Comprehensive Analysis of Gut Microbiota Alteration in the Patients and Animal Models with Polycystic Ovary Syndrome

        Zhou Jing,Qiu Xuemei,Chen Xuejing,Ma Sihan,Chen Zhaoyang,Wang Ruzhe,Tian Ying,Jiang Yufan,Fan Li,Wang Jingjie 한국미생물학회 2023 The journal of microbiology Vol.61 No.9

Polycystic ovary syndrome (PCOS) is a common disease of endocrine–metabolic disorder, and its etiology remains largely unknown. The gut microbiota is possibly involved in PCOS, while the association remains unclear. The comprehensive analysis combining gut microbiota with PCOS typical symptoms was performed to analyze the role of gut microbiota in PCOS in this study. The clinical patients and letrozole-induced animal models were determined on PCOS indexes and gut microbiota, and fecal microbiota transplantation (FMT) was conducted. Results indicated that the animal models displayed typical PCOS symptoms, including disordered estrous cycles, elevated testosterone levels, and ovarian morphological change; meanwhile, the symptoms were improved after FMT. Furthermore, the microbial diversity exhibited disordered, and the abundance of the genus Ruminococcus and Lactobacillus showed a consistent trend in PCOS rats and patients. The microbiota diversity and several key genera were restored subjected to FMT, and correlation analysis also supported relevant conclusions. Moreover, LEfSe analysis showed that Gemmiger, Flexispira, and Eubacterium were overrepresented in PCOS groups. Overall, the results indicate the involvement of gut microbiota in PCOS and its possible alleviation of endocrinal and reproductive dysfunctions through several special bacteria taxa, which can function as the biomarker or potential target for diagnosis and treatment. These results can provide the new insights for treatment and prevention strategies of PCOS.

Dissimilatory Azoreduction of Orange I by a Newly Isolated Moderately Thermophilic Bacterium, Novibacillus thermophilus SG-1

        Zhen Yu,Xuemei Zhou,Yueqiang Wang,Guiqin Yang,Shungui Zhou 한국생물공학회 2015 Biotechnology and Bioprocess Engineering Vol.20 No.6

Dye wastewater normally is discharged at high temperature, but thermophilic bacteria capable of decolorizing azo dyes have rarely been isolated. Here we report a newly isolated moderately thermophilic bacterium, Novibacillus thermophilus SG-1, which had a remarkable ability to decolorize the azo dye Orange I by utilizing a large variety of organic substrates as electron donors. When Orange I served as the sole electron acceptor, almost complete decolorization occurred at 50ºC and pH 8.0 with acetate as the electron donor after anaerobic incubation of strain SG-1 for 24 h. The decolorization process followed the pseudofirst- order kinetics. The complete reduction of 0.3 mM Orange I was accompanied by a stoichiometric consumption of 0.17 mM acetate over time. The measured molar ratio (1.76) of Orange I reduced to acetate oxidized was close to the theory value of 2.0, suggesting that most of the electrons released by acetate had been transported to Orange I. Simultaneously energy generated from the electron transfer process was used to support cell anaerobic growth, which meant that azoreduction by strain SG-1 is an azorespiration process. To our knowledge, this is the first report of a thermophilic bacterium capable of azorespiration, which increases the limited number of bacteria for treating hightemperature azo dye wastewater.

Temporal expression profiling of long noncoding RNA and mRNA in the peripheral blood during porcine development

        Gu, Yiren,Zhou, Rui,Jin, Long,Tao, Xuan,Zhong, Zhijun,Yang, Xuemei,Liang, Yan,Yang, Yuekui,Wang, Yan,Chen, Xiaohui,Gong, Jianjun,He, Zhiping,Li, Mingzhou,Lv, Xuebin Asian Australasian Association of Animal Productio 2020 Animal Bioscience Vol.33 No.5

