        배양한 흰쥐 대뇌세포의 저산소증 모델에서 황금(黃芩)이 유전자 표현에 미치는 영향

        Objectives : The purpose of this investigation is to evaluate the effects of Scutellaria baicalensis GEORGI on alteration in gene expression in a hypoxia model using cultured rat cortical cells. Methods : E18 rat cortical cells were grown in a Neurobasal medium containing B27 supplement. On 12 DIV, Scutellaria baicalensis GEORGI(20 ug/ml) was added to the culture media and left for 24 hrs. On 11 DIV, cells were given a hypoxic insult $(2%\;O_2/5%\;CO_2,\;37^{\circ}C,\;3\;hrs)$, returned to normoxia and cultured for another 24 hrs. Total RNA was prepared from Scutellaria baicalensis GEORGI-untreated (control) and -treated cultures and alteration in gene expression was analysed by microarray using rat 5K-TwinChips. Results : For most of the genes altered in expression, the Global M values were between -0.5 to +0.5. Among these, 1143 genes increased in their expression by more than Global M +0.1, while 1161 genes decreased by more than Global M -0.1. Effects on some of the genes whose functions are implicated in neural viability are as follows: 1) The expression of apoptosis-related genes such as Bad (Global M = 0.39), programmed cell death-2(Pdcd2) (Global M = 0.20) increased, while Purinergic receptor P2X(P2rxl) Global M = -0.22), Bc12-like1(Bc1211)(Global M = -0.19) decreased. 2) The expression of 'response to stress-related genes such as antioxidation-related AMP-activated protein kinase subunit gamma 1 gene (Prkag1) (Global M = 0.14), catalase gene (Global M = 0.14) and Heme Oxygenase(Hmoxl) increased. 3) The expression of Fos like antigen 2 (Fos12) expressed in neurons that survive ischemic insult increased (Global M = 0.97). Conclusions : these data suggest that Scutellaria baicalensis GEORGI increases the expression of antiapoptosis- and antioxidation- related genes in a way that can not yet be explained.

        Integrated analysis of copy number alteration and RNA expression profiles of cancer using a high-resolution whole-genome oligonucleotide array

        Recently, microarray-based comparative genomic hybridization (array-CGH) has emerged as a very efficient technology with higher resolution for the genome-wide identification of copy number alterations (CNA). Although CNAs are thought to affect gene expression, there is no platform currently available for the integrated CNA-expression analysis. To achieve high-resolution copy number analysis integrated with expression profiles, we established human 30k oligoarray-based genome- wide copy number analysis system and explored the applicability of this system for integrated genome and transcriptome analysis using MDA-MB-231 cell line. We compared the CNAs detected by the oligoarray with those detected by the 3k BAC array for validation. The oligoarray identified the single copy difference more accurately and sensitively than the BAC array. Seventeen CNAs detected by both platforms in MDA-MB-231 such as gains of 5p15.33-13.1, 8q11.22-8q21.13, 17p11.2, and losses of 1p32.3, 8p23.3-8p11.21, and 9p21 were consistently identified in previous studies on breast cancer. There were 122 other small CNAs (mean size 1.79 mb) that were detected by oligoarray only, not by BAC-array. We performed genomic qPCR targeting 7 CNA regions, detected by oligoarray only, and one non-CNA region to validate the oligoarray CNA detection. All qPCR results were consistent with the oligoarray-CGH results. When we explored the possibility of combined interpretation of both DNA copy number and RNA expression profiles, mean DNA copy number and RNA expression levels showed a significant correlation. In conclusion, this 30k oligoarray- CGH system can be a reasonable choice for analyzing whole genome CNAs and RNA expression profiles at a lower cost.

