      • A Phase 3 Evaluation of Daclatasvir plus Asunaprevir in Treatment-Naive Patients with Chronic HCV Genotype 1b Infection

        Background/Aims: Treatment-naive GT 1b-infected patients from mainland China, South Korea and Russia were assessed for SVR at follow-up week 12 (SVR12) after receiving daclatasvir (60 mg, QD) and asunaprevir (100 mg, BID) (DCV+ASV). Methods: Patients were randomized 3:1 to receive DCV+ASV (24 weeks; immediate treatment [IM]) or 12 weeks of placebo followed by DCV+ASV (24 weeks; placebo-deferred treatment [PD]). The primary endpoint was to evaluate SVR12 in the IM arm to the historical rate for peginterferon/ribavirin (70%). Secondary endpoints included overall safety and safety comparisons between the treatment arms during the first 12 weeks. Results: 207 patients were randomized to IM (n=155) or PD (n=52); Asian (86%), female (59%), IL28B CC genotype (68%) and median age 49 (range 18-73) years; cirrhosis (13%), HCV RNA ≥6x106 IU/mL (53%). SVR12 in the IM arm was 92% and broadly unaffected by most baseline factors assessed (Figure); SVR12 was higher in patients without (96%) baseline NS5A-L31M/V or Y93H polymorphisms. There were 6 virologic breakthroughs, 6 relapses and 1 detectable HCV RNA at end-of-treatment in the IM arm. Safety was mostly comparable between the two arms during the first 12 weeks. The most frequent adverse events (AEs; ≥5%) during DCV+ASV (24 weeks) treatment in both arms were aminotransferase, bilirubin and INR elevations, hypertension, fatigue and respiratory tract infections; the most frequent treatment-emergent grade 3/4 laboratory abnormalities were aminotransferase (≤4.5%) and hematologic, lipase and total bilirubin abnormalities (≤2%); one patient (IM) discontinued DCV+ASV for aminotransferase elevations, nausea and jaundice (all reversible); one patient PD) discontinued DCV+ASV for a fatal AE unrelated to treatment. Conclusions: These data demonstrate that DCV+ASV is a highly efficacious and well tolerated treatment for treatment-naive HCV GT 1b-infected patients. Those treated immediately with DCV+ASV achieved a 92% SVR12 rate which was unaffected by factors known to attenuate response to interferon.

        True Digestibility of Phosphorus in Different Resources of Feed Ingredients in Growing Pigs

        To determine the true digestible phosphorus (TDP) requirement of growing pigs, two experiments were designed with the experimental diets containing five true digestible P levels (0.16%, 0.20%, 0.23%, 0.26% and 0.39%) and the ratio of total calcium to true digestible P (TDP) kept at 2:1. In Experiment 1, five barrows (Duroc${\times}$Landrace${\times}$Yorkshire) with an average initial body weight of 27.9 kg were used in a $5{\times}5$ Latin-square design to evaluate the effect of different dietary P levels on the digestibility and output of P and nitrogen. In Experiment 2, sixty healthy growing pigs (Duroc${\times}$Landrace${\times}$Yorkshire) with an average body weight (BW) of 21.4 kg were assigned randomly to one of the five dietary treatments (12 pigs/diet), and were used to determine the true digestible phosphorus (TDP) requirement of growing pigs on the basis of growth performance and serum biochemical indices. The results indicated that the true digestibility of P increased (p<0.05) linearly with increasing dietary TDP level below 0.26%. The true P digestibility was highest (56.6%) when dietary TDP was 0.34%. Expressed as g/kg dry matter intake (DMI), fecal P output increased (p<0.05) linearly with increasing P input. On the basis of g/kg fecal dry matter (DM), fecal P output was lowest for Diet 4 and highest (p<0.05) for Diet 5. The apparent digestibility of crude protein (CP) did not differ (p>0.05) among the five diets, with the average nitrogen output of 12.14 g/d and nitrogen retention of 66% to 74% (p>0.05), which suggested that there was no interaction between dietary P and CP protein levels. During the 28-d experimental period of Experiment 2, the average daily gain (ADG) of pigs was affected by dietary TDP levels as described by Eq. (1): $y=-809,532x^4+788,079x^3-276,250x^2+42,114x-1,759$; ($R^2=0.99$; p<0.01; y = ADG, g/d; x = dietary TDP, %), F/G for pigs by Eq. (2): $y=3,651.1x^4-3,480.4x^3+1,183.8x^2-172.5x+10.9$ ($R^2=0.99$; p<0.01; y = F/G; x = dietary TDP, %), and Total P concentrations in serum by Eq. (3): $y=-3,311.7x^4+3,342.7x^3-1,224.6x^2+195.6x-8.7$ (R2 = 0.99; p<0.01; y = total serum P concentration and x = dietary TDP, %). The highest ADG (782 g/d), the lowest F/G (1.07) and the highest total serum P concentration (3.1 mmol/L) were obtained when dietary TDP level was 0.34%. Collectively, these results indicate that the optimal TDP requirement of growing pigs is 0.34% of the diet at a total Ca to TDP ratio of 2:1.