Objective: We investigated the temporal expression profiles of long noncoding RNA (lncRNA) and mRNA in the peripheral blood of pigs during development and identified the lncRNAs that are related to the blood-based immune system. Methods: Peripheral blood samples were obtained from the pigs at 0, 7, 28, and 180 days and 2 years of age. RNA sequencing was performed to survey the lncRNA and mRNA transcriptomes in the samples. Short time-series expression miner (STEM) was used to show temporal expression patterns in the mRNAs and lncRNAs. Gene ontology and Kyoto encyclopedia of genes and genomes analyses were performed to assess the genes' biological relevance. To predict the functions of the identified lncRNAs, we extracted mRNAs that were nearby loci and highly correlated with the lncRNAs. Results: In total of 5,946 lncRNA and 12,354 mRNA transcripts were identified among the samples. STEM showed that most lncRNAs and mRNAs had similar temporal expression patterns during development, indicating the expressional correlation and functional relatedness between them. The five stages were divided into two classes: the suckling period and the late developmental stage. Most genes were expressed at low level during the suckling period, but at higher level during the late stages. Expression of several T-cell-related genes increased continuously during the suckling period, indicating that these genes are crucial for establishing the adaptive immune system in piglets at this stage. Notably, lncRNA TCONS-00086451 may promote blood-based immune system development by upregulating nuclear factor of activated T-cells cytoplasmic 2 expression. Conclusion: This study provides a catalog of porcine peripheral blood-related lncRNAs and mRNAs and reveals the characteristics and temporal expression profiles of these lncRNAs and mRNAs during peripheral blood development from the newborn to adult stages in pigs.

Optical Encryption Scheme with Multiple Users Based on Computational Ghost Imaging and Orthogonal Modulation

        Sheng Yuan,Xuemei Liu,Xin Zhou,Zhongyang Li 한국광학회 2016 Current Optics and Photonics Vol.20 No.4

For the application of multiusers, the arrangement and distribution of the keys is a much concerningproblem in a cryptosystem. In this paper, we propose an optical encryption scheme with multiple usersbased on computational ghost imaging (CGI) and orthogonal modulation. The CGI encrypts the secret imageinto an intensity vector rather than a complex-valued matrix. This will bring convenience for post-processingand transmission of the ciphertext. The orthogonal vectors are taken as the address codes to distinguishusers and avoid cross-talk. Only the decryption key and the address code owned by an authorized userare matched, the secret image belonging to him/her could be extracted from the ciphertext. Therefore, thereare two security levels in the encryption scheme. The feasibility and property are verified by numericalsimulations.

SYNTHESIZING A WELL-ALIGNED CARBON NANOTUBE FOREST WITH HIGH QUALITY VIA THE NEBULIZED SPRAY PYROLYSIS METHOD BY OPTIMIZING ULTRASONIC FREQUENCY

        ZHI YANG,HUAGUI NIE,XUEMEI ZHOU,ZHEN YAO,SHAOMING HUANG,XIAOHUA CHEN 성균관대학교(자연과학캠퍼스) 성균나노과학기술원 2011 NANO Vol.6 No.4

Nebulized spray pyrolysis (NSP) offers ease of scaling into an industrial scale process method for large-scale CNT synthesis. In the method, ultrasonic frequency plays an important role in the formation of well-aligned carbon nanotubes (CNTs). In this work, we obtain well-aligned CNTs with high quality by optimizing ultrasonic frequency via the NSP method. SEM, TEM and HRTEM results show that the aligned CNTs obtained under an ultrasonic frequency of 1.8 MHz have good alignment, uniform nanotube length (~200 μm), high density and no trace of other carbon nanoparticles. The Raman spectra result further confirms that the aligned CNTs have a high graphitization degree. Furthermore, we find that ultrasonic frequency is an important factor in determining the nanotube diameter. The higher the ultrasonic frequency, the smaller the nanotube diameter. This rule may be very helpful in the preparation of large-scale well-aligned CNTs with high quality and diameter-controlled at low cost.

Cloning and expression of a Chk1 gene in Daphnia pulex during different modes of reproduction

        Xiaoge Guo,Shanliang Xu,Xuemei Yan,Wei Zhou,Xinyu Dai,Xiu Zou,Chunlin Wang,Danli Wang,Yunlong Zhao 한국유전학회 2015 Genes & Genomics Vol.37 No.9

