        Numerical Study on the Influence of the Width of the SBM Disc Cutter on Rock Breaking

        The study of the rock-breaking pattern of shaft boring machine (SBM) cutters is the basis for cutter design. In order to reveal the influence of the cutter blade width on the rock-breaking effect, this study established a discrete-continuous coupled rock mass numerical model. By setting different blade widths and types, the variation patterns of cutter forces, rock-breaking area, and specific energy with the blade width were revealed. The influence of the blade type on the breaking coefficient was further quantified according to the fractal dimension. The results show that under the cutting action, the rock mass can be divided into dense core, fractured zone, and elastic deformation zone, with the fractured zones mainly located on the lower side of the cutter's central axis. As the width of the cutter increases, the cutter penetration force and side force increase linearly with the increase in blade width, the breaking area increases and then tends to remain constant, the specific energy decreases first and then increases, and the rock fragmentation factor increases linearly. After scaling the width of the flat blade up to 18 mm and the arc blade to 24 mm, further increase in the blade width does not improve the breaking efficiency but only results in a smaller slag granularity.

        Poly(vinly alcohol)/Nano-Sized Layered Double Hydroxides Nanocomposite Hydrogels Prepared by Cyclic Freezing and Thawing

        In this paper, nano-sized MgAl-layered double hydroxides (LDH) were synthesized via a very fast nucleation and slow aging method that can be suspended in water. Poly(vinyl alcohol) (PVA)/LDH nanocomposite hydrogels were then prepared using a cyclic freezing/thawing method. The mechanical properties of PVA hydrogels were significantly enhanced by incorporation of LDH nanoplatelets. After 7, 9, and 11 freezing/thawing cycles, the tensile strength of PVA nanocomposite hydrogels containing 0.5 wt% LDH was remarkably increased by 100%, 100%, and 300%, respectively, while the Young’s modulus was increased by 25%, 22%, and 149%, respectively. More interestingly,the nanocomposite hydrogels exhibited larger elongation at the break than neat PVA hydrogels. However,after addition of the LDH nanoplatelets, the water-swelling ability of PVA hydrogels decreased. The microstructures and morphologies of PVA/LDH nanocomposite hydrogels were further investigated by scanning electron microscopy,transmission electron microscopy, and differential scanning calorimetry. The results showed that well-dispersed LDH nanoplatelets played an important role in increasing physical cross-linking density by inducing PVA crystallization and the formation of more uniform polymer networks. As a result, the mechanical properties of PVA hydrogels were greatly improved, even at very low LDH loading levels.

        Efficacy and safety of olmesartan medoxomil/amlodipine fixed-dose combination for hypertensive patients uncontrolled with monotherapy

        This study aimed to evaluate the efficacy andsafety of olmesartan medoxomil (OM)/amlodipine (AML)20/5 mg fixed-dose combination tablet in Chinese mild tomoderately hypertensive patients with inadequate bloodpressure (BP) control on monotherapy. Two multicenter,randomized, double-blind, double-dummy, active-controlled,parallel group clinical trials were conducted. Afterscreening and a 2-week placebo run-in period, patients with95 mmHg B seated diastolic blood pressure (SeDBP) \110 mmHg received monotherapy with OM 20 mg (inStudy 1) or AML 5 mg (in Study 2), once daily for4 weeks. Patients with 90 mmHg B mean SeDBP \110 mmHg at the end of the monotherapy period wererandomized to receive OM/AML 20/5 mg treatment orcontinue with the monotherapy, once daily for 8 weeks. OM/AML (20/5 mg) treatment significantly lowered bothsystolic and diastolic BP at 4 and 8 weeks compared to40 mg olmesartan or 5 mg AML. The incidence of drugrelatedadverse effects did not differ significantly betweenthe groups. OM/AML 20/5 mg was superior to OM 40 mgor AML 5 mg monotherapy in lowering BP in Chinesemild to moderately hypertensive patients with inadequateBP control on monotherapy. No new or unexpected safetyissues were identified with OM/AML combination therapycompared to monotherapy.

        Using low-coverage whole genome sequencing technique to analyze the chromosomal copy number alterations in the exfoliative cells of cervical cancer