        Dietary Supplementation of Magnesium Sulfate during Late Gestation and Lactation Affects the Milk Composition and Immunoglobulin Levels in Sows

        This experiment was conducted to investigate the effects of dietary supplementation of magnesium sulfate ($MgSO_4$) during late gestation and lactation on sow and litter performance, fecal moisture, blood biochemistry parameters, immunoglobulin levels and milk composition in sows. Forty-eight sows ($Yorkshire{\times}Landrace$, 4th to 5th parity) were randomly allocated to 1 of 4 dietary treatments supplemented with 0, 200, 400, or 600 mg/kg $MgSO_4$ (n = 12). The experiment started on day 90 of gestation and continued through day 21 of lactation. Blood samples were collected on day 107 of gestation, day 0 (farrowing) and 21 (weaning) of lactation for the analyses of the blood biochemistry parameters and immunoglobulin levels. The colostrum and milk samples were obtained on day 0 and 14 of lactation, respectively. Fecal samples were collected from the sows on day 107 of gestation as well as day 7 and 20 of lactation to determine fecal moisture content. The results showed that the survival percentage of piglets and the litter weight at weaning were decreased linearly (p<0.05) and other parameters of the sow or litter performance were not influenced (p>0.05) by $MgSO_4$ supplementation. The fecal moisture content of the sows were increased (p<0.05) linearly as dietary $MgSO_4$ increased on day 7 and 20 of lactation. Supplementation with $MgSO_4$ increased the plasma magnesium (Mg) level linearly (p<0.05) and had a trend to increase total protein level (p>0.05 and p<0.10). However, an increase in the dietary $MgSO_4$ level resulted in a linear decrease in the colostrum fat content (p<0.05). Dietary $MgSO_4$ supplementation enhanced the immunoglobulin G (IgG) level (linear, p<0.05) in plasma on day of farrowing and immunoglobulin A (IgA) level in colostrum (quadratic, p<0.05) and milk (linear, p<0.05) of the sows. These results indicated that supplementation with $MgSO_4$ during late gestation and lactation may have the potential to prevent sow constipation, but may also result in some negative effects.

        Enhanced energy storage properties in PbZrO3 thin films via the incorporation of NiO

        In this study, NiO–PbZrO3 composite films were deposited on SiO2/Si substrates buffered with LaNiO3 films via the sol-gel coating technique. The effects of NiO addition in PZO thin films on the microstructure, dielectric properties, leakage mechanism, ferroelectric properties and energy storage properties have been discussed. The dielectric constant increased with the addition of NiO, while the leakage current density decreased. Compared with pure PZO films, the maximum polarization of the composite films was improved. For the composite films prepared using the NiO precursor solution with 0.05 mol/L, the recoverable energy storage density of the NiO-PZO composite film is up to 19.6 J/cm3 under the electric field of 1038 kV/cm, which is 30% higher than that of the pure PZO film under the same conditions. Also, the energy storage efficiency of the composite film reaches 48%. Accordingly, we demonstrate a simple and convenient method by adding NiO to fabricate thin films with high energy storage performance.