The full-length cDNA of a Chk1 gene (DpChk1) was cloned from Daphnia pulex using RACE method. This cDNA is 1767 bp in length, consisting of a 1497 bp open reading frame encoding 498 amino acid protein. The deduced DpChk1 protein sequence contains phosphorylation sites, three conserved Ser-Gln and Thr- Gln motifs and shares 51–55 % similarity with A. pisum, M. rotundata, D. melanogaster, A. echinatior, B. terrestris, R. pulchellus and M. occidentalis, respectively. Gene expression analysis showed that Chk1 is differentially expressed in D. pulex during different reproductive modes. DpChk1 expression in sexually-reproducing females and males is significantly higher than in parthenogenetic Daphnia. Whole-mount in situ hybridization revealed that DpChk1 was expressed during two kinds of reproductive modes. In parthenogenetic females, hybridization signals were found in the antennae and thoracic limb, whereas expression levels in the corresponding sites of sexuallyreproducing females were relatively strong, DpChk1 could not be detected in the gonads of ephippial and parthenogenetic females. Taken together, these different reproductive stages and sex specific expression patterns are regulated temporally and spatially. We speculate that DpChk1 may be involved in conversion between different stages of reproduction and in sexual differentiation in D. pulex.

INVESTIGATION OF HOMOLOGOUS SERIES AS PRECURSORY HYDROCARBONS FOR ALIGNED CARBON NANOTUBE FORMATION BY THE SPRAY PYROLYSIS METHOD

        ZHI YANG,HUAGUI NIE,XUEMEI ZHOU,ZHEN YAO,SHAOMING HUANG,XIAOHUA CHEN 성균관대학교(자연과학캠퍼스) 성균나노과학기술원 2011 NANO Vol.6 No.3

The precursory carbon source is one of the key parameters which govern the formation of carbon nanotubes (CNTs). In this work, by selecting four homologous series, namely n-pentane, n-hexane, n-heptane and n-octane, as investigated targets, we comparatively study the relationship between thermodynamic properties of the precursory carbon source and formation of aligned CNTs. We find that all of these alkanes are favored for the growth of aligned CNTs in a suitable growth environment. But only n-heptane can yield the aligned CNTs with relatively high quality, high yield and narrow diameter distribution. Furthermore, after considering the link between thermodynamic properties of the precursory carbon source and the morphology characteristic of the nanotube samples, we find that the Gibbs free energy and formation enthalpy of precursory carbon sources play critical roles in the nanotube formation. In additions some possible explanations are proposed to better understand these phenomena. These rules will be very helpful in making clearer the formation mechanism from the precursory carbon sources to the CNTs, and preparing large-scale aligned CNTs with diameter control at low cost.

Experimental study of TiO2 hollow microspheres removal on elemental mercury in simulated flue gas

        Jiang Wu,Xian Li,Jian-Xing Ren,Xuemei Qi,Ping He,Bu Ni,Chong Zhang,Chengzhen Hu,Jun Zhou 한국공업화학회 2015 Journal of Industrial and Engineering Chemistry Vol.32 No.-

TiO2 hollow sphere was synthesized by hydrothermal method using trifluoroacetic acid (TFA) andTi(SO4)2 as raw materials, and it was applied to photocatalytic oxidization of elemental mercury (Hg0) inthe simulated flue gas. The prepared samples were well characterized by X-ray diffraction (XRD),scanning electron microscopy (SEM), transmission electron microscopy (TEM), X-ray photoelectronspectroscopy (XPS), UV–vis diffuse reflectance spectra (UV-vis DRS) and nitrogen adsorption/desorption. The results showed that only anatase phase TiO2 hollow sphere was obtained. The average diameter ofTiO2 hollow spheres was about 800 nm and the shell thickness was about 200–400 nm. When M, themolar ratio of TFA to Ti(SO4)2, was higher than a certain value, the photocatalytic activity of preparedTiO2 hollow spheres began to reduce. When M = 2, addition TFA content was the best quantity, which gotthe best TiO2 hollow spheres and provided the most appropriate F decomposed from TFA during thesample preparation, which was helpful to its photocatalytic ability. In the experimental temperaturerange, the photocatalytic oxidation efficiency decreased with the increasing of reaction temperature, andthe intensity of UV irradiation had an important effect on the photocatalytic reaction. When UVirradiation intensity was 303.45 mW cm 2 and reaction temperature was 55 8C, TiO2 hollow spheresample prepared at M = 2 had the highest photocatalytic ability and the mercury removal efficiencyreached up to 82.75%.
