A 2,656 bp fragment of chicken ghrelin gene was cloned and SNPs were detected by PCR-RFLP and Allele Specific PCR (ASP) in 12 Chinese indigenous chicken breeds and a commercial chicken population. The results showed that there were 23 base variations and an amino acid change (Gln->Arg) in cloned chicken ghrelin gene. Three SNPs were confirmed in 13 populations and associations between this gene and growth traits of Tibetan chicken (TC) and Recessive White chicken (RW) were investigated. The results of haplotype analysis revealed that 26 haplotype genotypes were composed of eight haplotypes. The results of x2 tests indicated that there were significant differences between genotypes or haplotype genotype frequencies in some of the breeds or sexes at 0.05 or 0.01 levels. The results of ANOVA revealed that there were significant differences between genotypes or haplotype genotypes on some growth traits of TC and RW chicken breeds at 0.05 or 0.01 levels. Multiple comparisons showed that there were significant associations between genotype CT at site 71 and some growth traits of two chicken breeds and between genotype AG at site 1,215 and body weight at 16 wk of two chicken breeds, and there was a significant association between haplotype genotype CAA/CAG and body weight and shank girth at 16 wk of two chicken breeds.

Located at the important tumor suppressor locus, 3p22, PLCD1 encodes an enzyme that mediates regulatory signaling of energy metabolism, calcium homeostasis and intracellular movements. We identified PLCD1 as a downregulated gene in aerodigestive carcinomas through expression profiling and epigenetic characterization. We found that PLCD1 was expressed in all normal adult tissues but low or silenced in 84% (16/19) gastric cancer cell lines, well correlated with its CpG island (CGI) methylation status. Methylation was further detected in 62% (61/98) gastric primary tumors, but none of normal gastric mucosa tissues. PLCD1 methylation was significantly correlated with tumor high stage. Detailed methylation analysis of 37 CpG sites at the PLCD1 CGI by bisulfite genomic sequencing confirmed its methylation. PLCD1 silencing could be reversed by pharmacological demethylation with 5-aza-2′-deoxycytidine, indicating a direct epigenetic silencing. Ectopic expression of PLCD1 in silenced gastric tumor cells dramatically inhibited their clonogenicity and migration, possibly through downregulating MMP7 expression and hampering the reorganization of cytoskeleton through cofilin inactivation by phosphorylation. Thus, epigenetic inactivation of PLCD1 is common and tumor-specific in gastric cancer, and PLCD1 acts as a functional tumor suppressor involved in gastric carcinogenesis.Oncogene (2009) 28, 2466–2475; doi:10.1038/onc.2009.92; published online 18 May 2009
R&D Status of High-current Accelerators at IFP

High-current accelerators have many important applications in Z-pinches, high-power microwaves, and free electron lasers, imploding liners and radiography and so on. Research activities on Zpinches, imploding liners, radiography at the Institute of Fluid Physics (IFP) are introduced. Several main high-current accelerators developed and being developed at IFP are described, such as the Linear Induction Accelerator X-Ray Facility Upgrade (LIAXFU, 12 MeV, 2.5 kA, 90 ns), the Dragon-I linear induction accelerator (20 MeV, 2.5 kA, 60 ns), and the Primary Test Stand for Z-pinch (PTS, 10 MA, 120 ns). The design of Dragon-II linear induction accelerator (20 MeV, 2.5 kA, 3 × 60 ns) to be built will be presented briefly.
Reproducing kernel based evaluation of incompatibility tensor in field theory of plasticity

This paper employs the reproducing kernel (RK) approximation for evaluation of field theory-based incompatibility tensor in a polycrystalline plasticity simulation. The modulation patterns, which is interpreted as mimicking geometrical-type dislocation substructures, are obtained based on the proposed method. Comparisons are made using FEM and RK based approximation methods among different support sizes and other evaluation conditions of the strain gradients. It is demonstrated that the evolution of the modulation patterns needs to be accurately calculated at each time step to yield a correct physical interpretation. The effect of the higher order strain derivative processing zone on the predicted modulation patterns is also discussed.

It has been known that mandible ramus flexure is an important morphologic trait for sex determination. However, it will be unavailable when mandible is incomplete or fragmented. Therefore, the anthropometric analysis on incomplete or fragmented mandible becomes more important. The aim of this study is to investigate the sex-discriminant potential of mandible ramus flexure on the Korean three-dimensional (3D) mandible models with anthropometric analysis. The sample consists of 240 three dimensional mandibular models obtained from Korean population (M:F; 120:120, mean age 46.2y), collected by The Catholic Institute for Applied Anatomy, The Catholic University of Korea. Anthropometric information about 11 metric was taken with Mimics, anthropometry libraries toolkit. These parameters were subjected to different discriminant function analyses using SPSS 17.0. Univariate analyses showed that the resubstitution accuracies for sex determination range from 50.4 to 77.1%. Mandibular flexure upper border (MFUB), maximum ramus vertical height (MRVH), and upper ramus vertical height (URVH) expressed the greatest dimorphism, 72.1 to 77.1%. Bivariate analyses indicated that the combination of MFUB and MRVH hold even higher resubstitution accuracy of 81.7%. Furthermore, the direct and stepwise discriminant analyses with the variables on the upper ramus above flexure could predict sex in 83.3 and 85.0%, respectively. When all variables of mandibular ramus flexure were input in stepwise discriminant analysis, the resubstitution accuracy arrived as high as 88.8%. Therefore, we concluded that the upper ramus above flexure hold the larger potentials than the mandibular ramus flexure itself to predict sexes, and that the equations in bivariate and multivariate analysis from our study will be helpful for sex determination on Korean population in forensic science and law.

<P>Amphiphile selection is a crucial step in membrane protein structural and functional study. As conventional detergents have limited scope and utility, novel agents with enhanced efficacy need to be developed. Although a large number of novel agents have been reported, so far there has been no systematically designed comparative study of the protein stabilization efficacy of stereo-isomeric amphiphiles. Here we designed and prepared a novel class of stereo-isomeric amphiphiles, designated butane-1,2,3,4-tetraolbased maltosides (BTMs). These stereoisomers showed markedly different behaviour for most of the targeted membrane proteins depending on the chirality of the linker region. These findings indicate an important role for detergent stereochemistry in membrane protein stabilization. In addition, we generally observed enhanced detergent efficacy with increasing alkyl chain length, reinforcing the importance of the balance between hydrophobicity and hydrophilicity in detergent design. The stereo-isomeric difference in detergent efficacy observed provides an important design principle for the development of novel amphiphiles for membrane protein manipulation.</P>
