      • High Sustained Virologic Response with Daclatasvir plus Asunaprevir in HCV GT-1b Chinese, Korean and Taiwanese without Baseline NS5A Polymorphisms

        Aims: Daclatasvir (DCV) plus asunaprevir (ASV) has demonstrated highsustained virologic response (SVR) in HCV genotype (GT-)1b infection.NS5A-Y93H and NS5A-L31 resistance-associated polymorphisms(RAPs) to DCV are known to impact DCV+ASV response in GT-1b-infectedJapanese. The effect of RAPs on SVR at posttreatment week12 (SVR12) to DCV+ASV was explored in mainland Chinese, Korean,and Taiwanese.Methods: Pooled data from 2 studies of DCV (60 mg daily) + ASV(100 mg capsule, twice-daily) for 24 weeks in GT-1b-infected interferon/ribavirin-naive and -experienced patients from mainland China,Korea, and Taiwan. Similar Japanese data (4 studies; n=445) werepooled for comparison. SVR12 with versus without baseline Y93Hand/or L31 RAPs was compared by age (<65 vs ≥65 years), cirrhosisstatus, and baseline HCV-RNA.Results: SVR12 and baseline NS5A sequences were available for 282patients (126 mainland Chinese [45%〕, 80 Koreans [28%〕, 76Taiwanese [27%〕). NS5A-Y93H and/or -L31 RAPs were observed pretreatmentin 8% mainland Chinese, 14% Korean, and 18%Taiwanese patients, compared with 19% in Japanese. SVR12 in allnon-Japanese patients is shown (Figure); rates were broadly similarbetween countries and with Japanese data (Japanese: 96% overallwithout RAPs, 41% with RAPs). Responses were lower among patientswith baseline RAPs. By contrast, SVR12 in patients without RAPs washigh (92-100%), irrespective of cirrhosis, age, or baseline HCV-RNA.Conclusions: At least 95% of HCV GT-1b-infected patients from mainlandChina, Korea or Taiwan without baseline NS5A-Y93H or -L31polymorphisms who had HCV-RNA ≤7 log10 IU/mL achieved SVR12on DCV+ASV, regardless of cirrhosis status and age.

        Gentiana straminea supplementation improves feed intake, nitrogen and energy utilization, and methane emission of Simmental calves in northwest China

        Objective: Native plants can be used as additives to replace antibiotics to improve ruminant feed utilization and animal health. An experiment was conducted to evaluate the effects of Gentiana straminea (GS) on nutrient digestibility, methane emissions, and energy metabolism of Simmental calves. Methods: Thirty-two (5-week-old) male Simmental clves, with initial body weight (BW) of 155±12 kg were fed the same basal diet of concentrates (26%), alfalfa hay (37%), and oat hay (37%) and were randomly separated into four treatment groups according to the amount of GS that was added to their basal diet. The four different groups received different amounts of GS as a supplement to their basal diet during whole experiment: (0 GS) 0 mg/kg BW, the control; (100 GS) 100 mg/kg BW; (200 GS) 200 mg/kg BW; and (300 GS) 300 mg/kg BW. Results: For calves in the 200 GS and 300 GS treatment groups, there was a significant increase in dry matter (DM) intake (p<0.01), average daily gain (ADG) (p<0.05), organic matter intake (p<0.05), DM digestibility (p<0.05), neutral detergent fibre (NDF) digestibility (p<0.05), and acid detergent fibre (ADF) digestibility (p<0.05). Dietary GS supplementation result in quadratic increases of DM intake (p<0.01), ADG (p<0.05), NDF intake (p<0.05), and ADF intake (p<0.05). Supplementing the basal diet with GS significantly increased nitrogen (N) retention (p<0.001) and the ratio of retention N to N intake (p<0.001). Supplementing the basal diet with GS significantly decreased methane (CH4) emissions (p<0.01), CH4/BW0.75 (p<0.05) and CH4 energy (CH4-E) (p<0.05). Dietary GS supplementation result in quadratic increases of CH4 (p<0.01) and CH4/DM intake (p<0.01). Compared with 0 GS, GS-supplemented diets significantly improved their gross energy intake (p<0.05). The metabolizable energy and digestive energy intake were significantly greater for calves in the 100 GS and 200 GS calves than for 0 GS calves (p<0.05). Conclusion: From this study, we conclude that supplementing calf diets with GS could improve utilization of feed, energy, and N, and may reduce CH4 emissions without having any negative effects on animal health.         R&D Status of High-current Accelerators at IFP

