      • Verticillium sp.가 생산하는 cellulase의 정제 및 그 성질

        The cellulase of verticillium sp. was purified to investigate its some properties. The enzyme purification was carried out by ammonium sulfate fractionation, and column chromatography of Sephadex G-50, G-100 and DEAE Sephadex A-25, with its specific activity increased to 17 fold. The molecular weight was estimated to be 46,000 by gel filtration. The optimum pH and temperature were pH 7 and 50℃, respectively. The enzyme was stable over the range of pH5-7 at 50℃ for 1 hour. But half of its activity was lost after 2 hour. The km value was calculated to be 1.2mg of CMC/㎖.

        The methods of the Computed Tomograph and the High-speed Speech analysis system were used for the comparative study of French and Korean oral vowels. Three male native speackers of French and three Korean speakers were selected for these articulatory and acoustic characteristics. The speech materials were the simple French oral vowels / i, e, ε, a, u, o, ??, a / and the Korean oral vovels / i, e, ε, a, w, u, o /, respectively. The results of these studies are as follows : (1) In the case of articulation, the aperture of Korean closed front unrounded vowels is larger than that of French vowels and the Korean back closed vowel /u/ and the half-closed vowel /o/ did not show significant differences with the corresponding French high vowel /i/. But the aperture of the Korean half-open vowel /??/ is smaller than that of French vowel and in this case, the place of articulation for Korean vowel /??/ is more advanced than that of French vowel /??/ (2) By means of spectrographic analysis of the French and Korean vowels, the first formant of the Korean high vowel /i/ is higher than that of French high vowel /i/. This means that the aperture of the Korean high vowel is larger than that of French high vowel. The second formant of French back vowels are lower than those of Korean back vowels. This result shows that the projection of French back vowels is greater than that of Korean back vowels.

        한국판 20항목 Toronto 감정표현불능증 척도(TAS-20K)의 개발과 타당도

        최근 감정표현불능증은 정서조절의 장애(disorder of emotion regulation) 또는 정신생물학적 조절이상(psychobiological dysregulation)으로 재개00념화되고 있으며, 다양한 측정 방법들이 개발되고 있다. 그러나 현재까지 이러한 장애를 검사할 타당도와 신뢰도를 인정받은 도구가 없는 실정이었다. 그래서 TAS가 개발되고 뒤이어 TAS-R이 개발되었으며, 새로이 TAS-20이 개발되어 여러 나라에서 신뢰도 및 타당도를 인정받았다. 그리하여 저자는 TAS-20을 문화와 언어가 다른 한국의 실정에 맞게 번역을 하여 한국에서의 타당도를 검정하고자 본 연구를 시도하였다. 이 연구에서는 한국판 20항목 감정표현불능증 척도(TAS-20K)의 특징적인 세가지 요인에 따른 감정표현 불능증의 전반적 차원을 확정적 요인 분석으로 검사하였다. 대상은 경북대학교 의과대학에 재학중인 의학과와 간호학과 학생 388명(남자 216명, 여자 172명)을 대상으로 하였다. 이 연구의 확정적 요인 분석에서 TAS-20K의 타당도가 독일, 미국, 캐나다에서의 연구 결과보다 더욱 높게 나왔다. 다만 #5 항목은 우리 나라의 문화적 특성을 고려할 때 “나는 어떤 문제를 단순히 논리적으로 설명하는 것보다는 그 문제의 이유를 이해할려고 노력하는 편이다.”라는 내용으로 변경하여 사용하는 것이 바람직하다고 생각된다. 그리고 이러한 검사가 더욱 다양한 연령군이나 사회경제적 배경 및 다양한 종류의 질환군을 표본으로 시행되어야 할 것으로 생각된다. 이러한 한국형 척도의 개발을 위해서는 동서양의 문화적 차이 또는 우리 나라의 문화적 특성을 고려하는 것이 매우 중요하다는 점이 본 연구를 통하여 강조되었다. Alexithymia has been reconceptualized as a disorder of emotion regulation or psychobiological dysregulation in humans, and various measures have been developed. However, these measures have been found to lack adequate reliability and /or validity. So, Taylor and others initially developed the TAS, and later developed an improved version of the TAS(the TAS-R). More recent studies led to further refinement of the TAS-R,and to the introduction of a 20-item version(the TAS-20), which has demonstrated its reliability and validity in several countries. The aim of the this study was to translate the English questionnaire of the TAS-20 into Korean, so that it will be applicable to Korean society. Next, the study attempted to examine the validity of the TAS-20K. This study tested a general dimension of alexithymia with a three factor-model in the TAS-20K, using confirmatory factor analysis. The subject were 388 Korean undergraduate students: 216 males and 172 females in the medical and nursing schools of Kyungpook National University. In this study, we found the Korean version of the TAS-20 to be replicable and valid. By us-ing confirmatory factor analysis, the study revealed more valid results than the ones obtained in Germany, Canada, and the United States. Finally, this research should also assess samples of various age groups or various kind of patients The author stress that it is very important to consider cultural difference between the East and West and characteristic of Korean culture in developing a korean version of foreign scales.