        Objectives: We analyzed the chromosomal-arm-level copy number alterations (CNAs) in the cervical exfoliative cell and tissue samples by using the low-coverage whole genomic sequencing technique. Methods: In this study, we retrospectively collected 55 archived exfoliated cervical cell suspension samples and the corresponding formalin-fixed and paraffin-embedded tissue section samples including 27 invasive cervical cancer and 28 control cases. We also collected 19 samples of the cervical exfoliative cells randomly from women to verify the new algorithm model. We analyzed the CNAs in cervical exfoliated cell and tissue samples by using the low-coverage next generation of sequencing. Results: In the model-building study, multiple chromosomal-arm-level CNAs were detected in both cervical exfoliated cell and tissue samples of all cervical cancer cases. By analyzing the consistency of CNAs between exfoliated cells and cervical tissue samples, as well as the heterogeneity in individual patient, we also established a C-score algorithm model according to the chromosomal-arm-level changes of 1q, 2q, 3p, 7q. The C-score model was then validated by the pathological diagnosis of all 74 exfoliated cell samples (including 55 cases in model-building group and 19 cases in verification group). In our result, a cutoff value of C-score >6 showed 100% sensitivity and 100% specificity in the diagnosis of cervical cancer. Conclusion: In this study, we found that CNAs of cervical exfoliated cell samples could robustly distinguish invasive cervical cancer from cancer-free tissues. And we have also developed a C-score algorithm model to process the sequencing data in a more standardized and automated way.

        Zearalenone regulates key factors of the Kelch-like erythroid cell-derived protein with CNC homology-associated protein 1-nuclear factor erythroid 2-related factor 2 signaling pathway in duodenum of post-weaning gilts

        Objective: This study explored the mechanism of the Kelch-like erythroid cell-derived protein with CNC homology-associated protein 1 (Keap1)-nuclear factor erythroid 2-related factor 2 (Nrf2) signaling pathway under conditions of zearalenone (ZEA)-induced oxidative stress in the duodenum of post-weaning gilts. Methods: Forty post-weaning gilts were randomly allocated to four groups and fed diets supplemented with 0, 0.5, 1.0, or 1.5 mg/kg ZEA. Results: The results showed significant reductions in the activity of the antioxidant enzymes total superoxide dismutase and glutathione peroxidase and increases the malondialdehyde content with increasing concentrations of dietary ZEA. Immunohistochemical analysis supported these findings by showing a significantly increased expression of Nrf2 and glutathione peroxidase 1 (GPX1) with increasing concentrations of ZEA. The relative mRNA and protein expression of Nrf2, GPX1 increased linearly (p<0.05) and quadratically (p<0.05), which was consistent with the immunohistochemical results. The relative mRNA expression of Keap1 decreased linearly (p<0.05) and quadratically (p<0.05) in the duodenum as the ZEA concentration increased in the diet. The relative mRNA expression of modifier subunit of glutamate-cysteine ligase (GCLM) increased quadratically (p<0.05) in all ZEA treatment groups and the relative mRNA expression of quinone oxidoreductase 1 (NQO1) catalytic subunit of glutamate-cysteine ligase decreased linearly (p<0.05) and quadratically (p<0.05) in the ZEA1.0 group and ZEA1.5 group. The relative protein expression of Keap1 and GCLM decreased quadratically (p<0.05) in the duodenum as the ZEA concentration increased in the diet, respectively. The relative protein expression of NQO1 increased linearly (p<0.05) and quadratically (p<0.05) in all ZEA treatment groups in the duodenum. Conclusion: These findings suggest that ZEA regulates the expression of key factors of the Keap1-Nrf2 signaling pathway in the duodenum, which enables resistance to ZEA-induced oxidative stress. Further studies are needed to examine the effects of ZEA induced oxidative stress on other tissues and organs in post-weaning gilts.

        Optimization of APE1547-catalyzed Enantioselective Transesterification of (R/S)-2-methyl-1-butanol in an Ionic Liquid

        Aeropyrum pernix esterase (APE1547) was successfully used to catalyze the enantioselective transesterification of (R/S)-2-methyl-1-butanol in an ionic liquid (IL). Effects of various reaction conditions on the synthetic activity of the enzyme as well as enantioselectivity, including the type of IL, acyl donor, temperature, water activity,and substrate molar ratio were inverstigated. APE1547showed good catalytic performance (activity > 0.8 μmol/min/mg, E > 25), and the enzyme-IL mixture was recycled five times with only a slight decrease in catalytic performance.

        Effects of zearalenone on the localization and expression of the growth hormone receptor gene in the uteri of post-weaning piglets

