      • Laser Raman detection of platelet as a non‐invasive approach for early and differential diagnosis of Alzheimer's disease

        <P><B>Abstract</B></P><P>Early and differential diagnosis of Alzheimer's disease (AD) is a problem that puzzled many doctors. Reliable markers in easy‐assembling samples are of considerable clinical diagnostic value. In this work, laser Raman spectroscopy (LRS) was developed a new method that potentially allows early and differential diagnosis of AD from the platelet sample. Raman spectra of platelets isolated from different ages of AD transgenic mice and non‐transgenic controls were collected and analyzed. Multilayer perceptron networks (MLP) classification method was used to classify spectra and establish the diagnostic models. For differential diagnosis, spectra of platelets isolated from AD, Parkinson’s disease (PD) and vascular dementia (VD) mice were also discriminated. Two notable spectral differences at 740 and 1654 cm<SUP>–1</SUP> were revealed in the mean spectrum of platelets isolated from AD transgenic mice and the controls. MLP displayed a powerful ability in the classifying of early, advanced AD and the control group, and in differential diagnosis of PD and advanced AD, as well as VD and advanced AD. The results suggest that platelet detecting by LRS coupled with MLP analysis appears to be an easy and accurate method for early and differential diagnosis of AD.         Preparation and Characterization of Magnetic Cobalt Ferrites/SBA-15 Nanocomposite Adsorbents and the Removal of Methylene Blue

        In this paper, ordered mesoporous SBA-15 silica was synthesized by the hydrothermal method, and then a series of CoFe2O4/SBA-15 nanocomposites were synthesized by a facile impregnation method. X-ray diffraction and N2 adsorption–desorption isotherms were used to characterize the microstructure and morphology of SBA-15 and CoFe2O4/SBA-15 nanocomposites. CoFe2O4 nanoparticles presented spinel phase structure and existed in the mesopores of SBA-15. The magnetic response of CoFe2O4/SBA-15 nanocomposites was characterized with vibrating sample magnetometer (VSM). The adsorption efficiency of CoFe2O4/SBA-15 nanocomposites for methylene blue increased firstly with the increasing CoFe2O4 content, and then decreased. Sample-2 (SBA-15: CoFe2O4 = 1: 0.1 in the precursor) not only presented the best adsorptive performance, but also could be separated and retrieved effectively by magnetic separation technique.

      • Surface-enhanced Raman scattering using monolayer graphene-encapsulated Ag nanoparticles as a substrate for sensitive detection of 2,4,6-trinitrotoluene

        <P>We developed a surface-enhanced Raman scattering (SERS)-active substrate based on chemical vapor deposition (CVD)-grown monolayer graphene-encapsulated silver nanoparticles (Ag NPs) and used it for the detection of 2,4,6-trinitrotoluene (TNT). Monolayer graphene-encapsulated Ag NPs can be used as a stable substrate for up to 2 months under aerobic exposure. The single layer of graphene can effectively stabilize the Ag NPs against oxidation and corrosion in the presence of H2O2, Na2S and HNO3. Due to the combination of the Ag NPs with graphene, the Raman signals of probe molecules were dramatically enhanced. This platform exhibits extraordinarily high sensitivity and excellent stability for the detection of TNT, with a limit of detection as low as 6.6 x 10(-10) mol L-1. This work provides a simple approach to fabricate graphene-encapsulated Ag NPs for sensitive SERS sensing of TNT after Fenton oxidation degradation.</P>

      • In-situ high-energy synchrotron X-ray diffraction study of micromechanical behavior of multiple phases in Ni<sub>47</sub>Ti<sub>44</sub>Nb<sub>9</sub> shape memory alloy

        High-energy synchrotron X-ray diffraction technique was used to in-situ characterize microstructure, lattice strain, and phase transition behavior of a Ni<SUB>47</SUB>Ti<SUB>44</SUB>Nb<SUB>9</SUB> shape memory alloy. Phase transformation kinetics and deformation mechanisms were studied under a uniaxial tension at three testing temperatures, i.e., -70<SUP>o</SUP>C, 25<SUP>o</SUP>C, and 150<SUP>o</SUP>C. At a testing temperature of -70<SUP>o</SUP>C, a complicated phase transformation with four stages of micromechanical deformation was identified which is associated with changes of martensite substructures. At room temperature of 25<SUP>o</SUP>C, there was no stress-induced selection process of variants of B19' phases observed. Whereas at a testing temperature of 150<SUP>o</SUP>C, there was no any phase transformation observed. It is verified that β-Nb phase, an effective stabilizer for the austenite, delays the process of martensitic transformation and relaxes the strain energy without strengthening the matrix. This new finding is important to understand the relationship between the micromechanical deformation behavior and phase transformations in the Ni<SUB>47</SUB>Ti<SUB>44</SUB>Nb<SUB>9</SUB> SMA.