      • 울금(Curcuma Longa)으로부터 유효성분 분리와 생산국 및 지역별 유효성분 함량 분석에 따른 커쿠민 동정연구

        Chinese herbal medicine Curcuma longa is used in food, Chinese herbal medicine and parts of medicine. In particular, Curcuma longais also used as spice due to its unique fragrance, and curcumin substance, a Curcuma longa substance, is known as an outstanding anti-cancer substance. Thus, it is a Chinese herbal medicine that is attracting the attention of numerous researchers. This research conducted an experiment, separating substances that are found in Curcuma longa. In particular, curcumin(CUR), activation material,and other effective substances were subjected to extraction, separation and purification to verify effective substance index by production site by leveraging a method of analysis that uses HPLC. Moreover, this research separated herb medicine substances such as imported material and Curcuma aromatica to conduct comparative analysis of substances. Thus, this research was attempted to develop base data for the medicine development as well as identification of substances that may be used as additive for beverages, and to differentiate Korean made from imports. Towards this end, each chemical combination of separated substances are subjected to physical property, IR, ¹H-NMR, GC Mass, HPLC spectrum analysis for publication.

      • 알코올의 섭취가 흰쥐의 혈액과 간의 지질대사에 미치는 영향

        To investigate the effects of ethanol on the lipid metabolism in blood and liver, twenty one male Sprague Dawley rats had orally taken the 25% alcohol and 50% alcohol by 5g per Kg of weight during 3 weeks after the adjusting period. For the energy balance, 85% sucrose and 45% sucrose was added in control group and 25% alcohol group each. The weight was reduced by alcohol treatment according to the levels, otherwise the weight of liver and heart was little increased in alcohol groups rather than in control without significance. Moreover, RBC, WBC, hemoglobin and hematocrit were increased by alcohol treatment according to the levels without significance. Otherwise, adjusted bilirubin levels by 100g of weight was significantly increased by alcohol treatment dose dependently. Alcohol induced RBC degradation and increased circulated bilirubin because of liver damage. For blood lipid profiles, blood cholesterol was increased in 50% of alcohol group compared to that of 25% and control groups, otherwise, blood triglycerides (TG) was increased by 30% in 25% alcohol groups rather than in control but not significantly different. The hepatic lipid composition, cholesterol levels in alcohol treatment groups was lower than those in control. The hepatic TG was not influenced by alcohol treatment. The two reasons of decreasing hepatic lipids were suggested by 85% sucrose of control for energy balance more synthesized cholesterol and TG than those in 25 or 50% alcohol groups, and by cholesterol and TG could not be synthesized because of liver damage from alcohol.

        Quercetin-3-O-${\beta}$-D-glucuronide isolated from Polygonum aviculare inhibits cellular senescence in human primary cells

        Cellular senescence is known to contribute to tissue aging, a variety of age-related diseases, tissue regeneration, and cancer. Therefore, aging intervention might be useful for prevention of aging as well as age-related disease. In this study, we investigated compounds from Polygonum aviculare to determine if they inhibited cellular senescence in human primary cells, human dermal fibroblasts (HDFs) and human umbilical vein endothelial cells (HUVECs). Ten compounds from P. aviculare were purified and their inhibitory effects on adriamycin-induced cellular senescence were measured by observing senescence-associated ${\beta}$-galactosidase (SA-${\beta}$-gal) activity and reactive oxygen species. Among them, compound 9 (quercetin-3-O-${\beta}$-D-glucuronide) showed inhibitory effects against cellular senescence in HDFs and HUVECs treated with adriamycin. Additionally, compound 9 rescued replicative senescence in HDFs and HUVECs. These data imply that compound 9 represses cellular senescence in human primary cells and might be useful for the development of dietary supplements or cosmetics that ameliorate tissue aging or aging-associated diseases.