        Dieckol Suppresses CoCl2-induced Angiogenesis in Endothelial Cells

        Dieckol is a polyphenol compound isolated from brown algae that has anti-oxidant, anti-inflammatory, and anti-tumor activity. We examined the anti-angiogenic effects of dieckol in endothelial cells under hypoxic conditions. Treatment with CoCl2, a hypoxic mimetic agent, increased proliferation, adhesion, migration, and tube formation in HUVECs, as well as vessel sprouting in rat aortic rings, which correlated well with increased expression of hypoxia-inducible factor 1-alpha (HIF1α) and β1-integrin. Dieckol suppressed CoCl2-induced adhesion, migration, and tube formation in HUVECs and vessel sprouting in rat aortic rings. Dieckol treatment decreased CoCl2-induced overexpression of HIF1α and its downstream signaling molecules, including β1-integrin/Fak, Akt/eNOS, and p38 MAPK. These results suggest that dieckol is a novel angiogenesis inhibitor and a potential treatment for angiogenesis-dependent diseases in humans, such as malignant tumors.

        카레분 및 향신료 추출물의 항 돌연변이 효과

        Antimutagenic effects of extracts from curry powder and its individual fourteen kinds of spices, were investigated by Ames test. The antimutagenic effects against a direct mutagen, 2-nitrofluorene(2-NF), and two indirect mutagens, 2-anthramine(2-AT) and 2-acetamidofluorene (2-AF) in the S. typhimurium TA98 were tested. For the 2-NF, the antimutagenicity of cinnamon, feungreek, fennel, ginger, clove, turmeric and celery seed were determined as 42, 38, 32, 28, 24, 23 and 20%, respectively. The antimutagenicity of clove against the 2-AT was the highest (116%), and followed by the order of celery seed (103%), cardamon (100%), red pepper (99%), cinnamon(92%), cumin(83%), ginger(82%), fennel(82%), coriander (71%), nutmeg(68%) and turmeric (55%). The results also showed that the antimutagenic effect of clove against the 2-AF was superior to other spices. In case of curry powder among more than 10 kinds of spices, the antimutagenenicity against the 2-AT and 2-AF showed 23% and 6%, respectively, but no effect was observed against the 2-NF.

        음식물 폐기물을 이용한 황토볼 제조시 적정 혼합비 연구

        Experiments were conducted to propose manufacturing methods to make loess ball using food wastes. Food wastes were used to improve porosity and increase specific surface area of loess ball. Red clay, food wastes and clay were used as main raw materials in making loess ball and these were mixed with varying ratio in order to find out optimum mixing ratio. The optimum mixing ratio of food wastes is evaluated to amount to 30%. In this case, 33.61 kg/㎠ of compressive strength, 65.13% of porosity, 50.04% of absorbing ratio, and 6.302 ㎡/g of specific surface area are obtained. When evaluating cross section and the appearance of red clay ball made up of 30% food wastes through a visual inspection and SEM photograph, it can be observed that a lot of pores are formed across the red clay ball due to the volatilization of moisture and organic matter in food wastes during the high-temperature burning. Taking into account compressive strength, porosity, absorbing ratio, specific surface area, and SEM photograph altogether, the optimum mixing ratio of loess, food wastes, and clay in the loess ball were 30%, 50%, and 20%, respectively. When evaluating the cross-section and outer appearance of the loess ball that comprise 30% of food wastes by SEM photography, it can be concluded that the optimum mixing ratio of loess, food wastes, and clay in the loess ball amounts to 30%, 50%, and 20%, respectively. Compressive strength of the manufactured loess ball amounts to 42.52 kg/㎠, which is 26.5% higher than the values obtained in the condition when the loess ball made without food wastes. However, the values of porosity, absorbing ratio, and specific surface area are somewhat lower than the corresponding values obtained in the condition when the loess ball was manufactured without food wastes.

