      • H3K27 methylation and H3S28 phosphorylation-dependent transcriptional regulation by INHAT subunit SET/TAF-Iβ

        Significant progress has been made in understanding the relationship between histone modifications and 'reader' molecules and their effects on transcriptional regulation. A previously identified INHAT complex subunit, SET/TAF-Iβ, binds to histones and inhibits histone acetylation. To investigate the binding specificities of SET/TAF-Iβ to various histone modifications, we employed modified histone tail peptide array analyses. SET/TAF-Iβ strongly recognized PRC2-mediated H3K27me½/3; however, the bindings were completely disrupted by H3S28 phosphorylation. We have demonstrated that SET/TAF-Iβ is sequentially recruited to the target gene promoter ATF3 after the PRC2 complex via H3K27me recognition and may offer additive effects in the repression of the target gene.

      • The Use of Collagen Content as Determined by Spectral Domain Polarization-Sensitive Optical Coherence Tomography to Assess Colon Anastomosis Healing in a Rat Model

        <P>Abstract</P><P><B><I>Background/Purpose:</I></B> Many studies have been undertaken to prevent anastomosis leakage of the colon, and several methods have been used to assess anastomosis healing, such as measurement of bursting pressure or hydroxyproline (a marker of collagen) content at the anastomosis site. However, these methods are inappropriate for comparing anastomosis healing at two time points in the same animals. In the present study, we measured the collagen level by spectral domain polarization-sensitive optical coherence tomography (SD-PS-OCT) to assess anastomosis healing. <B><I>Methods:</I></B> Sprague-Dawley rats were divided into groups C (saline-administered controls; study group) and M [a 5-fluorouracil (5-FU)-administered experimental group]. Immediately after end-to-end anastomosis of the colon, SD-PS-OCT images of anastomoses were taken (baseline). Animals were administered saline or 5-FU for 7 days. On the 7th postoperative day, SD-PS-OCT images were acquired, a histopathologic exam was performed, and hydroxyproline levels as well as mRNA expressions of collagen-1 and collagen-3 were measured at the anastomosis site. <B><I>Results:</I></B> Fibroblast proliferation and inflammatory cell infiltration were greater in group C than in group M. The mRNA expressions of collagen-1 and collagen-3 were substantially higher in group C. Hydroxyproline levels were higher in group M than in group C. Though collagen levels measured by SD-PS-OCT at 7 days were elevated compared with baseline in group C, no such changes were observed for group M. <B><I>Conclusion:</I></B> Collagen levels at the colon anastomosis site, measured with SD-PS-OCT, were not increased at 7 days postoperatively versus baseline when 5-FU was injected, but were increased in saline-treated controls. The measurement of collagen content by SD-PS-OCT was found to provide a good means of assessing anastomosis healing, because it allows in situ assessment of collagen contents at baseline and during the postoperative period.</P><P>© 2014 S.         The kinematics of the Seyfert 2 galaxy NGC 5728 circumnuclear region

        <P>ABSTRACT</P><P>We secured spectral images of the circumnuclear, 12 × 10 arcsec<SUP>2</SUP> region of the Seyfert 2 galaxy NGC 5728 with the Optically Adaptive System for Imaging Spectroscopy at the Canada–France–Hawaii Telescope 3.6-m telescope. The radial velocity distribution of the two bright zones, main central (C) core and north-west (NW) core, and strategically important points along the position angles, PA 33°, 90°, 123° and 143°, are analysed. The large-scale reconstructed monochromatic images of the Hα, Hβ, [O <SMALL>III</SMALL>] and [N <SMALL>II</SMALL>] lines show a bipolar morphology along PA ∼ 123°. The prominent elliptical ring is seen in the Hα and Hβ monochromatic images. Our analysis implies that this ring is rotating in the same clockwise direction as that of the galactic disc; this elliptical ring is in fact a projected result of a circular ring of radius ∼5.4 arcsec, with <I>i</I>∼ 50° (nearly aligned with the host galactic plane).</P><P>We used the [O <SMALL>III</SMALL>]5007 line profiles for a detailed analysis of kinematics in the circumnuclear region of the NGC 5728. The [O <SMALL>III</SMALL>]5007 line profiles reveal distinct kinematical structures. (1) The strongest component of Doppler peaks: two core components, consisting of the receding main C-core and the approaching NW-core with the radial velocities, <I>V</I><SUB>r</SUB>≃+250 and −240 (or −250) km s<SUP>−1</SUP>, respectively. These two cores consist of a roughly spherically symmetric distribution of the gas with radii of <I>r</I>∼ 1.0 arcsec (at <I>V</I><SUB>r</SUB>≃+240 km s<SUP>−1</SUP>) and 0.9 arcsec (at <I>V</I><SUB>r</SUB>≃−240 km s<SUP>−1</SUP>), respectively. Hints of the presence of inflows around these cores are observed. (2) The next strong line profile component: this component corresponds to the bipolar cone aligned PA ∼ 123°. (3) The reverse S-shaped kinematical boundary which runs through the double-peaked zone of 6–7 arcsec long in the galactic major axis direction is likely to be caused by a combination of the disc rotation of the host galaxy and the revolution of the two central cores.</P>

        Inhibition of Staphylococcus aureus by antimicrobial biofilms formed by competitive exclusion microorganisms on stainless steel

