        Background: Vitronectin is one of major cell- adhesive glycoprotein in mammalian serum and plasma ; the other is fibronectin. It is a mixture of 65 and 75kDa protein in plasma which promotes spreading of a variety of cultured cells ,inhibits the cytotoxicity of membrane attacks complex C5b-7 and modulates thrombin-antithrombin III activity. Human plasma and serum contain 10-40mg of vitronectin per 100ml,but only a few mg or less of vitronectin can be isolated with a 0.5-20% recovery efficiency through very long processes or with expensive,commercially available monoclonal antibody. In this study, we purified the vitronectin from human plasma and produced anti-vitronectin polyclonal antibody using purified vitronectin. Methods: Vitronectin was purified from the human plasma by heparin-sepharose column and its efficacy was measured by cell spreading assay. Anti-vitronectin polyclonal antibody produced and confirmed by ELISA and immunoblotting. Results: This procedures produced about 0.8mg vitronectin from 100ml human plasma within 2 days. 1) Purified vitronectin promoted spreading of HepG2 hepatoma cells on substrates with a half maximal activity at 0.lug/ml. 2) In SDS-PAGE analysis of purified protein, 2 bands were found and their molecular weights were 75kDa and 65kDa,respectively. 3) The immunoblotting assay showed that the bands of molecular were same site as SDSPAGE analysis. Conclusion: Simple,rapid purified vitronectin by heparin-sepharose column and anti-vitronectin antibody may facilitate the elucidation of vitronectin function and action mechanism in human body.

        Radiotherapy-associated Furin Expression and Tumor Invasiveness in Recurrent Laryngeal Cancer

        <P>Background/Aim: Recurrent laryngeal cancer often shows an aggressive phenotype after radiotherapy and does not respond to conventional therapeutic strategies. In this study, we investigated the contribution of furin to cellular invasiveness in radio-resistant laryngeal cancer. Materials and Methods: Using previously established AMC-HN-3 and AMC-HN-8 cell lines from laryngeal carcinoma patients, recurrent laryngeal cancer models were generated by cumulative irradiation (AMC-HN-3-70Gy and AMC-HN-8-70Gy). Immunocytochemistry and western blotting were used to determine the epithelial-mesenchymal transition (EMT). Invasion capacity was assessed using an in vitro invasion assay. Zymography was used to assess metalloproteinase-2 (MMP-2) activity. Tumor xenografts were developed to compare growth rate and furin expression in vivo. Furin expression in 35 patients (45 samples) with salvage total laryngectomy after radiation-based treatment was assessed by laryngeal cancer tissue microarray. Results: Both AMC-HN-3-70Gy and AMC-HN-8-70Gy cell lines underwent EMT following radiation. However, AMC-HN-3-70Gy cells showed increased cellular invasiveness, whereas AMC-HN-8-70Gy cells showed no difference. AMC-HN-3-70Gy cells also exhibited elevated furin expression with up-regulated expression of the active form of membrane type 1-matrix metalloproteinase (MT1MMP)/MMP-2, whereas AMC-HN-8-70Gy cells did not show significant changes. After administration of a furin inhibitor (chloromethyl ketone (CMK)), AMC-HN-3-70Gy cells showed a significant decrease in MT1-MMP/MMP-2 expression and cellular invasiveness. Nine of 22 samples (40.9%) from salvage total laryngectomy and one of 13 pre-radiation samples (7.7%) had high furin expression. Post-radiation, furin expression increased in seven of 10 patients whose pre- and post-radiation samples were available; all-cancer mortality (three patients) was observed in this group. Conclusion: Together with EMT, furin activity may serve as an indicator of an aggressive cancer phenotype, suggesting that furin is a potentially useful target for recurrent laryngeal cancer.</P>

        Preoperative vitamin D level does not affect the short-term functional outcome after total knee arthroplasty in elderly women

