. Catalase and EDTA could inhibit the enzyme inactivation by 50-80%. Small-molecular(Mr< -10,000) fraction of CSF inactivated the exogenous GS, but large-molecular(Mr> -10,000) fraction did not. The GS inactivation by the small-molecular fraction was also markedly inhibited by catalase and EDTA. These results suggested that metal-catalyzed oxidation is involved in the GS inactivation by the small-molecular fraction of CSF. Dithiothreitol(DTT) was shown to inhibit almost completely the oxidative inactivation of GS by CSF. However, DTT inhibited only partially the oxidative inactivation of GS caused by small-molecular fraction of CSF. When large-molecular fraction of CSF was separated by anion-exchange HPLC chromatogrpahy, there was a peak of antioxidant activity inhibiting the small-molecular fraction-induced GS inactivation in the presence of DTT. The antioxidant activity was neutralized by monoclonal antibodies to transthyretin. Purified transthyretin was found to efficiently inhibit ascorbate/Cu^(2+)-induced GS inactivation in the presence of DTT. Uric acid and glucose did not show any protective effect on the GS inactivation in the same condition. The above results suggest that metal-catalyzed oxidation occurs normally in human CSF, and that transthyretin may play an important role as a CSF antioxidant in protecting proteins from metal-catalyzed oxidation.

        척수손상·비손상인의 신체부위별 신체구성 및 골밀도 비교

        This study was designed to compare segmental body composition and bone mineral density in spinal cord-injured(SCIS; n=9, SCIA; n=9) and non-injured control(AB; n=9) subjects. Dual-energy X-ray absorptiometry(DEXA) was used to measure %body fat, body fat mass, lean body mass, bone mineral contents, and bone mineral density in total and various body regions(arms, legs, trunk). The present results showed that body fat mass and %body fat were significantly higher and lean body mass was significantly lower in the SCIS group compared to AB group, while there was no difference between the SCIA and AB group in those of parameters. In BMC, there was no difference among groups and in BMD, SCI group was significantly lower that AB group, but there was no difference between SCIA group and AB group. Based on these results, in can be suggested that long-term regular exercise can reduce the negative changes in body composition, bone mineral density by spinal cord injury.

        다중 중합효소 연쇄반응을 이용한 반코마이신 내성 장구균의 신속 검출

        일반적으로 임상검사실에서 vancomycin resistant enterococci(VRE)를 검출하는 일은 어렵고, 시간이 많이 들며, 검체처리 비용도 많이 든다. 따라서 본 실험은 임상검체에서 분리된 세균으로부터 VRE를 신속하게 확인하고, 진단하기 위한 방법으로서 다중 중합효소 연쇄반응을 확립하였다. 본 실험에 사용된 primer는 장구균에 특이한 유전자인 vanA, vanB, vanC-1, vanC-2/3각각의 염기서열을 기초로 primer를 제작하고, 다중 중합효소 연쇄반응을 실시하여 임상검체로부터 분리된 VRE 유전자의 type 및 분포율을 조사하고자 하였다. 국내에서 분리된 75주의 장구균을 대상으로 다중 중합효소 연쇄반응을 실시한 결과 36주의 분리균주에서 vancomycin에 대해 높은 저항성을 보이는 vanA 유전자를 가진 것으로 나타났다. 그리고 18주에서는 vancomycin에 낮은 저항성을 내성을 보이는 vanC-1또는 vanC-2/3유전자를 보유한 것으로 나타났다. 따라서 본 실험에서 확립한 다중 중합효소 연쇄반응 기법은 신속한 VRE 진단 방법으로 이용할 수 있을 것이다. It is generally difficult, time-consuming, and expensive for the clinical laboratory to detect vancomycin resistant enterococci (VRE). The aim of this study was to develop and evaluate the multiplex polymerase chain reaction (PCR) assay system as a diagnostic tool for the rapid detection of VRE from clinical samples and/or for the identification of VRE from the bacterial strains isolated from clinical specimens. Specific primers, designed from the nucleotide sequences respectively encoding the vanA, vanB, vanC-1, vanC-2/3 genes in enterococci, were coupled in a multiplex PCR assay system. With this multiplex PCR assay system, we investigated the incidence rates and types of VRE isolated from clinical samples. A total of 75 strains of enterococci were isolated in 3 general hospitals in Korea. Of these isolates, 36 strains showed a pattern of highlevel vancomycin resistance which associated with vanA gene, whereas 18 strains showed lowlevel vancomycin resistance associated with vanC-1 or vanC-2/3 gene. Thus, multiplex PCR assay method established in this study could be applied for the rapid detection of VRE.

      • 신경병증성 통증 모델쥐 뒤뿌리신경절의 Nitric Oxide Synthase 아형 및 NADPH-diaphorase 발현에 관한 연구

        Injury to peripheral nerve somtimes lead to neuropathic pain. Nitric oxide(NO) is increased in the dorsal root ganglia (DRG) of neuropathic pain models, and L-NAME, a nitric oxide synthase (NOS) inhibitor, reduces neuropathic pain. However, it is not known that underlying mechanism of NO involvement in development and maintanence of neuropathic pain. It was found that there were three NOS isoforms-nNOS, eNOS, and iNOS. It is also necessary to investigate NOS isoforms in neuropathic pain. The aim of this study is to investigate NOS isoforms and NADPH-diaphorase expressions and underlying mechanism of NO involvement in neuropathic pain. The 5^th lambar (L5) DRG were obtaind after tight ligation of L5 an L6 spinal nerves for 7 days. Toluidine blue stain, immunohistochemistry for nNOS, eNOS, and iNOS, and NADPH-diaphorase histochemistry were performed. All specimens were observed under light microscope. nNOS expression was increased significantly in the cytoplasm of the DRG of ligated side at 7 day post-ligation (7D PL). eNOS expression was also increased in the blood vessel, a few neurons or satellite cells, and cell membrane of Schwann cells in the DRG of ligated side at 7D PL. iNOS expression was increased in a few blood vessels and cytoplasm of small neurons in the DRG of ligated side at 7D PL. NADPH-diaphorase expression was increased significantly in the neurons of the DRG of ligate side at 7D PL, but NADPH-diaphorase expressions were weak or absent in the blood vessel and Schwann cell in the contralateral DRG and the DRG of ligated side at 7D PL. It is suggested that NOS isoforms may be closely related to development and mantanence of neuropathic pain in order of nNOS>eNOS>iNOS.
