        LncRNA A2M-AS1 lessens the injury of cardiomyocytes caused by hypoxia and reoxygenation via regulating IL1R2

        Background: Myocardial ischemia and reperfusion injury (MI/RI) is a complex pathophysiological process, which can lead to severe myocardial injury. The long noncoding RNA alpha-2-macroglobulin antisense RNA 1 (A2M-AS1) has been revealed to be abnormally expressed in MI, However, its function in MI and the potential mechanism are still unclear. Objective: To evaluate the functional role of A2M-AS1 in hypoxia/reoxygenation (H/R)-induced neonatal cardiomyocytes and its potential molecular mechanism. Methods: Dataset GSE66360 was obtained from GEO database for analyzing the RNA expression of A2M-AS1 and interleukin 1 receptor type 2 (IL1R2). KEGG pathway enrichment analysis of the genes that co-expressed with A2M-AS1 was performed. Human neonatal cardiomyocytes were subjected to H/R to construct in vitro models. QRT-PCR and Western blot were adopted to test the levels of mRNA and protein. The viability and apoptosis of cardiomyocytes were tested by CCK-8 and flow cytometry assays, respectively. Results: The expression of A2M-AS1 was notably downregulated in H/R-treated cardiomyocytes. Overexpression of A2M-AS1 can notably enhance the cell viability of H/R-damaged cardiomyocytes, whereas knockdown of A2M-AS1 showed the opposite outcomes. Besides, a negative correlation was showed between A2M-AS1 and IL1R2 expression. In H/R-treated cardiomyocytes, overexpression of IL1R2 weakened the promoting proliferation and anti-apoptosis effects caused by overexpressing A2M-AS1, however, IL1R2-knockdown abolished the anti-proliferation and pro-apoptosis effects caused by silencing A2M-AS1. Conclusion: This study demonstrates the potential regulatory role of A2M-AS1/ IL1R2 axis in cardiomyocytes suffered from H/R, and provides insight into the protection of MI/RI.

        Fever in cancer patients is often due to the following causes: evil qi and toxity stagnancy, disorders of qi and blood, deficiencies of zang and fu organs, and the disorder of yin and yang. The treatments given to cancer patients with a fever are according to five: (a) Excessive inner heat and toxicants: remove heat and the toxicant, induce purgation. We use Cheng-Qi-Tang plus Qing-Wen-Bai-Du-Yin. (b) Tangle of damp and heat, and qi stagnancy: remove damp and heat, smooth the qi channel. We use Gan-Lu-Xiao-Du-Dan or San-Ren-Tang. (c) Obvious blood and heat stagnancy: remove heat and blood stasis. We use Xue-Fu-Zhu-Yu-Tang. (d) Deficiency of spleen qi, inner heat caused by a yin deficiency: nourish spleen qi and yin to remove the inner heat. We use Bu-Zhong-Yi-Qi-Tang or Xiao-Jian-Zhong-Tang. (e) Prominent yin deficiency and hectic fever: replenish yin and remove inner heat. We use Qing-Hao-Bie-Jia-Tang or Chai-Qian-Mei-Lian-San. The pathogenesis of fever in cancer patients is complicated. We can see both deficiency and excess in one differentiation. Therefore, we must make sure of it, then we can get the most effective treatment.

        Effects of chelated Zn/Cu/Mn on redox status, immune responses and hoof health in lactating Holstein cows

        To evaluate the effects of chelated Zn/Cu/Mn on redox status, immune responses and hoof health in lactating Holstein cows, 48 head in early lactation were divided into healthy or lame groups according to their gait score. Cows were fed the same amount of Zn/Cu/Mn as sulfate salts or in chelated forms for 180 days, and foot-and-mouth disease (FMD) vaccine was injected at day 90. The results showed that lame cows had lower antioxidant function, serum Zn/Mn levels, hair Cu levels, and hoof hardness. Moreover, increased antioxidant status, FMD antibody titers, serum and hair levels of Zn/Cu/Mn, and hoof hardness and decreased milk fat percent and arthritis biomarkers were observed in cows fed chelated Zn/Cu/Mn. In summary, supplementation with chelated Zn/Cu/Mn improved antioxidant status and immune responses, reduced arthritis biomarkers, and increased accumulation of Zn/Cu/Mn in the body and hoof hardness in dairy cows.