      • 六一順氣湯 抽出物이 생쥐의 全腦虛血에 미치는 影響

        생쥐의 全腦虛血 모델에서 potassium cyanide 誘發 昏睡時間 및 生存時間, 減壓性 無酸素 負荷時 生存時間을 測定하여 全腦虛血時 腦循環代謝 改善 效果를 觀察하였다. KCN 誘發 昏睡時間의 短縮, 致死量의 KCN에 대한 生存時間의 延長, 減壓에 의한 無酸素 負荷時 生存時間의 延長 效果가 나타났다. The effect of Yukilsunki-tang extracts on global cerebral ischemia were investigated in this study. The multiple parameters of global cerebral ischemia assessed in mice included the duration of KCN-induced(1.8㎎/㎏ i.v.) coma, the survival time of KCN-induced(3.0㎎/㎏ i,v.) coma the survival time exposed to hypoxia induced by vacuum pump. In the case of global cerebral ischemia International Cancer Research mice were used and divided into two groups at random. Group A, normal control, was treated after oral administration of normal saline. Group B, experimental control, was treated after oral administration of 13.2㎎/20g of Yukilsunki-tang extracts. Each treatment was KCN-induced(1.8㎎/㎏ i.v.) coma, KCN-induced(3.0㎎/㎏ i.v.) coma and exposure to hypoxia induced by vacuum pump. The results were obtained as follows : In global cerebral ischemia, Yukilsunki-tang extracts significantly prolonged the duration of KCN-induced(1.8㎎/㎏ i.v.) coma, the survival time of KCN-induced(3.0㎎/㎏ i.v.) coma and the survival time of exposure to hypoxia induced by vacuum pump in mice. Conclusion Yukilsunki-tang extracts had a significant effect on Global cerebral ischemia.

        삼화탕(三化湯)이 중간대뇌동맥 결찰로 유발된 뇌허혈에 미치는 영향

        The purpose of this investigation is to evaluate the effects of Samwha-tang(三化湯) Extracts on reversible forebrain ischemia experimentally induced from the occlusion of middle cerebral artery. The volume of cerebral ischemia, the volume of cerebral edema, and the change of pyramidal neuron of the CA1 area in hippocampus through light microscopy were investigated. we obtained the following results. The volume of the control group, which had ischemic damage was 21%, and the volume of the sample group, which had ischemic damage, was 16%. The ratio of the volume of the right/left hemisphere was 117.2 in the control group, and 108.8 in the sample group. Also, the light microscopy revealed that the pyramidal cells of CA1 area in hippocampus had many damages like changes into discontinuous and unsystematic forms. But, in the sample group, the cells were less damaged compared with the control group.