        <P> <B>Aim:</B> To assess and compare the efficacy and safety of liraglutide with those of glimepiride, both in combination with metformin for the treatment of type 2 diabetes in Asian population from China, South Korea and India.</P><P> <B>Methods:</B> A 16‐week, randomized, double‐blind, double‐dummy, four‐arm, active control trial was carried out. In total, 929 subjects with type 2 diabetes with a mean (±s.d.) age of 53.3 ± 9.5 years, HbA<SUB>1c</SUB> of 8.6 ± 1.0% and body weight of 68.1 ± 11.7 kg were randomized (liraglutide 0.6, 1.2 or 1.8 mg once daily or glimepiride 4 mg once daily all in combination with metformin: 1 : 1 : 1 : 1). One subject withdrew immediately after randomization and before exposure.</P><P> <B>Results:</B> HbA<SUB>1c</SUB> was significantly reduced in all groups compared with baseline. Treatment with liraglutide 1.2 and 1.8 mg was non‐inferior to glimepiride (mean HbA<SUB>1c</SUB> reduction: 1.36% points, 1.45% points and 1.39% points, respectively). No significant difference was shown in the percentage of subjects reaching American Diabetes Association HbA<SUB>1c</SUB> target <7% or American Association of Clinical Endocrinologists target ≤6.5% between liraglutide 1.2 and 1.8 mg and glimepiride. Liraglutide was associated with a 1.8–2.4 kg mean weight reduction, compared with a 0.1 kg mean weight gain with glimepiride. Liraglutide led to a significantly greater reduction in systolic blood pressure (SBP) compared with glimepiride. Two subjects in the glimepiride group reported major hypoglycaemia while none in the liraglutide groups. Liraglutide was associated with about 10‐fold lower incidence of minor hypoglycaemia than glimepiride. Gastrointestinal disorders were the most common adverse events (AEs) for liraglutide, but were transient and resulted in few withdrawals.</P><P> <B>Conclusions:</B> In Asian subjects with type 2 diabetes, once‐daily liraglutide led to improvement in glycaemic control similar to that with glimepiride but with less frequent major and minor hypoglycaemia. Liraglutide also induced a significant weight loss and reduced SBP and was generally well tolerated. The most frequently reported AE was transient nausea. The effect of liraglutide in this Asian population is comparable to the effects seen in Caucasian, African American and Hispanic populations in global liraglutide phase 3 trials.</P>

        FOXO protects against age‐progressive axonal degeneration

        <P><B>Summary</B></P><P>Neurodegeneration resulting in cognitive and motor impairment is an inevitable consequence of aging. Little is known about the genetic regulation of this process despite its overriding importance in normal aging. Here, we identify the Forkhead Box O (FOXO) transcription factor 1, 3, and 4 isoforms as a guardian of neuronal integrity by inhibiting age‐progressive axonal degeneration in mammals. FOXO expression progressively increased in aging human and mouse brains. The nervous system‐specific deletion of <I>Foxo</I> transcription factors in mice accelerates aging‐related axonal tract degeneration, which is followed by motor dysfunction. This accelerated neurodegeneration is accompanied by levels of white matter astrogliosis and microgliosis in middle‐aged <I>Foxo</I> knockout mice that are typically only observed in very old wild‐type mice and other aged mammals, including humans. Mechanistically, axonal degeneration in nerve‐specific <I>Foxo</I> knockout mice is associated with elevated mTORC1 activity and accompanying proteotoxic stress due to decreased Sestrin3 expression. Inhibition of mTORC1 by rapamycin treatment mimics FOXO action and prevented axonal degeneration in <I>Foxo</I> knockout mice with accelerated nervous system aging. Defining this central role for FOXO in neuroprotection during mammalian aging offers an invaluable window into the aging process itself.</P>

        Polymorphism of Ghrelin Gene in Twelve Chinese Indigenous Chicken Breeds and Its Relationship with Chicken Growth Traits