        User Commitment to Blockchain-Based Social Media Platforms from the Perspective of Perceived Justice Regarding the Token Reward System: the Mediating Role of Psychological Ownership

        Purpose – In this study, we aimed to theorize blockchain-based social media platform users’ commitment by examining the impact of their perceived justice of the token reward system. In addition, this study applied psychological ownership theory to verify the underlying mechanism between users’ perceptions of justice and their commitment to the platforms. Research design, data, and methodology – To empirically test our conceptual framework in the study, we collected data through a web-based survey approach from the responses of 385 users who had experience with blockchainbased social media platforms. We employed a structural equation modeling approach to empirically test our proposed hypotheses. Result – The results indicated that distributive justice and informational justice have positive effects on user commitment. The results also showed that psychological ownership plays an important role in mediating the relationship between users’ sense of distributive justice and commitment, and between procedural justice and commitment. The findings provided a better understanding of the sense of justice and user commitment in a blockchain-based social media environment. Conclusion – This study represents a preliminary attempt to theorize and empirically examine blockchain-based social media platform users’ commitment. This study provided important contributions to the literature on how the effect of users’ sense of justice in a reward system affects their commitment to blockchain-based social media platforms.

      • Enhancement of the thermoelectric performance of bulk SnTe alloys via the synergistic effect of band structure modification and chemical bond softening

        <P>SnTe alloys, which have the same crystal structure as PbTe, have attracted increasing attention. Here, we demonstrate that the synergistic effect of band structure modification and chemical bond softening can be realized simultaneously in In & Mn doped SnTe bulk alloys. The Seebeck coefficient and power factor are synergistically improved by co-doping of In and Mn. In doping is known to introduce a resonance level. Mn doping reduces the separation of light- and heavy-valence bands. The combination of these effects significantly enhances the Seebeck coefficient at room temperature owing to around a factor of five times increase in the band effective mass. The reduction of thermal conductivity is from the decrease of both the electronic and phononic parts. The electronic thermal conductivity is decreased by the increase in defect scattering, as can be confirmed by the carrier mobility. The force constant of the bonds around the Te site is decreased due to the co-doping of In & Mn, which indicates that the chemical bonds are softened, which leads to low sound velocity and lower lattice thermal conductivity. As a result, the peak thermoelectric figure of merit,<I>zT</I>= 1.03 has been achieved for Sn0.89In0.01Mn0.1Te at 923 K. This strategy of using the synergistic effect of band structure modification and chemical bond softening could be applicable to other thermoelectric materials.</P>

        Annealing Characteristics of Planar Waveguides in Lanthanum Gallium Silicate Crystal by Oxygen Implantation

        Planar waveguides have been fabricated in z-cut lanthanum gallium silicate (LGS) crystal by 6.0 MeV oxygen-ionsimplantation with a dose of 6×10^15 ions/cm². The waveguides were characterized by the prism-coupling method. The dark modes are measured before and after annealing at 600, 700, 750 and 800℃ for different times in air. The refractive index profiles are reconstructed by using a reflectivity calculation method. TRIM'98 code is used to simulate the process of the oxygen-ion implantation into LGS. It is found that the refractive-index change may be partly due to the damage induced by nuclear collision, and the waveguide has thermal stability below 600℃.         The Positive Association between Subclinical Hypothyroidism and Newly-Diagnosed Hypertension Is More Explicit in Female Individuals Younger than 65

        Background: Subclinical hypothyroidism (SCH) is the most common thyroid dysfunction, and its relationship with blood pressure(BP) has been controversial. The aim of the study was to analyze the association between SCH and newly-diagnosed hypertension. Methods: Based on data from the Thyroid disease, Iodine nutrition and Diabetes Epidemiology (TIDE) study, 49,433 euthyroid individuals and 7,719 SCH patients aged ≥18 years were enrolled. Patients with a history of hypertension or thyroid disease were excluded. SCH was determined by manufacturer reference range. Overall hypertension and stage 1 and 2 hypertension were diagnosedaccording to the guidelines issued by the American College of Cardiology/American Heart Association in 2017. Results: The prevalence of overall hypertension (48.7%), including stage 1 (28.9%) and 2 (19.8%) hypertension, increased significantly in SCH patients compared with euthyroid subjects. With elevated serum thyroid stimulating hormone (TSH) level, the hypertension prevalence also increased significantly from the euthyroid to different SCH subgroups, which was more profound in femalesor subjects aged <65 years. The age- and sex-specific regression analysis further demonstrated the same trends in the general population and in the 1:1 propensity matched population. Similarly, several BP components (i.e., systolic, diastolic, and mean arterial BP)were positively associated with TSH elevation, and regression analysis also confirmed that all BP components were closely relatedwith SCH in female subjects aged <65 years. Conclusion: The prevalence of hypertension increases for patients with SCH. SCH tends to be associated with hypertension and BPcomponents in females younger than 65 years.