        사회적 자본이 지식 축적에 미치는 영향: 업종의 조절효과를 중심으로

        본 연구는 공급사슬환경에서 구매자-공급자 관계 사이 사회적 결과물인 사회적 자본이 공급자 관점에서공급사슬 관계자인 두 주체 간에 일어나는 지식축적에 미치는 영향과, 이러한 과정이 업종에 따라 나타나는조절효과의 영향 검토를 목적으로 한다. 특히 사회적 자본은 기존에 연구된 구매자 관점과 달리 공급자 관점에더욱 중요한 의미를 나타내기 때문에, 구매자-공급자 관계에서 어떻게 구현되고 있는지 실증하고자 하였다. 본 연구의 목적은 공급자의 관점에서 공급사슬 관계에 있어 사회적 자본과 지식축적의 영향 관계를 실증적으로 분석하는 데에 있다. 구체적 연구 목적은 다음과 같다. 첫째, 공급자 관점에서 사회적 자본을 구조적 자본, 인지적 자본 그리고 관계적 자본으로 세분화하여 지식 축적에 어떻게 영향을 미치는가 파악하고자 한다. 이는기존에 연구가 부족하였던 공급자 관점의 사회적 자본과 지식 축적간의 관계 파악을 목적으로 한다. 둘째, 제조업과 정보통신업을 중심으로 사회적 자본과 지식 축적 간의 관계를 정(+)적으로 조절할 수 있는 조절요인으로 업종을 분석하고자 한다. 분석결과, 공급사슬환경에서 사회적 자본의 구조적 자본, 인지적 자본, 관계적 자본 요인 중 관계적 자본이지식축적에 가장 중요한 요인으로 나타났으며, 구조적 자본과 인지적 자본이 다음으로 지식축적에 높은 영향을미치는 변수로 나타났다. 또한, 사회적 자본과 지식 축적의 관계에 대한 업종의 조절효과는 제조업의 경우사회적 자본 중 구조적 자본과 지식축적을 제외하고, 인지적 자본과 지식축적, 관계적 자본과 지식축적의관계를 정(+)적으로 조절하였다. 아울러, 정보통신업은 구조적 자본과 지식축적, 인지적 자본과 지식축적, 관계적 자본과 지식축적의 관계를 정(+)적으로 조절하였다. 본 연구를 통하여 공급사슬환경의 공급자 관점에서 사회적 자본이 지식축적에 영향을 미치는 과정에 대한이해를 넓히고, 업종에 따라 사회적 자본과 지식축적의 관계에 대한 차별화된 경영전략과 지속적인 성과 향상에도움을 줄 수 있는 공급사슬관리의 전략적 방향성이 요구된다는 이론적, 실무적 시사점을 도출하였다. This study is to examine the effect of social capital, a study result between buyer and supplier in supply chain environment, on knowledge accumulation between two suppliers and suppliers, and the effect of moderating effects of these processes depending on industry. Especially, since societal capital has more important meaning in the supplier's perspective than the previous buyer's perspective, this study tried to prove how this relationship is realized within the buyer-supplier relationship. The purpose of this study is to empirically analyze the relationship between social capital and knowledge accumulation in supply chain relationship from the viewpoint of suppliers. First, from the supplier's perspective, social capital is divided into structural capital, cognitive capital, and relational capital to understand how it affects knowledge accumulation. Second, this study aims to analyze the industry as a moderating factor that can positively control the relationship between social capital and knowledge accumulation centering on manufacturing and information and communication industries. For this purpose, a survey was conducted on workers in manufacturing and information and communication industries in Seoul and Gyeonggi Province to collect data. The measurement data were statistically analyzed on the validity of the measurement items and the suitability of the measurement model through descriptive statistics, exploratory factor analysis, correlation analysis, and confirmatory factor analysis.In addition, the research hypothesis was verified through path analysis and multiple group analysis on the research model. As a result of the analysis, 195 manufacturing companies were 50.9%, and 188 information and communication companies were 49.1%. In addition, 247 companies with an average sales of less than 50 billion won accounted for 64.6%, while 292 companies with less than 500 regular workers accounted for 76.3%. In addition, the trading period with buyers was 177 with less than 10 years, 46.2%, and 53.8% with 206 with more than 10 years. As a result of the analysis, among the structural capital, cognitive capital, and relational capital factors of social capital in the supply chain environment, relational capital was the most important factor in knowledge accumulation, and structural capital and cognitive capital were the second most influential variables on knowledge accumulation. In addition, the moderating effect of the industry is social capital, which significantly controls the relationship between cognitive capital, knowledge accumulation, relational capital and knowledge accumulation except for structural capital, but the information and communication industry significantly controls the relationship between structural capital and knowledge accumulation, cognitive capital and knowledge accumulation, relational capital and knowledge accumulation. The implications of this study are as follows: First, the role and importance of social capital and knowledge accumulation in the situation where efficient management strategies of the supply chain participating organizations are required are empirically verified. Second, this study proved the effect of social capital on knowledge accumulation from the viewpoint of suppliers, unlike previous studies focused on buyers. Third, it verified the moderating effect of social capital and knowledge accumulation by industry. Through this study, it was found that the understanding of the process of social capital affecting knowledge accumulation from the supplier's point of view of supply chain environment was broadened, and the strategic direction of supply chain management was required to help the differentiated management strategy and continuous performance improvement of the relationship between social capital and knowledge accumulation according to the industry.