        Background: We aimed to evaluate the effect of vitamin D levels on the functional outcome of elderly women who underwent total knee arthroplasty (TKA). Methods: Seven hundred and four patients (1013 knees) who underwent primary TKA were included in our retrospective study. Preoperative vitamin D levels were measured and the relationship analyzed between these and age, weight, height, body mass index, and bone mineral density. Two hundred and twenty patients (220 knees) who received unilateral TKA and were followed up for more than 1 year after operation were divided into two groups: Group 1, serum 25-hydroxyvitamin D3 (25(OH)D3) level < 20 ng/ml; and Group 2, 25(OH)D3 level ≥ 20 ng/ ml. Both groups were evaluated for the relationship between vitamin D levels and postoperative Visual Analogue Scale (VAS) score, Knee Society Knee Score (KSKS), Knee Society Function Score (KSFS), and Western Ontario and McMaster Universities Arthritis Index (WOMAC) score. Results: The number of vitamin D-deficient patients (< 20 ng/ml of serum 25(OH)D3 level) was 556 (79.0%). In the correlation analysis, the vitamin D level was negatively correlated with weight only (p = 0.033). No significant differences were observed between the groups in terms of postoperative VAS score, KSKS, KSFS, and WOMAC score. Conclusions: Vitamin D deficiency was highly prevalent in patients who underwent TKA. Vitamin D levels negatively correlated with weight. Low vitamin D level was not a risk factor for unsatisfactory TKA outcome in elderly women.

        Endothelial dysfunction induces atherosclerosis: increased aggrecan expression promotes apoptosis in vascular smooth muscle cells

        Endothelial dysfunction-induced lipid retention is an early feature of atherosclerotic lesion formation. Apoptosis of vascular smooth muscle cells (VSMCs) is one of the major modulating factors of atherogenesis, which accelerates atherosclerosis progression by causing plaque destabilization and rupture. However, the mechanism underlying VSMC apoptosis mediated by endothelial dysfunction in relation to atherosclerosis remains elusive. In this study, we reveal differential expression of several genes related to lipid retention and apoptosis, in conjunction with atherosclerosis, by utilizing a genetic mouse model of endothelial nitric oxide synthase (eNOS) deficiency manifesting endothelial dysfunction. Moreover, eNOS deficiency led to the enhanced susceptibility against pro-apoptotic insult in VSMCs. In particular, the expression of aggrecan, a major proteoglycan, was elevated in aortic tissue of eNOS deficient mice compared to wild type mice, and administration of aggrecan induced apoptosis in VSMCs. This suggests that eNOS deficiency may elevate aggrecan expression, which promotes apoptosis in VSMC, thereby contributing to atherosclerosis progression. These results may facilitate the development of novel approaches for improving the diagnosis or treatment of atherosclerosis. [BMB Reports 2019; 52(2): 145-150]

      • ICT Convergence Culture System using Automated Container Type Auricularia auricula-judae Mushroom Production Technology

        Information and communication technology(ICT) around the world in the 21st century presents a new vision of agriculture. Time, place, and the high-tech industry to overcome barriers to the fusion of the so-called “smart agriculture” is changing the landscape of agriculture. Precision Agriculture’s core container production for the mushroom cultivation temperature, humidity, irradiation, self-regulation, such as carbon dioxide, the optimal environment for mushroom cultivation was implemented. Auricularia auricula-judae, known as the Jew's ear, wood ear, jelly ear or by a number of other common names, is a species of edible Auriculariales fungus found worldwide. The fruiting body is distinguished by its noticeably ear-like shape and brown colouration. The fungus can be found throughout the year in temperate regions worldwide, where it grows upon both dead and living wood. We controlled different light source (Blue-Red-White combined LED, blue LED, red LED and fluorescent light) with different intensity of LED irradiation (1.5, 10.5 and 20.5 μmol/m2s for LEDs) to compare growth and development. Lights were treated with 12 hour on/ 12hour off cycle maintained in a controlled room with 19 ~ 21oC temperature, 85~95% humidity, and 1000ppm CO2 atmosphere concentration for 30 days. Growth and development differed from the LED color source and intensity of LED irradiation. Growth and development was most effective in 1.5 μmol/m2s irradiation for blue LED. All LED light sources showed less growth and development in highest intensity of irradiation, which indicates that higher than 20.5 μmol/m2s for LED is not effective. After harvesting fruit bodies, we measured their weight, length, width, thickness and chromaticity of fruit bodies. 1.5 μmol/m2s blue LED group was the best result of harvest with average individual weight (5.38g), length (65.37mm), width (56.87mm) and thickness (13.51mm) with fine chromaticity. This results show us that 1.5 μmol/m2s blue LED was the best effect on growth and development of Auricularia auricula-judae mushroom’s ICT system container type environment.