        Intramuscular Administration of Zinc Metallothionein to Preslaughter Stressed Pigs Improves Anti-oxidative Status and Pork Quality

        This study was conducted to determine the effects of exogenous zinc-metallothionein (Zn-MT) on anti-oxidative function and pork quality. After feeding a corn-soybean meal-based diet for two weeks, 48 pigs ($Duroc{\times}Landrace{\times}Chinese\;Black Pig$) were assigned randomly to four groups. Pigs in Group 1 were maintained under non-stress conditions, whereas pigs in Groups 2, 3 and 4 were aggressively handled for 25 min to produce stress. Pigs in Groups 1, 2, 3, and 4 received intramuscular administration of saline (control group; CON), 0 (negative control group; NCON), 0.8 (low dose group; LOW), and 1.6 (high dose group; HIGH) mg rabbit liver Zn-MT per kg body weight, respectively. Pigs were slaughtered at 3 and 6 h post-injection. Zn-MT treatment increased (p<0.05) the activities of superoxide dismutase (SOD) and glutathione-peroxidase (GSH-PX) while decreasing the concentration of malondialdehyde (MDA) in liver. These responses were greater (p<0.05) at 6 h than at 3 h post Zn-MT injection. Zn-MT treatment increased (p<0.05) hepatic SOD mRNA levels in a time and dose-dependent manner and decreased (p<0.05) serum glutamate-pyruvate transaminase and lactate dehydrogenase activities (indicators of tissue integrity). Zn-MT administration decreased (p<0.05) lactate concentration and increased (p<0.05) pH and water-holding capacity in the longissimus thorasis meat. Collectively, our results indicate that intramuscular administration of Zn-MT to pre-slaughter stressed pigs improved tissue anti-oxidative ability and meat quality.

        Fluorescent bioimaging of pH: from design to applications

        <P>Protons play crucial roles in many physiological and pathological processes, such as receptor-mediated signal transduction, ion transport, endocytosis, homeostasis, cell proliferation, and apoptosis. The urgent demand for pH imaging and measurement in biological systems has incited the development of fluorescent pH probes. Numerous fluorescent probes have been reported, but many lack the abilities needed for biological applications. Hence, the development of new pH probes with better biocompatibility, sensitivity, and site-specificity is still indispensable. This review highlights the recent trends in the development of fluorescent materials as essential tools for tracing pH variations in the biological processes of diverse living systems.</P>

        Energy absorption behavior of metallic staggered double-sine-wave tubes under axial crushing

        A new type of double-sine-wave tube which is improved from a traditional square tube by embedding the sinusoid corrugation of bothtransvers and axial directions on the surfaces with phase differences between the adjacent ones is proposed in this paper. Attention isfocused on the energy-absorption behavior of the tube under axial crushing and to verify that it can significantly reduce the initial peakload and the magnitude of the fluctuant wave in load-displacement curve. A theoretical model based on rigid, perfectly plastic materialidealization is adopted to predict the mean crushing load. Besides, the numerical simulation of the staggered double-sine-wave-tube underaxial crushing is operated by using Hypermesh and Ls-dyna. The numerical results are good agreement with the theoretical predictions. Two typical examples of using the staggered double-sine wave tube to attenuate impact force are provided for verification of itsgood attenuation performance as an energy absorption structure.

      • Modelling of catalytically oxidative decomposition of carbon tetrachloride on a ZnS nanocluster using density functional theory

        Using density functional theory (DFT), adsorption of O-2 and co-adsorption of O-2 and CCl4 on (ZnS)(n) clusters (n = 6, 7, 11, 12, 13, 15, 16, 17, 25, 26) and the mechanism of oxidative decomposition of CCl4 on a ZnS nanocluster were investigated. The decomposition of CCl4 without the ZnS cluster was also studied for comparison. The ZnS cluster achieved a significant catalytic effect during the oxidative decomposition of CCl4. It is found that CCl4 in gas phase undergoes a homolytic dissociation process with the activation by ZnS, reacting with O-2 to produce a CCl3 center dot radical and a ClOO center dot radical first. CO2 and Cl-2 are identified as the final neutral reaction products, while COCl2, CO and Cl2O are produced as intermediates. The released energy of the total reaction, 149.83 kcal mol(-1), is sufficient to afford the transition energy of the cataluminescence, based on which a novel and sensitive gas sensor for the determination of CCl4 could be constructed.