        <P>The goal of this study was to develop a desiccation resistant antimicrobial surface using biofilm of competitive exclusion (CE) microorganism inhibitory to Staphylococcus aureus. We isolated 161 microorganisms from soils, foods, and food-contact surfaces that are inhibitory to S. aureus. Among them, three CE microorganisms (Streptomyces spororaveus strain Gaeunsan-18, Bacillus safensis strain Chamnamu-sup 5-25, and Pseudomonas azotoformans strain Lettuce-9) exhibiting strong antibacterial activity and high growth rates were selected for evaluation. These isolates formed biofilms within 24 h on stainless steel coupons (SSCs) immersed in Bennet's broth and tryptic soy broth at 25 degrees C. Cells in these biofilms showed significantly (P <= 0.05) enhanced resistance to a desiccation (43% relative humidity [RH]) compared to those attached to SSCs but not in biofilms. The antimicrobial activities of biofilms formed by these isolates on SSCs against S. aureus at 25 degrees C and 43% RH were determined. Compared to SSCs lacking biofilms formed by CE microorganisms, populations of S. aureus on SSCs harboring CE biofilms were significantly lower (P 0.05). Results indicate that persistent antimicrobial activity against S. aureus on stainless steel surfaces can be achieved by the presence of biofilms of CE microorganisms. This information will be useful when, developing strategies to improve the microbiological safety of foods during storage, processing, and distribution by facilitating the development of effective antimicrobial food-contact surfaces.         Quantification of main wall erosion/deposition pattern in n = 2 RMP KSTAR H-mode plasmas

        <P><B>Abstract</B></P> <P>Erosion/deposition pattern was studied using cavity sample at KSTAR main wall region. The cavity sample has a different two slit opening directions. The amorphous carbon film inside the cavity structure was installed on mid-plane manipulator system and was exposed to H-mode plasmas with n = 2 RMP. The most of characterized amorphous hydrocarbon film in cavity structure was eroded after exposing to H-mode plasmas. The erosion rate of bottom plate sample was in a range from 0.32 nm s<SUP>−1</SUP> to 0.77 nm s<SUP>−1</SUP> in a different slit opening directions, respectively. But the upper samples were deposited and the maximum deposition rate of it was 0.42 nm s<SUP>−1</SUP>. Thickness profiles measured by ellipsometer were compared with simulated profile using Monte Carlo Ballistic Simulation. The main parameter for simulation was the incident angle of particle. The incident angle of particle influenced the shape and width of simulated profile. The incident angle used in simulation was in the range of 80°–89° and it was well matched with 81.9° derived from EFIT.</P>

        Performance and carrier transport analysis of In<sub>0.7</sub>Ga<sub>0.3</sub>As quantum-well MOSFETs with Al<sub>2</sub>O<sub>3</sub>/HfO<sub>2</sub> gate stack

        In this paper, we have fabricated and characterized In<SUB>0.7</SUB>Ga<SUB>0.3</SUB>As quantum-well (QW) metal-oxide-semiconductor field-effect-transistors (MOSFETs). We have employed the gate dielectric of the Al<SUB>2</SUB>O<SUB>3</SUB>/HfO<SUB>2</SUB> (0.6/2nm) bi-layer stack by ALD. The fabricated device with L<SUB>g</SUB>=4μm exhibits a record maximum transconductance (g<SUB>m_max</SUB>) in excess of 520μS/μm at >1μm region, and reasonably good electrostatic integrity, such as SS=110mV/decade and DIBL=43mV/V. Also, we have investigated the gate length scaling behavior in terms of output, transconductance, and transfer characteristics. In particular, our devices feature very uniform values of the electrostatic integrity, such as SS=100-110mV/decade, V<SUB>T</SUB>=-0.25V to -0.2V and DIBL=40-50mV/V, as L<SUB>g</SUB> decreases from 10μm to 4μm. Furthermore, we have explored the impact of source resistance (R<SUB>S</SUB>) onto the device characteristics of the InGaAs QW MOSFETs. In doing so, we have modeled both measured extrinsic transconductance (g<SUB>m_ext</SUB>) and intrinsic transconductance (g<SUB>m_int</SUB>) as a function of L<SUB>g</SUB>.

      • Biochemical, pharmaceutical and therapeutic properties of long-acting lithocholic acid derivatized exendin-4 analogs

        Alterations in the physicochemical characteristics of peptide drugs can transform their biological and pharmaceutical features. In the present study, we explored the potentials of lithocholic acid (LCA)-modified exendin-4 derivatives as novel long-acting GLP-1 receptor agonists. Exendin-4 was modified with lithocholic acid at two lysine residues to produce three derivatives that were obtained by reverse-phase HPLC separation, namely, Lys<SUP>12</SUP>-LCA-exendin-4 (LCA-M2), Lys<SUP>27</SUP>-LCA-exendin-4 (LCA-M1), and Lys<SUP>12,27</SUP>-LCA-exendin-4 (LCA-Di)). The biological, pharmacological, and physicochemical characteristics of these three exendin-4 analogues were then investigated. Although slight reductions in the GLP-1 receptor binding capacity and insulinotropic activity of exendin-4 were observed after derivatization, the mono-LCA substitutions, especially LCA-M1, well-preserved antidiabetic activity in type 2 diabetic mice when administered subcutaneously or intraperitoneally. Furthermore, the pharmacokinetic characteristics were dramatically enhanced, that is, absorption was delayed and elimination half-life was increased (1.6+/-0.4 and 9.7+/-1.4h by exendin-4 and LCA-M1, respectively). The enhanced long-acting characteristics of the derivative was found to be due to albumin binding and nanoparticle formation, and these were verified by the restoration of normoglycemia in type 2 diabetic mice after single injection (>24h, >10nmol/kg, s.c.) and daily injections (15nmol/kg/day) maintained normoglycemia for the 4-week administration period. Furthermore, antidiabetic potentials, such as, glucose clearance kinetics and percentage areas occupied by pancreatic β-cells were also enhanced by long-term LCA-M1 administration. The present study demonstrates that the derivatization of exendin-4 with LCA offers a possible means of producing a long-acting GLP-1 receptor agonist.