        Dieback Reality of Apple Trees Resulting from Soil-Borne Fungal Pathogens in South Korea from 2016 to 2019

        Recently, the severe dieback of apple trees resulting from soil-borne diseases has occurred in South Korea. The casual agents of dieback were surveyed on 74 apple orchards that had been damaged nationwide in 2016–2019. The number of apple orchards affected alone by Phytophthora rot, violet root rot, and white root rot was 31, 34, and 3, respectively. Also, the total number of mixed infection orchards was 6. Out of 9,112 apple trees affected by dieback, the trees damaged by Phytophthora rot, violet root rot, and white root rot were 3,332, 3,831, and 44, respectively. Moreover, the total number of mixed infection apple trees was 1,905. The provinces mainly affected were Gyeongnam, Gyeongbuk, Chungbuk, and Jeonbuk. The survey on these infected apple orchards will be available to form management strategy for the dieback that had been in- creased by soil-borne fungal pathogens.

        Effect of Irrigation Frequency on Growth and Functional Ingredient Contents of Gynura procumbens Cultivated in Hydroponics System

        Gynura procumbens (Family Asteraceae) is a medicinal plant commonly found in tropical Asian countries such as Malaysia, Indonesia, Thailand, China, and Vietnam. It is widely used as a traditional medicine in many different countries for the treatment of a variety of health ailments such as kidney discomfort, hypertension, constipation, diabetes mellitus, and rheumatism. The objective of this study was to investigate the effect of irrigation on growth and functional ingredients of Gynura procumbens. Gynura procumbens were planted in a greenhouse on April 24 (2019), and irrigation was carried out throughout the entire growth period at 12, 24, 48, 72hour cycles using drip hydroponics system. The total height of Gynura procumbens became higher with shorter irrigation cycle, whereas a smaller height was recorded with the longer periods. The maximum yield was recorded for 12 - 24 h irrigation cycle, but it tended to decrease as the irrigation cycle became longer. The leaf marketability based on the irrigation cycle, leaf length, leaf width, leaf weight, and leaf thickness were best for the 24 h irrigation cycle treatment. The total polyphenols was higher for the 72 h irrigation cycle treatments compared to the others. Vitamin C concentrations were high at 48 and 72 h irrigation cycle treatment. Hence, the growth of Gynura procumbens based on irrigation cycle was maximum at 24-hour, and total polyphenol and vitamin C content was highest at 72-hour irrigation cycle treatment.

        Effect of Shading on Growth and Functional Ingredient Contents of Gynura procumbens Cultivated in Hydroponics System

        Gynura procumbens (Family Asteraceae) is a medicinal plant commonly found in tropical Asian countries such as China, Thailand, Indonesia, Malaysia, and Vietnam. It is widely used as a traditional medicine in many different countries for the treatment of a variety of health ailments such as kidney discomfort, rheumatism, diabetes mellitus, constipation, and hypertension. The objective of this study was to investigate the effect of shading on growth and functional ingredients of Gynura procumbens. Gynura procumbens was planted in a greenhouse on April 24 (2019), and shading treatments were carried out throughout the entire growth period at 0%, 35%, 55%, and 75% using a black net. The total plant height of Gynura procumbens was highest for the 75% shading treated ones. Additionally, the stem diameter and yield was higher for all the shading treated plants compared to the non-shaded control plants. The DPPH radical scavenging activity of the 0% and 35% shading treated plants were significantly higher than the plants exposed to 55% and 75% shading treatment at 0.5% and 2.5% sample concentrations. The total flavonoids and vitamin C concentrations had also shown the same trend, whereas rutin content was highest for 0% shading treatment and the lowest was for 55% and 75% shading treatments. Hence, this study suggests that 35% shading treatment on Gynura procumbens can be considered as an advantageous condition for their growth and functional ingredients.
