        Analysis of elliptical Hertz contact of steel wires of stranded-wire helical spring

        A stranded-wire helical spring is formed of a multilayer and coaxial strand of several wires twisted together with the same direction ofspiral. Because of the multilayer structure, the wear on the local area of the steel wires’ surface is related to the elliptical contact betweenadjacent wires during working process. Based on Boussinesq potential functions and elastic half-space model, the contact area and surfacepressure of elliptical Hertz contact were investigated. Moreover, these surface contact quantities are used to expand the calculationof subsurface stress relevant to elliptical contact. It is found that the greater contact angle of two contact wires, the smaller contact areaand greater maximum contact pressure. Meanwhile, the magnitude of subsurface von Mises stress is much higher when the contact angleincreases.

        MiR-124 regulates apoptosis in hypoxia-induced human brain microvessel endothelial cells through targeting Bim

        Human brain microvessel endothelial cells (HBMECs) are crucial for brain vascular repair and maintenance. The high-expressed expressions of micro- RNA-124 (miR-124) in brain have been investigated and revealed in many researches. In this work, we aimed to investigate the role of miR-124 in apoptosis of HBMECs and the underlying mechanism. Here, we found the lowexpressed miR-124 in hypoxia-induced HBMECs using qRT-PCR analysis. MiR-124 targeting 30-untranslated region (30-UTR) of Bim mRNA was predicted by Targetscan database. Importantly, the decreased miR-124 expression and increased Bim expression, an opposite trend, were obtained in hypoxia-induced HBMECs. The further confirmation of the correlation between miR-124 and Bim was conducted by miR-124 overexpression and dual luciferase reporter assays. The inhibitory role of miR- 124 in Bim expression was evidenced by results obtained from miR-124 overexpression analysis. Luciferase reporter assay further proved that miR-124 directly targeted the two conserved seed sites in the Bim 30-UTR. The inhibited apoptosis of HBMECs was observed under both miR-124 overexpression and Bim knockdown condition in flow cytometry analysis. Collectively, these findings outline that miR-124 regulates apoptosis in hypoxia-induced HBMECs through targeting Bim, providing a better understanding of the role of miR-124 in apoptosis of HBMECs.

        Research on torsional fretting wear behaviors and damage mechanisms of stranded-wire helical spring

        Wear on the local area of steel wires’ surface is attributed to torsional fretting on the working process of stranded-wire helical spring. A mathematical model to calculate normal contact force and angular displacement amplitude among the wires is established first when the spring is impacted. With the experimental parameters obtained from the model, the torsional fretting test, which stimulates torsional fretting among the wires in the working process of the spring, is realized successfully on a newly developed fretting tester. Torsional fretting behaviors are strongly dependent upon normal contact force, angular displacement amplitude, and number of cycles. There are three basic types of T-θ curves (short for torque), angular displacement curves during the process of torsional fretting, namely, parallelogram,elliptic, and linear T-θ curves. To analyze the damage mechanisms, distribution maps of oxygen in the wear scar of spring wires under different working conditions are revealed. The damage gets slight in the partial slip region, mainly with the abrasive wear and the slight oxidative wear, whereas the wear mechanisms are mainly the abrasive wear, the oxidative wear, and the delamination, accompanied with obvious plastic deformation in the mixed fretting and slip regions.

        Analysis of Free Amino Acids during Fermentation by Bacillus subtilis Using Capillary Electrophoresis

        A high performance capillary electrophoresis (HPCE) method was presented to identify and quantitate free amino acids during fermentation by Bacillus subtilis. Amino acids, pre-column derivatized with phenylisothicyanate,were separated and characterized by HPCE. In order to optimize separation conditions, the assay was developed by varying the β-cyclodextrin concentration and pH of the background electrolyte. A buffer system comprising 30 mM phosphate and 3 mM β-cyclodextrin at pH 7.0, voltage of 20 kV and detection wavelength of 254 nm showed the best results, with 17 out of 20phenylthioncarbamyl amino acids in a solution adequately separated. For quantification, p-aminobenzoic acid was added as an internal standard. Analysis of free amino acids in Bacillus subtilis culture medium using this method revealed good consistency with the values obtained using conventional ninhydrin-based amino acid analyzer. Four free amino acids (aspartic acid, glutamic acid, proline, and tyrosine) concentration in an extracellular matrix during fermentation by Bacillus subtilis were mainly monitored using this method.

        Mutations of ARX and non-syndromic intellectual disability in Chinese population

        Mutations of Aristaless-related homeobox (ARX) gene were looked as the third cause of non-syndromic intellectual disability (NSID), while the boundary between true disease-causing mutations and non-disease-causing variants within this gene remains elusive. To investigate the relationship between ARX mutations and NSID, a panel comprising six reported causal mutations of the ARX was detected in 369 sporadic NSID patients and 550 random participants in Chinese. Two mutations, c.428_451 dup and p.G286S, may be disease-causing mutations for NSID, while p.Q163R and p.P353L showed a great predictive value in female NSID diagnosis with significant associations (X2 = 19.60, p = 9.54e−6 for p.Q163R; X2 = 25.70, p = 4.00e−07 for p.P353L), carriers of these mutations had an increased risk of NSID of more than fourfold. Detection of this panel also predicted significant associations between genetic variants of the ARX gene and NSID (p = 3.73e−4). The present study emphasized the higher genetic burden of the ARX gene on NSID in the Chinese population, molecular analysis of this gene should be considered for patients presenting NSID of unknown etiology.