        High-current accelerators have many important applications in Z-pinches, high-power microwaves, and free electron lasers, imploding liners and radiography and so on. Research activities on Zpinches, imploding liners, radiography at the Institute of Fluid Physics (IFP) are introduced. Several main high-current accelerators developed and being developed at IFP are described, such as the Linear Induction Accelerator X-Ray Facility Upgrade (LIAXFU, 12 MeV, 2.5 kA, 90 ns), the Dragon-I linear induction accelerator (20 MeV, 2.5 kA, 60 ns), and the Primary Test Stand for Z-pinch (PTS, 10 MA, 120 ns). The design of Dragon-II linear induction accelerator (20 MeV, 2.5 kA, 3 × 60 ns) to be built will be presented briefly.

      • Ternary solvent for CH3NH3PbI3 perovskite films with uniform domain size

        <P>A ternary solvent system consisting of dimethyl sulfoxide (DMSO), g-butyrolactone (GBL) and N-methyl-2-pyrrolidone (NMP) has been developed to improve the uniformity of CH3NH3PbI3 (MAPbI(3)) perovskite domains. Compared to MAPbI(3) perovskite films prepared using a binary solvent consisting of DMSO and GBL, the surface roughness and uniformity of MAPbI(3) films fabricated by using the ternary solvent system are greatly improved. The thermogravimetric analysis reveals that a NMP-PbI2-MAI intermediate, a DMSO-PbI2-MAI intermediate and MAPbI(3) crystals co-existed in the as-cast MAPbI(3) films. Furthermore, it is found that the thermal stability of intermediate phases and the solvent evaporation rate are critical for the nucleation of the perovskite crystals during the thermal annealing treatment. The thermally stable intermediates prepared with the ternary solvent are converted to MAPbI(3) films with a highly uniform and smooth surface. The film forms an intimate contact with the charge transporting layer when the layer is applied as a photoactive layer in the solar cell. As a result, the power conversion efficiency of ternary solvent processed solar cells is enhanced by 38.2% compared to that of the binary solvent processed one. Furthermore, the stability of the ternary processed perovskite solar cells is greatly improved, as well. This investigation provides a better understanding about the role of different processing solvents or additives in effecting the perovskite film quality.</P>

        Effective protection of sequential solution-processed polymer/fullerene bilayer solar cell against charge recombination and degradation

        Both charge recombination and degradation in sequential solution processed polymer/fullerene bilayer organic photovoltaics (OPV) are effectively reduced by the insertion of a TiO<SUB>2</SUB> inter-layer between the bilayer and Al electrode. The polymer/fullerene bilayer composed of a poly(3-hexylthiophene) (P3HT) bottom-layer and a [6,6] phenyl C61-butyric acid methyl ester (PCBM) top-layer shows significant change in morphology due to the substantial inter-penetration of P3HT and PCBM during the thermal annealing process. Consequently, the bilayer surface becomes P3HT rich resulting in significant charge recombination at the bilayer/Al interface of the bilayer OPV. The charge recombination rate of the bilayer OPV is reduced by one order of magnitude upon the insertion of a TiO<SUB>2</SUB> nanoparticle inter-layer between the bilayer and the Al electrode after the thermal annealing process. In contrast, when the thermal annealing process is conducted after insertion of the inter-layer, the effect of the TiO<SUB>2</SUB> inter-layer becomes insignificant. The V<SUB>OC</SUB> and efficiency of the bilayer OPV is greatly enhanced from 0.37 to 0.66V and 1.2% to 3.7%, respectively by utilizing the properly constructed TiO<SUB>2</SUB> inter-layer in the bilayer OPV. Additionally, insertion of the TiO<SUB>2</SUB> inter-layer significantly improves the stability of the bilayer OPV. The bilayer OPV with a TiO<SUB>2</SUB> inter-layer maintains 51% of its initial PCE after storage under dark ambient conditions for 700h without encapsulation, whereas the bilayer OPV without a TiO<SUB>2</SUB> inter-layer did not show any solar cell performance after 200h under the same conditions.