        Objective: In this study, we investigated the adverse effects of dietary zearalenone (ZEA) (0.5 to 1.5 mg/kg diet) on the localization and expression of the growth hormone receptor (GHR) in the uteri of post-weaning gilts and explored alternative mechanism of the reproductive toxicity of ZEA on piglets. Methods: A total of forty healthy piglets (Duroc×Landrace×Large White) aged 28 d were selected for study. Piglets were transferred to single cages after 10 days’ adaptation on an obstetric table. The animals were allocated to one of four treatments: a normal basal diet supplemented with 0 (Control), 0.5 (ZEA0.5), 1.0 (ZEA1.0), or 1.5 (ZEA1.5) mg/kg purified ZEA, and fed for 35 d after the 10-d adaptation. Analyzed ZEA concentrations in the diets were 0, 0.52±0.07, 1.04±0.03, and 1.51±0.13 mg/kg, respectively. At the end of the feeding trial, piglets were euthanized after being fasted for 12 h. Two samples of uterine tissue from each pig were rapidly collected, one of which was stored at –80°C for analysis of the relative mRNA and protein expression of GHR, and the second was promptly fixed in Bouin’s solution for immunohistochemical analysis. Results: The relative weight of the uteri and thickness of the myometrium and endometrium increased linearly (p<0.001) and quadratically (p<0.001) with an increasing level of ZEA. The results of immunohistochemical analysis indicated that GHR immunoreactive substance was mainly localizated in the cytoplasm of uterine smooth muscle, glandular epithelial, luminal epithelial, stromal, and vascular endothelial cells. In contrast, nuclear staining was rarely observed. The immunoreactive integrated optic density of GHR in the myometrium, luminal epithelium, glandular epithelium, and whole uteri of weaning gilts increased linearly (p<0.001) and quadratically (p<0.05) with an increasing level of ZEA. The mRNA and protein expression of GHR in the uteri of weaning gilts increased linearly (p<0.001) and quadratically (p<0.05) with an increasing level of ZEA. Conclusion: In conclusion, ZEA at a concentration of 0.5 mg/kg was sufficient to significantly thicken the myometrium and endometrium, and at a concentration of 1.0 mg/kg induced a high level of GHR expression to promote growth and development of the uteri. This revealed an alternative molecular mechanism whereby ZEA induces growth and development of the uteri and provides a theoretical basis for the revision of Chinese feed hygiene standards.

      • Relationship Between Expression of Gastrokine 1 and Clinicopathological Characteristics in Gastric Cancer Patients

        The aim of the study was to clarify the role of gastrokine 1 in the process of formation and development of gastric cancer. The expression of gastrokine 1 in gastric cancer and corresponding non-cancerous gastric tissues of 52 gastric cancer patients was assessed with the real-time fluorescence quantitative polymerase chain reaction (RT-PCR) and immunohistochemistry. We also analyzed the relationship between the expression level and clinicopathological characteristics. Gastrokine 1 gene and protein expression in gastric cancer tissues was in both cases significantly lower than in corresponding non-cancerous gastric tissues (both P<0.01), but no significant relationship was found with clinicopathological parameters including tumor location, depth of invasion, differentiation, lymph node metastasis, stage, gender, age and carcinoembryonic antigen (CEA), and carbohydrate antigen 19-9 (CA19-9) level in peripheral blood preoperation of patients (P>0.05, respectively). Furthermore, gastrokine 1 gene expression was markedly lower in gastric cancer tissues of Helicobacter pylori (HP)-positive patients than negative ones (P<0.05). The result of the study showed that gastrokine 1 might play a significant role in the process of formation and development of gastric cancer as an anti-oncogene. Its effect might be weakened by HP infection.

        Dynamic Studies on Physiology and Biochemistry in American Seng Seed During Stratification - Part I. Embryo Ratio, Dry Weight Ratio and respiration Rate

        Dynamic parameters of physiology including embryo ratio (ER), embryo length to endosperm length, dry weight ratio between embryo and endosperm (DWR) and respiration rate (RR) in American send (Panax quinquefolium L.) seed were investigated. According to the changes of ER during seed stratification, the duration of embryo afterripening could be divided into three stages as \circled1 embryo slow growth stage (ESGS), ER increased from 7.31% to 20.48% (0.16% day-1): \circled2 embryo rapid growth stage (ERGS), ER increased to 80.98% (0.61% day-1) (75G5+ ERGS=morphological afterripening stage (MP,5)) and \circled3pysiological afterripening stage (PAS), ER Increased to 88.50% (0.094 day-1) only. DWR Increased slowly from 0.20% to 2.76clp (0.016% day-1) in MAS and rapidly to 8.81% (0.061% day-1) in PAS. The RR correlated significantly with ER as well (r=0.8934 > rot, 0.6610). The steep increment of both DWR and RR in PAS indicated that the PAS was not a static stage although the ER was not changed too much. All of these may provide some information for understanding the dormancy mechanisms of American sting seed.

        충적과정중 미국인삼 종자의 생리 및 생화학적 지표에 관한 동태학적 연구 I

        Dynamic parameters of biochemistry including the contents of soluble carbohydrate (SC) , crude fat (CF) , fatty arid (FA) and soluble protein (SP) in American send (Panax qufnquelolium L.) seed are reported in the present paper. When embryo ratio (ER) increased from 7.31 to 20.48%, the SC content decreased from 4.08 to 1.16%. After that, SC content changed little. The CF content decreased successively from 34.50 to 14.40% from the beginning to the end during the embryo afterripening. The changes of FA content did not correlate with that of ER and the changing range of SP content was not large during the seed stratification. All of these may provide some information for understanding the dormancy mechanisms of American seng seed.