      • A New Cell Counting Method to Evaluate Anti-tumor Compound Activity

        Determining cell quantity is a common problem in cytology research and anti-tumor drug development. A simple and low-cost method was developed to determine monolayer and adherent-growth cell quantities. The cell nucleus is located in the cytoplasm, and is independent. Thus, the nucleus cannot make contact even if the cell density is heavy. This phenomenon is the foundation of accurate cell-nucleus recognition. The cell nucleus is easily recognizable in images after fluorescent staining because it is independent. A one-to-one relationship exists between the nucleus and the cell; therefore, this method can be used to determine the quantity of proliferating cells. Results indicated that the activity of the histone deacetylase inhibitor Z1 was effective after this method was used. The nude-mouse xenograft model also revealed the potent anti-tumor activity of Z1. This research presents a new anti-tumor-drug evaluation method.

      • Preparation and Antitumor Activity of a Tamibarotene-Furoxan Derivative

        Multi-target drug design, in which drugs are designed as single molecules to simultaneously modulate multiple physiological targets, is an important strategy in the field of drug discovery. QT-011, a tamibarotene-furoxan derivative, was here prepared and proposed to exert synergistic effects on antileukemia by releasing nitric oxide and tamibarotene. Compared with tamibarotene itself, QT-011 displayed stronger antiproliferative effects on U937 and HL-60 cells and was more effective evaluated in a nude mice U937 xenograft model in vivo. In addition, QT-011 could release nitric oxide which might contribute to the antiproliferative activity. Autodocking assays showed that QT-011 fits well with the hydrophobic pocket of retinoic acid receptors. Taken together, these results suggest that QT-011 might be a highly effective derivative of tamibarotene and a potential candidate compound as antileukemia agent.

        Pharmacokinetic Behavior of Gentiopicroside From Decoction of Radix Gentianae, Gentiana Macrophylla After Oral Administration in Rats: a Pharmacokinetic Comaprison with Gentiopicroside after Oral and Intravenous Administration Alone

        The pharmacokinetics in rats of gentiopicroside (GPS) from orally administered decoctions of Radix Gentianae (DRG) and Gentiana macrophlla (DGM) were compared with that of GPS alone administered at 150 mg/kg orally and 30 mg/kg intravenously. The metabolic profile of GPS after intravenous injection could be fitted to two-compartment model whereas oral administration decoctions DRG or DGM, or GPS alone, could all be fitted to a one-compartment model. After oral administration of GPS alone, GPS was absorbed quickly and reached a maximum plasma concentration ($C_{max}$) value, 5.78 ${\pm}$ 2.24 ${\mu}g/mL$ within 0.75 ${\pm}$ 0.62 h. The plasma level of GPS declined with a $T_{1/2ke}$, 3.35 ${\pm}$ 0.76 h. After oral administration of decoctions DRG and DGM, GPS was absorbed and reached significantly higher maximum concentrations of 10.53 ${\pm}$ 3.20 ${\mu}g/mL$ (p < 0.01) and 7.43 ${\pm}$ 1.64 ${\mu}g/mL$ (p < 0.05) at later time points 1.60 ${\pm}$ 0.76 (p < 0.01) and 2.08${\pm}$0.74 h (p < 0.05), for DRG and DGM respectively, compared with oral GPS alone. Significantly larger AUC values were found for decoctions of GPS (83.49 ${\pm}$ 20.8 ${\mu}g{\cdot}h/mL$ for DRG and 59.43 ${\pm}$ 12.9 ${\mu}g{\cdot}h/mL$ for DGM) compared with oral GPS alone (32.67 ${\pm}$ 12.9 ${\mu}g{\cdot}h/mL$. The results demonstrate that the bioavailability of GPS was markedly improved when administered as a decoction than as purified GPS. The decoction from Radix Gentianae provided 2.5 times better bioavailability and that from Gentiana macrophlla 1.8 times higher. The study confirms the importance of careful pharmacokinetic analysis in the characterization of herbal medicines when applied for future clinical applications.