      • Patterns of somatic alterations between matched primary and metastatic colorectal tumors characterized by whole-genome sequencing

        Colorectal cancer (CRC) patients have poor prognosis after formation of distant metastasis. Understanding the molecular mechanisms by which genetic changes facilitate metastasis is critical for the development of targeted therapeutic strategies aimed at controlling disease progression while minimizing toxic side effects. A comprehensive portrait of somatic alterations in CRC and the changes between primary and metastatic tumors has yet to be developed. We performed whole genome sequencing of two primary CRC tumors and their matched liver metastases. By comparing to matched germline DNA, we catalogued somatic alterations at multiple scales, including single nucleotide variations, small insertions and deletions, copy number aberrations and structural variations in both the primary and matched metastasis. We found that the majority of these somatic alterations are present in both sites. Despite the overall similarity, several de novo alterations in the metastases were predicted to be deleterious, in genes including FBXW7, DCLK1 and FAT2, which might contribute to the initiation and progression of distant metastasis. Through careful examination of the mutation prevalence among tumor cells at each site, we also proposed distinct clonal evolution patterns between primary and metastatic tumors in the two cases. These results suggest that somatic alterations may play an important role in driving the development of colorectal cancer metastasis and present challenges and opportunities when considering the choice of treatment.

        Optimized finite element model updating method for damage detection using limited sensor information

        Limited, noisy data in vibration testing is a hindrance to the development of structural damage detection. This paper presents a method for optimizing sensor placement and performing damage detection using finite element model updating. Sensitivity analysis of the modal flexibility matrix determines the optimal sensor locations for collecting information on structural damage. The optimal sensor locations require the instrumentation of only a limited number of degrees of freedom. Using noisy modal data from only these limited sensor locations, a method based on model updating and changes in the flexibility matrix successfully determines the location and severity of the imposed damage in numerical simulations. In addition, a steel cantilever beam experiment performed in the laboratory that considered the effects of model error and noise tested the validity of the method. The results show that the proposed approach effectively and robustly detects structural damage using limited, optimal sensor information.

        The Safety and Efficacy of a Dietary Herbal Supplement and Gallic Acid for Weight Loss

        The objective of this study was to test the safety and efficacy of NT, a dietary herbal supplement made fromrhubarb, ginger, astragulus, red sage, and turmeric, combined with gallic acid (GA) to reduce food intake and cause weightloss. A total of 105 healthy subjects, 1860 years old with a body mass index of 2535 kg/m2 and on no chronic medication,were randomized to a 300 mg/1.2 g NT-GA combination, a 600 mg/2.4 g NT-GA combination, or placebo in three divideddoses daily for 24 weeks. Food intake was measured at baseline and 2 weeks, and safety parameters were followed regularly.Pharmacokinetic studies of a 200 mg/800 mg NT-GA combination and 800 g GA alone were performed with and withoutfood. There was no dose-related weight loss or reduction in food intake at the 8-week analysis, and the study was terminatedearly. Pharmacokinetic studies showed plasma levels of GA did not increase above 10 .M and were not dose-related. TheNT-GA at all concentrations was well tolerated, but was ineffective in causing weight loss or in suppressing food intake. Phar-macokinetics suggested that GA plasma levels were limited by oral absorption, and may be the reason for lack of efficacy.