        A 2,656 bp fragment of chicken ghrelin gene was cloned and SNPs were detected by PCR-RFLP and Allele Specific PCR (ASP) in 12 Chinese indigenous chicken breeds and a commercial chicken population. The results showed that there were 23 base variations and an amino acid change (Gln->Arg) in cloned chicken ghrelin gene. Three SNPs were confirmed in 13 populations and associations between this gene and growth traits of Tibetan chicken (TC) and Recessive White chicken (RW) were investigated. The results of haplotype analysis revealed that 26 haplotype genotypes were composed of eight haplotypes. The results of x2 tests indicated that there were significant differences between genotypes or haplotype genotype frequencies in some of the breeds or sexes at 0.05 or 0.01 levels. The results of ANOVA revealed that there were significant differences between genotypes or haplotype genotypes on some growth traits of TC and RW chicken breeds at 0.05 or 0.01 levels. Multiple comparisons showed that there were significant associations between genotype CT at site 71 and some growth traits of two chicken breeds and between genotype AG at site 1,215 and body weight at 16 wk of two chicken breeds, and there was a significant association between haplotype genotype CAA/CAG and body weight and shank girth at 16 wk of two chicken breeds.

        R&D Status of High-current Accelerators at IFP

        High-current accelerators have many important applications in Z-pinches, high-power microwaves, and free electron lasers, imploding liners and radiography and so on. Research activities on Zpinches, imploding liners, radiography at the Institute of Fluid Physics (IFP) are introduced. Several main high-current accelerators developed and being developed at IFP are described, such as the Linear Induction Accelerator X-Ray Facility Upgrade (LIAXFU, 12 MeV, 2.5 kA, 90 ns), the Dragon-I linear induction accelerator (20 MeV, 2.5 kA, 60 ns), and the Primary Test Stand for Z-pinch (PTS, 10 MA, 120 ns). The design of Dragon-II linear induction accelerator (20 MeV, 2.5 kA, 3 × 60 ns) to be built will be presented briefly.

      • Derivatization reaction-based surface-enhanced Raman scattering (SERS) for detection of trace acetone

        <P>A facile method was developed for determination of trace volatile acetone by coupling a derivatization reaction to surface-enhanced Raman scattering (SERS). With iodide modified Ag nanoparticles (Ag IMNPs) as the SERS substrate, acetone without obvious Raman signal could be converted to SERS-sensitive species via a chemical derivatization reaction with 2,4-dinitrophenylhydrazine (2,4-DNPH). In addition, acetone can be effectively separated from liquid phase with a purge-sampling device and then any serious interference from sample matrices can be significantly reduced. The optimal conditions for the derivatization reaction and the SERS analysis were investigated in detail, and the selectivity and reproducibility of this method were also evaluated. Under the optimal conditions, the limit of detection (LOD) for acetone was 5 mg L-1 or 0.09 mM (3 sigma). The relative standard deviation (RSD) for 80 mg L-1 acetone (n=9) was 1.7%. This method was successfully used for the determination of acetone in artificial urine and human urine samples with spiked recoveries ranging from 92% to 110%. The present method is convenient, sensitive, selective, reliable and suitable for analysis of trace acetone, and it could have a promising clinical application in early diabetes diagnosis.         Effects of Maturity Stages on the Nutritive Composition and Silage Quality of Whole Crop Wheat

        The changes in yields and nutritive composition of whole crop wheat (Triticum aestivum L.) during maturation and effects of maturity stage and lactic acid bacteria (LAB) inoculants on the fermentation quality and aerobic stability were investigated under laboratory conditions. Whole crop wheat harvested at three maturation stages: flowering stage, milk stage and dough stage. Two strains of LAB (Lactobacillus plantarum: LAB1, Lactobacillus parafarraqinis: LAB2) were inoculated for wheat ensiling at $1.0{\times}10^5$ colony forming units per gram of fresh forage. The results indicated that wheat had higher dry matter yields at the milk and dough stages. The highest water-soluble carbohydrates content, crude protein yields and relative feed value of wheat were obtained at the milk stage, while contents of crude fiber, neutral detergent fiber and acid detergent fiber were the lowest, compared to the flowering and dough stages. Lactic acid contents of wheat silage significantly decreased with maturity. Inoculating homofermentative LAB1 markedly reduced pH values and ammonia-nitrogen ($NH_3$-N) content (p<0.05) of silages at three maturity stages compared with their corresponding controls. Inoculating heterofermentative LAB2 did not significantly influence pH values, whereas it notably lowered lactic acid and $NH_3$-N content (p<0.05) and effectively improved the aerobic stability of silages. In conclusion, considering both yields and nutritive value, whole crop wheat as forage should be harvested at the milk stage. Inoculating LAB1 improved the fermentation quality, while inoculating LAB2 enhanced the aerobic stability of wheat silages at different maturity stages.