        Identification of Plant Extracts that Inhibit Cellular Senescence in Human Fibroblasts, Endothelial Cells, and Vascular Smooth Muscle Cell

        To identify plant extracts capable of inhibiting cellular senescence, the effects of plant extracts on adriamycin-induced cellular senescence in human fibroblasts, human umbilical vein endothelial cells (HUVECs), and vascular smooth muscle cells were investigated. After adriamcyin treatment, the effects of plant extracts on cellular senescence were evaluated by measuring senescence-associated ${\beta}$-galactosidase activity of the cells. Among 799 plant extracts, those from Rhei Rhizoma, Cirsii Radix, and Plantagnis Semen repressed adriamycin-induced cellular senescence in fibroblasts. Extracts of Cinnamomi Cortex and Cinnamomi Cortex Spissus were effective in endothelial cells. Extracts of Euonymi Lignum Suberalatum, Salicis Radicis Cortex, Polygoni aviculari Herba, and Chaenomelis langenariae Radix reduced senescence in vascular smooth muscle cells. These results suggested the effectiveness of plant extracts in reducing cellular senescence of human cells. These plant extracts could be used to develop dietary supplements or cosmetics for modulating tissue aging or aging-associated diseases.

        Background: This study aimed to identify the clinical characteristics and prognosis of primary gastric lymphoma detected by screening upper endoscopy in high prevalence area of Helicobacter pylori ( H. pylori) infection. Methods: Between October 2003 and May 2013, consecutive subjects who were diagnosed with primary gastric lymphoma by screening upper endoscopy were retrospectively enrolled at Seoul National University Hospital Healthcare System Gangnam Center, Seoul, Korea. Results: During the study period, a total of 101,103 subjects received 207,114 screening upper endoscopy. Among them, primary gastric lymphoma was detected in 49subjects. They were 53.0 ± 10.5 years of age, and 72.9% (35/49) were female. Histologic type of primary gastric lymphoma was predominantly (98.0%, 48/49) extranodal marginal zone B-cell lymphoma of mucosa-associated lymphoid tissue (MALT lymphoma), and one remaining case (2.0%) was diffuse large B-cell lymphoma. Initial stage was IEa in 95.9% (47/49), and H. pylori was positive in 91.8% (43/49). Among 41 H. pylori-positive patients with gastric MALT lymphoma stage IEa, H. pylori eradication achieved complete remission in 95.1% (39/41). There were three cases of recurrence with (n = 2) or without (n = 1) reinfection of H. pylori. Conclusions: Upper endoscopy as a screening for upper gastrointestinal malignancymay detect gastric MALT lymphoma in early stage, which can be successfully treated with H. pylori eradication, especially in H. pylori endemic area. For those who achieved complete remission, it is needed to undertake surveillance for recurrence.

        Prevalence of serum allergen-specific immunoglobulin E for canine atopic dermatitis in Korea

        Canine atopic dermatitis (CAD) is an allergic skin disease with characteristic clinical features associated with immunoglobulin E (IgE) antibodies. Identification of the causative allergens is the diagnostic goal, which is essential to treat and manage CAD patients. CAD is commonly associated with environmental allergens surrounding the patients. For this reason, it is important for diagnostic tests to select allergens that are related to the environment of each country and each province. There are two main allergen-specific tests, serological IgE test (SAT) and intradermal skin test (IDT). SAT did not show direct cutaneous reaction but did show serological reaction against allergens. However, SAT is simpler and more convenient than IDT in small animal practice. In this study, we selected domestically prevalent allergens for SAT, including 60 food allergens and 60 inhalant allergens, and tested eight dogs tentatively diagnosed with CAD based on Favrot’s criteria. Furthermore, IDT was performed on four dogs from the SAT group for comparison of SAT and IDT, and the results were very similar. In SAT, four types of mites (Bloomia tropicalis, Glycophagus domesticus, Euroglyphus maynei, and mite mixture 1 Korea; house dust mites), four types of molds (Botrytis cinerea, Alternaria alternata, mold fungi mixture 11, mold fungi mixture), and one type of pollen (tree pollen mix 3 Korea) induced a reaction in more than half of dogs tested. In IDT, all four dogs reacted positively to Dermatophagoides farinae, and three reacted positively to Dermatophagoides pteronyssinus and house dust. The mean agreement rate between SAT and IDT in this study was 76.3%. This is the first trial to apply local allergens for SAT in Korean veterinary medicine, and it might play an important role for diagnoses and management of animal allergic diseases.
